Study on Deodorizing Effects of Mume Fructus, Eriobotryae Folium, Acanthopanacis Cortex and Angelicae Dahuricae Radix for the Development of a Gargle Solution (구강함수제 개발을 위한 오매, 비파엽, 오가피, 백지의 구취억제효과 연구)

  • Objectives: The aim of this study was to investigate deodorizing effectsof medicinal herbs (Mume Fructus, Eriobotryae Folium, Acanthopanacis Cortex, Angelicae Dahuricae Radix) for development of a gargle solution. Methods: 1. The antimicrobial effects of medicinal herbs were evaluated with the minimal bactericidal concentration (MBC) and the change of the number of viable cells in the herb extracts(1%) for 48 hrs against P. gingivalis 2561 and Pr. intermedia ATCC 25611. 2. Deodorizing activity of each herb and Garglin $Mint^{(R)}$gainst methyl mercaptan were analyzed by gas chromatography (GC). 3. We used the malodor modeling of the salivary sediment system with a Halimeter. 4. In the preliminary clinical study, the baseline concentration of VSC in the oral cavity of each subject was measured by Halimeter. Subjects would gargle for 30 seconds with cysteine. After 4 minutes subjects would gargle for 30 seconds with Garglin and herb extracts (2%). Subsequently, concentration of VSC were measured at 0, 4, 8, 12 and 20 minutes. Results: 1. MBC of Mume Fructusfor P. gingivalis 2561 was determined to be <1% and MBCs of Eriobotryae Folium for P. gingivalis 2561 and Pr. intermedia ATCC 25611 were determined to be <2% and <1%, respectively. Mume Fructus (1%) completely suppressed the P. gingivalis cell viability from 5 hrs and Eriobotryae Folium (1%) completely suppressed the Pr. intermedia cell viability from 48 hrs. 2. In GC analysis, deodorizing activities were 91.54% with Mume Fructus, 87.97% with Eriobotryae Folium, 100% with Acanthopanacis Cortex, 72.36% with Angelicae Dahuricae Radix and 40.54% with Garglin $Mint^{(R)}$. 3. In malodor modeling of the salivary sediment system, each of the medicinal herbs had significantly inhibitory effect on malodor formation (p<0.05). 4. In the preliminary clinical study, the concentration of VSC of the herb groups was significantly lower than of the control group, but not in Garglin $Mint^{(R)}$. Conclusions: Mume Fructus, Eriobotryae Folium, Acanthopanacis Cortex and Angelicae Dahuricae Radixhave deodorizing activities and potential as an effective mouthwash against oral malodor.

Antimicrobial Effect on the Periodontal Pathogens and Anti-inflammatory Effect of Eriobotryae Folium (비파엽(枇杷葉)의 치주염세균에 대한 항균효과 및 항염효과)

  • Objective: The purpose of this study was to evaluate on the antimicrobial effect on the periodontal pathogens and anti-inflammatory effect of Eriobotryae folium. Eriobotryae folium are constituent herbs of Gagamgamroum, which has been used for a long time in oriental medicine as a herbal medicine for treating halitosis and toothache. Method: Eriobotryae folium was prepared by extracting medicinal herb with water. We investigated antimicrobial activity by the minimum inhibitory concentration (MIC) test. We also investigated inhibition of $IL-1{\beta}-induced$ collagenase (mmp-1), stromelysin-1 (mmp-3), interleukin-6 gene expression in human gingival fibroblasts using RTPCR analysis. Result: The antimicrobial effects of Eriobotryae folium was evaluated with MIC against periodontopathogens; Porphyromonas gingivalis 2561, W50, A7A1-28, 9-14K-1, Prevotella intermedia 28, and Actinobacillus actinomycetemcomitans Y4. MICs of Eriobotryae folium were 1.25 mg/ml, 2.5 mg/ml, 0.625 mg/ml, 1.25 mg/ml, 10 mg/ml and 10 mg/ml. The anti-inflammatory effect of Eriobotryae folium was evaluated with influence of herbs on the $IL-1{\beta}-induced$ expression of mmp-1, mmp-3, and interleukin-6. $IL-1{\beta}$ increased mmp-1, mmp-3, and interleukin-6 mRNA levels. Eriobotryae folium significantly inhibited $IL-1{\beta}-induced$ mmp-1, mmp-3, and interleukin-6 gene expressions in a dose-dependent manner. Conclusion: These results suggested that Eriobotryae folium might reduce the excessive proteolytic capacity of the gingival fibroblast during inflammation and could be developed as a new drug for periodontitis.

Antibacterial activity from medicinal plant extracts on the Staphylococcus aureus (수종 한약재 추출물의 Staphylococcus aureus에 대한 항균활성)

  • Medicinal plant extracts including Rubus coreanus, Sanguisorba officinalis, Eriobotrya japonica, Prunus mume, Crataegus pinnatifida, Rosa leavaigate Prunus persica, Prunus japonica var. nakaii and Spiraea blumei were prepared for the test of antibacterial activity. Tryptic soy broth (TSB) containing $0{\sim}10mg/ml$ of medicinal plant extracts was inoculated with $10^6$ cells/ml of Staphylococcus aureus and incubated at $35^{\circ}C$ for 24 hours. The plate counting method and clear zone test were used to test inhibitory effect of the extracts. Minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) was derived from the survival curves of S. aureus. The order of antibacterial activities of medicinal plant extracts on the S. aureus was Rubus coreanus > Sanguisorba officinalis > Eriobotrya japonica > Prunus mume > Crataegus pinnatfida. Minimum inhibitory concentration of Sanguisorba ofEcmalis on the Staphylococcus aureus was 2.5mg/ml and minimum bactericidal concentration of Rubus coreanus was 1.0%. Inhibition zone of Rubus coreanus, Sanguisorba officinalis, Eriobotrya japonica, Prunus mume, and Crataegus pinnatifida was 16.5mm, 14.3mm 11.0mm, 14.0mm and 12.7mm, respectively. The morphology of S. aureus cells treated with medicinal plant extracts showed damage of cell wall and cytoplasmic membrane. Severely damaged cells of S. aureus lost electron dense material and cytoplasm. This result suggests that medicinal plant extracts can be used as an effective natural antibacterial agent in food.

