Immunocytochemical Study on the Enteroendocrine Cells in the Gastrointestinal Tracts of the Korean Snakes (한국산 뱀목 위장관 점막이 장내분비세포에 대한 면역세포화학적 연구)

  • This study attempts to investigate several enteroendocrine cells in the gastrointestinal epithelia of the Korean snakes (Dinodon rufozonatum rufozonotum Rhabdophis tigrina tigrina, Enhydris rufodorsata, Agkistrodon blomhoffii brevicaudus, Agkistrodon saxatilis, Agkistodon calginosus). For a light-microscopical examination of immunocytochemistry, the paraffin sections (5 $\mu$ m) of tissue specimens taken from the various parts of the gastrointestinal tract were stained immunocytochemically by PAP procedure with 10 antisera. The frequency of enteroendocrine cells per unit area (mm$^2$) of each mucosa were counted and the shapes of the cells were observed. In Dinodon rufozonatum rufozonatum, Rhobdophis tigrina tigrina, Enhydris rufodorsata, Agkistrodon saxatilis and Agkistrodon caliginosus, cholecystokinin (CCK)-8, gastrin, pancreatic polypeptide (PP) and serotonin cells were observed. But the freuqency of these immunoreactive cells differ trom each portion of gastrointestinal tracts of all species, respectively. In Agkistrodon blomhoffii brevicaudus, CCK-8, gastrin and serotonin cells were observed. CCK-8 and serotonin cells were found in whole gastrointestinal tracts and gastrin cells were observed in pylorus and mucosa of small intestine. The frequency of these cells was different from each portion. The shapes of CCK-8, gastrin, PP and serotonin cells were pyramidal or oval and closed type in stomach. A large number of these cells were spindle in shape and open type in small intestine and anterior pant of large intestine, whereas some cells were closed type. In posterior part of large intestine and rectum, these cells were oval in shape and closed type.

Quality and Nutrition Labeling Study of Domestic Fruit (Plum) (국내산 청과물 (자두)의 품질 영양표시에 관한 연구)

  • In order to develop a model on the quality and nutrition label of Korean fruit plum, we first examined the quality and nutritional characteristics of Korean Plum, ChooHee. The contents of moisture, crude protein, crude fat, crude fiber, crude ash and N-free extinct were 92% 0.74% 0.64% 1.65%, 0.32% 4.29% respectively. Thus the total calorie of the sample was 32 kcal/100 g. The content of sugar, acidity and Vitamin C were $33{\pm}0.85 Bx^{\circ}$, $1.08{\pm}0.12%$ and $7.037{\pm}0.317mg%$ respectively. The principle minerals were S, K, Ca, Na, and Mg. Major free sugars were fructose, glucose and sucrose and major organic acid were succinic acid, malic acid and citric acid. The difference of contents of total phenols between peel and flesh pare were considerable. The total phenolic contents of whole fruit were $75.55{\pm}0.73mg%$. On the above results, the quality and nutrition label of Korean plum were developed in the first time. We expect that this study on labeling could play an important role to the quality control and marketing of Korean agricultural products.

Response of Turnip to Botrytis cinerea Infection and Their Relationship with Glucosinolate Profiles (잿빛곰팡이병에 대한 순무잎의 반응과 Glucosinolate 함량변화)

  • In order to determine the response of turnip leaves to various races of Botrytis cinerea, fourteen different races were infected to the leaf surface and measured the lesion size. Based on lesion size of the pathogen on the leaves, turnip showed susceptible response to four races, moderate to seven races and resistant to three races. Four glucosinolate (GLS) compounds, such as butenyl GLS, indole-3ylmethyl GLS (I3M), 4-methoxy-indole-3-ylmethyl GLS (4MOI3M) and normal methoxy-indole-3ylmethyl GLS (NMOI3M) were identified in turnip leaves infected with Botrytis cinerea. Leaves infected with resistant races showed higher GLS contents as compared with the leaves infected with susceptible races. Contents of I3M in the leaves with resistant races were 2.5 times as high as those in uninfected leaves, whereas I3M in the leaves with susceptible races showed lower contents than those in untreated leaves. Leaves infected with resistant races showed 4MOI3M and NMOI3M contents 2.3 and 2.7 times as high as those in uninfected leaves, respectively. GLSs in the infected leaves were most abundant at 5~10 mm area from center of the lesion. However, the GLSs in 5~10 mm area were rapidly degradated at leaves with susceptible races which resulted in continuous expansion of the lesion on the leaves, whereas no degradation was obseveved at leaves with resistant races. These results suggest that accumulation and degradation of GLS compounds in turnip leaves closely related to the susceptibility and resistance of turnip leaves to Botrytis races.

