Runoff Characteristics of Non-point Source Pollutants in apple cultivation (사과재배지의 비점오염물질 유출특성)

  • 본 연구에서는 사과재배지에서 발생하는 비점오염물질 유출 및 수질특성을 살펴보고자 2014년 5월부터 9월까지의 총 15회의 강우사상에 대해 유출특성을 분석하고 오염물질별 유량가중평균농도(Event Mean Concentration, EMC) 및 오염부하를 산정하였다. 모니터링 기간동안 3.2~80.3 mm의 강우가 발생하였으며, 조사된 총 15회의 event 중 3회 유출이 발생하였다. 강우강도는 0.39~4.46 mm/hr의 범위로 나타났으며, 선행무강우일수는 0.3~20.1일, 총 유출량은 $2.7{\sim}100.8m^3$, 유출율은 0.02~0.3의 범위로 나타났다. 강우모니터링 결과, EMC는 TOC 7.1~18.7 mg/L(평균 12.0 mg/L), BOD 4.3~7.8 mg/L(평균 5.9 mg/L), COD 16.9~30.2 mg/L(평균 22.3 mg/L), SS 35.6~738.1 mg/L(평균 293.1 mg/L), T-N 2.342~11.043 mg/L(평균 6.563 mg/L) 그리고 T-P 1.563~2.563 mg/L(평균 1.961 mg/L)의 범위로 나타났으며, 각 강우사상에 대한 단위면적당 오염부하는 TOC 0.12~2.4 kg/ha, $BOD_5$ 0.03~1.0kg/ha, $COD_{Mn}$ 0.12~7.1 kg/ha, SS 0.34~173 kg/ha, T-N 0.068~1.478 kg/ha, T-P 0.011~0.601 kg/ha의 범위로 산정되었다. 사과재배지의 경우 대부분 투수지역으로 강우 초기에 지하로 침투하는 양이 많기 때문에 초기세척효과는 크게 발생하지 않는 것으로 나타났으며, 대체적으로 30 mm 이하의 강우에서는 유출이 발생하지 않는 것으로 분석되었다. 하지만, 사과재배지에서 발생하는 비점오염물질의 유출특성은 기상조건 및 영농활동 조건등에 따라 상이하게 나타나기 때문에 강우시 비점오염원 유출특성을 보다 정량적으로 파악하기 위해서는 체계적이고 지속적인 연구가 필요할 것으로 판단된다.

  • The effect of medium composition on PLB formation and multiplication and shoot regeneration was studied to establish the micropropagation system of the tropical orchid Phalaenopsis. The highest frequence of PLB formation resulted from the VW medium with 1.2 times ion concentration, 1% sucrose, 1.5g/L PVP or 2.5g/L active charcoal, apple and potato extract and 4g/L gellan gum. The highest ratio of PLB multiplication was obtained from the VW medium with 2% sucrose, apple and potato extract and cotton plate. The shoot regeneration was the most effective with the hyponex medium with 3% apple, 3% potato and 4% banana extract.

Kanamycin Concentration for Selection of 'Mcintosh Wijcik' Transgenic Apple (사과 'McIntosh Wijcik' 형질전환체 선발을 위한 Kanamycin 농도)

  • Effects of kanamycin concentration on regeneration and rooting of transgenic 'McIntosh Wijcik' were investigated to establish the efficient Agrobacterium-mediated transformation system. Relatively high regeneration frequency of explants appeared even at the high concentration of $150mg{\cdot}L^{-1}$ kanamycin, but the regeneration frequency and the number of normal shoots decreased significantly at a concentration of higher than $100mg{\cdot}L^{-1}$ kanamycin in the gelrite-gellifying medium. Rooting response varied with the transgenic lines in the agar-solidifying medium supplemented with the different concentrations of kanamycin and they were grouped with the inhibition level at $30mg{\cdot}L^{-1}$ concentration. No correlation between copy number and root response was observed. The optimum concentrations of kanamycin for the regeneration of 'McIntosh Wijcik' apple in the medium gellified with gelrite and for indirect-selection of putative transformants in the rooting medium solidified with agar were found to be $100mg{\cdot}L^{-1}$ and $30mg{\cdot}L^{-1}$ respectively.

  • This study was conducted to investigate the effect of culture media and environment on PLB proliferation by using PLBs produced from leaf segments excised from shoot of Phalanopsis flower stalk. The fresh weight of PLBs propagated was higher in MS medium than in NDM (New Dogashima medium) or VW, but the condition of PLB was better in NDM medium. Natural additives of Coconut water, potato and apple were absolutely required for the PLB propagation. PLB propagation was better in solid medium than in liquid medium including cotton as support. Optimal sucrose concentration for proliferation was 10 g/L. PLB proliferation was very effective condition 14.3 $\mu$mol.s$^{-1}$ m$^{-2}$ in PPFD and $25^{\circ}C$.

