Differences in Rooting Rates and Callus Formation According to the Cutting Dates and Soil Conditions of Sageretia theezans (상동나무(Sageretia theezans) 삽목시기와 토양조건이 발근 및 캘러스형성에 미치는 영향)

  • 본 연구는 최근 암세포 생장억제 효과가 입증되어 약용소재로서의 활용가치가 높은 상동나무 (Sageretia theezans)의 대량증식 연구의 기반을 마련하기 위해 삽수묘를 시기별로 채취하여 삽목실험을 실시하였다. 삽수묘는 제주 대정읍, 전남 완도 보길도, 청산도 인근에서 수집하였으며, 상동나무 삽목은 시기별로 2019년 3월 21일(춘계), 7월 22일(하계), 9월 2일(추계)에 실시하였다. 토양조건은 모래, 원예용 상토, 모래와 원예용 상토를 1 : 1 배율로 섞은 토양에 각각 치상하였다. 삽목 후 산림약용자원연구소 온 실에 약 100일간 보관하였으며 1일 2회 관수하여 관리하였다. 그 결과 춘계에 삽목한 삽수묘에서 66.4% 의 발근율로 나타났으며, 뿌리는 평균 17.5mm의 길이로 측정되었다. 하계는 27.4%의 발근율을 보였으 며, 뿌리는 평균 12.6mm의 발달을 보였다. 추계에는 29.6%의 발근율로 13.2mm의 뿌리 발달을 보였다. 상동나무는 춘삽이 추삽에 비해 발근율이 높고, 숙지삽이 상동나무 발근에 있어 유리한 것으로 사료된 다. 또한 토양은 모래에 삽목한 삽수묘가 60.3%로 가장 발근율이 높았으며, 그 다음으로 모래와 원예용 상토를 1 : 1 배율로 섞은 토양이 42.7%의 발근율을 보였다. 마지막으로 원예용 상토에서 25.4%의 발근 율을 보였다. 이는 상동나무 삽목시 배수가 양호한 상태가 뿌리 및 캘러스형성에 유리하게 작용하는 것 으로 사료된다.

  • In sweet potato cultivars 'Mokpo #29' and 'Sanchunza', shoots from extplants were formed 100% on the MS medium with 0.1 ㎎/L NAA and 2.0 ㎎/L BA after 30 days of culture and roots produced from the base of stem at frequencies of 66.7% ('Mokpo #29') and 69.2% ('Sanchunza'), respectively, The media with 0.5∼4.0 ㎎/L BA were produced the greatest frequency of multiple shoot and the most of shoots developed rapidly into normal plantlets with rooting within 60 days of culture. Whereas the cultivar 'Keumsi' failed to produce normal shoot multiplication on the medium with cytokinins alone because of callusing of adventitious shoots. When single shoots with 1 to 2 nodes were excised from the multiple shoot or shoots covered with callus devoid of root and transferred to MS medium with 4.0 ㎎/L BA or kinetin. Host divided shoots showed the callus induction at the stem base and it was enable to obtain regenerated plantlets with shoot and root normally.

  • 밀은 세계 3대 작물로 국내 1인당 소비량은 지속적으로 증가하고 있다. 하지만 국내 밀의 유전적 배경 확대와 기후변화 신속 대응을 위한 국내 밀 유전자 풀의 확장은 밀의 질적 향상을 위해 매우 중요한 목표이다. 국내 자생 갯그령(Leymus mollis)은 해안가에서 번식하는 영년생 식물로 뿌리줄기를 이용한 왕성한 번식력을 가지고 있다. 또한, 해안가의 뜨겁고 염에 대한 적응성과 저항성을 가지고 있다. 이러한 특성은 밀 유전자 풀의 확장에 매우 유용할 것이다. 국내 밀 재배지 한계 극복을 위한 간척지 재배가 가능한 내염성 강화 밀 자원 개발을 위하여 모본인 보통 밀(Triticum aestivum L., Chinese Spring)과 부본인 갯그령을 원연교배하였다. 갯그령과 보통 밀의 원연 교배를 통한 종자 형성은 매우 어려우나 불가능한 것은 아니며, 최종적으로 10개의 교배 종자를 얻어 F1 식물체로 생장하는 과정에서 5 식물체는 고사하였고, 나머지 5 식물체는 영년생 특성인 뿌리줄기에 의해 새로운 줄기가 출현하는 것을 확인하였다. 또한, 갯그령의 DNA를 이용한 genomic in situ hybridization 방법으로 F1 식물체에서 갯그령의 염색체가 밀의 유전적 배경에 이입된 것을 확인하였다. F2 식물체는 모본인 보통 밀보다 긴 수장과 간장을 나타내고 이삭 수는 많았지만, 출수기는 보통 밀보다 3주 이상 늦어지는 것을 확인하였다. 내염성 평가를 위하여 F2 종자를 2% 소금물에서 발아시켜 생육이 좋은 식물체를 선발하여 50 cm 투명 아크릴 원통에 이식하고 2% 소금물을 지속적으로 관개하였다. 내염성 강화 F2 식물체는 염에 감수성을 보인 식물체에 비하여 상대적으로 긴 이삭과 종자 형성을 보였으며, 감수성 식물체는 종자 형성이 이루어지지 않았다. 또한, 내염성 강화 F2 식물체는 감수성 식물체에 비하여 좋은 뿌리의 신장과 천근성을 보였다. 이러한 갯그령의 식물학적 특성이 이입된 계통은 기후변화 대응, 환경 적응성 강화, 및 근권 강화에 좋은 작물 소재로 이용할 수 있다고 생각한다.

