Adsorption Characteristics of PES-BSA Affinity Membrane Prepared by Electrospinning (전기방사로 제조된 PES-BSA 친화막의 흡착특성)

  • The adsorption characteristics of L-tryptophan of PES-BSA affinity membranes prepared by using electro-spinning were investigated. It was found that the fiber diameters could be controlled by the comparisons of fiber diameters prepared by various spinning conditions through FESEM and Image Analyzer. Darcy's permeability constants, mechanical properties and hydrophobic properties were enhanced since micro-fibers increased by increasing the composition ratio of HFB and BSA. The elution capacity of L-tryptophan in borate-DMSO buffer solution was higher than that in tris-HCl buffer solution, while the elution capacity of A 7906 type BSA was higher than A 8022 type BSA. This is due to the characteristics of BSA type, i. e. higher purity and uniform molecular weight and better pH stability of A 7906 type BSA than those of A 8022 type BSA.

  • Polyurethane foams are polymeric material with repeating groups of urethane and urea. When these are heated with ethylene glycol and K acetate catalyst at $200^{\circ}C$, the transesterification of them leads to soluble products. The mechanisms of the reaction were investigated from the molecular weight and the component distributions of the products by GPC and IR analysis. The degradation of the urethane groups was faster than that of urea groups in transesterification reaction. K acetate catalyst accelerated the rate of the transesterification because it had a high ionization tendency. Each reaction, using K or Sr acetate as a catalyst, progressed in the same reaction path but yielded different compositions in products because of the difference of the reaction rate.

Moisturizing Property and Physiological Activity of Chitosan Oligosaccharide (키토산 올리고당의 보습성과 생리활성에 관한 연구)

  • Chitosan oligosaccharide having physiological activity, such as cell proliferation and wound healing, was prepared by NaNO$_2$ oxidation-NaBH$_4$ reduction from natural chitosan. After deamination by NaNO$_2$ oxidation, the reducing-end residue from NaBH$_4$ was converted to alditol unit, and the reduction was checked by MBTH reagent. The resulting chitosan oligosaccharide had a degree of polymeration of 2-6 from HPLC analysis. From moisture absorption test at relative humidity of 43% and 81%, the moisture absorption ability was 63% and 57%. Moisture retention ability at relative humidity of 43%, silica gel emvironment, was 98% and 97% respectively. Cell proliferation was showed In the range of 0.000032~0.01%, wound healing effect was also appeared in the concentration of 2% and 20%. Antioxidative effect ($SC_{50}$/) was 3213 ppm. Chitosan oligosaccharide was compatible with most of ingredients used in cosmetic products.

Relationship between Molecular Structure of Acid-Hydrolyzed Rich Starch and Retrogradation (산처리 쌀전분의 분자구조와 노화속도)

  • The relationship between the molecular structure of acid-hydrolyzed rice starch and the retrogradation rate of starch gel was investigated. The molecular structure of starch was modified by acid hydrolysis with 1 N HCl at $35^{\circ}C$. The molecular weight of starch decreased as acid hydrolysis time was increased. At the early stage of hydrolysis up to 3 hr, the branching point of amylopectin was degraded and thereafter both ${\alpha}-1,4\;and\;{\alpha}-1,6$ linkages were hydrolyzed. The starch gel (50%) stored at $20^{\circ}C$ revealed that the rapid retrogradation occurred during 4 hr of storage which was more pronounced as the hydrolysis time increased. The degree of retrogradation of starch gels after 4 hr storage showed a linear relationship with the yield of hydrolyzate. These results suggested that the retrogradation of starch gel was accelerated by degradation of ${\alpha}-1,6$ linkages with acid.

  • Kinetics of solution photopolymerization of acrylonitrile(AN) with sensitizer, such as $NaSCN,\;KSCN,\;Ba(SCN)_2,\;NH_4SCN,\;ZnCl_2$ and $Na_2SeO_3$, were studied using UV crosslinker at various monomer concentrations($1.8{\sim}7.58mo1/1$), sensitizer concentrations($10{\sim}60%$), reaction temperature($10{\sim}70^{\circ}C$), energy intensities($1,000{\sim}9,900{\mu}J/cm^2$) at isothermal condition under nitrogen atmosphere. Under the irradiation of high pressure mercury lamp(${\lambda}=365nm$). High conversion and uniform molecular weight were obtained compare to thermal polymerization at reaction temperature of $50^{\circ}C$, reaction time of 3hr and 50% NaSCN without any initiator. Their kinetic model was as follows : $R_p=0.0142[M]^{0.82}[I]^{0.49}[S]^{0.52}$ exp(-1.33/RT).

