Effect of Chitosan Treatment on the Quality of Dried Lavers (건조김 제조시 키토산처리가 품질에 미치는 영향)

  • Chitosan was used to treat lavers during the production of dried lavers. In the procedure of washing the harvested lavers, the lavers were treated with various solutions of 0.1 and 0.01% of 30 cp chitosan and of 0.005 and 0.04% of 90 cp chitosan. Then treated lavers were processed as usual for the production of dried lavers. Significant differences between chitosan-treated and non-treated lavers were observed in flavor, in total microbial counts, and in the number of coliform microorganisms. Off-flavor of dried lavers was reduced significantly by treating with chitosan. Total microbial count was $3.7{\times}10^{4}$ cells/g in the non-treated dried lavers, but they were $5.7{\times}10^{3}$ cells/g and $2.1{\times}10^{3}$ cells/g, respectively, in the dried lavers treated with 0.01% and 0.1% of 30 cp chitosan. The number of coliform microorganisms was 240 cells/g in non-treated dried lavers. However, coliform microorganisms were not detected in the dried lavers treated with 0.1% of 30 cp chitosan. When the dried lavers were used to make Kimbab (rice rolled with laver), the number of total microorganisms increased very slowly during storage time at $20^{\circ}C$ in the Kimbab rolled with chitosan-treated dried lavers compared to that in the Kimbab rolled with non-treated dried lavers.

Development of Broad-range and Specific 16S rRNA PCR for Use in Routine Diagnostic Clinical Microbiology (임상미생물 검출을 위한 광대한 범위와 특이도를 가지는 16S rRNA PCR법 개발)

  • Broad-range and specific 16S rRNA gene PCR is used for detection and identification of bacterial pathogens in clinical specimens from patients with a high suspicion for infection. We describe the development of a broad-range and specific PCR primer, based on bacterial 16S rRNA, for use in routine diagnostic clinical microbiology services. The primers were designed by using conservative regions of 16S rRNA sequences from 10 strains. Ninety-eight clinical strains were isolated from clinical patient specimens. A total of 98 strains of bacteria were identified by phenotypic methods; PCR with newly designed primers and universal primers. All purified PCR products were sequenced using both forward and reverse primers on an automated DNA analyzer. In this study, we evaluated the usefulness of the newly designed primers and the universal primers for the detection of bacteria, and both these techniques were compared with phenotypic methods for bacteria detection. When we also tested 98 strains of clinical isolates with newly designed primers, about 778 bp DNA fragments were amplified and identified from all strains. Of the 98 strains, 94 strains (95.9%) correspond in comparison with phenotypic methods. The newly designed primers showed that the identities of 98 (100%) strains were the same as those obtained by universal PCR primers. The overall agreement between the newly designed primers and universal primers was 100%. The primer set was designed for rapid, accurate, and cheap identification of bacterial pathogens. We think the newly designed primer set is useful for the identification of pathogenic bacteria.

Determination and Monitoring of Grayanotoxins in Honey Using LC-MS/MS (LC-MS/MS를 이용한 벌꿀 중 grayanotoxin 분석법 연구 및 실태조사)

  • This study was performed to establish analysis methods, and evaluated for grayanotoxin in domestic/foreign honey and wild honey. The molecular weight of grayanotoxins I, II and III, excluding grayanotoxin III that has been commercialized, were analyzed by LC-MS/MS. Then, the molecular structure of grayanotoxins I and II were analyzed by NMR. A total 111 samples (25 Korean honey, 21 Korean wild honey, 13 Korean honeycomb honey, 44 foreign honey, 8 foreign wild honey) were examined to determined whether or not each sample contained grayanotoxins I, II, and III. The honey samples were mixed with methanol and loaded into a tC18 cartridge, the filtrate was diluted with water, and the mixture was then analyzed by ESI triple-quadrupole LC-MS/MS. Grayanotoxins were only found in the foreign wild honey and were not detected in Korean honey, Korean honeycomb honey, or Korean wild honey. Three of the samples contained grayanotoxin I, II, and III, and one sample contained only grayanotoxins I and III. The lowest level for grayanotoxin I was 3.13 ${\pm}$ 0.00 mg/kg, and the highest level was 12.93 ${\pm}$ 0.01 mg/kg. The levels of grayanotoxin II were 0.84 ${\pm}$ 0.01 mg/kg, 0.92 ${\pm}$ 0.00 mg/kg and 1.08 ${\pm}$ 0.01 mg/kg, respectively. The lowest level of grayanotoxin III was 0.25 ${\pm}$ 0.01 mg/kg and the highest level was 3.29 ${\pm}$ 0.74 mg/kg. Through this study, safety management for foreign wild honey has been enabled.

