Effect of the Dietary Composition (quantitative ratio between carbohydrate and protein) and the Rearing Temperature during the 4th Moulting Period on Proteinase Activity of Digestive Juice and Sucrase Activity of Midgut Tissue in the Silkworm, Bombyx mori L. (가잠유충의 소화액 Proteinase 및 중장조직 Sucrase의 활성에 미치는 사료조성(당과 단백질과의 량비) 및 4 면기보호온도의 영향)

  • This study has been carried out to investigate proteinase activity of digestive juice and sucrase activity of midgust tissue in the 5th day of the 5th instar influenced by the dietary composition (quantitative ratio between carbohydrate and protein) and tile rearing temperature during the 4th moulting period. The larvae grew on three kinds of semi-synthetic diet. The A-diet has more carbohydrate than the others, the B-diet has carbohydrate in 1 : 2 with protein, and the C-diet has more protein than the others. All the diets were kept at 16$^{\circ}C$, 25$^{\circ}C$ and 33$^{\circ}C$ during the 4th moulting period. Proteinase activity of digestive juice at the 5th day of the 5th instar was analyzed by Anson's hemoglobin method. Sucrase activity of midgut tissue at the 5th day of 5th instar was analyzed by Somogyi-Nelson's method. The results were as follows. 1. The dietary composition influencing contents of blood sugar was not related to the rearing temperature during the 4th moulting period. The contents of blood sugar appeared to increase in A-diet, B-diet and C-diet order, while proteinase and sucrase activity were stronger in C-diet, B-diet and A-diet order. 2. All kinds of diets showed almost the same fact that proteinase activity at 16$^{\circ}C$ was stronger than that at 32$^{\circ}C$. 3. It was found that sucrase activity became gradualy stronger at 32$^{\circ}C$, 25$^{\circ}C$ and 16$^{\circ}C$ in order in all kinds of diets. 4. There was an interaction in proteinase activity between the dietary composition and the rearing temperature in male larval digestive juice during the 4th mouiting period. On the other hand, there was an inter-acion in sucrase activity between the dietary composition and the rearing temperature in both female and male larval midgut tissue during the 4th moulting period.

Analysis of Nutrient Cycling Structure of a Korean Beef Cattle Farm Combined with Cropping as Affected by Bedding Material Types (깔개물질의 종류에 따른 한우-경종 결합 농가의 양분순환 구조 분석)

  • In this study, we analyzed nutrient cycling structure of a small farm (cattle of 100 heads and arable lands of 2.5 ha) in Jeonnam province to investigate the effects of nutrients input by the addition of bedding materials (sawdust and rice hull) and nutrients loss before the application to the soils (the period during manure storage in the feedlot and composting process) on nutrient cycling structure. Sawdust and rice hull added as bedding materials increased N by 1.6% and 14.2% and $P_2O_5$ by 3.1% and 27.4%, respectively, relative to the amount of nutrients produced by excretion. This result suggests that the addition of nutrients via bedding materials should be considered for better estimation of nutrient balance. The most significant characteristics of the nutrient cycling structure was loss of mass and nutrients during the storage (21 days) and composting period (90 days). During this period, 78.4% of N and 9.5% of $P_2O_5$ was lost from sawdust compost; meanwhile, the percentages of loss for rice hull compost were 81.6% and 10.3%, respectively. A lower percentage of nutrients loss in sawdust compost than that in rice hull compost was attributed to the relatively slow decomposition rate of organic materials in the sawdust compost which has higher C/N ratio and lignin contents. Therefore, it was concluded that estimation of nutrient balance should be conducted based on nutrient contents in the final compost being applied to the lands rather than the amount of nutrients contained in the livestock excretion. In addition, the effects of bedding materials on nutrient losses should be also taken into account.

