Plant Parasitic Nematodes in Soybean in Korea and Their Importance Rating (국내 콩 기생성선충의 중요도 등급)

  • Plant parasitic nematodes were isolated from 274 soil samples collected from soybean fields in Korea. Nematode importance rating in soybean is proposed based on this study and by reviewing other reports. Soybean cyst nematode, Heterodera glycines is the most important nematode species and rated as $1^{st}$ because it detected from 38%(range 25-51%) of soil samples with high density except Jeju province. Root-knot nematode, Meloidogyne spp. is rated $2^{nd}$ and is more widely distributed in southern provinces, Gyeongnam, Jeonnam, and Jeju province (detection rate ranged 16-44%). Pratylenchus is rated $3^{rd}$ and is more frequently detected from northern provinces such as Gyeonggi and Gangwon (detection rate ranged 10-13%). Helicotylenchus is rated $4^{th}$ and is particularly important in Jeju province (detection rate is 62% and numbers averaged 571 nematodes/$300cm^3$ soil), which is the main production area for sprouting soybean in Korea. Tylenchorhynchus, Paratylenchus, Criconema, Criconemoide, Mesocriconema, Ogma, Xiphinema, Paratrichodorus, and Trichodorus occur in low frequency and density, thus they are rated to $5^{th}$. We propose to delete following nematode genus from the list of soybean parasitic nematode in Korea because their parasitism on soybean is unconfirmed or negligible; Tylenchus, Aphelenchus, Aphelenchoides, Ditylenchus, Hirschmanniella immamuri, Basiria graminophila, Psilenchus, and Pseudhalenchus. Therefore, we revised 30 species in 16 genera as soybean parasitic nematodes in Korea. Importance rating is $1^{st}$ Heterodera glycines, $2^{nd}$ Meloidogyne spp., $3^{rd}$ Pratylenchus spp. and $4^{th}$ Helicotylenchus spp. especially in Jeju province.

Soil Nematode Fauna in Dokdo Island of Korea (독도의 토양 선충상)

  • Nematode fauna in Dokdo island was investigated for two years between 2008 and 2009. Population density and biomass were $239,500m^2$ (26,000-836,000) and $103.9mg/m^2$ (3.0-388.6), respectively. Total of 31 species of nematodes were identified : Acrobeloides is the most important genus with prominence value (PV) of 117.8, followed by Rhabditidae (PV = 81.9), Prismatolaimus (PV = 39.0), and Aphelenchoides (PV = 31.4). Among feeding group, bacteriovorus nematode composed 77% of density and 62% of biomass followed by plant-parasitic and fungivorus nematodes. A species of Pratylenchus having distinctive tail terminus is under study as a new species. Biological indices, Shannon and Wiener index (2.46), Simpson's diversity index (0.86), Species evenness (0.73), Species richness (1.30) indicate that nematode fauna in Dokdo island is diverse and unique. This study may give a general idea of nematode fauna at the Dokdo island.

Rapid Methods to Distinguish Heterodera schachtii from Heterodera glycines Using PCR Technique (PCR 기법을 이용한 사탕무씨스트선충과 콩씨스트선충의 간이동정)

  • The purpose of this study was to develop rapid methods for distinguishing between Heterodera schachtii and H. glycines detected from chinese cabbage fields of highland in Gangwon, Korea. To do this, we performed PCR-RFLP and PCR with the primers set developed in this study for GC147, GC408 and PM001 population, H. schachtii, and YS224, DA142 and BC115 population, H. glycines. Eight restriction enzymes generated RFLP profiles of mtDNA COI region for populations of H. schachtii and H. glycines, repectively. As a result, treatment of two restriction enzymes, RsaI and HinfI, were allowed to distinguish H. schachtii from H. glycines based on the differences of DNA band patterns. The primer set, #JBS1, #JBG1 and #JB3R, amplified specific fragments with 277 and 339 bp of H. schachtii, 339 bp of H. glycines, respectively, while it did not amplify fragments from three root-knot nematodes and two root-lesion nematodes. Thus, the primer set developed in this study could be a good method, which is used to distinguish between H. schachtii and H. glycines.

