A Study on the Effects of Retardation with Pb in the Biofilter (생물여과층에 의한 납의 이동억제효과에 관한 연구)

  • The purpose of this study was to show the application of the Biofilter for treatment of the soil contaminated by lead and to investigates the effect of the biofilter on the retardation of lead with pilotplants that were simulated with different media and the number of bed. and to testify the inoculation by seeding microbes. The ratio of the degradation of soil contaminant was verified as CODcr/TOC in order to find a variation of the stabilization index in soil. The Biofiltration was one of biological processing methods for treatment. The contaminants were transported through the biofilter that was filled with the media. The surface of media formed biofilm which was surrounded by microbes and through its boundary, some materials were exchanged and migrated into the cell of microbes in an orderly manner. To investigate the effect of the Biofiltration, contaminated soil with lead nitrate of 1000mg/kg in dry was made artificially. The tests were simulated such as compost, Bioceramic and compost with bioceramic by 7:3 in weight. The bed consisted of three layers in order to find effect of the number of bed. Aspergillus niger was used as a biosorbent could probe the effect on the retardation of lead.

Temperature Sensitivity of Sigma Background Is Suppressed by the Disruption of ScKNS1 in Saccharomyces cerevisiae (ScKns1 결손에 의한 Saccharomyces cerevisiae ${\Sigma}1278b$ 균주의 온도 민감성 억제 효과)

  • The Saccharomyces cerevisiae S288c strain does not show haploid and diploid filamentous growth, and biofilm formation, because it has a flo8 nonsense mutation unlike ${\Sigma}1278b$ strain which has a FLO8 gene. During the heat stress experiments to investigate the role of ScKns1, LAMMER kinase in S. cerevisiae, we found that ${\Sigma}1278b$ strain revealed heat sensitivity at $37^{\circ}C$, a mild heat stress in contrast to S288c strain. We also found that the disruption of ScKns1 and the addition of sorbitol suppress heat sensitivity of ${\Sigma}1278b$ strain. These results suggest the possibility that Flo8 and ScKns1 may interact to transducer a signal for regulating heat stress through a novel signaling pathway.

Serodiagnosis of Typhoid Fever by Enzyme-Linked Immunosorbent Assay(ELISA) (효소면역측정법에 의한 장티푸스의 혈청학적 진단)

  • Serum samples from 51 patients with clinically suspected typhoid fever were tested for immunoglobulin G (IgG), IgM and IgA antibodies against the whole bacteria antigen of Salmonella typhi by an enzyme-linked immunosorbent assay. The levels of IgG and IgA antibody to-whole bacteria antigen were higher in the culture-proven patients than in controls. The levels of IgM antibody to- whole bacteria antigen showed better discrimination between culture negative patients and controls than those of IgG or IgA antibody to-whole bacteria antigen. The enzyme-linked immunosorbent assay was much more sensitive than the Widal test. It would be a useful tool for the diagnosis of typhoid fever with a single serum sample.

Isolation of Dispersed Mutants from Wild Myxobacteria. (분산 돌연변이 점액세균의 분리)

  • Clumping of cells is one of the major obstacles to culture wild myxobacterial strains in liquid media. In an effort to solve this problem, we tried a method isolating spontaneous mutants that grow dispersed in liquid media from a wild myxobacterial strain. Myxococcus stipitatus KYC1001, a newly isolated strain from Gyearyongsan National Park in Korea, clumps and sticks to the surface of culture vessels as other wild myxobacteria behave in liquid media. Taking an advantage of the characteristics that dispersed mutant cells would grow dispersed while most other wild type cells would clump and stick to the surface of culture vessels, spontaneous dispersed mutants were enriched by repeated subculturing of culture supernatant. A resultant mutant, KYC2001, did not form any clumps nor stick to the surface of culture flasks, but grew completely dispersed in liquid. Meanwhile, three other spontaneous mutants, KYC2002, KYC2003, and KYC2004, shelved partially dispersed phenotype. A major portion of the cells grew dispersed in liquid but they still formed some clumps.