Quality Comparison of Chuncheon Dakgalbi Made from Korean Native Chickens and Broilers (토종닭과 육계로 만든 춘천닭갈비의 품질비교)

  • This study was conducted to evaluate a quality comparison between Chuncheon Dakgalbi made from Korean native chickens (KNC) and that made from commercial broilers. Two Korean native chickens including Woorimatdag (KNCWoori) and Hanhyup3 (KNC-Hanhyup), and two commercial broilers including grades of 18 (Broiler-18) and 13 (Broiler-13) were slaughtered at 110, 70, 38, and 31 d of ages. Chuncheon dalkalbi was prepared by mixing/dipping the meat in chili pepper sauce; it was then packed with air-packaging (Air-P) and 30% $CO_2$-MAP (0% $O_2$/30% $CO_2$/70% $N_2$), and stored at $5^{\circ}C$ for 10 d. The results showed that the KNC group had a lower pH but a higher cooking loss compared with the broiler group (p<0.05). In a texture analysis, KNC-Woori had the highest shear force value among the breeds (p<0.05). For the fatty acid composition of the thigh, the KNC-Woori contained more total saturated acids, myristic acid, palmitic acid and stearic acid, but less total unsaturated fatty acids, linoleic acid and linolenic acid than other breeds (p<0.05). Also, the n6/n3 ratios of the KNC group (19.24 and 16.77) were higher than those of the broiler group (14.02 and 14.77) (p<0.05). The total acceptability scores of Dakgalbi made from the KNC group were decreased by sensory panelists. The Dakgalbi with 30% $CO_2$-MAP delayed the protein deterioration (Volatile basic nitrogen) and lipid oxidation during storage. However, no clear evidence was observed of $CO_2$-MAP on the effect of different chicken materials. It is suggested that 30% $CO_2$-MAP instead of Air-P is used for methods for Chuncheon Dakgalbi. Furthermore, it might be unfavorable to use Korean native chickens as raw material for Chuncheon Dakgalbi from a practical quality point of view.

Effect of Feeding High Carbohydrate-Low Fat Fermented Feed on the Meat Quality Characteristics in Finishing Pigs (저탄수화물 고지방 발효사료 급여가 돈육의 육질 특성에 미치는 영향)

  • A total of ninety swine ($79.0{\pm}2.2\;kg$) were employed for 58 d to determine the meat quality of pigs fed fermented agricultural by-products (FAB) mainly consisting of brewers grain shell. FAB was replaced with commercial feed at dietary levels of 20%, 40%, 60%, 80%, and 100% (T1) and 30%, 60%, 100%, 100%, and 100% (T2) at 1, 2, 3, 4 and 5-9 wk, respectively. Compared with the control (CON) feed, FAD feed had lower moisture and nitrogen-free extract content, and higher crude fat, crude fiber, and total calorie (p<0.05). The protein content, amino acid profile, and pH values of pork loin were not affected by dietary treatment. However, higher moisture, crude ash, and meat cholesterol, and lower fat, were found in CON compared with treatment (p<0.05). FAB treatment significantly improved drip loss and cooking loss value (p<0.05), and increased the CIE $L^*$ values of loin and back-fat surface, whereas it decreased the CIE $a^*$ values of loin surface (pp<0.05). The results indicate that dietary FAB affected meat cholesterol and fat content, and improved drip loss and cooking loss, but had no affect on amino acid composition.

Tumor Suppressive Effect of Zoledronic Acid on Human Osteosarcoma Cells in Vivo (인간 골육종 세포주에서 Zoledronic acid의 종양 억제에 대한 생체내 실험)