A Study on the Pollution of Polycyclic Aromatic Hydrocarbons(PAHs) in the Surface Sediments Around Gwangyang Bay (광양만 주변해역 표층퇴적물에서의 다환방향족탄화수소류(PAHs)의 오염에 관한 연구)

  • PAHs(Polycyclic Aromatic Hydrocarbons) are widespread contaminants in the marine environment. They are of mainly anthropogenic origin from urban runoff, oil spill and combustion of fossil fuels. Some PAHs are potentially carcinogenic and mutagenic to aquatic organism The contamination of PAHs in the coastal environments has not been well known yet in Korea. This study was carried out to survey the contamination of PAHs in sediments around Gwangyang bay. The Yeosu petrochemical industrial complex, POSCO(Pohang steel company) and Gwangyang container harbor are located around the bay. PAHs in sediment samples were extracted in soxhlet extractor and were identified and quantified by GC-MS(Gas Chromatography-Mass Spectrometry) TOC(Total Organic carbon) and textural parameters in sediment samples were also analyzed 13 species of PAHs were detected at all of the surface sediments. Total PAHs concentrations in the surface sediments ranged from 171.40 to $1013.54{\mu}g/kg$ dry wt.. In most of the surface sediments, Naphthalene was the highest in the range of 14.08 to $691.39{\mu}g/kg$ dry wt. and Anthracene was the lowest in the range of 0.49 to $22.66{\mu}g/kg$ dry wt.. The correlation coefficients between individual PAHs and Total PAHs in the surface sediments were relatively higher in the low molecular compounds such as Naphthalene and Phenanthrene. In the relationship of the P/A(Phenanthrene/Anthracene) ratio and F/P(Fluoranthene/Pyrene) ratio, P/A ratio was generally above 10 and F/P ratio was shown to be above 1 in all sediment samples. These data indicate that PAHs in sediments around Gwangyang bay seem to be of both pyrolytic and petrogenic origin. Total PAHs in the surface sediments were correlated with TOC and textural parameters. The values of PAHs in the surface and core sediments were lower than the biological effect guidelines.

  • The health benefits associated with consumption of fresh produce have been clearly demonstrated and encouraged by international nutrition and health authorities. However, since fresh produce is usually minimally processed, increased consumption of fresh fruits and vegetables has also led to a simultaneous escalation of foodborne illness cases. According to the report by the World Health Organization (WHO), 1 in 10 people suffer from foodborne diseases and 420,000 die every year globally. In comparison to other processed foods, fresh produce can be easily contaminated by various routes at different points in the supply chain from farm to fork. This review is focused on the identification and characterization of possible sources of foodborne illnesses from chemical, biological, and physical hazards and the applicable methodologies to detect potential contaminants. Agro-chemicals (pesticides, fungicides and herbicides), natural toxins (mycotoxins and plant toxins), and heavy metals (mercury and cadmium) are the main sources of chemical hazards, which can be detected by several methods including chromatography and nano-techniques based on nanostructured materials such as noble metal nanoparticles (NMPs), quantum dots (QDs) and magnetic nanoparticles or nanotube. However, the diversity of chemical structures complicates the establishment of one standard method to differentiate the variety of chemical compounds. In addition, fresh fruits and vegetables contain high nutrient contents and moisture, which promote the growth of unwanted microorganisms including bacterial pathogens (Salmonella, E. coli O157: H7, Shigella, Listeria monocytogenes, and Bacillus cereus) and non-bacterial pathogens (norovirus and parasites). In order to detect specific pathogens in fresh produce, methods based on molecular biology such as PCR and immunology are commonly used. Finally, physical hazards including contamination by glass, metal, and gravel in food can cause serious injuries to customers. In order to decrease physical hazards, vision systems such as X-ray inspection have been adopted to detect physical contaminants in food, while exceptional handling skills by food production employees are required to prevent additional contamination.

