Characteristics of Bacterial Community for Biological Activated Carbon(BAC) by Culturable and Unculturable Methods. (배양적 및 비배양적 방법에 의한 생물활성탄 부착세균 군집 특성)

  • The Biological Activated Carbon (BAC) process in the water treatments represents a kind of biofiltration process which capabilities of bacteria to remove organic matters are maximized. It enables to eliminate organic matters and effectively reduce microbial regrowth potentials. As attached bacteria employ natural organic matter as a substrate, they are significantly dependent on indigenous microorganisms. In this study, characteristics of bacterial community by culturable and unculturable Methods have been conducted in a pilot plant using SAC in water treatment process at the downstream of the Nakdong River. Based on the results, HPC and bacterial- production for coal-based activated carbon material were $1.20{\sim}56.2{\times}l0^7$ cfu/g and $1.2{\sim}3.7\;mgC/m^{3}h$, respectively, in the SAC process. The highest level of attached bacteria biomass and organic carbon removal efficiency was found in the coal-based activated carbon. The genera Pseudomonas, Flavobacterium, Alcaligenes, Acilzetobacter, and Spingomonas were identified for each activated carbon material. Pseudomonas vesicularis was the dominant species in the coconut- and coal-based materials, where as Pseudomonas cepacia was the dominant species in the wood-based material. The Scanning Electron Microscope (SEM) observation of the activated carbon surface also found the widespread distribution of rod form and coccus. The community of attached bacteria was investigated by performing Fluorescent in situ hybridization (FISH) analysis. a group was dominant in coal, wood and coccunt-based materials, ${\alpha},\;{\beta}\;and\;{\gamma}$ group ranged from 27.0 ${\sim}$ 43.0%, 7.1 ${\sim}$ 22.0%, 11.3 ${\sim}$ 28.6%, respectively. These results suggest that a group bacterial community appears to be regulated removal efficiency of organic material in water treatment process.

Adhesion and Biofilm Formation Abilities of Bacteria Isolated from Dental Unit Waterlines (치과용 유니트 수관에서 분리한 세균의 부착 및 바이오필름 형성 능력)

  • The purpose of our study is to compare the adhesion and biofilm formation abilities of isolates from water discharged from dental unit waterlines (DUWLs). Bacteria were isolated from a total of 15 DUWLs. Twelve isolates were selected for the experiment. To confirm the adhesion ability of the isolates, each isolate was attached to a glass coverslip using a 12-well plate. Plates were incubated at $26^{\circ}C$ for 7 days, and the degree of adhesion of each isolate was scored. To verify the biofilm formation ability of each isolate, biofilms were allowed to form on a 96-well polystyrene flat-bottom microtiter plate. The biofilm accumulations of all isolates formed at $26^{\circ}C$ for 7 days were identified and compared. A total of 56 strains were isolated from 15 water samples including 12 genera and 31 species. Of the 56 isolates, 12 isolates were selected according to the genus and used in the experiment. Sphingomonas echinoides, Methylobacterium aquaticum, and Cupriavidus pauculus had the highest adhesion ability scores of +3 among 12 isolates. Among these three isolates, the biofilm accumulation of C. pauculus was the highest and that of S. echinoides was the third-most abundant. The lowest biofilm accumulations were identified in Microbacterium testaceum and M. aquaticum. Most isolates with high adhesion ability also exhibited high biofilm formation ability. Analysis of adhesion and biofilm formation of the isolates from DUWLs can provide useful information to understand the mechanism of DUWL biofilm formation and development.

Quantitative analysis of mutans streptococci adhesion to various orthodontic bracket materials in vivo (다양한 교정용 브라켓 원재료에 부착하는 mutans streptococci 양의 비교분석)

  • Objective: To estimate the effects of bracket material type on enamel decalcification during orthodontic treatment, this study analyzed the adhesion level of mutans streptococci (MS) to orthodontic bracket materials in vivo. Methods: Three different types of orthodontic bracket materials were used: stainless steel, monocrystalline sapphire, and polycrystalline alumina. A balanced complete block design was used to exclude the effect of positional variation of bracket materials in the oral cavity. Three types of plastic individual trays were made and one subject placed the tray in the mouth for 12 hours. Then, the attached bacteria were isolated and incubated on a mitis salivarius media containing bacitracin for 48 hours. Finally, the number of colony forming units of MS was counted. The experiments were independently performed 5 times with each of the 3 trays, resulting in a total of 15 times. Mixed model ANOVA was used to compare the adhesion amount of MS. Results: There was no difference in colony forming units among the bracket materials irrespective of jaw and tooth position. Conclusions: This study suggested that the result of quantitative analysis of MS adhesion to various orthodontic bracket materials in vivo may differ from that of the condition in vitro.