  • Purpose: Bisphosphonates (BPs) are the analogues of endogenous pyrophosphates: they have been used in the treatment of skeletal diseases such as Paget's disease, osteoporosis, and tumorinducing ostelysis, and are used in treatment of osteolytic metastasis of breast cancer recently. They are also used as one of the therapeutic agents for metastasis of prostatic cancer of which metastasis makes the mixed nature of osteolysis and ostegenesis. Although the action mechanism of BPs are well known for diseases with excessive osteoclastic bone resorption, the direct effect of BPs has not been known yet. This study was intended to see the tumor suppression capability of Zoledronic acid(ZOL) using nude mouse with osteosarcoma. Materials and Methods: MG-63 and HOS osteosarcoma cell lines were used and the transforemed MG-63-GFP and HOS-GFP cells, which were made for detection under fluorescent light, were subcutaneously injected to make osteosarcoma. The five 6-week male mice were used for the experiment at each group. After the injection, mice were cultivated until tumor pieces grow up to $3{\times}3{\times}3$ $mm^3$ and ZOL of 120 ug/kg was subcutaneously injected twice a week. Sizes of tumor were measured twice a week and photographed under fluorescent light. Results: In in vivo test with HOS osteosarcoma cell lines, mean size of tumors was 2,520 $mm^3$ in control group and was 131 $mm^3$ in ZOL group, which showed 94% of reduction comparing with the control ; with MG-63 osteosarcoma cell lines, mean size of tumors was 2,866 $mm^3$ in control group and was 209 $mm^3$ in test group with 72% of reduction (p<0.05). Conclusion: In in vivo tests with nude mice, we suggest that ZOL has direct effect on osteosarcoma cells and it would be used as one of the therapeutic agents for osteosarcoma, especially to ZOL-sensitive osteosarcoma cells.

Effect of Water Temperature on Ammonia Excretion of Juvenile Pacific Cod Gadus macrocephalus (대구 Gadus macrocephalus 치어의 암모니아 배설에 미치는 수온의 영향)

  • A study was carried out to examine the effect of water temperature on daily pattern and rate of total ammonia nitrogen (TAN) excretion in juvenile Pacific cod Gadus macrocephalus (mean body weight: $36.5{\pm}0.8\;g$) under fasting and feeding conditions. Fish were acclimated over 10 days under three different water temperatures (9, 11 and $13^{\circ}C$), and transferred to TAN measuring system under each water-temperature condition. After 72 hours of starving, fasting TAN excretion was measured at each temperature. To investigate post-prandial TAN excretion, fish were hand-fed with a commercial diet containing 40.6% crude protein for 7 days, two times daily at 08:00 and 16:00 h. Water was sampled from both the inlet and outlet of the fish chamber every 2 h over a 24-h period. Both fasting and post-prandial TAN excretion increased with increased water temperatures (p<0.05). Mean fasting TAN excretion rates at 9, 11 and $13^{\circ}C$ were 9.3, 11.0 and $11.9\;mg\;TAN\;kg\;fish^{-1}\;h^{-1}$, respectively. The value of $9^{\circ}C$ was lower than those of 11 and $13^{\circ}C$ (p<0.05), but there was no significant difference between $11^{\circ}C$ and $13^{\circ}C$. Mean post-prandial TAN excretion rates at 9, 11 and $13^{\circ}C$ were 23.0, 31.6 and $45.4\;mg\;TAN\;kg\;fish^{-1}\;h^{-1}$, respectively. A peak value of post-prandial TAN excretion rate occurred after 2 h from each feeding, and the second value is always higher than the first value. Maximum post-prandial TAN excretion rate occurred after 10 h from the first feeding at $9^{\circ}C$ (mean $38.0\;mg\;TAN\;kg\;fish^{-1}\;h^{-1}$), $11^{\circ}C$ ($52.9\;mg\;TAN\;kg\;fish^{-1}\;h^{-1}$) and $13^{\circ}C$ ($77.5\;mg\;TAN\;kg\;fish^{-1}\;h^{-1}$), respectively. The TAN loss for ingested nitrogen at $9^{\circ}C$ (43.9%) was lower than those of $11^{\circ}C$ (46.4%) and $13^{\circ}C$ (48.4%). The overall results indicate that water temperature exhibits a significant effect on the nitrogen excretion of juvenile Pacific cod.