An Experimental Study on the Treatment of Phenolic Wastewater Using Rotating Biological Contactors (회전원판법(RBC)에 의한 페놀성 폐수의 처리에 관한 실험적 연구)

  • This study is an experimental research on the treatment of phenolic wastes by Rotating Biological Contactors(RBC). The objective of this study is to determine the optimum range of influent phenol concentration and organic loading rate. Organic removal rates were analyzed with increasing organic loading and influent phenol concentration, together with the observation of microorganism. Biomass, SCOD, and phenol concentration were measured under the steady state after a step change of influent phenol concentration. As the result, at the phenol concentration less then 98.8 mg/L there were no evidence of substrate inhibition. As the results, organic removal rates in each stage at various organic loading, were decreased with increasing phenol concentration. First order kinetic was observed on the removal of SCOD for which phenol concentration is within the range of substrate inhibition. And also, microorganisms were changed with influent phenol concentration. Namely, at low influent phenol concentration, thin biofilm with filamentous growth was produced. To the contrary, thick biofilm with nonfilamentous growth was produced at high influent phenol concentration.

The Inactivation Effects of UV Light on Bacteriophage f2 (박테리오파지 f2에 대한 자외광선의 살균효과)

  • The effects of ultraviolet light on bacteriophage f2 were investigated to determine the inactivation kinetics and its mechanism. The 260nm light showed a little higher inactivation rate than the one of 300 nm. In this work, our main concern was whether structural and/or conformational changes in the protein capsid could occur by UV irradiation. The inactivation for the first 20 minutes irradiation was rapid with a loss of about 4 logs and followed by a slower rate during the next 40 minutes with no survival noted in the samples irradiated for 90 minutes or longer. The structural change of the protein capsid was examined by optical spectroscopic techniques and electron microscopy. The absorption spectra of the UV irradiated phages showed no detectable differences in terms of the spectral shape and intensity from the control phage. However, the fluorescence emission spectroscopic data, i.e. 1) fluorescence quenching of tryptophan residues upon irradiation of 300 nm light, 2) enhancement of fluorescence emission of ANS (8-aniline-1-naphthalene sulfonate) bound to the intact phages compared to the one in the UV-treated phages, and 3) decrease of energy transfer efficiency from tryptophan to ANS in the UV-treated samples, presented remarkable differences between the intact and UV-treated phages. Such a structural alteration was also observed by electron microscopy The UV-treated phages appeared to be broken and empty capsids. Therefore, the inactivation of the bacteriophage f2 by UV irradiation is thought to be attributed to the structural change in the protein capsid as well as damage in the viral RNA by UV irradiation.

The Bacterial Community Structure in Cheonho Reservoir Dominated by Cyanobacteria (봄철 Cyanobacteria 가 우점한 천호지에서 세균군집구조의 변화)

  • The composition of bacterial communities was detected in surface water of Cheonho Reservoir dominated by cyanobacteria, using fluorescent in situ hybridization (FISH) method. Total bacterial numbers were very high ranging from 0.6~$1.3{\times}10^7 \cells{\cdot}ml^-1$, whereas the ratio of Eubacteria to total bacteria was 29.8~45.8%, which was lower than that in other freshwater ecosystems. On average only 2.1% of DAPI-stained bacteria were detected by FISH with probes for $\alpha$, $\beta$, and $\gamma$-groups, respectively. Unknown eubacteria which was not bound to any probes except EUB 338, was relatively high. On the other hand, the Cytophaga-Flavobacterium group increased following the change of dominant species from Anabaena sp. to Microcystis sp. This result showed that bacterial communities could be affected by phytoplanktons, especially cyanobacteria.

