  • The aims of this study was to elucidate the correlation between cotyledon numbers and seedling growth and some other traits of Pinus koraiensis. The results are summurized as follows; 1. A highly significant correlations between numbers of coty ledons and juvenile needles was found but no significances between cotyledon numbers and juvenile needle length, width and stomata row numbers. 2. The accessory resin canals did not begin to appear before June but began to appear only small numbers of seedling after July. It was observed that the number of accessory resin canals of juvenile leaves at various growing stages were not related with the number of cotyledons. 3. In the case of adult leaves, there was a significant difference in the number of leaves and the number of cotyledons, but no significant difference between the number of cotyledons and needle length, needle width and stomata row. 4. There were significant differences in the number of cotyledons and top height growth, and root caliper growth among individuals and due to ages. 5. Seedlings with more than 12 cotyledons showed better growth than those with 9 to 11 cotyledons.

  • In order to observe the degree and response of drought-resistance and its physiological mechanism in barley and wheat seedling stage, 5 species (16 cultivars) were tested for the changes of nitrate reductase and protease activity and the accumulation of free proline, by being subjected to water stress by withholding watering for 8 days at 10 days (at the 3rd leaf stage) after emergence and by imposing water stress to the excised first leaf by polyethyleneglycol solution (osmotic potential, -20 bars) for 48 hours. The average rate of decrease of all cultivars was 42% in nitrate reductase activity and 73% in protease activity. But proline content in water stress was increased 10 folds more than that of control. The decrease4 rate of nitrate reductase activity in 5 species was in the order of wheat < rye < covered barley < naked barley < two-row barley: wheat being the lowest. The decreased rate of protease activity in 5 species was in the order of wheat > rye > two-row barley > covered barley > naked barley: wheat being the" heighest. The accumulated amount of free proline in 5 species by water stress was in the order of wheat > covered barley > rye > naked barley > two-row barley. And the increased ratio (folds) of free proline of water stress to control was in the order of rye(13) > wheat. covered barley(11) > naked barley(99) > two-row barley(7): rye being the highest. In terms of the enzymatic activity and the physiotically adaptive metabolism during the processing leading to drought-resistance, the degree of drought-resistance of 5 species to water stress at seedling stage was shown to be in the order of wheat > rye > covered barley > naked barley > two-row barley.

  • Variations in photosynthetic capacities of leaves differing in thickness were explained on the basis of relationships between gas exchange and internal leaf structure. The relative importance of gas diffusion and of biochemical processes as limiting for leaf photosynthesis was also determined. Mesophyll cell surface was considered to be the limiting internal site for gas diffusion. and cell volume to be indicative of the sink capacity for CO$_2$ fixation. Increases in cell surface area were assumed to reduce proportionately mesophyll resistance to the liquid phase diffusion of CO$_2$. Increased cell volume was thought to account for a proportional increase in reaction rates for carboxylation, oxygenation. and dark respiration. This assumption was tested using chamber-grown Glycine max (L.) Merr. cv. Amsoy plants. Plants were grown under 200, 400, and 600 ${\mu}$mol photons m$\^$-2/ s$\^$-1/ of PAR to induce development of various leaf thickness. Photosynthetic CO$_2$ uptake rates were measured on the 3rd and 4th trifoliolate leaves under 1000 ${\mu}$mol photons m$\^$-2/ s$\^$-1/ of PAR and at the air temperature of 28 C. A pseudo -mechanistic photosynthesis model was modified to accommodate the concept of cell surface area as well as both cell volume and surface area. Both versions were used to simulate leaf photosynthesis. Computations based on volume and surface area showed slightly better agreement with experimental data than did those based on the surface area only. This implies that any single factor, whether it is photosynthetic model utilized in this study was suitable for relating leaf thickness to leaf productivity.

  • The bacterial isopentenyl transferase (ipt) gene involved in cytokinin biosynthesis was fused with 35S promoter of cauliflower mosaic virus (CaMV) and introduced into tobacco plants (Nicotiana tabacum L. cv. Samsun) via Agrobacterium-mediated transformation. As expected, ipt gene was constitutively expressed in all tissues of transgenic plants. Several primary transgenic plants were obtained that expressed different level of transcripts for ipt gene. Three of transgenic plants with different expression level of ipt gene were selected and selfed to obtain homozygous line for further analysis. A number of interesting phenotypic changes such as viviparous leaves, delayed senescence, larger axillary shoots, an abundance of tiny shoots at the apex and a release of lateral buds, were observed in transgenic plants. Chlorophyll content was 1.5- t.o 4-fold higher in transgenic plants as compared with non-transformed plants. These results indicate that the cytokinin synthesized in transgenic plants could improve forage crop yield by delay of leaf senescence and increase of leaf number.

  • Fifty three diseased root samples of Chinese cabbage and 22 soil samples were taken from six alpine areas in Kangwon province to examine the organisms causing root rot of Chinese cabbage. Among thirteen microorganisms detected from diseased roots samples, Pythium ultimum, Pythium echinocarpum and Rhizoctonia solani were detected frequently and were pathogenic to Chinese cabbage. Five to 100% of young Chinese cabbage plants grown in the sample soils taken from alpine cabbage fields were infected either alone or together with P. ultimum, P. echinocarpum and R. solani, depending upon the or gin of sample soils. When relative density of the three organsims in the sample soils was estimated based on the infection frequency on the plants grown in those sample soils, P. ultimum was prevalent only in Chahang-i-li area. P. echinocarpum was prevalent in Yongsan-il-li, Hoengwe-li, and Munmaek areas. R. solani was solely found in Maebongsan area where both P. ultimum and P. echinocarpum were not detected. In the three areas where both P. ultimum and P. echinocarpum were present, density of P. echinocarpum was generally greater than that of P. ultimum. Seedlings of thirty major Chinese cabbage varieties were inoculated independently with the three organisms in the greenhouse. Six varieties were resistant to P. ultimum. Three and two varieties were moderately resistant to P. ultimum and P. echinocarpum, respectively. All varieties were highly susceptible to R. solani.