Effects of Dietary Beef Tallow, Soybean Oil and Squid Liver Oil on Growth and Body Composition of the Korean Rockfish Sebastes schlegeli, and Biochemical Changes with Starvation (사료 지질원으로 우지, 대두유 및 오징어 간유 첨가에 따른 조피볼락 Sebastes schlegeli의 성장 및 체성분 변화와 절식시 체내 대사)

  • The effects of different dietary lipids on growth, body chemical composition, and nutrient partition of the Korean rockfish (43 g of initial mean weight) were studied. Fish were fed three experimental diets containing 1) $4.5\%$ squid liver oil, 2) $1\%$ squid liver oil and $3.5\%$ beef tallow, and 3) $1\%$ squid liver oil and $3.5\%$ soybean oil. Growth and feed efficiency were detarmined during a 19-week feeding period at water temperature of $15.5^{\circ}C$, and at the end of which body chemical compositions were analyzed. Remained fish were fasted for 45 days, and four fish were taken at 6, 12 and 24 hours, and 3, 15 and 45 days after the fasting to determine tissue compositions and hematological changes. Growth, feed efficiency, protein and lipid retention efficiency, hepatosomatic index(HSI), viscerosomatic index (VSI), and chemical composition of whole body and dorsal muscle were not affected by the different dietary lipids. Liver moisture content of fish fed squid liver oil diet was higher than that of fish fed beef tallow or soybean oil diet. Liver lipid content of fish fed beef tallow diet was higher than that of fish fed the other diets. Fatty acid composition of dorsal muscle and liver were affected by the different dietary lipids ; high levels of 20 : 5n-3 and 22 : 6n-3 from fish fed the diet containing squid liver oil, 18 : 1 from fish fed the diet containing beef tallow, and high 18 : 2n-6 and 18 : 3n-3 from fish fed the diet containing soybean oil were observed. Both HSI and VSI of fish fed three diets decreased with time after the begining of starvation. Liver glycogen did not change during the first 15 days of starvation and decreased thereafter, and that was not affected by the different dietary lipids. Lipid and protein contents in the dorsal muscle of fish decreased up to 15 days of starvation and remained unchanged thereafter, these were not different from each other. Glucose, free fatty acid, triglyceride and phospholipid concentrations in Fish serum were varied for the first 15 days of starvation, after that the concentrations of fish serum remained relatively stable in all the treatment groups with prolonged starvation. The results indicate that Korean rockfish can utilize fish oil, animal fat or vegetable oil equally as energy source when n-3HUF A is adquate.

Effects of Salinity on Spontaneous Maturation and Induced Spawning in Seabass, Lateolabrax japonicus (실내사육 농어, Lateolabrax japonicus의 성 성숙 및 산란 유도를 위한 염분의 영향)

  • To study the effect of salinity on maturation and spawning, two series of experiments were carried out during November, 1998- July, 1999 and January, 1999- March 2000. In the first series, the control group (C) was reared at 14.3$^{\circ}C$ and 35.1$\textperthousand$ S, but the experimental group was reared at 33.8$\textperthousand$ S. In the second series, they were also exposed to approximately the same temperature but the experimental group was reared at still lowered salinity of 2p.6$\textperthousand$. Survival, food intake and growth of the experimental groups in either series reared at lower salinities were higher than those of their respective control groups. At lower salinity, larger number of females attained spontaneous sexual maturity and successfully spawned. Hatching success of the eggs spawned by the experimental females reared at lower salinities was also higher (54-56$\textperthousand$) than that (~19%) of the control group reared at higher salinity.

Effects of Scutellaria baicalensis Water Extract on Lipid Metabolism and Antioxidant Defense System in Rats Fed High Fat Diet (황금추출물이 고지방 식이를 급여한 흰쥐의 지질대사 및 항산화방어계에 미치는 영향)