Diversity of Myxobacteria in Soil Samples from Asansi and Uponeup in Korea (아산시와 우포늪 토양의 점액세균 다양성)

  • Diversity of myxobacteria in five soil samples from Asansi and Uponeup in Korea was explored by means of polymerase chain reaction (PCR) using primers that specifically bind 16S rDNA of myxobacteria. DNA sequence analysis of 76 PCR fragments containing myxobacterial 16S rDNA revealed five putative novel myxobacterial genera whose 16S rDNA sequences shared <95% sequence identity with those of the type strains. This finding indicates the presence of many uncultured and unidentified myxobacterial species in Korean soil.

The Characteristics of Microbial Community for Biological Activated Carbon in Water Treatment Plant (생물활성탄 공정에서 활성탄 재질에 따른 부착미생물 군집특성)

  • The purpose of this research is to survey characteristics of microbial community and the removal efficiency of organic materials for biological activated carbon in water treatment plant. Coal based activated carbon retained more attached bacterial biomass on the surface of the activated carbon than the other activated carbon with operating time and materials. The heterotrophic plate count(HPC), eubacteria(EUB) and 4,6-diamidino-2-phenylindole(DAPI) counts were ranged from $0.95{\times}10^7$ to $52.4{\times}10^7$ CFU/g, from $3.8{\times}10^8$ to $134.2{\times}10^8$ cells/g and from $7.0{\times}10^8$ to $250.2{\times}10^8$ cells/g, respectively. The biomass of EUB and DAPI appeared to be much more $10^2$ than HPC, which were increasing in bed volume of 20,000 at the stage of steady-state. The change of microbial community by analyzing fluorescent in situ hybridization(FISH) method with rRNA-targeted oligonucleotide probes, the dominant group was $\alpha$-proteobacteria($\alpha$ group) and high G+C content bacteria(HGC) the lowest distributing rate before reaching the bed volume of 20,000. After reaching the bed volume of 20,000, $\alpha$ group and other groups of bacteria became decreased, on the other hand, the proportion of both $\beta$-proteobacteria($\beta$ group) and $\gamma$-proteobacteri($\gamma$ group) were increasing. Coconut and wood based activated carbons had similar trend with coal based activated carbon, but the rate of $\alpha$ group on coal based activated carbon had gradually increased. Bacterial production with the operating period appeared highest in coal based activated carbon at the range of $1.2{\sim}3.4\;mg-C/m^3{\cdot}h$ while the coconut and wood based activated carbon were ranged from 1.1 to 2.6 $mg-C/m^3{\cdot}h$ and from 0.7 to 3.5 $mg-C/m^3{\cdot}h$ respectively. The removal efficiency of assimilable organic carbon(AOC) showed to be highly correlated with bacterial production. The correlation coefficient between removal efficiency of AOC and bacterial production were 0.679 at wood based activated carbon, 0.291 at coconut based activated carbon and 0.762 at coal based activated carbon, respectively.

  • Acinetobacter sp.가 외부환경보다 많이 발견되었다는 것으로 보아 이 균이 섬유류 유물의 훼손에 영향을 많이 미칠 것으로 보인다 Co교nebacterium sp., Bacillus sp.등은 외부환경이나 전시실 등에서 전혀 발견되지 않았으나 유물보관함에서 발견되었다. 때문에 이 세균들은 외부환경으로부터 유입된 균이 아니라고 추정되며, 이 균들이 섬유류 유물에 어떠한 영향을 미치는지 연구해 볼 필요성이 있다. 섬유의 부착세균으로는 공중 부유에서 대다수로 검출되었던 Acinetobacter spp., Pseudomonas spp., Neisseria spp.,등으로 공기를 통해 섬유류 유물이 오염된다는 것을 확인할 수 있었다.

Inhibition of Plaque Formation on the Titanium Surface by Anti-bacterial Varnish (항균제 처리한 titanium 표면의 치태형성 억제효과)