  • In this study, the possibility of a quartz crystal micro-balance(QCM) modification of crystallization of L-Penicillamine and D-Penicillamine with a Vapor Diffused Molecular Assembly Technique and its application to the R-(-)-Mandelic acid and S-(+)- Mandelic acid measurement was investigated. The 3-dimensional structures of L-Penicillamine and D-Penicillamine on the surface of QCM were verified to be different from each other through QCM and AFM analyses. The D-Penicillamine modified QCM had specific recognition to the R-(-)-Mandelic acid, but L-Penicillamine modified QCM had no specificity to the R-(-)-Mandelic acid and S-(+)- Mandelic acid. From these results, it was known that the QCM could be modified with various selective meterials via VDMA, and the chiral isomer such as a Mandelic acid isomer could be detected by using a modified QCM.

A Study on Monte Carlo Simulation in Resin of New Austria Tunnel Method by admixture for Shrinkage Compensating Concrete (무수축 콘크리트 혼화제를 활용한 New Austria Tunnel Method 수지에서 Monte Carlo 시뮬레이션에 관한 연구)

  • The influences of scatterer and absorber in turbid material by light scattering in concrete admixture were interpreted for the scattered intensity and wavelength. The molecular properties have been studied by Monte Carlo simulation in resin of New Austria Tunnel Method. It has been found that the effects of optical properties in scattering media could be investigated by the optical parameters(${\mu}_s$, ${\mu}_a$,${\mu}_t$). Monte Carlo Simulation method for modelling of light transport in the civil engineering and construction field was applied. The results using a phantom were discussed that the admixture for shrinkage compensating concrete in NATM-rasin from source to detector is measured, and scattering intensity is stronger with those obtained through Monte Carlo Simulation. It may also aid in designing the best model for coatings and corrosion for the durability of metal constructions.

  • 현재 화합물 반도체 나노구조는 광학적, 전기적 특성을 기반으로 하는 단전자 트랜지스터, 적외선 검출기, 레이저, 태양전지와 같은 분야에 응용하기 위한 많은 연구가 진행되고 있다. 특히 양자점은 3차원으로 구속되어 있는 상태 밀도를 갖고 있어 레이저 응용 시 낮은 문턱 전류 밀도, 높은 이득, 높은 열적 안정성을 기대되고 있지만 양자점의 운반자 수집과 열적 안정성의 한계가 여전히 존재한다. 이러한 문제를 해결하기 위해 다양한 방법이 연구되고 있으며, 그 중 단층 양자점에 비해 운반자 수집과 열적 안정성이 뛰어난 다층 양자점이 결합된 구조에 대한 연구가 활발히 이루어지고, 다층으로 성장된 양자점 구조는 양자점의 크기 분포 조절이 용이하고 양자점 층간의 전기적 결합력이 강한 특성이 있다. 본 연구에서는 분자 선속 에피 성장법(Molecular Beam Epitaxy; MBE)과 원자 층 교대 성장법(Atomic Layer Epitaxy; ALE)으로 CdTe/ZnTe 다층 양자점을 ZnTe 장벽층의 두께를 변화하면서 성장 후 광학적 특성을 연구하였다. 저온 광루미네센스 측정(Photoluminescence; PL)을 통하여 ZnTe 장벽층 두께가 증가할수록 양자점의 PL 피크가 높은 에너지로 이동함을 알 수 있었는데, 이는 ZnTe 장벽층의 두께가 증가할수록 양자점 층간의 결합력이 감소하면서 양자점의 크기가 작아졌기 때문이다. 그리고 ZnTe 장벽층의 두께가 증가할수록 PL 세기가 커지는 것을 알 수 있었는데, 이는 ZnTe 장벽층의 두께가 증가할수록 더 많은 운반자가 양자점으로 구속되기 때문이다. 또한 온도 의존 광루미네센스 측정 결과 ZnTe 장벽층의 두께가 증가할수록 열적 활성화 에너지가 커지는 것을 관찰하였고, 시분해 광루미네센스 측정을 통해 ZnTe 장벽층의 두께에 따른 운반자 동역학에 대해 연구하였다. 이와 같은 결과 CdTe/ZnTe 다층 양자점 구조에서 장벽층의 두께에 따른 광학적 특성에 대해 이해 할 수 있었다.

