Bacterial Reverse Mutation Test of Wild Ginseng Culture Extract (산삼배양추출물의 세균을 이용한 복귀돌연변이시험)

  • To evaluate the bacterial reverse mutation of wild ginseng culture extract, the in vitro Ames test using Salmonella typhimurium (TA100, TA1,535, TA98, TA1,537) and Escherichia coli (WP2 uvrA) were performed with wild ginseng extract at the concentrations 0, 1.6, 8, 40, 200, 1,000, 2,500 and $5,000{\mu}g/ml/plate$. Wild ginseng culture extract was negative in Ames test with both Salmonella typhimurium or Escherichia coli with and without rat liver microsomal enzyme (S-9 fraction). According to these results, we concluded that wild ginseng culture extract did not cause bacterial reverse mutation.

Effects of the Artificial Culture Medium of Wild Ginsengs on Rumen Fermentation Characteristics In Vitro (산삼배양액 이용에 관한 반추위 미생물 대사 연구)

  • This study was conducted to determine the effects of the artificial culture medium of wild-ginsengs on in vitro fermentation characteristics. NH$_3$-N concentration was showed the highest in 3% WGM treatment among all treatments and control. In addition, microbial protein synthesis was significantly different in all treatments throughout the incubation time, and WGM 3% treatment was the highest at the 9 h incubation(P〈0.05). Protozoa numbers within rumen were decreased in all WGM treatments at 9 h incubation time, whereas WGM 3% treatment was always decreased throughout the incubation(P〈0.05). NDF and ADF digestibility were proportionally increased as the incubation time in both control and treatments. NDF digestibility showed no significantly difference between control and the 3% treatment, and ADF digestibility was similar in all. Total volatile fatty acid(VFA) concentrations of WGM treatments without 5% were significantly higher than control (P〈0.05). No differences were observed in total VFA, acetate, propionate and butyrate concentration among the WGM treatments. Acetate/Propionate ratio of WGM treatments was higher than control after 12 h incubation(P〈0.05). As a result of the artificial culture medium of wild-ginseng on rumen fermentation characteristics in vitro, microbial protein synthesis of WGM treatment was higher than control, and WGM 3% was the highest in all treatments(P〈0.05). The effect of saponin in artificial culture medium of wild-ginseng tended to decrease NH$_3$-N concentration, while it increases the microbial synthesis in early incubation. Therefore, artificial cultures medium of wild-ginseng can increase utilization of feed by microbial and anti-protozoal effects of saponin, which may enhance microbial synthesis capacity in early fermentation period in rumen.

Effects of Dietary Supplementation of Fermented Wild-ginseng Culture By-products on Egg Productivity, Egg Quality, Blood Characteristics and Ginsenoside Concentration of Yolk in Laying Hens (발효 산삼 배양액 부산물 급여가 산란계의 산란율, 계란 품질, 혈액특성 및 난황내 진세노사이드 함량에 미치는 영향)

  • The present study was conducted to investigate the effects of dietary supplementation of fermented wild-ginseng culture by-product on egg production, egg quality and blood characteristics in laying hens. A total of 216 ISA Brown laying hens, 55 wk of age, were used in the 6-wk feeding trial. Experimental diets were consisted of basal diet (CON), 2.5% fermented wild-ginseng culture by-product replaced lupin in basal diet (WG1) and 5.0% fermented wild-ginseng culture by-product replaced lupin in basal diet (WG2). Birds were randomly allotted to 18 replicate pens. There were 6 replicates per treatment, and 12 laying hens per replicate. Through the 6-wk feeding trial, egg production was significantly increased in WG1 and WG2 treatments compared to CON (P<0.05). Egg weight was significantly higher in WG2 than CON (P<0.05). WG1 resulted higher yolk color than CON (P<0.05). Albumen height and Haugh unit were significantly improve in WG1 compared to WG2 (P<0.05). Red blood cell was significantly lower in WG2 than CON (P<0.05). LDL-cholesterol was significantly decreased in CON compared to WG2 (P<0.05). In conclusion, fermented wild-ginseng culture by-product could improve egg production and egg weight in laying hens.

The Antihypertensive and Vasodilating Effects of Adventitious Root Extracts of Wild Ginseng (산삼 배양근 추출물의 혈압강화 및 혈관이완 효과)

  • Nitric oxide (NO) is a potent antihypertensive and vasodilator which plays an important role in regulating vascular tones. In this study, we investigated the effects of adventitious root extracts of wild ginseng on NO production and NO linked physiological activities. When human endothelial cell line (ECV304) was incubated with either water extracts of wild ginseng adventitious root (WE) or aqueous fraction of butanol extracts of wild ginseng adventitious root (ABE), considerable amounts of NO were released by the cells. The level of endothelial nitric oxide synthase (eNOS) expression was unchanged and about 6% of the angiotensin converting enzyme (ACE) was inhibited with treatment of ABE. The vasodilating activities of pulmonary artery rings in response to different doses of extracts were shown as 44.8% and 91.3% in 2.5 mg/ml WE and 0.1 mg/ml ABE, respectively. The blood pressure lowering effect was observed from the oral administered spontaneously hypertensive rat (SHR) with the lowest blood pressure (154.5${\pm}$8.6 mmHg) after 8 h. The blood pressure was recovered to the initial level after 24 h.

Fatty Acid, Amino Acid Composition and Sensory Traits of Pork from Pigs Fed Artificial Culture Medium of Wild Ginseng (산삼 배양액 급여 돈육의 지방산, 아미노산 조성 및 관능적 특성)

  • A total of 120 pigs (Berkshire) were used to investigate the effect of dietary supplementation with artificial culture medium ot wild ginseng (CMWG) on the fatty acid composition, amino acid composition and sensory characteristics or pork About $60{\pm}3kg$ pigs were randomly assigned to one of four experimental diet groups[(both sexes)${\times}$(C: commercial diet feed; T: commercial diet+1 L CMWG per day for 70 days)]. Pigs were slaughtered at approximately 110 kg live weight, and fatty acid composition, amino acid composition and sensory characteristics were measured in pork loin. The monounsaturated and saturated fatty acid contents was greater in barrow fed a diet containing CMWG than those of the gilt pork and control groups, however the polyunsaturated fatty acid composition decreased. The EAA (essential amino acid) content was lower in pigs fed diets containing CMWG than that of the control groups, whereas the SAA (amino acid with sulfide) and FRAA (fragrant amino acid) contents were higher. The EAA and FAA (amino acid in relation to flavor) contents were lower for harrow than for gilt, however the SAAA (amino acid in relation to saccarinity), SAA and FRAA the contents were higher. Regarding the sensory evaluation of fresh meat, the color of gilt pork increased with diets containing CMWG relative to the control group. Drip loss and the marbling score for pigs fed with diets containing CMWG were higher in barrow than in gilt. The flavor and overall acceptability of cooked meat from the control group was higher for barrow than for silt.