Control of the Photo-induced Discoloration of the Handmade Korean Paper with Polyethylene Glycol (Polyethylene Glycol에 의한 수록한지(手漉韓紙)의 광(光) 변색(變色) 억제(抑制))

  • Classified handmade Korean paper which has been commonly used as the material for the paper cultural assets divide into four classes. We irradiated xenon arc lamp, which has almost the same spectrum distribution as natural sunlight, to test photo-induced of discoloration according to PEG molecular weight and concentration. So, we reach the followings. 1) Unbleached handmade Korean paper with PEG showed rapid decrease of brightness for 10 hours after photo-irradiation, regardless of molecular weight of PEG, but, after 10 hours, it showed gradual increase of brightness. From this, it proved that there was an effect of preventing discoloration of photo-irradiation. 2) Chemical bleached handmade Korean paper showed increase of brightness after 20 hours. Though time was delayed in increase of brightness, there was also an effect of preventing discoloration of photo-irradiation in chemical bleached paper. 3) Handmade Korean paper made with abet-mosk showed the lowest of brightness when it was tested on 400-20% PEG. Before photo-irradiation, the brightness of paper, applied it, was the lowest, but it showed the least difference of brightness after photo-irradiation. From this, it proved that there was the greatest effect of preventing discoloration in this paper. 4) Preventing yellowish, the first phenomena by photo-irradiation ageing with applying PEG was proved that there was the decrease of value of $b^*$ +, and increase value of $L^*$, which could fairly prevent decrease of brightness.

Effect of Ozonation and Coagulation on NOM Molecular Distribution and Activated Carbon Adsorption Capacity (응집과 오존처리가 NOM 분자량 분포특성과 활성탄 흡착능에 미치는 영향)

  • The main goals of this work are to investigate the effect of molecular weight distribution(MWD) and activated carbon adsorption capacity after conventional coagulation and enhanced coagulation. The ozonation was very effective to decompose the NOM to smaller size and to remove molecular smaller than 1,000. The concentration of DOC was reduced 0.25mg/L and 0.56mg/L by the conventional coagulation and the enhanced coagulation, respectively The conventional coagulation was not effective to remove NOM. However, the enhanced coagulation was effective to remove MW bigger than 10,000. The higher MW was shifted to smaller weight by ozonation in the raw water and the after conventional coagulation. After enhanced coagulation the MW had not changed significantly by ozonation. Also, it was observed that the ozone dosage did not have significant impact on MW shifting to smaller size. The adsorption capacity simulated by IAST comparing K values showed that the adsorption capacity was not impacted by ozone doses. There was very strong correlation between MW smaller than 10,000 and the mid- and strongly adsorbable fractions.

  • Poly($\small{L}$-Lactide)(PLLA) was prepared by a ring-opening polymerization of $\small{L}$-Lactide with various polydimethylsiloxane(PDMS) based copolymers as a stabilizer in supercritical carbon dioxide($scCO_2$). The block copolymeric stabilizers were synthesized by group transfer polymerization (GTP) by using PDMS macroinitiator. PLLA was found to be produced with fairly low molecular weight distribution as confirmed by gel permeation chromatography(GPC) analysis. Scanning electron microscopy (SEM) results showed that sub-micron size Poly($\small{L}$-lactide)(PLLA) particles were formed by suspension polymerization.

  • In HTPB/AP propellants, zirconium(Zr) addition to formulation was shown to be less specific impulse than aluminum(Al) by the theoretical calculation because of the lower flame temperature and higher molecular weight of Zr oxide. It was found that the burning rate was faster with the finer size of Zr and the more content of $2{\mu}m$ Zr the faster burning rate is in HTPB/AP/Zr propellants caused by the more conduction energy transfer from Zr flame to the burning surface. Also the burning rate of HTPB/AP/Zr propellant could be reduced by addition of 150nm Al, depending on AP size distribution in formulation with Butacene and $1{\mu}m$ AP.

  • Nitric Oxide Synthase(NO synthase: EC.1.14.13.39)는 생체내에서 L-arginine을 기질로 하여 nitric oxide(NO)와 L-citrulline의 생성을 매개하는 효소로서 뇌, 간장, 신장, 체장등 대부분의 주요장기와 근육세포, 신경세포 등 거의 모든 조직에 분포하고 있다. NO synthase에 의해 생성되는 NO는 혈관이완작용, 신경전달 물질로서의 작용, 면역 담당세포에서의 세포 독작용 등 많은 생리현상에 중요한 역할을 하는 것으로 알려져 있다. 특히 체장에서는 췌외분비 기능의 항진에 있어 세포내 cGMP level의 변동이 NO와 연관된다는 사실에 주목하고 있으며 본연구실에서도 이에 관한 연구가 진행중이다. 따라서 본 연구에서는 소 췌조직의 100,000$\times$g cytosol을 효소원으로 하여 다음과 같이 NO synthase의 분리, 정제를 시행하였다. Ammonium sulfate로 30%(176g solid ammonium sulfate/$\ell$) 포화, 침전 후 2',5'-ADP agarose 및 calmodulin-agarose affinity chromatography를 연속적으로 시행하여 NO synthase를 분리하였으며 electrophoresis상에서 약 160kd의 분자량을 나타내었다.
