The peptidyl prolyl sis-trans isomerase (PPIase, EC 5.2.1.8) from bacillus stearothermophilus was extracted from the cells treated with by lysozyme. PPIase was purified from the cell extracts by heat treatment, ammonium sulfate precipitation, ion exchange chromatography and finally gel filtration, sodium dodecyl sulfate polyacrylamide gel electrophoresis(SDS-PAGE). The molecular weight of the purified PPIase was estimated as 18kDa by SDS-PAGE. The 39 amino acid residues from the N-terminus were determined by the protein sequencer. The enzyme showed the optimum pH at 8.0 and was stable at the range of pH 7.0∼8.0. The enzyme was considerably stable after heat treatment at 60$\^{C}$ for 30minutes, and the enzyme was quite stable up to 65$\^{C}$. The presence of the PPIase in the refolding solution accelerated the isomerization rate of the assay peptide. PPIase gene of Bacillus stearothermophilus was screened from a genomic library by plaque hybridization using the A-l primer as a probe. A PPIase positive plaque contained a 3.0kb insert of the chromosomal DNA. A 3.0kb fragment was subcloned into pUC18, resulting pPI-40. A DNA fragment encoding the N-terminal portion of the PPIase in pPI-40 was amplified by polymerase chain reaction(PCR) method using the A-1 and B-2 primers. The amplified fragment was cloned into the Sma I site of pUC18 and recombinant plasmid was designated as pSN-18. The nucleotide sequence of 167bp fragment was determined. The deduced amino acid sequence of PPIase was completely matched with the determined N-terminal amino acid sequence of PPIase B. stearothermophilus.

Focal epithelial hyperplasia(FEH) is Human papilloma virus - induced, localized proliferation of oral squamous epithelium. FEH usually occurs in the childhood, but occasionally affects the young and middle-aged adults. Sites of the greatest involvement include the labial, buccal and lingual mucosa, but lesions of gingiva or tongue have also been reported. This disease is typically characterized by multiple soft, non-tender flattened papules and plaques. Occasional lesions show a slight papillary surface change. Individual lesions are small, discrete and well demarcated. The histopathologic hallmark of FEH is acanthosis of the oral epithelium. Cells demonstrating viral cytopathic changes including koilocytes or mitosoid cells may be present. The 5-year-old female of this case visited Department of Pediatric Dentistry, College of Dentistry, Yonsei University with a chief complaint of exophytic lesions on gingiva. Sessile papillary papules were detected by clinical examination on buccal gingiva at the maxillary left and right second deciduous molars. The patient did not complain of pain by palpation. An excisional biopsy was carried out for a histological examination and acanthosis was observed. The lesions were diagnosed as FEH. FEH would regress spontaneously after several months or years. Conservative excision may be performed for diagnostic or esthetic purpose. The risk of recurrence after this therapy is minimal, and there is no malignant transformation.
To investigate the effect of resin cement, which had been known to increase the adhesive capacity of the cast gold inlay, on the gingival marginal fit and whether the tin-planting of the beveled area affects the marginal fit, Class II cast gold inlays were made on the 25 sound molars. Control group(ZPC goup) was cemented with the ZPC by conventional method. Experimental groups were cemented with the resin cement(Super-hond & $Panavia_{EX}$) and subdivided further by the existence or nonexistence of the tin-plating of the beveled area(ST & PT groups: with plating, SNT & PNT groups: without plating). So, each group was consisted of 5 teeth and the gingival margin of each specimen was mesiodistally sectioned by 3 times and the marginal and internal gap were evaluated by the Stereo Microscope (${\times}180$) and the Scanning Electron Micrascope(${\times}5,000$) was used for examining the adhesive relationship of the resin cement to the cavity wall and to the cast gold surface. The results were as follows : 1. Marginal gap was less than internal gap in all groups. 2. ZPC and SNT(bevel without tin-plating) groups showed the least gap and gap in PNT(bevel without tin-plating) group, ST(bevel with tin-plating) group, PT(bevel with tin-plating) group showed the greater value in order in evaluation of the both internal gap and marginal gap. 3. With the exception of the relationships between ZPC and SNT groups, ST and PNT groups, relationships between any other groups showed the statistical significance in the internal gap(p<0.05). 4. In the marginal gap, all relationships between groups showed the statistical significance (p<0.05) except the relationships between ZPC and SNT groups, ST and PNT groups, ZPC and PNT groups. 5. ZPC group showed more soluble phenamena than the resin groups(ST, SNT, PT, PNT). 6. Resin cement showed the void spaces in spite of good penetration into the micro-irregularities on both the tooth surface and the cast surface. The void was shown more in PT and PNT groups than in the ST and SNT groups. 7. After the treatment of heat and desiccation for SEM specimen, resin cements were detached more easily from the tooth surface than from the cast surface.