Increased Expression of Adhesion Molecules on Human Umbilical Vein Endothelial Cells by Orientia tsutsugamushi Infection (Orientia tsutsugamushi 감염에 의한 사람 제대정맥 내피세포에서 부착분자 발현의 변화)

  • Scrub typhus is caused by Orientia tsutsugamushi characterized by fever, headache, lymphadenopathy and eschar formation. Infiltration of inflammatory cells around blood vessels and within the affected organs isS known to be pathologic hallmark of the scrub typhus. Recently, expression of adhesion molecules on vascular endothelial cells was implicated as an important pathogenic mechanism in rickettsial disease. This study was performed to examine the expression of adhesion molecules and to investigate its role in the pathogenesis of O. tsutsugamushi infection. The expression of adhesion molecules on human umbilical vein endothelial cells (HUVEC) was measured by flow cytometry and indirect immunofluorescence. Expression of E-selectin, ICAM-1 and VCAM-1 was significantly increased 4 hours after the infection and persisted at least for 24 hours. Expression of those molecules was not induced by killed O. tsutsugamushi. Adhesion of polymorphonuclear cells and mononuclear cells to HUVEC was increased after the infection with O. tsutsugamushi. In conclusion, adhesion molecules are expressed on HUVEC during the infection of live O. tsutsugamushi and those molecules can contribute to the infiltration of inflammatory cells during the infection.

Secretion of Cytokine Stimulating Intercellular Adhesion Molecule-l Expression from THP-l Cells Infected with Human Cytomegalovirus (HCMV에 감염된 TBP-1 세포에서 세포간 부착분자-1 발현 촉진물질의 방출)

  • Human cytomegalovirus (HCMV) stimulates the expression of intercellular adhesion molecule (ICAM-l) on the surface of monocytic THP-1 cells. Stimulation of ICAM-l did not require HCMV gene expression since UV-inactivated HCMV (UV-HCMV) was able to induce ICAM-l expression. ICAM-l expression was also stimulated in uninfected THP-l cells which were fed with culture supernatant of HCMV-infected THP-l cells. Co-culture experiment using trans-well insert supported that HCMV-infected THP-l cells secreted some cytokine(s) stimulating ICAM-l expression. The stimulation of ICAM-l by HCMV-infected cell culture supernatant appears to involve $NF-{\kappa}B$ pathway. Culture supernatant from THP-l cells infected with UV-HCMV, whose gene expression was abrogated, failed to stimulate ICAM-l expression on naive THP-l cells. Thus, HCMV gene expression seems to be required in secretion of cytokine(s) stimulating ICAM-l expression.

Relationship between Germ Tube Formation, Adherence to Human Buccal Epithelial Cells and Virulence of Candida albicans (Candida albicans의 상피세포에 대한 부착능과 병원성과의 상관관계에 관한 연구)

  • This study investigated whether a correlation exists between environmental physical and biochemical factors and adherence of Candida albicans to human buccal epithelial cells by using normal and UV-irradiated strains. The results were as follows: 1. The percentage of germ tube forming activities of normal Candida albicans was 91.5% and UV-irradiated Candida albicans was 15.0%. The $LD_{50}$ of normal strains in mice were $1.0{\times}10\;cells/ml$, but could not be observed in the UV-irradiated strains even with $1.0{\times}10\;cells/ml$. It demonstrated that the virulence is decreased in the UV-irradiated strain. 2. The adherence of normal Candida albicans to human buccal epithelial cells($166{\pm}29{\sim}207{\pm}17\;cells$/100 epithelial cells) was significantly greater than UV-irradiated Candida albicans($99{\pm}21{\sim}131{\pm}25\;cells$/100 epithelial cells). 3. Candida albicans cultured at $37^{\circ}C$ adhered to buccal epithelial cells($166{\pm}16{\sim}207{\pm}17\;cells$/100 epithelial cells) in greater numbers than cultured at $25^{\circ}C$($80{\pm}15{\sim}143{\pm}22\;cells$/100 epithelial cells). 4. On comparison of the adherence of viable and nonviable(heat-killed) Candida albicans to human buccal epithelial cells, the nonviable Candida albicans demonstrated poorer adherence than viable Candida albicans. 5. Adherence in vitro of Candida albicans to human epithelial cells appeared to be effected by the pH. The adherence ability was maximum increased at pH 7.0($187{\pm}22\;cells$/100 epithelial cells) other than experimental pH. 6. The adherence was proportional to the incubation time and the Candida cell concentration in the suspension. 7. A strong correlation was shown between germ tube forming activity and increased adherence of Candida albicans to human epithelial cells, indicating that germ tube forming activity were responsible for candidal virulence.
