Antibacterial Activity of Continentalic Acid from Aralia continentalis (독활(獨活)(Aralia continentalis)추출물 Continentalic Acid의 항균활성 연구)

  • 독활(Aralia continentalis)로부터 항균활성 물질을 찾아내기 위한 일환으로 항균실험과 분리실험을 병행하여 실시하였다. 항균활성물질 분리는 독활을 클로로포롬으로 추출하여 실리카겔($SiO_2$)과 분취용액체크로마토그래피(prep-HPLC)법으로 2종의 화합물을 분리하여 핵자기공명(NMR) 등 분광학적인 기법으로 이용하여 구조동정을 하였다. 이때 2종의 화합물은 (-)-pimara-8(1),15-diene-19-oic acid와 (24E)-stagmasta-5,22-dien-$3{\beta}-ol$임이 확인되었다. 2종의 화합물에 대한 메티실린 내성 황색포도상구균(MRSA) 및 메터실린 감응 황색포도상구균(MSSA)의 표준균주와 임상분리균주(MRSA)에서의 최소억제농도(MIC)가 $8-16{\mu}g/mL$로 나타났다. 그러므로 본 연구의 결과로부터 화합물(-)pimara-8(1),15-diene-19-oic acid은 항생제 내성균에 대한 치료제로서 개발 가능성을 확인할 수 있었다.

Precision measurements of radiometric aperture area by laser spot scanning along the edge of the aperture (레이저 스폿의 칼날주사 방법에 의한 복사계 개구 면적의 정밀측정)

  • The uncertainty of the detector-based candela scale is limited by the area measurement uncertainty of radiometric apertures. The apertures were fabricated with a diamond-turning machine which trimmed the edge of the apertures as sharply as a knife edge. The positions of the apertures were controlled by a digital feedback algorithm to scan the laser spot with the beam waist less than 5 ${\mu}{\textrm}{m}$. The knife edge scan yielded a set of coordinates on the edges of the aperture. The areas of the apertures were obtained by fitting the coordinates to the ellipses. The relative standard uncertainty of the measurement was estimated to be 8${\times}$10$^{-5}$.

  • The electrochemical spectral sensitization of a $SnO_2$ electrode with eosin B, a Xanthene dye, has been studied in acetonitrile. Measurements of the photocurrent have been carried out in the presence of supersensitizers such as thiourea, 1-allyl-2-thiourea, NaSCN, and NaI. The magnitude of the supersensitized photocurrent was greater than that of the sensitized photocurrent for all of the supersensitizers studied. However, the long time span of irradiation causes a significant decrease of the supersensitized photocurrent as well as the absorbance. These results, together with infrared spectra and fluorescence spectra, are taken into account to elucidate the mechanism of photoreaction between eosin B and supersensitizers in acetonitrile.

  • The analytical method to determine europium(III) ion in aqueous solution by fluorescence spectroscopy based upon the conformational change of calmodulin in the presence of the analyte has been studied. The fiber optic chemical sensor used in this study was constructed by entrapping a fluorescein-labeled calmodulin solution, EGTA, buffer solution at the common end of a bifurcated fiber optic bundle by means of a dialysis membrane. The calibration curve to determine europium(III) ion was obtained when concentration of calmodulin, concentration of EGTA, Tris-HCl buffer solution, pH, excitation wavelength and fluorescence wavelength were $5.0{\times}10^{-5}M$, 0.50 mM, 5.0 mM, 7.0, 495 nm and 520 nm, respectively. The detection limit was $1.0{\times}10^{-11}M$ and the working range of the calibration curve for the sensor was $1.0{\times}10^{-11}M{\sim}1.0{\times}10^{-9}M$. The response time was 15 minutes. For the determination of europium(III) ion by the present method, $Na^+$ and $K^+$ ions did not interfere but $Ca^{2+}$ ion seriously interfered.

