Immuno-stimulating Polysaccharides from the Fruiting Bodies of Fomitella fraxinea (II) -Isolation and characterization of hot-water extracted polysaccharides- (Fomitella fraxinea로부터 분리한 면역활성 다당류 (II) -열수추출 다당류의 분리 및 특성 -)

  • Polysaccharide FHW was extracted from the fruiting bodies of Fomitella fraxinea with hot-water treatment and then fractionated into FHW-I and FHW-II on DEAE-Cellulose chromatography. FHW-I and FCW-II were further purified into FHW-Ia and Ib, FHW-IIa and IIb on gel permeation chromatography, respectively. A small amount of uronic acid was detected and glucose, galactose, fucose, and mannose were found to be main sugars in the polysaccharides. Protein was detected in FHW-Ia, FHW-IIa, and FHW-IIb, but not in FHW-Ib. FHW-Ia was identified to be a fuco-gluco-mannogalactan with molecular weight of 19,000 and FHW-Ib was a gluco-fuco-mannogalactan of 15,000. FHW-IIa and FHW-IIb were galacto-mannoglucan and their molecular weights were estimated to be 31,000 and 9,000, respectively. Both FHW-Ib and FHW-IIb did not show an absorption band characteristic of the ${\beta}-glycosidic$ linkage in IR spectra. FHW-IIb showed a strong immuno-stimulating activity but the other three polysaccharides showed a weak activity.

Efficient Treatment Methods for Reducing Escherichia coli Populations in Commercially-Available Red Pepper Powder in Korea (국내 유통 고춧가루의 병원성 대장균 오염 및 대장균 저감화 방법)

  • This study was conducted to investigate the level of contamination of pathogenic Escherichia (E.) coli in 50 types of red pepper powders collected domestically. Pathogenic E. coli was confirmed using real-time PCR to confirm the 4 types of EAEC, EPEC, EHEC and ETEC. One sample out of 50 was contaminated with pathogenic E. coli. The type of pathogenic E. coli detected in the sample was EAEC. This study was also conducted to determine the effect of alcohol treatment on the reduction of E. coli populations in red pepper powder. The amount of E. coli in the control was $1.2{\times}10^6$ cfu/mL. The amount of E. coli in 10 minutes immersion treatment with 10% alcohol was $1.1{\times}10^6$ cfu/mL. In samples treated with over 20% alcohol, E. coli was not detected. This showed that 10 minutes of immersion in over 20% alcohol might be effective to reduce E. coli. This study was also conducted to determine the effect of UV irradiation on E. coli reduction. The number of E. coli in the control group was $5.0{\times}10^5$ cfu/mL. However, the number of E. coli in 45 min of the UV irradiated sample decreased to $1.0{\times}10^3$ cfu/mL, by $10^2$ cfu/mL. In contrast, E. coli was not detected in an over 60 min UV irradiated sample in $10^{-2}dilution$. This study showed that over 20% alcohol treatment and UV irradiation for 60 min was effective to control E. coli in red pepper powder.

Development of an SH-SAW Sensor for Protein Measurement (단백질 측정용 SH-SAW 센서 개발)

  • We developed SH-SAW sensors to detect protein molecules in liquid solutions applying a particular antibody thin film on the delay line of transverse SAW devices. The antibody investigated was human-immune-globulin G (HigG) to hold the antigens (anti-HigG) in the protein solution. We fabricated the sensor generating 100 MHz with the piezoelectric single crystal LiTaO₃. We measured the frequency change of the sensor by adding the anti-body concentration on SAM (self assembled monolayer) deposited on the Au layer. The sensor showed stable response to the mass loading effects of the anti-HigG molecules with the sensitivity up to 10.8 ng/ml/Hz at noise level 400 Hz below.

  • Methyl mercaptan oxidase was isolated and purified from Thiobacillus thioparus TK-m for the detection of mercaptans. The procedure of purification involved DEAE-Sephacel and Superose 12 column chromatographies with recovery yields of 47.5 and 48.5 %, and specific activity of 374 and 1240.8 units/mg-protein, respectively. The molecular weight of purified methyl mercaptan oxidase was determined to be 66.1 kDa by SDS-PAGE. Optimum temperature for activity was observed at $55\;^{circ}C$. This enzyme was activated by $(NH_4)_2SO_4$ and NaCl and inhibited by $NH_4Cl$.

