Determination of Ultimate Biodegradability and Multiple Decay Rate Coefficients in Anaerobic Batch Degradation of Organic Wastes (유기성폐기물의 회분식 혐기성 최종생분해도와 다중분해속도 해석)

  • A new graphical method was developed to separate two distinctive decay rate coefficients($k_1$ and $k_2$) at their respective degradable substrate fractions($S_1 and $S_2$). The mesophilic batch reactor showed $k_1$ of $0.151\;day^{-1}$ for wasted activated sludge(WAS), $0.123\;day^{-1}$ for thickened sludge(T-S), $0.248{\sim}0.358\;day^{-1}$ at S/I ratio of $1{\sim}3$ for sorghum and $0.155{\sim}0.209\;day^{-1}$ at S/I ratio $0.2{\sim}1.0$ for swine waste, whereas their long term batch decay rate coefficients($k_2$) were $0.021\;day^{-1}$, $0.001\;day^{-1}$, $0.03\;day^{-1}$ and $0.04\;day^{-1}$ respectively. At least an order of magnitude difference between $k_1$ and $k_2$ was routinely observed in the batch tests. The portion of $S_1$, which degrades with each $k_1$ appeared 71% for WAS, 39% for T-S, 90% for sorghum, and $84{\sim}91%$ at S/I ratio of $0.2{\sim}1.0$ for swine waste. Ultimate biodegradabilities of 50% for WAS, 40% of T-S, $82{\sim}92%$ for sorghum, and $81{\sim}89%$ for swine waste were observed.

Proteinase activity in the isolates of Trichomonas vaginalis according to their pathogenicity (질트리코모나스의 병원성과 단백 분해 효소와의 상관성)

  • Ten axonic isolates of Trichomonns uaginolis were subcutaneously injected to the BALB/c mice in order to assess their pathogenicity by means of so-called "mouse assay" method. All the isolates revealed neutral and acid proteinase activities both in their Iysates and in culture media, but the specific activities of both proteinases in the severely pathogenic group were significantly higher than the mildly pathogenic group (p < 0.05). In the SDS-PAGE system in which the electrophoretic gels contained 0.4% gelatin as the substrate, five different handing patterns of trichomonal proteinases were detected, and the patterns were closely related with the pathogenicity of the isolates of T. vosinalis. All five bands might be regarded as cysteine proteinases group in the inhibitor assays. The cytotoxicity of the Iysates of T. vaginalis to the target Chinese hamster ovarian (CHO) cell line was also significantly different according to the pathogenicity of the isolates, and generally lower in the Iysates treated with cysteine proteinase inhibitors than in the control Iysates. In summarizing the results, it might be considered that the proteinases of T.vaginalis showing five electrophoretic banding patterns are closely related with the pathogenicity and cytotoxicity of the isolates of T. voginolis.

  • 최근 기후변화로 인한 기상이변이 빈번히 발생하면서 그로 인한 피해도 점점 증가하고 있다. 이를 최소화하기 위해서는 기후변화가 강수에 미치는 영향에 대한 연구가 필요하며, 특히 강수의 기후변화를 고려한 장기적인 변동에 대한 예측이 매우 중요하다. 그 중, 기후변화로 인한 강수현상의 변화를 분석하기 위한 방법 중 하나로 강수 현상이 주변 기후 요소의 분포에 영향을 받는다는 가정 하에 기상인자를 통하여 강수를 예측하는 방법이 있다. 우리나라에 영향을 미치는 주변 기상인자들과 강수 간의 상관관계를 분석하여 상관관계가 높게 나타나는 기상인자를 통해 우리나라 강수량을 예측하면 장기적인 관점에서 강수 예측의 정확도를 높일 수 있다. 하지만 상관관계 분석에 있어서 강수 원 자료 와 기상인자간의 상관관계를 비교할 경우 원 자료가 가지는 큰 변동성으로 인해 정확한 상관관계 분석이 이루어지지 않을 가능성이 크다. 따라서 강수자료를 분해하여 분해된 요소별로 상관관계를 분석하여 분석의 정확도를 높일 필요가 있다. 다양한 자료 분해 방법중 경험적 모드분해법(Empirical Mode Decomposition, EMD)을 사용할 경우 자료의 분해에 있어서 주기성, 경향성에 따라 분해가 가능하며, 비정상성을 가지고 있는 시계열에 대해 효과적으로 분해가 가능한 장점이 있다. 본 연구에서는 30년 이상의 자료기간을 가지는 지점의 강수량 자료를 바탕으로 경험적 모드분해법을 이용하여 강수자료를 분해하고, 이를 다양한 기상인자와의 상관관계를 분석함으로써, 우리나라 강수량 변동과 연관이 있는 기상인자들을 선별하였다. 선별된 기상인지를 바탕으로 다중회귀분석을 수행하여 기상인자를 독립변수로 하는 강수 예측식을 구축하여 우리나라 강수의 예측 가능성을 살펴보고자 한다.

