Selction of Useful Chemicals Reducing Soybean -Sprout Rot (콩나물 부패경감에 유용한 약제 선발)

  • This study was carried out to select useful chemicals to control pathogens inciting soybean-sprout rot. Pathogen strains were isolated from decaying soybean seedlings. The isolated strains were identified as Pseudomonas spp. (Pse. strains) through microbiological test, however, no rot-inciting fungus was isolated. Eight food additives were tested with different concentrations in controlling pathogens and harmfulness. Five chemicals(over 5% cone.) including sodium hypochlorite apparently inhibited the growth of Pse. strains on media plate, however, sodium hypochlorite was discarded due to its severe germination inhibition. Propionic acid and acetic acid inhibited the growth of Pse. strains more effectively than calcium propionate and sodium propionate relatively. As no harmful effect on seed germination and no growth retard of soybean sprouts were observed by those chemicals with lower concentration(0.5∼1.0%), these chemicals were considered to be applicable to sprout rot control judged by the effectiveness and permissible concentration as food additives.

Isolation and Utilization of Antagonistic Pseudomonas fluorescens from Soils for the Protection of Soybean Sprouts Rot (콩나물 부패병 방제를 위해 토양으로부터 분리한 길항균 Pseudomonas fluorescens의 이용)

  • Thirty-three bacterial and fungal strains were isolated from the rotten soybeans and soybean sprouts to isolate pathogenic microorganisms which cause soybean sprouts rot during soybean sprouts cultivation. In pathogenicity tests of the isolates on soybean sprouts, two isolates(K-17 and K-28) caused soybean sprouts rot and were identified as Erwinia carotovora and Fusarium sp., respectively. To isolate antagonists aganist K-17 and K-28 pathogens, bacteria were isolated from various soybean-cultivated soils and screened by the inhibition zone method. A bacterial isolate(J-232) which inhibited growth of both pathogens was identified as Pseudomonas fluorescens and further examined. The culture filtrate of P. fluorescens J-232 (dilution rate of 500 times) inhibited the growth of Erwinia carotovora K-17 and Fusarium sp. K-28 both on potato dextrose agar medium and on soybean sprouts cultivated in vessel. The development of soybean sprouts rots was observed during cultivation by inoculation of soybean seeds with culture filtrate of both pathogens. The combined inoculation of soybean seeds with culture filtrate of antagonistic bacterium and that of pathogens prevented soybean sprouts rot, and the growth of soybean sprouts was similar to that of control. The soybean sprouts inoculated with antagonists culture filtrate alone did not develop soybean sprouts rot, and the growth of the seedlings was shown to be slightly promoted as compared with that of control.

Identifications of a Sprout-Rot Pathogen Pseudomonas Species SN239 and Selection Resistant Soybean Line (콩나물 부패균 Pseudomonas sp. SN239 동정과 콩나물 부패병 내병성 계통 선발)

  • Control microbial contamination in pathogens to soy sprouts has always been highly concerned in soybean sprout industries because the soybean sprouts are consumed largely as a nutritious fresh vegetable around the world. However, pathogens in soy sprouts are little known. Here, we isolated a strain of Pseudomonas sp. SN239 that caused severer symptoms in sprouts of many soybean cultivars. In phylogenetic relationships using 16S ribosomal RNA sequences of the Pseudomonas species, the identified Pseudomonas sp. SN239 was grouped with P. putita, P. plecoglossicida, P. monteilii and P. mevalonii. Thus, the bacterial strain SN239 might be a newly identified Pseudomonas species which closely related to P. putida. Furthermore, we found that a Korean indigenous soybean (Glycine max) cultivar YNPCSS3-19 has strong resistance against the Pseudomonas sp. SN239.

