Effect of Permeability-Controlled Polyethylene Film on Extension of Shelf-life of Brined Baechu Cabbage (투과도 조절 폴리에틸렌 필름의 절임배추 보존기간 연장효과)

  • Brined Baechu cabbages were packed with different films of high density polyethylene (HDPE), aluminium polyethylene (Al/PE), nylon polyamide (Ny/PE), low density polyethylene (LDPE) and permeability-controlled polyethylene (Mirafresh (MF), US patent No. 5972815), and then stored at 4${^{\circ}C}$ for 4 weeks. Changes in quality characteristics of pH, acidity, total bacteria counts, lactic acid bacteria counts, E. coli counts, texture and $O_2$ concentration were determined during the storage. The pH of brined Baechu cabbage packed with Mirafresh (MF) film was 6.25 after 4 weeks from initial pH of 6.80. The acidity of all brined Baechu cabbages increased, however, the increase of the cabbage in MF was the lowest. The levels of total bacteria, lactic acid bacteria and E. coli in the cabbages packed with MF were also lower than the other films. After 4 weeks, of all brined springiness Baechu cabbages decreased, but MF showed relatively high springiness. The $O_2$ concentrations by its permeation through MF were 0.35%-1.00% at 4-25${^{\circ}C}$ after 1 week. In conclusion, MF was found to be the most effective packaging film for brined Baechu cabbage to extend shelf-life.

Antimicrobial Activity of the Solvent Extract from Different Parts of Orostachys japonicus (와송 부위별 추출물의 항균활성)

  • This study was to determine the inhibitory effect against food borne pathogens of ethanol and water extracts from leaf, stem and root of Orostachys japonicus. On the paper disc assay, no detectable bactericidal activity in the water extracts from leaf, stem and root of Orostachys japonicus and ethanol extracts form stem and root of Orostachys japonicus was shown. However, ethanol extract of Orostachys japonicus leaf showed the highest antimicrobial activity. Minimum inhibitory concentration (MIC) of ethanol extracts was determined to range from 0.05 to 0.1% in leaf of Orostachys japonicus against gram positive bacteria and yeast. Antimicrobial activity of ethanol extracts was stable by heating at $121^{\circ}C$ for 15 min, and not affected by pH $2{\sim}10$ except for B. subtilis. These findings suggest ethanol extract from leaf of Orostachys japonicus may be useful as natural preservative.

Antioxidant and Antimicrobial Effects of Solvent Fractions from Smilax china L. Leaves (청미래덩굴 잎(Smilax china L.) 용매 분획물의 항산화 및 항균 효과)

  • To utilize Smilax china L. leaves as a natural preservatives, solvent fractions from crude methanol extract were prepared and investigated their antioxidant and antimicrobial activities against Staphylococcus aureus. Antioxidant activities were examined by 1,1-diphenyl-1-picrylhydrazyl radical scavenging and ferric ion reducing antioxidant power assays. Ethyl acetate fraction from Smilax china L. leaves exhibited the strongest antioxidant and antimicrobial activities among the fractions (P<0.05), as well as the highest total phenolic and total flavonoid contents. Caffeic acid, ferulic acid, quercetin, and kaempferol contents in the ethyl acetate fraction from Smilax china L. leaves were determined by TLC and HPLC. In aqueous phase, as well as the n-butanol and n-hexane fractions, quercetin, ferulic acid, and kaempferol were detected in small amounts. Ferulic acid, which showed the highest content, is the major active compound from Smilax china L. leaves.

A REVIEW OF SOME ASPECTS OF THE YELLOWFIN TUNA FISHERY IN THE ATLANTIC OCEAN (대서양 황다랭이의 자원생물학적 연구)

  • Korea started the Atlantic tuna fishery from 1964 by means of longline, then added bait-boats in 1972. Both fisheries have given the top priority to catch yellowfin tuna(Thunnus albacares). The paper reviews available catch, effort and biological data, estimates some population parameters in order to understand the status of the Atlantic yellowfin tuna as a whole. The main findings are summarized as follows: 1. The total of 476 million hook-equivalent fishing effort was thrown to catch yellowfin tuna in 1974, among which one thired was shared by longliners. 2. The dominant age group becomes younger in both surface and longline fisheries. 3. The recent mortality coefficients were calculated as 1.5 for total mortality and 0.7 for fishing mortality. 4. The weight at recruitment was 2.7 kg in 1973 which was smaller than the regulation size(3.2kg) proposed by ICCAT. 5. The maximum sustained yield was calculated to 95-145 thousand metric tons, which was the level of recent catch. Therefore, it is apparent that the present yellowfin tuna fishery should continue to receive close attention.