  • New conducting polymers containing heterocyclic ring with carbazole, EDOT and benzobisthiazole were synthesized and characterized by organic spectroscopic methods. Potentiometric ion-selective membrane electrodes (ISMEs) have been extensively used for ion analysis in clinical, environmental, and industeial fields owing to its wide response range (4 to 7 orders of magnitude), no effect of sample turbidity, fast response time, and ease of miniaturization. Considerable attention has been given to alternative use of room temperature vulcanizing (RTV)-type silicone rubber(SR) owing to its strong adhesion and high thermal durability. Unfortunately, the high membrane resistance of SR-based (ISMs)(2 to 3 higher orders of magnitude compared to those of poly(vinyl chloride)(PVC)-based ones) has significantly restricted its application. Herein we demonstrate a new method to reduce membrane resistance via addition of new conducting polymer into the SR-based ISMs.

The characteristics of dye-sensitized solar cells using carbon nanotube in working and counter electrodes (작업전극과 상대전극에 탄소나노튜브를 이용한 염료감응 태양전지의 특성연구)

  • The effect of electrochemical characteristics of dye-sensitized solar cells (DSSC) upon employing multi-wall carbon nanotube (MWCNT) on both working electrode and counter electrode were examined with using EIS, J-V curves and UV-Vis absorption spectrometry. When 0.1 wt% of MWCNT was employed in the $TiO_2$-MWCNT composit on working electrode, the energy conversion efficiency increased about 12.5% compared to the $TiO_2$ only working electrode. The higher light conversion efficiency may attribut to the high electrical conductivity of MWCNT in $TiO_2$-MWCNT composite which improves the electron transport in the working electrode. However, higher amount of MWCNT than 0.1 wt% in the $TiO_2$-MWCNT composite decreases the light conversion efficiency, which is mainly ascribed to the decreased transmittance of light by MWCNT and to the decreased adsorption of dye onto $TiO_2$. The MWCNT employed counter electrode exhibited much lower light conversion efficiency of DSSC than the Pt-counter electrode, while the MWCNT-Pt counter electrode showed similar in light conversion efficiency to that of Pt-counter electrode.

Determination of L-Dopa by Spectrofluorimetry Using Co-fluorescence Enhancer (보조 형광증가제를 이용한 L-Dopa의 형광분광법적 정량)

  • Methods to determine L-dopa(L-3,4-dihydroxyphenylalanine) in aqueous solution by spectrofluorimetry based upon the ligand sensitized luminescence of Tb(III) ion L-dopa complex have been studied. Tb(III) ion and Lu(III) ion were used as ligand sensitized fluorescencer and co-fluorescence enhancer, respectively. The effects of excitation wavelength, pH, concentration of Tb(III) ion, concentration of Lu(III) ion and emission wavelength on the fluorescence intensity were investigated. The fluorescence intensity of the Tb(III) ion L-dopa complex was further increased with addition of Lu(III) ion. The calibration curve for L-dopa was linear over the range from 5.0 ${\times}$ $10^{-7}$ M to 1.0${\times}$ $10^{-4}$ M and the detection limit was 4.0 ${\times}$ $10^{-8}$ M under the optimal experimental conditions of 300 nm, 8.0, 1.0 ${\times}$ $10^{-4}$ M and 545 nm for excitation wavelength, pH, concentration of Tb(III) ion and emission wavelength, respectively. When Lu(III) ion was added to Tb(Ⅲ) ion L-dopa complex, the concentration range of linear response and detection limit were 1.0${\times}$$10^{-8}$ M to 2.0 ${\times}$ $10^{-4}$ M and 1.0 ${\times}$ $10^{-9}$ M, respectively under the optimal experimental conditions of 300 nm, 8.5, 1.0 ${\times}$ $10^{-5}$ M, 1.0 ${\times}$ $10^{-5}$ M, 545 nm for excitation wavelength, pH, concentration of Tb(III) ion, concentration of Lu(III) ion and emission wavelength, respectively.