  • This study was conducted to improve circulation system and the storage of garlic against the spoiling microorganisms during the storage. To fulfill this objective, the isolation and identification of spoiling microorganisms from horticultural products were performed at the first step. Antimicrobical activity of electrolyzed alkaline water (EAW) was tested against the spoiling microorganisms and MIC(minimun inhibitory concentration) was determined. As a result, it showed the formidable MIC value at near 2.0%(v/v) against the spoiling microorganisms.

16S rDNA-PCR and RFLP Analysis for rapid identification of Spoilage Bacteria from low Salt Cucumber Brine (저염 발효오이로부터 16S rDNA-PCR과 RFLP분석을 통한 부패균의 신속한 확인)

  • The aim of this study was to isolate and identify the spoilage bacteria in the low salt cucumber brine. The PCR amplicons comprising a portion of the 16S rRNA gene of the isolated colonies were directly sequenced and the untrimmed whole sequencing results of the unknown strains were aligned with the type strains using BLAST of NCBI. Then Sequence Aligner and Sequence Match of RDP confirmed the outcome. The identified isolates were eight species and belong to three genuses: Clostridium, Lactobacillus, and Bacillus. The RFLP pattern of the 16S rRNA gene of isolates verified the identified species. From now on the complex spoiling process of law salt fermented cucumber could be analyzed using the isolated species individually or with certain combinations.

  • 사람의 장내에 서식하는 장내세균은 사람의 건강을 지켜주는 유용균과 나쁜 영향을 주는 유해균이 있으며 이들 양자의 균형에 의하여 건강상태가 조절되고 있다. 사람의 건강에 대단히 중요한 역할을 하는 유용균의 대표적인 것이 Bifidobacterium이다. 이 Bifidobacterium은 장내에서 lactic acid 및 acetic acid를 생산하여 장내의 pH를 산성으로 유지시키고 부패균의 증식을 억제하는 역할을 하며 신체를 유해세균의 작용으로부터 방어하는 역할을 하고 있다. 현재 우리나라에서 사용하고 있는 Bifidobacterium은 대부분 외국에서 개발된 것으로 한국인에게 어느 정도 효과가 있는지에 대해서는 아직 정확한 보고가 없으며 한국인의 장내균총으로부터 유산균이 분리된다면 한국인에게 가장 잘 정착될 수 있는 균주일 것으로 생각된다.

The Study on Decomposition of Metal-working Fluids Against Microbes (미생물을 이용한 절삭유제의 부패성능 평가에 관한 연구)

  • Growth curves of microbes were examined to evaluate decomposition of metal-working fluids and decomposition properties of metal-working fluids were experimented using controled microbes such as E. coli and K. pnemoniae. According to the results of growth curve of microbes, the growth period depended on species of microbes, 2 h of E. coli, 3 h of K. pneumoniae, 4 h of P. aeruginosa and 3 h of P. oleovarans after incubation. The colony count of E. coli and K. pneumoniae controled to OD of 0.5 ranged from $4.4{\sim}10.0{\times}10^5CFU/mL$ and $1.8{\sim}9.5{\times}10^7CFU/mL$, respectively. The decomposition of metal-working fluids was excellently progressed in the range of pH 6~8 than below pH 4 and above pH 10. In the case of controled fluids to pH 6~8, the decomposition of the fluid containing ester group was more accelerated than that of the fluid containing ethylene glycol.

Microflora of Decayed Onion Bulbs and their Suppression by Fumigation Treatment (양파의 부패원인균 분포 및 훈증처리에 따른 억제효과)

  • Putrefactive microorganisms from infected onion bulbs collected at several areas were isolated and identified. The infecting microorganisms were mostly the species of Botrytis, Fusarium, Penicillium, Ericinia and Pseudomonas, among which the last was not pathogenic to onion bulbs. Fumigation of onion bulbs with Tetrachloroisophthalonitrile cut down decay rate by half of the control and the onion bulbs stored at 80% RH showed slow decay rate than those stored at 90% RH. The decay of onion bulbs was mainly caused by molds and the portions of them were 78-85% of Botrytis, 11-17% of Fusarium and 3-5% of Penicillium.