  • This study was carried out to investigate the effect of sulfuric fertilizers on allylisothiocyanate(allylNCS) content of upland Wasabia japonica Matsum cultivated in heating condition in the winter season. The rhizome's growth characters following soil application of sulfur dust were not significantly different compared to control, though its soil acidity and available sulfur content was higher than other treatments. Total sulfur content of plant was increased by soil application as compared to foliar application and allylNCS content of rhizome increased up to 22.8% in the sulfur dust treatment. The result showed that soil application was effective for supplying $SO_4^{-2}$ in upland Wasabia japonica Matsum when cultivated in heating condition. In the analysis of correlation coefficients between growth and quality characteristics, allylNCS content of rhizome showed the significantly positive correlation with total sulfur content of shoot, rhizome and allylNCS content of shoot.

  • This study was conducted to investigate the occurrence and characterization of tobacco mosaic virus(TMV) in Chinese foxglove isolated from the field of the Chonbuk province(Jinan, Jangsu, Jeongeup). TMV was detected in all three regions and confirmed positive reaction by ELISA test. In the host range test, Chenopodium amaranticola, Nicotiana glutinosa, N. tabacum cv. 'Bright yellow', N. tabacum cv. 'KY57, Datura stramonium were locally infected with the virus. The virus produced mosaic symptom on inoculated leaves of N. tabacum cv. 'Samson'. However, Chenopodium quinoa, Glycine max, Raphanus sativus, Cucumis sativus, Cucurbita moschata, Brassica rape and Lycopersion esculentum did not show any symptoms. TMV particles were revealed as a stiff rod shape by transmission electron microscopic(TEM) and measured as 300 nm in length with 18 nm in diameter. Total RNA was extracted from showing symptom loaves infected with TMV and the reverse transcriptionpolymerase chain reaction (RTPCR) obtained 531 bp DNA product of RNA with specific primer used. The capsid protein of TMVRE showed higher amino acid sequence homology(97.7%) with TMVTo than with TMVP(72.2%). The capsid protein of TMV152 showed same amino acid sequence homology with TMVF. The result of comparison of nucleotides sequence homology between TMVRE strain and other TMV strain showed 94% homology with others except TMVP(67.3%) and TMV C(68.6%).

  • This study was carried to select artichoke (Cynara scolymus L.) varieties that can be cultivated in non-heated greenhouse during winter in Jeju Island in Korea. Artichoke varieties namely, 'Purple Romagna' (Neseed Co.), 'Imperial Star' (Keithley Williams Seed Co.), 'Green Globe' (Park Seed Co.), 'Emerald' (Ferry-morse Seed Co.), 'Italian' (Unknown), and 'Carciofo Di Romagna' (Lortolano), were raised in a plug tray with 72 cells for two months and transplanted in a non-heated greenhouse on October 5, 2004. The first harvesting of 'Emerald' and 'Imperial Star' were the earliest and April 26 and 27, 2005 respectively. Most of the varieties were harvested from early May to June 2005. The number of head per plant was the highest in 'Green Globe' with more than five heads and the lowest were recorded in 'Italian' with one head or less. The weight of heads per plant was more than 228 g in 'Purple Romagna', 'Imperial Star' and 'Green Globe'. The highest yield was in 'Green Globe' with 2,172 kg/10a, followed by 'Imperial Star' with 1,947 kg/10a. Accordingly, it is considered that 'Green Globe' and 'Imperial Star' are adequate for the high-yield cultivation in non-heated greenhouse in Jeju Island. They can be harvested from the April to June when cultivated from the early October of the previous year.

  • The evaluate the effect of plant spacing on cured leaf of burley tobacco, the row spacings divided to 90, 105, 120cm and hill spacings to 30, 35, 40cm within each row. Growth amount per plant increased with thinner row and wider hill spacing in the same planting density. Relative light intensity increased with thinner row spacing in cutters and leaf and showed the positive correlation with quality. When the planting density was equal, the wider hill spacings, the more effective in utillization of solar radiation. The more plants per l0a were, the greater yield was obtained, and in the case of 3,200 plants per 10a (the most dense planting plot) was 267kg. But, quality, total-alkaloid and total-nitrogen content decreased with dense planting. Value per 10a was highest in the plots of 90 $\times$ 40cm and $105{\times}40cm$. In conclusion the optimum density level was 2,400 to 2,700 plants per 10a and spacing of tobacco either in 105 $\times$ 35 cm or 105 $\times$ 40cm seems to be most appropriate.

  • This experiment was conducted to evaluated the effect of CM (plant growth regulator) treatment on flowering, pod maturing and root yield components of Astragalus membramaceus Bunge. Three dilution rates (60, 70 and 80 times) were sprayed at $3{\sim}4$ leave stage. As the results, flowering was delayed $25{\sim}35$ days than August 13 of control and higher dilution rates was more effective to delay flowering. At the havesting time, be compared with non-treatment, plant height reduced $7.6{\sim}18.9\;cm$ and number of matured pods were decreased $50.6{\sim}76.1$ by increasing dilution rates. However, weight of dry root per plant was increased to 29% and dry root yield per 10a was increased to 28% in dilution rates 70 compared with non-treatment.