Characteristics of Eggshell Powder as Carriers of Probiotics (생균제의 부형제(운반체)로서의 난각분말의 특성)

  • Eggshell (ES) is a by-product of table eggs with high content of calcium carbonate which can be used as a calcium source in feed. In this study, we have first illuminated the potential application of ES as a novel carrier for probiotics. The carriers used in the study include a SBM (Soybean meal), ESL (Eggshell powder with large particles), ESF (Eggshell powder with fine particles), and the complex carriers (SBM+ESL, SBM+ESF). The structure of carriers absorbed by L. plantarum was confirmed by SEM image. Among these carriers, the complex carrier SBM+ESF showed the highest viability of L. plantarum with pH 7~8 during four weeks storage at room temperature. The SBM+ESF was further tested as a carrier for various probiotic strains at $4^{\circ}C$ or $30^{\circ}C$. All the probiotic strains showed high viability at $4^{\circ}C$ storage. However, a significant reduction of Lactobacillus cells was observed at $30^{\circ}C$ storage. B. lichenifomis maintained high viability whereas B. subtilis, B. amyloliquefaciens, and S. cerevisiae showed the reduction of $2{\log}_{10}$ (CFU/g). These results suggest that if the ESF as a calcium source in feed was mixed with SBM, it can be used as an effective complex carrier for improving the viability of some probiotics including B. licheniformis.

Recent Perspectives on Oncoplastic Breast Surgery in Korea (우리나라의 종양성형학적 유방암 수술에 대한 최신 동향)

  • Oncoplastic surgery (OPS) combines oncologically safe tumor resection with aesthetically satisfying reconstruction of defects using established plastic surgery techniques. OPS is characterized by initial excision as extensive as is beneficial for oncological safety, and, once sufficient resection is complete, displacement or replacement techniques are selected based on remnant volume. The size of the lesion and the individual patient are important factors when considering the appropriate approach, and when pre-operative imaging, including MRI, is used to determine the technique, the complete removal of cancer cells by permanent pathology is essential. A frozen section is used during the operation to reduce the reoperation rate, but it is difficult to cover the entire margin surface theoretically and even harder in practice. A recent report about adequate margins has empowered OPS in its oncological safety. Considering the patients to whom each modality could be applied, basic breast volume is an important factor, and this is influenced by ethnic differences. In Europe or the US, for example, the average breast size is 36D (600 ㎤) and reduction mammoplasty is predominantly used. However, the average size of patients in our institution is 33A (300 ㎤), and so quite different approaches are selected in most cases. New techniques involving radiofrequency and fluorescence have been proposed as safe and easily accessible ways of reducing complications.

A Novel PHKA1 Mutation in a Patient with Glycogen Storage Disease Type IXD (당원 축적병 9D (GSD9D) 환자의 신규 PHKA1 돌연변이)

  • Distal myopathy is a clinically and genetically heterogeneous group of degenerative diseases of the distal muscle. Glycogen storage disease type IXD (GSD9D) is a metabolic distal myopathy characterized by muscle deficiency of phosphorylase kinase, a key regulatory enzyme in glycogen metabolism. Affected individuals may develop muscle weakness, degeneration, and cramps, as well as abnormal muscle pain and stiffness after exercise. It has been reported that mutations in the PHKA1 gene which encodes the alpha subunit of muscle phosphorylase kinase cause GSD9D. In this study, we examined a Korean GSD9D family with a c.3314T>C (p.I1105T) mutation in the PHKA1 gene. This mutation has not been previously reported in any mutation database nor was it found in 500 healthy controls. The mutation region is well conserved in various other species, and in silico analysis predicts that it is likely to be pathogenic. To date, only seven mutations in the PHKA1 gene have been documented, and this is the first report of Korean GSD9D patients. This study also describes and compares the clinical symptoms and pathological conditions of previously reported cases and these Korean patients. We believe that our findings will be useful for the molecular diagnosis of GSD9D.