모호집합을 적용한 네트워크 신뢰도

  • 모호집합은 인간의 추론에 내재하는 모호성을 정형화한 것이다. 네트워크 신뢰도를 계산할 때, 각 링크의 확률이 모호수로 정의될 때의 신뢰도를 구하는 알고리즘을 설명하였다. 알고리즘은 분해법을 적용하는 것인데, 모호수의 연산을 포함한다. 분해법을 할 때 키스톤 부품을 시점으로 부터 하나씩 선택하여 분해한다. 이러한 방법은 키스톤 부품을 선택하는 기준이 필요 없으므로 간단하게 만든다. 분해에 의해서 두 개의 하위 문제가 생성되고 원 문제와 재귀관계를 수립할 수 있다. 재귀 알고리즘은 컴퓨터 프로그램을 간단하게 만든다.

Supercritical Water Hydrolysis of Waste Logs after Oak Mushroom Production (초임계수를 이용한 표고버섯 골목의 가수분해)

  • In order to investigate the possibility of waste logs after oak mushroom production as a source of an alternative energy and to obtain the fundamental data of supercritical water hydrolysis that has been paid attention as a new saccharification method of lignocellulosics, supercritical water hydrolysis of normal log woods (Quercus acutissima Carruth) and waste logs was carried out. With the increase of reaction time and temperature, the color of the degradation products has been dark and the degradation rate and the crystalline index increased. However the increase of reaction pressure affected the color of the degradation products and the degradation rate at only low reaction temperature. In the early stage of the reaction, the degradation of hemicellulose was progressed, while in the late stage, the cellulose was degraded. The increase of reaction time and reaction temperature (less than $415^{\circ}C$) improved the sugar yield, while at high temperature(more than $415^{\circ}C$), the sugar yield was decreased. Based on the result of the sugar yield, the optimal hydrolysis condition of Q. acutissima Carruth by supercritical water was determined to be $415^{\circ}C$, 60 seconds and 230 pressure bar with the sugar yield of 2.68% (w/w). At the optimal condition, the supercritical water hydrolysis of waste logs after the mushroom production was carried out and the sugar yield was increased to 358% (w/w). The major degradation products of waste logs by supercritical water hydrolysis were 1,1'-oxybis-benzene and 1,2-benzendicarboxylic acid by the GC-MS analysis. At the reaction condition with low degradation rate, the fatty acids such as pentadecanoic acid, 14-methyl-heptadecanoic acid were identified. With the increase of the reaction temperature and time, the amounts of phenol and benzene were increased, but the reaction pressure did not affect the kinds of degradation products. Holocellulose content was 60.6~79.2% in the water insoluble residue and the monosaccharide yield of the water insoluble residue was 49.2~675% by the acid hydrolysis. The monosaccharide yield of water-soluble portion was increased largely by the second hydrolysis using dilute acid.

  • A disassembility assessment has mostly depend on the subjective decision making from the qualitative element. The work of disassembly is already classified with given disassemble points from the symbolic chart method. It is not useful in the practical assessment because it is not specified. The new method of design for disassembility assessment(DFDA) is practical to introduce the fuzzy number as the conversion of quantitative element from qualitative. It is appled to compare the usefulness of air-cleaner in Korea-Japan passenger-vehicle.

Isolation of Protease Producing Microorganisms (단백질 분해효소 생산 균주 분리)

  • Protease producing microorganisms were isolated from many kinds of food waste and fermented foods, which contains high amount and variable kinds of degraded substances. Several microorganisms were identified by 16S rRNA full sequencing analysis methods. The activity of protease was analyzed and identified in variable conditions for the application. For industrial use for biowaste treatment some proteases were isolated, identified and selected from microbial cells. And the tests were carried for the further use. The protein degrading activity at low temperature is useful for the treatment of organic waste, which contains much proteins. By the protein degradation process the organic waste can be utilized in variable fields, for example from feedstuff supplement to fertilizer for agriculture. Bacterial cells with protease activity at low temperature were isolated and identified. The optimal conditions for microbial cultivation and protease production were studied.

