Lentinuls edodes has been used for traditional food and medicine around Asia, and a variety of biological effects have been reported. In this study, L. edodes water extract (LWE) was investigated for its anti-photodamage effect in HaCaT keratinocytes. To perform the necessary assays, L. edodes was extracted with distilled water for 8 hr at 40℃ in an extract tank. Anti-photodamage activity was assessed using a scratch wound healing assay, cell proliferation, and a reactive oxygen species (ROS) scavenging test and by measuring the mRNA and protein expression levels of matrix metalloproteinases (MMPs) and type I procollagen. MMPs and collagen expression are major markers of UV-induced photodamage in skin. Prior to photodamage analysis, the total polyphenol and β-glucan contents of the LWE were evaluated and found to be 4.64 mg GAE/g DW and 165.96 mg/g, respectively. Treatment with LWE induced cell migration and cell proliferation in UV-irradiated HaCaT cells, and LWE effectively scavenged the ROS induced by H2O2 and UVB irradiation in HaCaT cells. UVB irradiation induced ROS generation and led to increased production of MMP-1 and MMP-9 and to decreased collagen production in human keratinocytes. Treatment with LWE upregulated the expression levels of MMP-1, MMP-9, and type I procollagen in UVB-irradiated HaCaT cells. This study suggests that LWE could be used to develop cosmetic materials with anti-photodamage effects.
From their general natures of peraluminous, S-type and ilmenite-series granites, two-mica granites in the Cheongsan, Inje-Hongcheon, Yeongju and Namwon areas were originated from crust-derived granitic magma and solidified under reducing condition. Each two-mica granite in Inje-Hongcheon and Namwon districts was differentiated from the the residual magma of porphyric biotite granite and high Ti/Mg biotite granite, respectively. The genetic relationships between two-mica granite and porphyritic biotite granite in Chenongsan district and between two-mica granite and biotite granodiorite in Yeongju district are ambiguous. In Namwon district granitic magmas were water-saturated and possible water solubilities in magmas were more than 5.8wt.%. In Yeongju district two-mica granitic magma was nearly water-saturated and showed possible water solubilities between 2.4~5.8wt.%. Two-mica granitic magmas in Cheongsan and Inje-Hongcheon districts were water-undersaturated. Pressure-dependent minimum melt compositions (0.5~2kb) and petrographic textures of two-mica granites in Inje-Hongcheon and Yeongju districts represent that the granites intruded and solidified at shallow level, whereas those in Cheongsan and Namwon districts exhibit relatively deeper level of granitic intrusion (2-3kb). The intersection of granite-solidus/muscovite stability indicates that magmatic primary muscovite can be crystallized from the water-saturated magma above 1.6kb (ca. 6km), but below the pressure muscovite can be formed by the subsolidus reaction. On the other hand, more pressure would be necessary for the crystallization of primary muscovite from the water-undersaturated magma. This pressure condition can explain the occurrence of primary and secondary muscovites from the two-mica granites in the areas considered. The experimental muscovite stability must be cautious of the application to examine the origin of muscovite. The muscovite stability can move toward high temperature field with adding of Ti, Fe and Mg components to the octahedral site of pure muscovite end member.
Food constituents analysis of Acer tegmentosum. Maxim.(Acer TM) stem was carried out according to AOAC method, and the antiradical activity on DPPH and cytotoxicity on human cell lines (AGS, HepG2, A549, MCF-7 and Chang) for the 80% ethylalcohol(EtOH) extracts of Acer TM stem were studied. The antiradical activity on DPPH radical of the ethylacetate(EtOAc) fraction of the bark showed a higher activity than that of $\alpha$-tocopherol, ascorbic acid and BHT. The inhibition activity of the 80% EtOH extracts from Acer TM stem on human cancer cell lines by SRB assay indicated a dose-dependent growth inhibition on most human carcinoma cells. The growth inhibition rate of each human cancer cell line showed 91.3% to AGS, 75.0% to A549, 74.1% to HepG2, and 70.2% to MCF-7 cells, respectively, when the 80% EtOH extract(1 mg/ml) of Acer TM stem was added.
Polycystic ovary syndrome (PCOS) is hormonal imbalance condition as the endocrine and metabolic disorder that induces the infertility and various complications in reproductive age women. Estradiol valerate (EV) is used hormone replacement therapy in menopausal women and is reported that excessive administration of EV induces the PCOS. Nerve growth factor (NGF) is the factor to regulate the survival and maturation of developing neuronal cell and is also synthesized in ovary. And NGF is overexpressed in EV-induced polycystic ovary (PCO) as previously reported. Therefore, this study examined the possibility of NGF as can be used the biological marker in diagnosis of PCOS, the hormonal imbalance condition, using PCO and CHO (chinese hamster ovarian) cell lines. The concentration of EV treatment is optimized a 1 mg as not influence on the proliferation of CHO cell but 2 mg and 3 mg of EV treatment have the inhibition effect at initial stage. The morphological change was not observed in CHO cell after dose dependent manner treatment of EV. Expression of NGF mRNA and protein is significantly increased at 30 min after EV treatment in CHO cells compared to that of control. And NGF protein expression is strongly increased in PCO tissue, which observed many follicular cysts compared to normal ovary tissue. Taken together, overexpression of NGF may be act as a molecule to induce an abnormal development of follicle, suggesting that NGF can be used as a biological marker in diagnosis of PCOS.
