Laboratory studies have revealed that naturally transformable bacteria develop competence under in situ conditions. Thus, the occurrence of competent bacteria in the environment can be considered as a certainty The persistence of free DNA in natural habitats is influenced by nucleolytic degradation and protection from degradation by adsorption to minerals. Although DNA seeded into natural environment was hydrolysed at substantial rates, but was still detectable at low levels after even several weeks. Compared to the number of laboratory based studies, only a few data have been published dealing with transformation of bacteria in the field. Recently, the potential transfer of recombinant DNA (rDNA) from deliberately or accidentally released bacteria to indigenous microbes has raised biosafety issues, since the persistence of rDNA becomes independent of the survival of its original host and leads to unpredictable, long-term ecological effects. The aim of the present review is to summarise recent literature on horizontal gene transfer (HGT) by transformation among bacteria in both soil and aquatic habitat and special emphasis is placed on recent reports which have addressed HGT among bacteria in the field. Pseudomonas aeruginosa and Vibrio vulnificus are Gram-negative human pathogens, which exert their virulence through quorum sensing (QS) regulation. The infection of these pathogens have been known to be mediated by biofilm formation in many cases and this study carried out the time-course analysis of biofilm formation depending on the QS regulation in P. aeruginosa and V. vulnificus. In P. aeruginosa, our results demonstrated that QS-deficient mutant better attached to surface at initial stage of biofilm formation, but poorly proceeded to the maturation of the biofilm structure, while wild type less attached at initial stage but developed highly structured biofilm at late stage. Because of this, the quantitative comparison of biofilm formation between wild type and the QS mutant showed the reversion; the QS mutant formed more biofilm until 10 h after inoculation than wild type, but wild type formed much more biofilm after 10 h than QS mutant. V. vulnificus has been reported to form more biofilm with the mutation on QS system. When we performed the same time-course analysis of the V. vulnificus biofilm formation, the reversion was not detected even with prolonged culture for 108 h and the QS mutant always forms more biofilm than wild type. These results indicate that the QS regulation negatively affects the attachment at early stage but positively facilitates the biofilm maturation at late stage in P. aeruginosa, while the QS regulation has a negative effect on the biofilm formation throughout the biofilm development in V. vulnificus. Based on our results, we suggest that the developmental stage of biofilm and bacterial species should be considered when the QS system is targeted for biofilm control.
This study was carried out to investigate the effect of vinegar on pH, total bacterial count, texture and color of rice with vinegar before and after storage at $4^{\circ}C$ and $25^{\circ}C$. This results obtained were summarized as follows : The pH value and total bacterial counts were not significant changed under different storage conditions with sushi vinegar, but control without sushi vinegar was increased the number of bacteria from storage 1 days. and When rice with vinegar was stored at low temperature ($4^{\circ}C$), the tendency of textural changes such as hardness, springiness, cohesiveness, chewiness, brittleness and adhesiveness was almost the same as those of $25^{\circ}C$. However, the rate of change was much higher in rice with vinegar when stored at $4^{\circ}C$ than at $25^{\circ}C$. and The Hunter's color parameters such as L, a, and b, of rice with vinegar were not changed much during storage at both $25^{\circ}C$. However, L value was slightly increased during storage, but a value was increased negatively without changing b value of rice with vinegar during storage at $4^{\circ}C$.
Gill epithelia of normal rainbow trout fingerlings and abnormal ones suffering bacterial gill disease by experimental infection were examined by transmitting electron microscopy (TEM) and scanning electron microscopy (SEM). TEM observations revealed that Flavobacterium branchiophila consisted of slender rods measuring 0.5 by 5 to $8{\mu}m$, and they had which were long, thin, flexible filaments measuring approximately 4 nm by $1{\mu}m$, and packed together to organize into bundles. Morphological alterations of the diseased epithelia started at hypertrophy of the lamellar epithelium. F branchiophila attached to the gill surface of infected fish through pili with a regular distance, and did not invade into gill tissue. In SEM observations, normal surface ultrastructure of epithelial cell in the outermost layer were characterized by a typical labyrinth-like structure branching and anastomosing microridges on the cell surface. Hyperplastic lesions in experimentally infected gill were most serious at near the tips. Each filament exhibited a club-like, and fusion between the filaments was sometimes observed at their tips. On the surface of gill filaments, thread-like bacterial cells attached and were entangled. The bacterial cells almost covered the surface. After immersion in 5 % NaCl, the cell of F branchiophila, however, appeared to be indeterminate shape.