  • Since the enzymatic degradation of microbial poly(hydroxylalkanoate)s (PHAs), such as poly[(R)-3-hydroxybutyrate] and poly[(R)-3-hydroxybutyrate-co-3-hydroxyvalerate] initially occurs by a surface erosion process, their degradation behaviors can be controlled by the change of surface property. In order to control the rate of enzymatic degradation, plasma modification technique was applied to change the surface property of microbial PHAs. The surface hydrophobic and hydrophilic properties of PHA films were introduced by $CF_3H$ and $O_2$ plasma exposures, respectively. The enzymatic degradation was carried out at $37^{\circ}C$ in 0.1 M potassium phosphate buffer (pH 7.4) in the presence of an extracellular PHB depolymerase purified from Alcaligenes facalis T1. The results showed that the significant retardation of initial enzymatic erosion of $CF_3H$ plasma-treated PHAs was observed due to the hydrophobicity and the enzyme inactivity of the fluorinated surface layers while the erosion rate of $O_2$ plasma-treated PHAs was not accelerated.

  • Enzymatic hydrolysis with tuna pyloric caeca crude enzyme(TPCCE) was performed to recover a protein hydrolysate from hoki frame, fish processing by-product. Optimum hydrolytic conditions were pH 10.0, temperature $50^{\circ}C$, and incubation time 12 hrs, and then the degree of hydrolysis was about 60%. The yield of the hydrolysate from hoki frame by enzymatic hydrolysis was approximately 77% on a dry weight basis. The prepared protein hydrolysates were also fractionated through a series of 30, 10, 5 and 1 kDa molecular weight cut-off (MWCO) membranes in order to investigate the effect of their functionalities according to the difference of their molecular size. As the result of studying functionalities of the hydrolysates, 1 K hydrolysate showed the highest solubility over all pHs, and 30 and 10 K hydrolysate showed more excellent emulsifying property and whippability than the other hydrolysates.

  • 목적 - 기질금속단백분해효소(matrix metalloproteinase)는 조직의 염증 및 치유과정에서 숙주세포에서 생성, 분비되어 세포외기질(extracellular matrix)의 분해에 작용한다. 다양한 염증반응에 기질금속단백분해효소가 중요한 역할을 하는것으로 보고되고 있으나 치수 및 치근단 질환에서의 그 역할은 거의 알려져 있지 않은 상태이다. 본 연구에서는 염증이 있는 사람의 치수 및 치근단 조직을 채취하여 Enzymeimmunoassay 및 면역조직화학적 검색을 통해 제1형, 2형, 3형 기질금속단백분해효소의 수준 및 그 분포를 측정하여 치수 및 치근단 병소에서 이 효소의 작용을 알아보는 것을 목적으로 한다. 방법 - 연구재료는 근관치료를 위해 서울대학교 병원 치과 진료부 보존과에 내원한 환자를 대상으로 34개의 치아에서 통상의 근관치료 중 발수한 치수조직과 치근단 수술중 얻은 치근단 병소(n=10)를 이용하였다. 치수는 발수 전에 임상진단을 통해 급성 치수염(n=12), 만성 치수염(n=12), 정상 치수(n=10)로 구분하고 정상치수로 진단된 것을 대조군으로 설정하였다. 채취된 표본은 둘로 나누어 절반은 30분 이내에 5$\mu\textrm{m}$ 두께로 동결절단을 시행하여 조직표본을 제작하였고 deep freezer에 보관하였다가 헤마톡실린-에오신 염색 및 면역조직화학적 검색을 시행하였다. 나머지 조직은 ELISA를 위해 액체 질소에 보관하였다. ELISA를 시행하기전 표본의 단백질 정량을 시행하여 모든 표본의 단백질 양을 50mg/$\mu\textrm{l}$로 일치시키고 Amersham사의 ELISA kit를 사용하여 제1형, 2형, 3형의 기질금속단백분해효소의 양을 측정하였으며 그 결과를 Mann-Whitney U test를 사용하여 각 군간의 통계학적 유의성을 검증하였다. 결과 1. ELISA의 결과 제1형 기질금속단백분해효소의 농도는 모든 실험군에서 대조군보다 유의성있게 높게 나타났다.(p<.05). 또한 급성치수염군의 제1형 기질금속단백분해효소의 농도가 다른 실험군보다 유의성있게 높았다(p<.05). 2. 제2형 기질금속단백분해효소의 경우 급성치수염군과 대조군에서만 유의성있는 차이를 보였다(p<.05). 3. 제3형 기질금속단백분해효소의 경우 급성치수염군에서 대조군이나 만성치수염군보다 유의성 있는 높은 수치를 보였다(p<.05). 4. 면역조직화학검색 결과 염증성 치수에 존재하는 급성 및 만성염증세포 주위로 기질금속단백분해효소에 대한 면역 반응이 존재하였으며 주로 제1형과 제3형 기질금속단백분해효소의 경우 대식세포 및 림파구 주위로 강한 발색제의 침윤양상이 관찰되었다. 5. 치근단병소의 면역조직화학적 검색 결과 만성염증 세포 주변으로 미약한 발색제의 침윤양상이 관찰되었다.