  • 매식된 인공치아의 성공을 위해서는 적절한 교합과 수동적 적합성을 갖는 보철물의 제작과 구강내 노출 직후부터의 세균성 치태조절이 요구된다. 본 연구는 전처리(passivation과 tridodecyl - methyl - ammonium chloride(TDMAC) 처리)가 다른 타이타늄 표 면에 chlorhexidine varnish와 테트라사이클린 을 도포시 약제의 방출역학을 알아보고 구강내 치태형성의 억제정도를 평가하기 위하여 시행 되었다. 이를 위해 방출용액으로 인산완충액 성분의 인조타액을 1일${\sim}$1개월간 매일 교환하여 약제농도를 측정하고 타이타늄 박막에 잔류한 약제 활성을 측정하였으며 항균제 도포한 타이타늄 원판을 부착한 장치를 구강내 위치시킨 1일${\sim}$3주 후 원판을 제거하여 주사전자현미경으로 세균 부착상을 관찰하였다. 테트라사이클린은 TDMAC 처리된 표면에서 $10{\sim}18$일까지 유효농도로 방출되었고 표면의 유효 항균 활성은 $3{\sim}4$주간 유지되었으며, chlorhexidine varnish 도포 시에는 TDMAC 전처리시 초기에 $3{\sim}7$일 간 증가한 유효 항균 활성을 방출하여 매식지대치 등에 이러한 항균제도포 시 매식치 주위환경에 항균활성 공급원으로 작용할 수 있음을 보였다. 주사현미경적 관찰시 모든 타이타늄 표면에서 구강내 위치 30분 후에는 세균이 부착되어 있지 않고 타액 단백질 성분에서 유래한 것으로 보이는 피막물질이 표면을 부분 또는 전면에 걸쳐 덮고 있었다. 구강내 노출 2시간 후 항균제 미도포 표본들에는 약간의 구균이 단층으로, $1{\sim}3$일 후에는 부분적으로 두꺼운 세균층을 형성하였고 7일 후에는 표면전체에 걸쳐 세균층이 덮여있었으며 주로 구균과 약간의 간균이 주종을 이루었다. 항균제 도포시 구강내 노출 1주일 이전까지는 미도포군에 비해 치태형성이 지연되는 경향을 보였지만 2주 이후에는 세균 수나 치태형성 양상이 유사하였다. 이 연구로부터 항균제 도포시 1주일 이전의 초기 치태형성을 감소시킬 수 있음을 알 수 있었으며 이러한 연구결과는 타이타늄 임프란트 지대치 표면에 항균제의 도포가 임상적으로 유용할 수 있음을 시사하였다.

Hairs as Physical Barrier against Adhesion of Xanthomonas axonopodis pv. glycines on Soybean Leaf (콩 잎 엽모에 의한 불마름병균 부착 저해)

  • Bacterial pustule of soybean is caused by Xanthomonas axonopodis pv. glycines, one of the most important diseases in soybean. The symptom of bacterial pustule is mainly distributed around leaf veins. However, the reason has not been known. In order to determine pathosystem of bacterial pustule in leaf, soybean leaves were collected and observed using scanning electron microscopy (SEM) and light microscopy. Many hairs were observed at abaxial sides of the leaf, few hairs were observed at tissue around the leaf veins. In addition, unidentified bacterial cells and dusts at the no hair part near veins were observed. In the inoculation assays, the cells of X. axonopodis pv. glycines were observed near leaf veins. The imprint of underside of soybean leaves inoculated with X.axonopodis pv. glycines on PDA showed that the growth of bacteria around veins was observed but no bacterial growth at the part with leaf hairs. Our data demonstrated that soybean leaf hairs play an important role as a physical barrier for structural resistance of soybean against bacterial pustule pathogen.

Attachment of Pasteuria penetrans Endospores to Meloidogyne spp. Juveniles Affected by Temperatures and the Nematode species (선충기생세균(Pasteuria penetrans) 내생포자의 뿌리혹선충(Meloidogyne spp.) 유충 부착에 대한 온도와 선충종의 영향)

  • A greenhouse soil infested with an obligate nematode parasitic bacterium, Pasteuria penetrans, was used to test the effect of temperatures on the endospore attachment to root-knot nematode, Meloidogyne arenaria, juveniles (J2). Freshly hatched J2s were inoculated to the soil in petri dish and incubated under different temperatures of $20^{\circ}C$, $25^{\circ}C$, $30^{\circ}C$, and $35^{\circ}C$ for 7 days. The endospore attachment rates were 100% in all the temperatures, while the number of endospores attached per J2 was highest in $25^{\circ}C$ with 28.3 endospores/J2 followed by 20.2, 18.6, and 13.6 in $30^{\circ}C$, $20^{\circ}C$, and $35^{\circ}C$, respectively. When the soil was pre-treated under different temperatures before the J2 inoculation, the endospore attachment rates significantly decreased from 60% in room temperature to $25^{\circ}C$, 31.7, 8.3, 5.0, and 0% after the soil incubation in $-30^{\circ}C$, $4^{\circ}C$, $40^{\circ}C$, $50^{\circ}C$, and $100^{\circ}C$ for 10 days, respectively. The endospore numbers attached per J2 were 3.5, 4.3, 1, 1, and 0 when the soil was pre-treated in $-30^{\circ}C$, $4^{\circ}C$, $40^{\circ}C$, $50^{\circ}C$, and $100^{\circ}C$, respectively, which were lower than 5.3/J2 of room temperature treated soil. The P. penetrans isolate in the soil showed nematode species-specific endospore attachment characteristics with 100% attachment rate only on M. arenaria J2s while the rates were 0% on M. hapla and M. incognita J2s.