A Trial for Preparation of Jam using Sea Mustard Stem (미역줄기를 이용한 잼의 제조조건)

  • Preparation of sea mustard stem jam (SMSJ) with addition of commercial ingredients, sucrose, high methoxyl pectin (HM pectin) or low methoxyl pectin (LM pectin), organic acid (or its salt), and synthetic strawberry flavor to sea mustard stem paste was investigated. The sea mustard stem paste was produced by crushing sea mustard stem and homogenizing it with 2.5 times water of sea mustard stem, then followed by filtering ($30{\~}35 mesh$), The effects of those ingredients on the physical properties and the sensory score of SMSJ were evaluated. The SMSJ prepared with combination of $65{\%}$ (w/w) sugar, $1.0{\%}$ (w/w) pectin, and $0.3{\%}$ (w/w) citric acid or with combination of $65{\%}$ (w/w) sugar, $1.0{\%}$ (w/w) U pectin, and $0.08{\%}$ (w/w) calcium lactate, which finally adjusted to $62{\cdot}Brix$, was similar to commercial strawberry or apple jam in gel strength, Addition of over $0.06{\%}$ (v/w) synthetic strawberry flavor to sea mustard stem paste showed a good effect of masking the undesirable sea mustard flavor.

Monitoring Program of Organochlorine Pesticide Residues in Agricultural Products Provided for the Army (군납 농산물 중 유기염소계 농약의 잔류량)

  • This study was carried out for dietary safety based on the level of pesticide residues in 17 kinds of the agricultural products (apple, cabbage, cabbage kor, carrot, cucumber, pepper, onion, potato, radish, spring onion, pumpkin, lettuce, garlic, drapwort, spinach, leek crown daisy) consumed in Kyunggi-do and Inchon-city that provided for the army. From January 1998 to December 2001, ten organochlorine pesticides in 1075 samples were analyzed by using GC-ECD. According to the results, one kind of pesticides was detected in 135(12.5%) samples, and two kinds were in 7 (5.18%). While dicofol and tetradifon was not fecund. Detection rate of pesticides were 9.02% for $\alpha$, $\beta$-endosulfan, 2.8% far chlorothalonil, 0.46% for γ-BHC, 0.40% for chlorobenzilate, 0.33% for folpet, 0.28% far DDT, 0.15% for captafol, and 0.11% for captan. Dectection ranges of pesticides were from 0.001 to 0.092 ppm for $\alpha$, $\beta$-endosulfan, from 0.003 to 0.023 ppm for chlorothalonil, from 0.003 to 0.006 ppm far γ -BHC, from 0.001 to 0.003 ppm for DDT, from 0.009 to 0.095 ppm for folpet, 0.05 ppm for captan, 0.01 ppm for captafol, and 0.008 ppm far chlorobenzilatel respectively. Consequentlyl all detected pesticide levels in samples were below maximum residue limits(MRLs) in Korea and Codex.

Key Food Selection for Assessement of Oral Health Related Quality of Life among Some Korean Elderly (일부 한국 노인 구강건강 관련 삶의 질 평가를 위한 핵심 음식 선택)

  • Oral health can influence on diverse food intake, and food intake affect oral health related quality of life. The aim of this study was to select key foods to be able to represent oral health related quality of life in Korea. We used the data of 503 Korean older persons to participate in the oral health promotion programme in 2009. The low consumption or low intake foods with criteria in 2012 National Nutrition Statistics were eliminated among 30 foods of food intake ability (FIA) at first. Decision tree model, correlation analysis, factor analysis, and internal reliablity test were used for oral health related quailty of life (OHRQoL) key food selection. We selected 13 foods-hard persimmon, dried peanut, pickled radish, caramel, rib of pork, glutinous rice cake, cabbage kimchi, apple, yellow melon, boiled chicken meat, boiled fish, mandarin, noodles as OHRQoL Key Foods 13. Thirty foods of FIA and OHRQoL Key Foods 13 displayed the same pattern of variation among sociodemographic groups. In a regression model, both of 30 foods of FIA and OHRQoL Key Foods 13 influenced on oral health impact profile-14. The findings suggest that OHRQoL Key Foods 13 have good reliability and validity and be able to use in oral health survey.