  • Hemicycliophora minora Wu, 1966 and Pratylenchoides ivanovae Ryss, 1980 are newly collected from Korea. Five species of Hemicycliophora and 3 species of Pratylenchoides were reported in Korea and key is given. Two preciously described species, Pratylenchoides utahenis Baldwin, Luc & Bell and Rotylenchus feroxcis Eroshenko were observed under the scanning electron microscope.

Effects of Intercrops on Growth and Yield of Paeonia Lactiflora PALLAS. (간작물(間作物)이 작약(芍藥)의 생육(生育) 및 수양(收量)에 미치는 영향(影響))

  • This study was carried out to select proper intercrops in peony cultivation because peo­ny is damaged easily by disease and cannot make proper income with it's single crop system, so it needs rational intercropping system to raise it's productivity and income. The results of this study are as follows. The kinds of soil nematodes were Meloidogyne sp, Pratyienchus sp, Aphelenchoides sp, Ditylenchus sp, Xiphinema sp, and damaging dominant nematode was Meloidogyne sp. The density of Meloidogyne sp wa suppressed remarkably in the combination of peony+sesame, and peony + job's tears. This study shows that sesame and job's tear were most resistant crops to the IvIeloi­dogyne sp. The quanity of herbaceous peony was the best in the combination of peony+red pepper. The results of income analysis per lOa for three years shows next conclusion. Peony+red pepper combination and peony+ sesame combination were most effective when it's compared with single cropping of peony (1,490 thousand won). But red pepper was damaged easily by soil nematode, therefore, sesame was the most stable and high yield crop as a intercrop in the cultivation of herbaceous peony.

Development and validation of an analytical method for nematicide imicyafos determination in agricultural products by HPLC-UVD (HPLC-UVD를 이용한 살선충제 imicyafos의 시험법 개발 및 검증)

  • Imicyafos which is a nematicide for controlling root-knot nematodes has been registered in the Republic of Korea in 2012, and the maximum residue limits of imicyafos are set to watermelon and korean melon as each 0.05 mg/kg. Extremely reliable and sensitive analytical method is required for ensuring food safety on imicyafos residues in agricultural commodities. Imicyafos residues in samples were extracted with acetone, partitioned with hexane and dichloromethane, and then purified with florisil. The purified samples were analyzed by HPLC-UVD and confirmed with LC-MS. Linear range was between 0.1~5 mg/kg with the correlation coefficient ($r^2$) 0.99997. Average recoveries of imicyafos ranged from 77.0 to 115.4% at the spiked levels of 0.02 and 0.05 mg/kg with the relative standard deviations of 2.2~9.6%. Limit of detection and quantification were 0.005 and 0.02 mg/kg, respectively. An inter-laboratory study was conducted to validate the determination method in depth, and the results were satisfactory. All of the validation results revealed that the developed analytical method in this study is relevant for imicyafos determination in agricultural commodities and will be used as an official analytical method.

Weed Control Technology with Low Concentration Ethanol in Protected Cereal Crop Cultivation (비닐하우스 내 조 재배시 저농도 에탄올을 이용한 잡초억제 효과)

  • Weed control is of fundamental importance when planting cereal crops, particularly during the establishment phase. Weeds compete for nutrients, water and light, and can severely threaten the survival and early growth of newly planted crops. Field experiment was conducted to evaluate the effect of ethanol treatment on the weed control efficacy in cereal crops in protected vinylhouse. Five treatments like water, 25 L, 50 L, 75 L, and 100 L per $m^{-2}$ at 2% ethanol were applied and covered with vinyl on 10 days before foxtail millet seeding. After that, foxtail millet seeds were sown on June 20 at $60{\times}10cm$ planting distance and weed control efficacy was recorded at 30, and 50 days after seeding based on the dry weight. Soil oxidation-reduction potential was recorded -200~-400 mV after treatment 2 days. Weed control efficacy of at 75 L, and 100 L per $m^{-2}$ was observed 92%, 84% in 2% ETOH, respectively. Yields of foxtail millet at 75 L, and 100 L per $m^{-2}$ were 1,570 kg $ha^{-1}$, 1,230 kg $ha^{-1}$, respectively.