  • Flow-injection analysis (FIA) for on-line monitoring of glucose and acetate are described and employed in E.coli fermentation process. Glucose oxidase (GOD) for the detection of glucose is immobilized on epoxy polymer support, which is packed in a small cartirdge, and applied to a GOD-FIA system. The detection of acetate is based on the inhibition of acetate to the oxidation of sarcosine by sarcosine oxidase (SOD). SOD is also immobilized on epoxy polymer support and used for a SOD-FIA system. GOD-FIA system is characterized as well as SOD-FIA system by the investigation of the effects of pH, temperature and metabolites in samples on the peak height. GOD-FIA and SOD-FIA systems were also applied for on-line On-line measurements buy FIA measurements by FIA were in god agreement with off-line measurements.

  • In the present study, we performed the PCR assay using TOX2P/TOX2M primer targeting a specific region within mcyB gene to identify potential microcystin-producing cyanobacteria. TOX2P/TOX2M primer set was effective in amplifying mcy gene in the field samples containing Microcystis spp. of 1,000 cells per mL. Moreover, the results from the PCR assay agreed with those of the ELISA analysis. Consequently, this study demonstrated that TOX2P/TOX2M primer set can be used as a genetic probe for the early detection of cyanobacterial toxigenicity in Korean water bodies.

  • Recombinant Human serum albumin (rHSA) was purified to near homogeneity from H, polymorpha using heat treatment, ultrafiltratipn and Phenyl Sepharose CL-4B and Mono Q column chro - matographies with a recovery yield of 60% The molecular weight of the purified rHSA was estimated to be about 65,000 Da by denaturing SDS-PAGE The N terminal amino acid sequence of the purified HSA determined by Edman degradation was turned out to be Asp- Ala- His- Lys- Ser- Glu- Ala, suggesting that the rHSA expressed in H, polymorpha was efficiently secreted and correctly processed at the cleavage site of secretion signal sequence. The purified human albumin showed the pI value identical to that of authentic human serum albumin.

  • Carbonyl compounds, glycine, ammonia, carbon dioxide and hydrogen have been identified in gamma-irradiated oxygenated aqueous solutions of triglycine. The reactions occurring in this system are postulated only by representing the initial process and the final products. The triglycine molecule undergoes a reaction to form an intermediate giving rise to carbonyl compounds and acid amides. These compounds are decomposed to acetamide, glycine, glyoxylic acid, formic acid, oxalic acid, ammonia, and carbon dioxide by free radical attack in the primary and/or secondary reactions. It is also possible that the unrecognized reactions may give rise to products formed by radical-radical combinations. Satisfactory material balance for all the products have to wait until the completion of a study now in progress.ogress.

Molecular detection of infectious pathogens in honeybee colonies reared in eastern Gyeongbuk province, Korea (경북 동부지역 꿀벌에서 주요 병원체의 분자생물학적 검출)

  • The ecologically and economically important honeybee species are susceptible to infections by various pathogens. This study was investigated to detect infectious pathogens in honeybee colonies reared in eastern Gyeongbuk province by PCR in 2010~2011. A total of 11 infectious pathogens, including 6 viruses, 2 bacteria, 2 fungi, and 1 parasite, were investigated in honeybee colonies suffering from symptoms of sudden collapse, depopulation, or paralysis. The infectious pathogens and infection rates among 24 honeybee colonies detected were as follows: sacbrood virus (66.7%), deformed wing virus (4.2%), black queen cell virus (12.5%), Kashmir bee virus (29.2%), American foulbrood (41.7%), European foulbrood (12.5%), stonebrood (45.8%), chalkbrood (4.2%), and Nosema (33.3%), respectively. Since the coinfection rates of multiple pathogens were detected high in honeybee colonies reared in eastern Gyeongbuk province, large-scale investigation and appropriate control programs need to be established in this region.

  • The surface patterns and the structures of transverse section of the egg-shell of the sikmoth, Bombyx mandarina, have been described by scanning electron microscope. Three spatially differentiated cross section, called lamellar, conic pillar and cover layers, are found on the mature eg-shell. Silkmoth chorion proteins were detected more than 80 components from a single chorion by two-dimensional electrophoresis. Major protein components of the egg-shell have bee identified on the basis of their isoelectric points and molecular weights, pH 4-6 and 6-30 kd. Several protein components are found entirely or predominantly in th cover layers.