Effect of Eriobotrya folium on Local Fat via Regulation of Lipase Secretion (비파엽추출물의 지방분해효소 조절을 통한 국소 지방분해 효능)

  • Objectives: In this study, the lipolytic effects of Eriobotrya folium extract (EFE) on local fat was investigated in high fat diet (HFD)-induced obesity mouse and 3T3-L1 adipocytes. Methods: C57BL/6J mice (5 weeks) were fed HFD for 6 weeks to induce obesity. EFE (20 mg/ml, $100{\mu}l$) or saline ($100{\mu}l$) as a normal control was injected into left inguinal fat pad region, 3 times per a week for last 2 weeks. After sacrifice, body weight, and histological changes of the inguinal fat pad were evaluated. The expressions of hormone-sensitive lipase (HSL) and adipose triglyceride lipase (ATGL) in inguinal fat pad were analyzed by Western blotting. Also, lipid accumulation and lipases release were determined in 3T3-L1 adipocytes by oil red o staining. Results: EFE significantly reduced the weight of inguinal fat pad and the size of adipocytes in HFD-induced obesity mice compared to control. The treatment of EFE up-regulated the expressions of HSL and ATGL in inguinal fat pads of obesity mice, as well as 3T3-L1 adipocytes. In addition, EFE inhibited the lipid accumulation in 3T3-L1 adipocytes in a dose dependent manner. Conclusions: EFE showed lipolytic effect on local fat of HFD-induced obesity mice by up-regulation of the lipases secretion. This suggests that EFE could be considered as anti-obese substance with lipolytic property on local fat.

Effect of Combined Extract of Safflower Seed with Herbs on Blood Glucose Level and Biochemical Parameters in Streptozotocin-induced Diabetic Rats (홍화씨와 한약재 흔합추출물이 Streptozotocin으로 유도한 흰쥐의 혈당과 혈액 성분에 미치는 영향)

  • The aim of the present study was to investigate the effect of combined extract of safflower seed with herbs on the improvement of blood glucose, lipid peroxides, lipids in the plasma and liver of strpetozotocin-induced diabetic rats. Rats in the experimental group were orally administered with combined extract of safflower seed (100 mg, 200 mg/kg B.W.) with herbs (Ophiopogon japonicus Ker-Gaqler, Glycyrrhiza uralensis Fisch, Mori Folium, Poria cocos, Rehmannia glutinosa, Eriobtrya japonica, Aralia continentalis Kitagawa, Zizyphus jujuba var, Cornus officinalis, Paeonia suffruticosa, Trichosanthes kirilowii Maxim and Schizandra chinensis Baill) for 4 weeks. Body weight gain and food efficiency ratio were significantly lower in diabetic groups than those of control group. These were no protective effect of the supplementation of combined extract of safflower seed with herbs. Concentration of blood glucose was significantly higher in the diabetic groups than those in the control group. Blood glucose concentration was remarkably lower supplementation of combined extract of safflower seed (200 mg/kg B.W.) with herbs. There was no significant difference of plasma lipid peroxides among experimental groups, while liver lipid peroxides of diabetic group was significantly higher in control group. But supplementation of combined extract of safflower seed with herbs was induced markedly lower in liver lipid peroxides in diabetic rats. Diabetic groups had markedly higher levels in triglycerides, LDL-cholesterol and atherogenic index, while had lower HDL-cholesterol level. Triglyceride levels of plasma and liver were significantly lower with combined extract of safflower seed with herbs. But total cholesterol, phospholipid and free fatty acid were no differing significantly among experimental groups.

Experimental Studies on the Antitumor Effects of Jinryungtang Gagambang Extract (진령탕가감방의 항종양효과(抗腫瘍效果)에 관(關)한 실험적(實驗的) 연구(硏究))

  • The sprig of Jinryungtang Gagambang has been used for curing as a traditional medicine without any experimental evidence to support the rational basis for their clinical use. This experiment was carried out to evaluate the possible therapeutic or antitumoral effects of Jinryungtang Gagambang extract against cancer, and to study some mechanisms responsible for its effect. The cytotoxic and antitumor effects were evaluated on human cell liens (A549, hep3B, Caki-1, Sarcoma 180) after exposure to Jinryungtang Gagambang extract using in ILS, colony forming efficency and SRB assay which were regarded as a valuable method for cytotoxic and antitumor effects of unknown compound on tumor cell lines. The results obtained in this studies were as follows. 1. As a result of exposure to Jinryungtang Gagambang extract, the proliferation of A549, hep3B, Caki-1, good correlations were shown from the results of SRB assay and those of clogenetic assay. 2. The oral administration of Jinryungtang Gagambang extract showed significant effects of increase of MST(mean survival time) and ILS(increased life span) depending on the increasing concentration. 3. Against squamous cell carcinoma induced by MCA, Jinryungtang Gagambang decreased not only the frequency of tumor production but also the number and weight of tumors per tumor bearing mice(TBM). Jinryungtang Gagambang also significantly suppressed the development of 3LL cell-implanted tumors by frequency and their size, and some developed tumors were regressed by the continuous treatment of Jinryungtang Gagambang extract into TBM. 4. Jinryungtang Gagambang extract also increased NK cell activities. According to the above results, it could be suggested that Jinryungtang Gagambang extract has prominent antiutmor effect.