During the full-scale water treatment operation at "G" Water Treatment Plant in Seoul, we investigated changes in pore volume distribution and specific surface area of GAC with time. The pore volume of the used GAC decreased to the level below 0.6 cc/g while that of the brand new GAC was ranged 0.7~0.9 cc/g. The specific surface area of GAC pores changed within the range between $1100{\sim}1200m^2/g$ and $700{\sim}800m^2/g$. Bacteria attached to the surface of GAC shows a gradual increase ($0.4{\time}10^6{\sim}8.5{\time}10^6CFU/g$) under scanninig electron microscope (SEM). TOC removal was enhanced due to growth of the attached bacteria on GAC. It was found that TOC removal was closely related with physical parameters (pore volume, specific surface area) linearly under the investigated conditions. The used GAC need to be exchanged into new one or re-generated to remove organic matters (TOC) effectively from the finished drinking water.
The microbial immobilization media with curdlan and activated carbon which has great immobilization capacity has been developed. Characteristics of porosity and mechanical strength of this support media are dependent on manufacturing method. The support media showed the best cell immobilization performance when the ratio of curdlan and activated carbon was 30 g/L to 6 g/L in this study. The immobilization of iron-oxidizing bacteria on the supporting particles was photographed with a scanning electron microscope. Since cell concentration on the surface of supporting particle increased with the reaction time, the iron oxidation rate also increased.
For this study copper ions-containing bioactive glass nanoparticles commonly used in mineral trioxide aggregate (MTA) was developed to improve the mechanical properties and promote antibacterial effect of MTA with the original material. The mechanical properties and antibacterial activity of Cu-BGn incorporating varying amounts cooper incorporated bioactive glass nano particles(BGn) 0.5,1.0,2.0 and 4.0 wt% in MTA were characterized composition of the resulting were investigated. The compressive strength was calculated by weighing specimens with a diameter of 4 mm and a thickness of 6 mm according to ISO 6876 (2012). The antimicrobial effect was evaluated using two strains of S. mutans and E. faecalis. The mechanical properties of the test results was Cu-BGn increased no statistically significant difference was observed (p>0.05). Adhesion experiment results S. mutans in contrast to the control group Ortho MTA, 4.0 wt% of Cu-BGn added experimental group showed a significant difference was observed (p<0.05). Also, E. faecalis statistical analysis indicated a significant difference for antibacterial agents between control and Cu-BGn containing(p<0.05). It seems that this Cu-BGn proved that even a antibacterial effect was demonstrated. Therefore, it was suggest that it is necessary for in-depth research into various environments that can reproduce the oral environment.

In this study, the SELEX method was used to screen for and select aptamers with high selectivity and affinity for Streptococcus mutans, which is the causative agent of dental caries. Aptamers are single stranded oligonucleotides of DNA or RNA with high selectivity and affinity for target molecules because of their specific three-dimensional structures that are used to collect biometric information. When compared to antibodies in vitro, aptamers are more advantageous because of their ease of use in the screening process, low cost, chemical stability, and lack of restrictions on the target molecules. We sorted DNA aptamers, which contain 44 bp or 38 bp primer sequences in 5' and 3', 39 bp random sequences in the middle.We then analyzed the character and affinity to S. mutans. Aptamers of specific oligonucleotides that combine with S. mutans were selected and these results are selectively fused to S. mutans. The results confirmed that DNA aptamers can be used for rapid diagnosis and treatment of dental caries caused by bacteria of the oral cavity, namely S. mutans.
The purpose of the study was to gather basic bacteriological data regarding the quality of Korean surface waters. Total coliforms, fecal coliforms and fecal Streptococci survey in the water of Lake Eui-Am were undertaken by the membrance filter technique. A total of 37 samples were collected in August 14, 1970. Total coliforms were detected in all samples, fecal coliforms and fecal Streptococci were detected in 68% and 82% of samples, respectively. Bacterial densities of the lake water were varied by station and depth. The numbers of bacteria per 100ml of the lake water were; 8-225(65.3 average) for the total coliforms, 0-112(26.2 average) for the fecal coliforms, 0-77(25.8 average) for the fecal Streptococci, and 8-302 (91.1 average) for the total number of total coliforms and fecal Streptococci. These results suggest that the Eui-Am lake water is only lightly polluted and indicate that the lake water, properly maintained, is a source of raw water of good bacteriological quality. Three forms of fecal pollution bacteria tend to increase with depth. This is believed that the suspended matter with conglomerated bacteria plays an important role in regulating of bacterial densities in summer season.