Occurrence and Distribution of Weeds on Orchard Fields in Chungbuk Province of Korea (충북지역 과수원 발생 잡초 분포 현황)

  • This study was conducted to investigate the weed occurrence and distribution on the orchard fields (apple, pear, peach, grape) at 387 sites in Chungbuk province of Korea from May to September in 2015. From the result of this survey, 200 weed species in 47 families were identified and classified to 82 annuals, 40 biennials and 78 perennials. Based on the occurrence ratio, the most dominant weed species in Chungbuk province orchard fields were higher in order of Digitaria ciliaris (4.12%), Echinochloa crus-galli (3.60%), Stellaria aquatica (3.45%), Artemisia princeps (3.07%) and Chenopodium album (3.06%). The composition of major occurred weed families, Compositae, Poaceae, Leguminosae and Cyperaceae were 21, 9, 6 and 6%, respectively. Coefficient of similarity between a various orchards based on the degree of dominance were ranged from 68.7-91.8%. The most important weed species at apple, peach, grape was Digitaria ciliaris while these for pear was Poa annua. Fifty-one exotic weed species were also identified. The results of this study could be useful information for estimation of future weed occurrence, weed population dynamics and establishment of weed control methods on the orchard fields in Chungbuk province of Korea.

Effect of weight of radio-transmitters on survival of Red-tongue viper snake (Gloydius ussuriensis) and Short-tailed viper snake (Gloydius saxatilis)in the radio-transmitter implantation (발신기 삽입시 발신기 무게가 쇠살모사(Gloydius ussuriensis)와 까치살모사(Gloydius saxatilis)의 생존에 미치는 영향)

  • In order to study the home range of the red-tongue viper snake (Gloydis ussuriensis) and the short-tailed viper snake (Gloydius saxatilis) belonging to the viperidae of squamat in Korea, we implanted radio-transmitters which were weighed less than 5% of individual mass in them and traced their location by radio-tracking. Surgeries for transmitter insertion were performed on 5 red-tongue viper snakes and 6 short-tailed viper snakes (total 11 individuals) and the average ratio of transmitter mass to body mass were 4.2% and 2.2%, respectively. After radio-transmitter implantation, all short-tailed viper snakes survived but 4 out of 5 red-tongue viper snakes did not survive during the convalescence stage, showing only 20% of survival rate. The results suggest that the ratio of transmitter mass to body mass should be less than 3.6% at least in these species, although previous studies have recommended various ranges from 1% to 7% as the acceptable ratios.

Effects of Microspraying of Water and Coating by White Materials on Fruit Sunburn Occurrence for 'Fuji'/M.9 Apple Tree (미세살수와 흰색 코팅제 도포가 '후지'/M.9 사과나무 과실 일소 발생에 미치는 영향)

  • This study was carried out to compare and analyze control methods of the sunburn occurrence that affected 'Fuji'/M.9 planting systems in Korea from 2001 to 2003. It is very important to control temperature of the surface of fruits, because sunburn may occur when the temperature of fruit surfaces reaches $40^{\circ}C{\sim}45^{\circ}C$ due to high air temperature and sunlight during growing periods. As control methods of the sunburn occurrence, white coating materials such as $CaCO_3$ or kaolin were applied four times at two-week intervals from late June, and microspraying of water was conducted when air temperature was over $31^{\circ}C$ from late July to mid-August. Both methods were effective for preventing the sunburn occurrence and improving fruit quality by decreasing peel's temperature of fruits and increasing photosynthesis.

High-Performance Liquid Chromatographic Determination of Phenothrin and Silafluofen Residues in Crops with Mass Spectrometric Confirmation (고성능 액체크로마토그래피와 질량분석법을 이용한 농산물 중 Phenothrin 및 Silafuofen의 정밀 잔류분석법 개발)

  • A high-performance liquid chromatographic (HPLC) method was developed to determine residues of phenothrin and silafluofen, known as synthetic pyrethroids, in agricultural commodities. Insecticide residues were extracted with acetone from representative samples of four crops which comprised rice, apple, pepper and cabbage. The extract was purified serially by liquid-liquid partition and Florisil column chromatography. For rice and pepper samples, acetonitrile/n-hexane partition was additionally adopted to remove nonpolar interferences. Reversed phase HPLC using an octadecylsilyl column was successfully applied to separate two phenothrin isomers and silafluofen from sample co-extractives. Intact parent compounds were sensitively detected by ultraviolet absorption at 226 nm. Recovery experiment at the quantitation limit validated that the proposed method could apparently determine phenothrin and silafluofen residues at 0.02 and 0.01 mg/kg, respectively. Mean recoveries of phenothrin and silafluofen from four crop samples fortified at three levels in triplicate were in the range of 82.4~109.8% and 83.7~109.8%, respectively. Relative standard deviations of the analytical method were all less than 10%, irrespective of crop types and spiking levels. A selected-ion monitoring (SIM) LC/mass spectrometry (MS) with electrospray ionization was provided to confirm the suspected residue of phenothrin, even though no sufficient ionization of silafluofen was obtained. Both phenothrin and silafluofen could be successfully confirmed by gas chromatography/MS SIM with electron impact at 70 eV. The proposed method is sensitive, repeatable and rapid enough to apply to officially routine inspection of agricultural products.