  • Some factors of wounding methods, solidifying agents, origin of leaf explants, cone. of acetosyring-one, and MES affecting regeneration and transformation by Agrobacterium tumefaciens were investigated to establish an efficient transformation system of apple. Wounding by cutting the leaves merely showed the tendency of regeneration and transformation with higher efficiency compared with that of wounding by non-traumatic forcep when carrying out co-cultivation for three days after bacterial inoculation. While examining the solidifying agents of medium with the combination of agar (A)+Gelrit $e^{ }$ (G) in g. $L^{-1}$ , the higher concentration of Gelrit $e^{ }$ increased the efficiency of regeneration. However, there was no difference in the efficiency of transformation from the treatments of 2.5 G, 3.5 A+1.2 G, and 7.0 G. The origin of leaf explants showed no difference statistically in the efficiency of regeneration and transformation, but that from the shoots of proliferation medium showed the tendency with higher efficiency. The concentration of above 0.1 mM acetosyringon had an increase in the efficiency of regeneration and transformation and the concentration of 0.15 mM had the highest efficiency of transformation in the treatment of acetosyringone with different concentration. There was no effect of MES on regeneration and transformation.ion.

Analysis of Migration of the Oriental Fruit Moth, Grapholita molesta, in Apple-Cultivating Areas Based on Population Monitoring Using Sex Pheromone and RAPD Molecular Marker (성페로몬과 RAPD 분자지표를 이용한 사과 재배지 복숭아순나방(Grapholita molesta)발생 모니터링과 집단 이동 분석)

  • Local and seasonal populations of the oriental fruit moth, Grapholita molesta, were monitored with sex pheromone trapping and RAPD (random amplified polymorphic DNA) molecular marker to analyze their movement in apple orchards. To detect their movements among farms, pheromone traps were placed at regions between apple farms ('outside-farms') as well as within-farms ('inside-farms'). Four seasonal adult peaks were evident in apple-cultivating fields from April to October in both trappings of inside- or outside-farms. After overwintering generation, populations of inside-farms were significantly reduced with frequent insecticide applications, compared to populations of outside-farms. Within apple farms, G. molesta tended to be unevenly distributed because of significant sublocal preference. Active movements of local and seasonal populations of G. molesta were supported by gene flow analysis using RAPD marker. Monitoring data using sex pheromone and seasonal reduction in initial genetic differentiation detected in the overwintering populations suggest that there must be significant movement of G. molesta among different orchards in apple-cultivating areas.

Damage and Occurrence of Pratylenchus vulnus and Paratylenchus sp. on Spray Chrysanthemum, Dendranthema grandiflourm Kitamura (스프레이국화재배지의 사과뿌리썩이선충과 침선충의 발생 및 피해)

  • Root-lesion nematode (Pratylenchus vulnus) and pin nematode (Paratylenchus sp.) were detected with high population density at the spray chrysanthemum greenhouse in Gumi, Gyeongbuk. The average density of P. vulnus and Paratylenchus sp. was 667 and 716 nematodes per 100 g soil and P. vulnus density were distributed 87% to the depth of $0{\sim}30$ cm in greenhouse. When spray chrysanthemum cv Chopin, was transplanted in September 2004 in greenhouse, P. vulnus were 854 nematodes/100 g soil in planting and increased 14,985 nematodes/100 g soils and 1g root after 10 weeks in harvest. Shoot weight and shoot height of spray chrysanthemum decreased 24.8% and 8.0% compared with non-infested P. vulnus after 10 weeks in greenhouse.

Plant Regeneration from Cotyledon and Hypocotyl Culture in Apple Hybrid Seedling (P.16 $\times$ Malus prunifolia) (사과 P.16 $\times$ Malus prunifolia 교잡실생의 자엽 및 배축배양에 의한 식물체 재분화)

  • Adventitious shoots were induced from cotyledon and hypocotyl explants of apple hybrid seedlings (P.16$\times$ Malus prunifolia) on MS medium supplement with 2,4-D and various cytokinine (Kn. BA, TDZ) The shoot regeneration from the cotyledon culture was the highest on MS medium supplemented with 1.0 mg/L NAA and 2.0 mg/L BA. Whereas in case of hypocotyl culture, it was the highest on MS medium supplemented with 0.5 mg/L NAA and 0.5 mg/L BA. However, in the MS medium without BA there was no shoot regeneration. Hypocotyl culture seemed to be more effective than cotyledon culture in shoot regeneration. Specially, the top position of the hypocotyl found to be the best explant for shoot induction among the other segments of hypocotyls. Regenerated shoots were rooted on half-stength MS medium with 1.0 mg/L NAA. Above results suggest that Apple hybrid (P.16 $\times$ Malus prunifolia) can be multiplied via cotyledon or hypocotyl culture systems.

Genetic Differentiation of Overwintering Populations of Oriental Fruit Moth, Grapholita molesta, and Their Movement (월동세대 복숭아순나방(Grapholita molesta)의 지역적 분화 및 이동)

  • Spring phonology of the oriental fruit moth, Grapholita molesta, was monitored using sex pheromone traps in apple cultivating areas. Their occurrence was earlier in southern areas and their population sizes were significantly different among orchards even in a local cultivating zone. The overwintering populations appeared to move between local orchards, based on the fact that monitoring data obtained at the sites between orchards were similar to those of nearby orchards. However, within orchards, these adult movements appeared to decrease and showed skewed occurrences at the side of upwind direction or close to neighboring orchards. At initial occurrence peak(April 20-25), the ovenvintering populations of the different localities were collected and analyzed in their genetic distances. PCR-RAPD analysis indicated that there were significant genetic differences among the overwintering populations of G. molesta. This genetic differentiation of overwinterin populations may be due to genetic bottleneck following differential selection pressures against the subpopulations of G. molesta during winter on the basis of the RAPD analysis that each early spring population was significantly different to its previous fall population in the same locality.