  • The effects of plant growth regulators on callus formation and organogenesis in shoot tip explant of Belancanda chinensis were examined. Shoot tip explants cultured in full salt strength of MT(Murashige and Tucker) medium containing 2,4-D 1.0 or 2.0mg/l were vigorously formed callus. Full salt strength of MT medium and 1/2 MT medium supplemented with zeatin 1.0mg/l were more effective than that with combination treatments of 2,4-D on the formation of shoots from calli. When shoots regenerated from shoot tips were transplanted into 1/2 MT medium added with 1.0mg/l, 41% of shoots formed roots.

Effect of Space Limitation of Rhizosphere on Morphology and Development of Root System in Tobacco Seedlings (담배 육묘시 근권의 공간 제한이 근계의 형태와 발달에 미치는 영향)

  • This study was carried out to acquire the basic information of root growth under different pot size, imposing different space limitation on rhizosphere. Different size of pots that had same surface area but different depth, 5cm(Iength)$\times$5cm(width)$\times$30, 15, 5cm(depth), were used during the seedling stage of tobacco plant. Space limitation on rhizosphere affected not only the aerial growth, stem height, leaf area and shoot dry weight, but also root growth and root architecture. Aerial growth was highly related to growth of underground part, so space limitation on rhizosphere decreased aerial growth. Limitation on pot volume by reducing pot depth induced new rooting on crown. Root number and relative multiplication rate were higher in small pot that had 5cm depth than large pot, but total root length and mean extension rate showed reverse patterns. Root numbers of 1st order and 2nd order were increased as pot depth was increased, but the root number of 3rd order was increased in small pot. Root system of seedling grown in large pot distributed more horizontally than that in small pot at 20 days after temporary planting (DAT), but the root architecture of seedling was reversed at 25 DAT.

  • This study was conducted to get basic information on the occurrence of plant-parasitic nematodes for the establishment of nematode management strategy in major potato production areas in Korea. Nationwide soil collection was done in 11 areas of Cheju, Yesan, Gimchun, Goryoung, Hong chun, Pyungchang, Gimjae, Milyang, Namwon, Gangnung, and Inje in 2004-2005. Root-hot nematode juveniles(J2) were detected in 30 samples among the 50 samples. The average density was 12-69 J2/100cc soil. Pratylenchus sp., Helicotylenchus sp., Ditylenchus sp., Tylenchus sp., and Tylenchorhynchus sp. were also detected in various locations, however, their densities were very low. Root-knot nematode females were collected from tomato roots inoculated with the potato field soils for PCR-RFLP identification. The females from Cheju, Milyang, and Goryung showed PCR products of 500 bp. And the Dra I restriction enzyme digestions showing 290 bp and 230 bp fragments confirmed their identity as Meloidogyne hapla.

  • To investigate the severity of crown gall disease on grapevine, the ratio of healthy vs. galled grapevines and the presence of the pathogen of the disease in soil were measured in Korean vineyards. In field and greenhouse cultivations, the crown gall incidence of the cv. Kyoho grapevines was $0.4{\sim}97.9%$ and $1.4{\sim}3.8%$ and those of cv. Campbell Early was $1.2{\sim}2.1%$ and $0{\sim}1.8%$, respectively. The higher populations of Agrobacterium spp. were isolated from soils of grapeveins with crown gall than from soils of noninfected vineyards. Based on the colony shapes and growth on plates, 480 isolates of Agrobacterium spp. from 21 soil samples were collected. Only 13 isolates out of 480 developed the gall on inoculated grapevines.