Analysis of Imidazoline Type Cationic Surfactants (Imidazoline계 양이온 계면활성제 분석)

  • Analysis for the imidazoline type cationic surfactants was performed by the gas chromatography(GC) and high performance liquid chromatography(HPLC). The composition of the alkyl chain distribution was investigated by GC after base/Acid hydrolysis of the imidazoline ring. The distribution of total alkyl chain was separated clearly by a Bondclone C18/NOVA-Pak C18 HPLC column using 50% acetonitrile in methanol containing 0.1M sodium perchlorate as a mobile phase. Alkyl chain distribution and average molecular weight of imidazoline type cationic surfactants were obtained based on these analytical methods. The agreement of results from GC and HPLC was good. The detection limit of imidazoline by HPLC method was 10ng without pretreatment.

  • 현재 화합물 반도체 나노구조는 적외선 검출기, 레이저, 발광 다이오드, 단전자 트랜지스터, 태양전지 등과 같은 고효율 광전자 소자에서의 응용을 위해 활발한 연구가 진행 되고 있다. 특히 양자점은 3차원으로 구속되어 있는 상태 밀도를 갖고 있어 레이저 응용 시 낮은 문턱 전류 밀도, 높은 이득, 높은 열적 안정성을 기대되고 있다. 하지만 양자점의 크기가 불규칙적이고 운반자 수집의 한계로 인하여 기대 이하의 온도 안정성을 갖고 있어 이를 극복하기 위해 양자점의 크기와 운반자 수집을 제어하기 위해 다양한 방법이 연구되고 있다. 본 연구에서는 분자 선속 에피 성장법(Molecular Beam Epitaxy; MBE)과 원자 층 교대 성장법(Atomic Layer Epitaxy; ALE)으로 크기가 다른 CdTe/ZnTe 이중 양자점을 ZnTe 장벽층의 두께에 변화하면서 성장 후 광학적 특성을 연구하였다. 저온 광루미네센스 측정(Photoluminescence; PL) 측정 결과 장벽층 두께가 작아질수록 작은 양자점의 광 루미네센스의 세기가 감소하면서 큰 양자점의 세기가 증가하는 것을 관찰할 수 있었는데, 이는 장벽층 두께가 작아질수록 작은 양자점의 운반자들이 큰 양자점으로 이동되는 양이 많아지기 때문이다. 또한 장벽층 두께가 작아질수록 큰 양자점의 반치폭(Full Width at Half Maximum; FWHM)이 단층 양자점의 반치폭 보다 감소하는 것을 관찰 할 수 있었는데 이는 작은 양자점과 결합된 큰 양자점이 작은 양자점의 strain을 받아 크기의 균일함이 증가했기 때문이다. 이와 같은 결과 두 양자점이 결합된 이중 양자점 구조가 단층 양자점의 한계인 운반자 수집과 크기의 균일함을 향상할 수 있는 좋은 구조임을 제시하고 있다.