Evaluation of Stability in the Purified Wood Vinegar and Its Hair Growth Effect (목초액의 안정성 및 모발 성장 촉진 효과)

  • Generally, it takes a long time to purify wood vinegar, and it contains toxic compounds such as tar, methanol, phenol and benzopyrene. To reduce the toxicity of wood vinegar itself, we have developed a new purification method of wood vinegar using an oxidation-cohesion reaction and distillation with an active carbon. We have investigated the physico-chemical change (pH, specific gravity, refractive index and dissolved tar), the change of amount of toxic compounds (carbonyl group, phenol, benzopyrene and residual solvents) and organic acids (formic acid (FA), acetic acid (AA), propionic acid (PA)) of the purified wood vinegar under the long term and accelerated storage conditions. Also, we have evaluated the effect of the purified wood vinegar on hair growth using an alopecia model of C57BL/6 mice. As a result, we could find out that the purified wood vinegar was stable and remained without decay under the storage conditions and benzopyrene, a carcinogenic agent, was not detected in the purified wood vinegar. After topical treatment of the purified wood vinegar solution or minoxidil (MXD) for 2 weeks to dorsal skin, the hair regrowth of the mice accelerated faster than that of the control, with no clinical signs. In conclusion, we could suggest a guideline for quality control of process to reduce the toxic compounds in wood vinegar and it might be a useful hair growth promoter in the treatment of baldness or alopecia.

Studies on the Mutagenicity, Lipids Peroxidation and Meat Structure of Cooked Pork in Relation to Storage and Reheating using a Microwave Oven (가열조리한 돼지고기의 저장${\cdot}$Microwave 재가열에 의한 변이원성과 지질과산화 및 육조직에 관한 연구)

  • Pork was cooked using three kinds of instrument [electric pill (EG) for 5min., microwave oven (MW) for 6min. and reheated using a MW] and then extracted with $80\%$ methanol. The Ames test was performed on the methanol extracts, employing the S. typhimurium tester strain, TA100. The methanol extract of cooked pork showed high mutagenicity ion the 5.0 mg/plate without the S9 mix, but a higher mutagenicity was induced with the S9 mix With increasing refrigeration $(4^{\circ}C)$ and freezing $(-18^{\circ}C)$ periods the extracts showed higher mutagenicities and TBA values, and the same results where shown with reheating. Correlations of the mutagenicity (-S9 mix) and rancidity of the pork cooked by EG, according to storage at and $-18^{\circ}C$ and reheated by MW (1 min), were r=0.85, 0.86, 0.98 and 0.83, respectively. When the MW was used for reheating, the refrigeration storage (r=0.98) showed a higher correlation coefficient than for that stored frozen (r=0.83). From the structure of cooked pork, as observed by SEM, many vapor pathways were viewed in the pork reheated using themicrowave oven.

Inhibition of Acid Production in Gel Type Yogurt by the Lactoperoxidase System (호상 요구르트 제조시 LP System에 의한 산생성 억제에 관한 연구)

  • This study was carried out to investigate the effect of the lactoperoxidase (LP) system on the acid production of gel type yogurt. The LP system was activated by adding freshly prepared solutions of 1 ppm (v/w) lactoperoxidase and three different concentrations (0.125 mM, 0.25 mM, 0.5 mM) in equimolar ratios of KSCN and $(H_{2}O_{2}$, In 0.25 mM treated samples for the 4 hr fermentation resulted in titratable acidity of $0.4{\%}$, pH of 5.06, lactic acid bacterial count of $3.0{\times}10^{8}\;CFU/ml and acetaldehyde concentration of 10.2 ppm, whereas the untreated samples were 1.0%, 4.54, $4.7{\times}10^{9}\;CFU/ml and 18.0 ppm, respectively. The residual amount of KSCN in 0.25 mM treated samples was determined during the experiments, which decreased to 4.4 ppm. There was no detectable $(H_{2}O_{2}$ for 6 hr fermentation. However, residual KSCN and $(H_{2}O_{2}$ concentrations in 0.5mM treated samples were 5.7 and 8.4 ppm, respectively. These results have indicated that the optimum concentration of $(H_{2}O_{2}$ and KSCN to activate the LP system was 0.25 mM each.