Background: Gram-negative bacterial endotoxin induced septicemia is known to be a leading cause in the development of adult respiratory distress syndrome(ARDS). The mechanism of endotoxin induced lung injury is mainly due to the activated neutrophils which injure the capillary endothelial cells by releasing oxidant radical and resulted in pulmonary edema. We studied the change of antioxidant enzyme in the case of large or small, intermittant dose of endotoxin infused rat lungs. Methods: Endotoxin was given to the rat through the peritoneal cavity in the dose of 7 mg/kg body weight in the large dose group and 1 mg/kg for 10 days in the small dose group. Bronchoalveolar lavage (BAL) was done and rats were killed at 6, 12, 24 hours after single endotoxin injection in the large dose group and 3, 7, 10 days after daily endotoxin injection for 10 days in the small dose group. The lungs were perfused with normal saline through the pulmonary artery to remove the blood and were homogenized in 5 volume of 50 mM potassium phosphate buffer containing 0.1 mM EDTA. After centrifuging at 100,000 g for 60 minute, the supernatent was removed and stored at $-70^{\circ}C$ until measuring for superoxide dismutase (SOD), catalase, glutathione peroxidase (GSH-Px) and protein. Results: We observed the following results. 1) The lung wet/dry weight ratio and albumin concentration in the BAL fluids were increased to peak at 12 hours and neutrophil number in the BAL fluids were peak at 6 hours after endotoxin injection in the large dose group. 2) Cu, Zn SOD (IU/mg protein) was significantly decreased after 6, 12 hours after endotoxin injection in the large dose group. 3) There were no singnificant change in the level of Mn SOD, catalase, GSH-Px after endotoxin injection in both groups. Conclusion: Endotoxin in the large dose group produced the acute pulmonary edema and decreased the Cu, Zn SOD in the lung tissue after injecting endotoxin at 6 and 12 hours. These phenomenon may be due to the cell membrane damage by endotoxin. Further research would be necessary whther giving SOD by intratracheal route or method to increase the synthesis of SOD may lessen the acute lung injury by endotoxin.
In this study, to biosynthesize PHA with properties more similar to polypropylene, a Bacillus sp. EMK-5020 strain that biosynthesized poly (3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV) was isolated from soil. Bacillus sp. EMK-5020 strain biosynthesized PHBV containing 1.3% 3-hydroxyvalerate (3HV) using reducing sugar contained in Makgeolli lees enzymatic hydrolysate (MLEH) as a single carbon source. As the amount of propionic acid, which was added as a second carbon source, increased, the content of 3HV also increased. PHBV containing up to 48.6% of 3HV was synthesized when 1.0 g/l of propionic acid was added. Based on these results, the strain was cultured for 72 hr in a 3 l fermenter using reducing sugar in MLEH (20 g/l) and propionic acid (1 g/l) as the main and secondary carbon sources, respectively. As a result, 6.4 g/l DCW and 50 wt% of PHBV (MLEH-PHBV) containing 8.9% 3HV were biosynthesized. Through gel permeation chromatography and thermogravimetric analysis, it was confirmed that the average molecular weight and the decomposition temperature of MLEH-PHBV were 152 kDa and 273℃, respectively. In conclusion, the Bacillus sp. EMK-5020 strain could biosynthesize PHBV containing various 3HV fractions when MLEH and propionic acid were used as carbon sources, and PHBV-MLEH containing 8.9% 3HV was confirmed to have higher thermal stability than standard PHBV (8% 3HV).