The purpose of this study was to determine the effect of post types and sizes on fracture resistance in immature tooth model with various restorative techniques. Bovine incisors were sectioned 8 mm above and 12 mm below the cementoenamel junction to simulate immature tooth model. To compare various post-and-core restorations, canals were restored with gutta-percha and resin core, or reinforced dentin wall with dual-cured resin composite, followed by placement of D.T. LIGHT-POST, ParaPost XT, and various sizes of EverStick Post individually. All of specimens were stored in the distilled water for 72 hours and underwent 6,000 thermal cycles. After simulation of periodontal ligament structure with polyether impression material, compressive load was applied at 45 degrees to the long axis of the specimen until fracture was occurred. Experimental groups reinforced with post and composite resin were shown significantly higher fracture strength than gutta-percha group without post placement (p < 0.05). Most specimens fractured limited to cervical third of roots. Post types did not influence on fracture resistance and fracture level significantly when cement space was filled with dual-cured resin composite. In addition, no statistically significant differences were seen between customized and standardized glass fiber posts, which cement spaces were filled with resin cement or composite resin individually. Therefore, root reinforcement procedures as above in immature teeth improved fracture resistance regardless of post types and sizes.
The purpose of the present study was to compare the influence of post-surface treatment with silane, hydrogen peroxide, hydrofluoric acid or sandblasting and to investigate the effect of silane in combination of the other treatments on the microtensile bond strength between fiber posts and composite resins for core build-up. Thirty-two glass-fiber posts (FRC Postec Plus, Ivoclar Vivadent, Schaan, Liechtenstein) were divided into eight groups according to the different surface pretreatments performed: silane application (S); immersion in 28% hydrogen peroxide (HP); immersion in hydrogen peroxide followed by application of silane (HP-S); immersion in 4% hydrofluoric acid gel (HF); immersion in hydrofluoric acid gel followed by application of silane (HF-S); sandblasting with aluminum oxide particles (SB); sandblasting followed by application of silane (SB-S). In control group, no surface treatment was performed. The composite resin (Tetric Flow, Ivoclar Vivadent, Schaan, Liechtenstein) was applied onto the posts to produce the composite cylinder specimen. It was sectioned into sticks to measure the microtensile bond strength. The data was analyzed with one-way ANOVA and LSD test for post hoc comparison (p < 0.05). Post pretreatment with sandblasting enhanced the interfacial strength between the fiber posts and core materials. Moreover, sandblasting followed by application of silane appears to be the most effective method that can improve the clinical performance of glass fiber posts.
Physical properties of composite resins such as strength, resistance to wear, discoloration, etc depend on the degree of conversion of the resin components. The purpose of this study was to evaluate the degree of conversion of the composite resins according to the thickness of tooth structure penetrated by light and applied light curing time. The coronal portions of extracted human teeth (one anterior tooth, three posterior tooth) was embedded by pink denture material. the mounted teeth were cut into three illumination sections (1mm thickness enamel section, 1mm thickness dentin section, 2mm thicknes dentin section) and one backing section with cutting wheel. Thin resin films were made by using 6kg pressure between slide glass during 5 minutes Thin resin film was light cured on coupled illumination section during 40sec, 80sec and 120sec. each illumination section was coupled as follows; no tooth structure(X), ename section(E), enamel section + 1mm dentin section(ED1), enamel section + 2mm dentin section(ED2), enamel section + 1mm dentin section + 2mm dentin section(EDD). To simulate the clinical situation more closely, thin resin films was cured against a backing section of tooth structure. The degree of conversion of carbon double bonds to single bonds in the resin films were examined by means of Fourier Transform Infrared Spectrometer. The results were obtained as follows ; 1 As curing time was increased, conversion rate was increased and as tooth thickness which was penetrated by curing light was increased, conversion rate was decreased. 2. At all tooth thickness groups, conversion rate between 80sec and 120sec was not significantly increased(P>0.05). 3. At 40sec group and 80sec, conversion rate between no tooth structure(X) group and 1mm enamel section(E) group was not significantly decreased(P>0.05). 4. At 80sec group and 120sec, conversion rate between 1mm enamel section(E) group and 1mm enamel section + 1mm dentin section(ED1) group was not significantly decreased(P>0.05).