Development of an Efficient Screening Method for Resistance of Chili Pepper Plants to Meloidogyne incognita (뿌리혹선충 Meloidogyne incognita에 대한 저항성 고추를 선발하기 위한 효율적인 검정법 확립)

  • Root-knot nematodes (Meloidogyne spp.) are major plant pathogens that cause reductions in yield and quality of several solanaceous crops, including pepper (Capsicum spp.). These losses can be averted through planting of resistant cultivars. Plants are defined as resistant when they suppress nematode reproduction. In this study, the resistance degrees of 102 commercial cultivars of chili pepper (Capsicum annuum) to a root-knot nematode, Meloidogyne incognita, were evaluated by comparing the number of egg masses on their roots to those of 'PR huimangchan', a highly susceptible cultivar that exhibited the most egg masses of the chili pepper cultivars evaluated. Among these cultivars, forty-four (43.1%) showed resistance to M. incognita and eighteen (17.6%) were moderately resistant. The other cultivars (39.3%) were determined to be susceptible. For further study, six chili pepper cultivars (i.e., Gangryeokjosenggeon, Shinsegae, Muhanjilju, PR Bulrocho, PR Huimangchan, and Jjang) with different levels of resistance to the nematode were selected. Changes in resistance of the six cultivars under several conditions, such as inoculum concentration, plant growth stage, and cultivation period after transplanting were investigated. We found that an efficient screening method for resistance of chili pepper to M. incognita is to transplant the chili pepper seedlings 7 days before inoculation, to inoculate 28-day-old plants with M. incognita by loading 5,000 eggs per plant into the pot of soil, to cultivate the plants in a greenhouse ($25{\pm}5^{\circ}C$) for 45-60 days, to measure the number of egg masses on roots of the seedlings, and then to determine the resistance response of the plants by comparing the number of egg masses on the roots with a reference-susceptible cultivar 'PR huimangchan'.

Comparison of Underground Root Growth Characteristics of Major Cool-Season Grasses according to Establishment Stages in Sports Turf Designed by the USGA Soil System (USGA 지반으로 설계된 스포츠 잔디밭에서 조성단계별 주요 한지형 잔디의 지하부 뿌리생육 특성 비교)

  • Research was initiated to investigate root growth characteristics of major cool-season grasses (CSG) and to collect basic information useful for sports turf design, construction and maintenance. Several turfgrasses were evaluated in the USGA (United States Golf Association) soil system. Turfgrass entries were comprised 3 blends and 3 mixtures of Kentucky bluegrass (KB, Poa pratensis L.), perennial ryegrass (PR, Lolium perenne L.), and tall fescue (TF, Festuca arundinacea Schreb.). Significant differences were found in root growth, rooting potential and rooting development. These characteristics increased with time after seeding, but varied with establishment stages. In early stage, root length was highest with PR, intermediate with TF and lowest with KB. Evaluation in a middle stage indicated that root growth was similar to early-stage evaluation, but decreased by 13 to 31% compared with early-stage values. Root growth of late stage increased by 34 to 85% over middle-stage root growth. Overall, thhere was not much difference in root length among treatments, with all except Mixture I reaching 22cm in root length. Rooting potential ranking was variable with establishment stage, being PR > KB > TF in early stage, PR > TF > KB in middle stage and TF > PR > KB in late stage. At the end of the study, TF was rated best for rooting development, followed by PR and finally KB. Our results showed that TF was the best species in regard to overall rooting characteristics. TF exhibited excellent rooting development with time after establishment. Bunch-type PR showed fast root growth in the early stage, but rooting quality characteristics decreased with time, especially for rooting development. By contrast, rhizomatous-type KB was poor in early-stage root growth, but rooting characteristics improved with time after establishment. These variations in rooting characteristics among CSGs were considered to arise from differences in establishment vigor, growth habit and genetic characteristics. Information on root growth, rooting potential and rooting development by establishment stages will be useful for sports turf design, construction and maintenance.

Studies on the Wet-injury Resistance of Wheat and Barley Varieties III. Effect of Various Moisture Levels on the Top and Root Growth of Barley Crop (맥류 내습성에 관한 연구)

  • This experiment investigated the effect of various moisture levels on the top and root growth of barley and its yield. Resistant varieties to excess moisture injury shortened plant height, but tillered more, having creeping plant type compared with ordinary one, and greater portion of root of this varieties occured at the deeper soil depths with longer root length. However, susceptible one to excess moisture injury showed an inversed trend when compared with the untreated one. Oversaturated treatment compared with the untreated increased a little bit soil temperature and Eh of soil. Increased root activity and increased yield were obtained with resistant varieties compared with the untreated. Lower root activity and less yield than those of the untreated were observed in both extreme treatments. Yield reduction in both the extreme treatments was mainly due to reduction of tiller number, grain number and 1, 000-grain weight. Highest yield reduction was marked at the stage of internode elongation in the oversaturated and at the booting stage in the overdried treatment.