Osteoarthritis (OA), the most common form of arthritis, is a result of both mechanical and biological events that destabilize the normal coupling of degradation and synthesis of articular cartilage chondrocytes and extracelluar matrix, and subchondral bone. Although it is likely that the molecular basis of pathogenesis is similar to that of other joints, additional study of the temporomandibular joint (TMJ) is required due to its unique structure and function. This study was carried out to evaluate the epidemiologic characteristics of TMJ osteoarthritis. The purpose of this study was to investigate the prevalence of TMJ OA in Patients with temporomandibular disorders (1405 men and 2922 women whose mean age was $30.2\; {\pm}\; 15.4$ and $33.1\;{\pm}\;15.2$ years, respectively) who had visited the TMJ and Orofacial Pain Clinic of Seoul National University Dental Hospital in 2007. Orthopantomograms, TMJ tomograms and transcranial radiographs were used to evaluate degenerative change of the mandibular condyle. The obtained results were as follows: 1. Degenerative change of the mandibular condyle was observed in 883 (20.4%) of 4327 subjects. The prevalence was significantly higher in women (706 patients, 24.1%) than in men (177 patients, 12.6%), and this significant difference between genders was observed in all age groups. 2. The prevalence of degenerative change of the mandibular condyle in TMD patients showed a gentle increase along with age. Such increase was statistically significant in women (P < 0.001), but not in men. 3. Sclerosis was observed the most frequently in all age groups and the mean age of the patients with osteophyte was the highest among four types of degenerative change. 4. Although men showed degenerative change in the left side more often and women showed degenerative change more frequently in both sides, the difference of distribution in sides between genders was not significant. In conclusion, the prevalence of degenerative change of the mandibular condyle in TMD patients is higher in women than in men, and increases steadily with aging, but not as dramatically as in other joints that show a steep increase in prevalence around the age of 45 years. It can be suggested that the epidemiologic characteristic of OA of the TMJ differs from those of other joints, and that a more extensive study based on the general population is necessary.
Soybeans released proteins when immersed in water at $50{\sim}60^{\sim}C$. We investigated the changes in the characteristics of soybean when soaked in water at different temperatures and studied the electrophoretic properties of soy proteins in recommended Korean soybean varieties after heat treatment. Soybean seeds were heated in soaking water at temperatures of 30, 40, 50, 60, $70^{\circ}C$ for 90 min, and also from 10 to 150min at $60^{\circ}C$. The pH value of the water decreased with heating time at $60^{\circ}C$, and the amount of soluble solids increased with temperature and heating time. The protein concentration of the solution increased with temperature and time. From SDS-PAGE of the proteins in soaking water, we detected two new bands of 16 kDa- and 31 kDa-proteins from the Korean soybean varieties on heat treatment.
The sorption of $AsO_4,\;SeO_3,\;CrO_4$ on schwertmannite and thermal analysis of sorbed samples were carried out. The results of sorption experiments showed that sorption characteristics of those three oxyanions on schwertmannite can be divided into two groups. The extent of sorption of $AsO_4$ and $SeO_3$ were 100% at up to 1 mM solution concentration, and they increased no more significantly. This can be interpreted as $AsO_4$ and $SeO_3$ substituting $SO_4$ in schwertmannite strucure by the ratio of 1 : 1. The extent of the sorption of $CrO_4$ was much lower than those of other two oxyanions. Thermal analysis was performed using two kinds of sorbed samples at 0.1 and 1.25 mM concentrations. The results of the thermal analysis showed that the samples sorbed by three different oxyanions have different thermal characteristics. The samples sorbed by $AsO_4$ showed smaller weight loss around $600^{\circ}C$ than the original loss of pure schwertmannite, and it is attributed to the substitution of $AsO_4$ for $SO_4$, which was caused by the loss of $SO_4$, than pure schwertmannite due to the substitution of $SO_4$ by $AsO_4$. It also showed additional weight loss around $600^{\circ}C$ due to the decomposition of $AsO_4$ at that temperature. The weight loss of samples sorbed by $SeO_3$ started at slightly lower temperature than that sorbed by $SO_4$ and kept that loss at wider temperature range, probably indicating that the decomposition of $SeO_3$ occurs at slightly lower temperature. However, for the samples sorbed by $CrO_4$, the weight loss caused by the decomposition of $SO_4$ was also smaller and there was no additional weight loss at higher temperature due to the thermal stability of $CrO_4$, indicating that $SO_4$ was also substituted by $CrO_4$ in schwertmannite. Sorption experiment and thermal analysis indicate that $CrO_4$ sorbs on schwertmannite by substiuting $SO_4$, but the affinity to $SO_4$ or instability of $CrO_4$ in scwertmannite structure probably prohibit perfect 1 : 1 substitution.
Environment-friendly agriculture (EFA) are aimed to reduce use of chemical inputs as possible the recycling of resources and the environment while preserving the local resources in the long term to ensure a constant productivity and profitability for producing safe food. This study was conducted to investigate characteristics on soil environment at paddy field of environment-friendly agriculture regions (EFARs) in Honam area. Survey point of EFARs Chungnam, Jeonbuk and Jeonnam, rice bran farming method in two districts, rice bran + snail farming method in eight districts, snail farming method in five districts and ducks farming method in three districts a total of 18 districts were selected. Annual of farming method, friendly-environment certification, amount of applied fertilizer, and history of cultivation to the farm household were surveyed. The content of available phosphate and silicate among the soil chemical properties in EFA paddy field were a little lower than optimum level, and those of agricultural methods fertilized with rice bran were a little lower than those of others. Hardness among the soil physical properties in EFA paddy field were a little lower than conventional practices, and that of agricultural methods fertilized with rice bran were a little lower than those of others. We showed fertilizer recommendation dose about soil nutritional shortages according to fertilization prescriptions index by crops.