목적 : 최근 개발된 bonding agent 중 일부는 다양한 함량의 filler를 포함하고 있으며 filler의 첨가는 bonding agent의 기계적인 물성을 향상시킴으로써 접착력의 향상에 기여한다는 주장이 있다. 본 연구에서는 다양한 함량의 filler를 포함한 adhesive를 실험적으로 만들어, filler의 함량이 전단접착강도에 미치는 영향을 살펴보고 임상적으로 가장 적절한 filler의 함량을 알아보고자 하였다. 또 adhesive의 간접인장강도를 측정하여 adhesive의 기계적인 물성과 접착력과의 상관관계를 알아보았다. 방법 : 발거된 건전한 70개의 대구치를 투명 레진에 매몰하고 상아질면을 노출시켰다. 3M사의 Scotchbond Multipurpose의 etchant와 primer를 제조사의 지시대로 적용하고 1$\mu\textrm{m}$크기의 barium glass filler를 0, 5, 10, 15, 20, 30, 45wt% 포함하도록 실험적으로 제작한 adhesive를 도포한 후 레진을 충전하여 시편을 완성하였다. Instron으로 0.5mm/min의 속도에서 전단접착강도를 측정하고 그 단면을 입체현미경으로 관찰하여 파절의 양상을 확인하였다. Filler함량에 따른 adhesive의 후경을 측정하기 위해 상기한 방법으로 시편을 제작하여 주사 전자현미경으로 관찰한 후 Sigmascan을 이용하여 그 후경을 측정하였다. 또, 지름 4mm 높이 6mm의 원통형 시편을 제작하여 Instron로 간접인장강도의 측정을 시행하였다. 얻어진 결과는 Kruskal-Wallis test와 Mann-Whitney test를 시행하여 분석하였으며, 상관관계를 분석을 위해 Pearson Product Moment Correlation Coefficient를 구하였다. 결과 : 1) Filler함유량에 따라 전단접착강도는 유의할 만한 차이를 보였다(p<0.05). 2) Filler함량의 증가에 따라 전단접착강도는 유의하게 증가하여 15% 수준에서 가장 높은 갈(19.9$\pm$1.38Mpa)을 보였으며 20% 이상의 수준에서는 유의하게 감소하였다(p<0.05). 3) Adhesive의 간접인장강도는 20% 수준까지는 증가하는 양상을 보였으나 통계적 유의성은 없었으며(p>0.05), 30% 이상에서는 유의할 만한 감소를 보였다(p<0.05). 4) Adhesive의 후경은 0% 수준에서 5.97$\pm$1.23$\mu\textrm{m}$부터 45%수준에서 73.37$\pm$11.7$\mu\textrm{m}$까지 유의하게 증가하였다(p<0.05). 5) Filler함량에 따른 Adhesive의 간접인장강도와 전단접착강도는 상관관계가 없었다.
The present study was performed to check the viability of eggs, filariform larvae and adults of Strongvloines venezueLensis exposed to various conditions for an in vitro maintenance. The eggs in the feces remained viable for about 25 days at $4^{\circ}C$ and 15 days at room temperature. However, the isolated eggs in sterile saline lost their viability within 24 hr at $4^{\circ}C$. The eggs in morula stage were very sensitive to air drying and rapidly lost their viability (=12 hrs. Filariform larvae survived for a maximum period of 45 days in fecal suspension and 28 days in 0.12% nutrient broth in polyvinyl culture bags maintained at $20^{\circ}C$. On the other hand, those isolated from nutrient broth cultures survived for a maximum period of 32 days in tap water and 22 days in sterile saline at $20^{\circ}C$. The mature adult worms obtained from experimentally infected rats survived maximally for 9 days in serum supplemented (10% rat-serum) 0.12% nutrient broth and 4 days in serum free nutrient broth at $37^{\circ}C$ while the culture media were changed at an alternate day. The adult female worms deposited fertile eggs in serum supplemented and serum free nutrient broth cultures, however, the hatched larvae (Ll) were not able to develop to the filariform stage in the culture media and found to die within 24 hr of maintenance. The present findings on an in vitro maintenance of different stages of 5. uenezueLetis may provide useful information for biological and biochemical studies with Strongyloines species. This study attempted to seek policy directions for development of sustainable urban industrial space through reviewing recent policies and projects of US cities that experienced urban industrial land replacement and industrial suburbanization. The policies and regulations for the urban industrial space preservation include the New York City's Industrial Business Zone, Chicago's Planned Manufacturing District, the no-net-loss framework in San Jose, and the subdivided and the state of the art in zoning systems in Philadelphia and Seattle. These policies and zoning systems have been effective for preventing the conversion of urban industrial lands to other uses, promoting business activities, and creating jobs for urban residents. In addition, the updated and innovative zoning systems allow for a wide variety of urban industrial spaces to meet newly emerging industrial demands. On the other hand, we suggested several policy implications through reviewing the cases of urban regeneration projects in Portland's Willamette Industrial Urban Renewal Area, Atlanta's BeltLine, and Milwaukee's Menomone Valley as follows: 1) updating and reinforcing spatial planning and policies for preservation and revitalization of urban industrial land; 2) considering existing businesses and local workers first; 3) setting up a participatory planning for local workers and residents; 4) preparing policies for brownfield cleanups.
ln order to manage efficiently many ancient wooden buildings, which have been preserved as cultural properties in Korea, the internal state of wood members should be evaluated exactly and periodically by a NDE (non-destructive evaluation) method. A research project was planned to develop an X-ray CT (computed tomography) system as a NDE method for wood, which could be easily applied in field. This paper includes the first part of this project. First of all, to establish a measuring procedure of wood density using X-ray radiography, the correlation between X-ray intensity and the film brightness was evaluated. Also, initial X-ray intensity was quantified with various radiate conditions controlled by the tube voltage and tube current. And then, the effects of density, annual ring angle, and thickness on the mass attenuation coefficient of wood were examined. Finally, Beer's law was modified with the above results and adopted to calculate the density of wood. As a result of this study, the measuring procedure of wood density was established using a portable soft X-ray device and this procedure was verified with some small wood specimens. This results will he used valuably for the following researches to develop a portable X-ray CT system.