  • The objective of this study was to determine the effects of a Scutellaria baicalensis water extract (SDWE) on lipid levels, lipid peroxidation and antioxidant enzyme activities on rats fed a high fat diet for 6 weeks. Thirty-two male Sprague-Dawley rats (4-weeks-old) were randomly divided into four groups: normal diet and deionized water (ND), normal diet and Scutellaria baicalensis water extract (NDS), high fat diet and deionized water (HFD), high fat diet and Scutellaria baicalensis water extract (HFDS). The food intakes were significantly lower, but the food efficiency ratios were significantly higher in the high fat diet groups than those in other groups. The level of HDL-cholesterol and HDL-cholesterol/total cholesterol ratio in plasma were significantly higher and AI (atherogenic index) in HFDS group was significantly lower than that in HFD group. The level of triglyceride in plasma was significantly decreased in SDWE groups. The triglyceride of liver was significantly increased in the high fat diet groups and the total cholesterol of liver in the HFDS group was significantly lower than that in the HFD group. The plasma and liver concentrations of thiobarbituric acid reactive substances (TBARS) in the NDS group were significantly lower than those in the ND group. The total antioxidant status (TAS) in plasma was significantly increased in the HFDS group compared to the HFD group. The activities of SOD, catalase and GST were significantly increased in SDWE groups compared to ionized water groups. The activity of GSH-Px and the concentration of GSH in liver in the HFDS group were significantly higher than those in the HFD group. These results suggest that a supplement of SDWE on rats fed high fat diet reduce levels of lipid and lipid peroxidation in plasma and liver and improve the antioxidant defense systems.

Studies on the Morphological Characteristics and Life History of the Chili Fish, Cultriculus eigenmanni (Jordan et Metz) (1) On the Egg Development and the Fry in the Larval Stage (치리(Cultriculus eigenmanni)의 형태 및 생활사에 관한 연구 I. 난발생과 부화자어에 대하여)

  • The Chili fish, one of the most common species of the freshwater fishes, is distributed in the river basins of the western coast of Korea. However, any studies on this fish, especially its development or life history, have not been made so far. The authors, therefore, carried out an experiment to reveal some characteristics of egg develoment and fry in this fish. For accomplshing this study, we caught some adult Chili fish by means of a casting net at raceway in the vicinity of Kunsan in June and July, 1983; then, we injected 0. 1 cc Gonadotrophic hormone 1,000 unit solution (Puberogen) in order to induce the spawning on the abdominal cavity of male and female respectively, Approximately 12 hours after injection, the artificially fertilized eggs by using of the dry method and the naturally fertilized eggs were observed simultaneously in the aquarium ($30{\times}50{\times}70cm$). The fertilized eggs, in spherical shape, were sperative, demersel, adhesive nature and $1.38{\sim}1.59\;mm$ in diameter. The light yellow yolk was opaque and $1.17{\sim}l.27\;mm$ in diameter. The hatching of eggs took place at 29.5 hours after fertilization under the condition of $23.5{\sim}25.0^{\circ}C$ water temperature. Basically there were not found so many different aspects in comparison with the common species of Cyprinidae in the development process of eggs : however, there were a few noticable characteristics, that is, pigment cells, eye balls and blood circulation are invisible, or the velocity of egg development is rapid, seemed similar to the Loach's egg. Newly hatched larvae with the yolk sac (2.39 mm in size) were $3.87{\sim}3.99\;mm$ in total length and their myotome number was expressed as 28+15=43. About 20 hours after hatching, a caudal and a pectoral fin as well as an anus began to orgin. In addition, a few malanophore showed on the upper regin of the yolk sac of the larvae whose size was 4.73 mm in total length. The larvae developed about 5.02 mm in total length, and the median fin fold in the abdomen extended forward to two thirds or beyond of the yolk sac at 50 hours after hatching. Simultaneously, the eye balls and mouth also appeared. After 3 days, the yolk sac nearly disappeared, and the larvae (5.4 mm in total length) with perfect eye balls began to swim for themselves for the first time. Also, the specialization between the gill arches and the ailmentary canal, the development of each fins except dorsal fin as well as the blood circulation obviously distinguished in this stage. The larvae whose total length was 5.68 mm was very brisk: also, the development of the dorsal fin rudiment, nearly perfect formation of each organs and the function of the upper and lower jaws were noticeably observed 5 days after hatching.