  • An electrochemical glucose sensor with enzyme-free was fabricated using Pt nanoparticles (Pt NPs) decorated CuO nanoflowers (CuO NFs). 3-D CuO nanoflowers film was directly synthesized on Cu foil by a simple hydrothermal method and Pt NPs were dispersed on the petal surface of CuO NFs through electrochemical deposition. This prepared sample was noted to Pt NPs-CuO NF. Morphology of the Pt NPs-CuO NFs layer was analyzed using scanning electron microscopy (SEM) and energy-dispersive X-ray spectroscopy (EDS). The electrochemical properties and sensing performances were investigated using cyclic voltammetry (CV) and chronoamperometry (CA) under alkaline condition. The sensor exhibited a high sensitivity, wide liner range and fast response time. Its excellent sensing performance was attributed to the synergistic effect of the Pt NPs and CuO nanostructure.

  • Among various metal oxides semiconductors, $TiO_2$ is the most studied semiconductor for environmental clean-up applications due to its unique ability in photocatalyzing various organic contaminants, its chemical inertness, and nontoxicity. $TiO_2$, however, has a few drawbacks to be solved such as reactivity mainly working under ultraviolet irradiation (${\lambda}$ < 387 nm) and electron - hole recombination on $TiO_2$. In this study, to extend the absorption range of $TiO_2$ into the visible range and enhance electron - hole separation, we synthesized platinum (Pt) deposited $C-TiO_2$. The presence of Pt as an electron sink has been known to snhance the separation of photogenerated electron-hole pairs and induce the thermal decomposition. The characterization of as-synthesized $Pt-C-TiO_2$ was performed by Transmission Electron Microscopic (TEM), the Brunuer-Emmett-Teller (BET) method, X-ray Diffractometer (XRD), UV-vis spectrometer (UV-DRS), and X-ray Photoelectron Spectroscopy (XPS). In order to estimate the photocatalytic activity of the synthesized materials, the photoelectron Spectroscopy (XPS). In order to estimate the photocatalytic activity of the synthesized materials, the photodegradation experiment of an azo dye (Acid Red 44; $C_{10}H_7N=NC_{10}H_3(SO_3Na)_2OH$)was carried out by using an Xe arc lamp (300 W, Oriel). A 420 nm cut-off filter was used for visible light irradiation. From the results, Pt-deposited $C-TiO_2$ showed a far superior phothdegradation activity to Degussa P25, the commercial product under the irradiation of visible light and enhanced photocatalytic activity of visible-working $C-TiO_2$. This is a useful result into the application for the purification system of dye wastewater using visible energy of sun light.

Determination of Eu(III) by Fluorescence Spectrometry (형광분광법에 의한 Eu(Ⅲ)의 정량)

  • Methods to determine EU(Ⅲ) ion in aqueous solution by fluorescence spectrometry based upon the ligand sensitized fluorescence of Eu(Ⅲ)-terephthalic acid (TPA) complex ion have been studied. The effects of excitation wavelength, pH, concentration of TPA and emission wavelength on the fluorescence intensity were investigated. The fluorescence intensity of the Eu(Ⅲ) complex ion was further increased with addition of trioctylphosphine oxide (TOPO). In this case Triton X-100 was used to dissolve TOPO in aqueous solution. The calibration curve for Eu(Ⅲ) was linear over the range from $1.0{\times}10^{-6}M\;to\;4.0{\times}10^{-4}M$ and the detection limit was $1.0{\times}10^{-6}M$ under the experimental conditions of 256 nm, 5.6, $3.5{\times}10^{-4}$M$ and 615 nm for excitation wavelength, pH, concentration of TPA and emission wavelength, respectively. When TOPO was added to the Eu(Ⅲ)-TPA system, the concentration range of linear response and the detection limit were $1.0 {\times}10^{-9}M\;to\;1.0{\times}10^{-4}M,\;1.0{\times}10^{-7}M,$ respectively under the experimental conditions of 284 nm, 4.4 and $1.0{\times} 10^{-4}M$ for excitation wavelength, pH and concentration of TOPO, respectively. Effects of interferences from various cations for the determination of Eu(Ⅲ) ion were also investigated.
