Mungbean sprout rot is one of the most serious problems of the commercial mungbean sprout industry. In this study, 70 strains of mungbean sprout rot pathogens were isolated from rotten sprouts at different time intervals. The pathogenicity of the isolated pathogens was tested. The highly pathogenic strain (YV-St-033) was identified as Pseudomonas sp. by 16S rRNA gene sequencing. In phylogenetic analysis, the YV-St-033 strain was grouped with P. mosselii, P. putita, P. fluorescens, P. entomophila, and P. lecoglossicida. The results of the 16S rRNA gene sequence analysis revealed that the YV-St-033 strain shared the highest sequence identity (more than 99%) with the P. mosselii R10 strain. The mungbean lines of Yeungnam University germplasm were screened against the YV-St-033 strain. Based on the growth rate of the sprouts after 3 days of inoculation with the pathogen, the YV148 line was highly resistant to the pathogen. The remaining lines were either partially or fully infected. The highly resistant line YV 148 is suitable for future breeding programs due to their thin sprouts and fast growing nature.
Since Tongil has been cultivated widely in farmers' fields, the variety is known to be highly resistant to existing races of Pyricularia oryzae in Korea. However, Tongil and the sister lines were moderately susceptible to race IA-65, which was isolated from Tongil at the blast nursery of the College of Agriculture, Seoul National University, 1972. In field tests of 1973, pathogenic reactions of IR lines at the isolated nursery, where IA-65 had been inoculated, were significantly higher than those at the farm nursery. When seedlings of 196 IR lines were inoculated with IA65, about $30\%$ of the IR lines including IR667-98 were moderately susceptible. Among the 30 isolates of P. oryzae obtained mostly from indicas, 12 were also differentiated in 1973 as IA group known to be the most prevalent races at IRRI in the Philippines, where Tongil has been highly susceptible. This IA group was also pathogenic to Tongil in greenhouse experiments. The race group causing moderately susceptible to Tongil should be new in Korea, since no occurrences of the race IA have ever been reported previously from the areas where japonica types of rice are cultivated. Because some races of P. oryzae present in Korea are pathogenic to Tongil and because these races may become prevalent in the fields, the reaction of Tongil must be continuously monitored.
Trichoderma disease of oyster mushroom has not been effectively detected in the field for testing its resistance against the disease with its varieties. In this study, we investigated the methods to detect its resistance in the laboratory by using media, which enables us to understand the relevant characteristics (e.g., lysis, toxin enzyme, mycelial growth rate). In coculturing with strains of Trichoderma and oyster mushroom, it is possible to observe the difference in the resistance of oyster mushroom against Trichoderma with the phenomena of barrage reaction, overgrowth and lysis. We also observed the inhibition of mycelial growth of oyster mushroom using the dilution method with 48-well plate, but could not observed the inhibition of mycelial growth using the filter paper method of cultural supernatant. In simultaneously culturing both Trichoderma and oyster mushroom, it was possible to detect the inhibition of the mycelial growth of oyster mushroom, but Trichoderma mycelium did not overgrow against oyster mushroom. We found that the pathogenicity was efficient in using solid medium with the phenomena of overgrowth and lysis by inoculating Trichoderma on top of mycelia of oyster mushroom. In conclusion, the methods (e.g., coculture method, dilution method with 48-well plate, post-inoculation method) are recommended to detect the resistance of oyster mushroom against Trichoderma disease.
This studies were investigated the occurrence of sclerotinia rot at the crisphead lettuce field in Uiryeong-Gun, Gyeongsangnam-Do from January to May in 2003. Average incidence rates of sclerotinia rot on crisphead lettuce was up to 21.9% at the five plastic houses. A total of 140 isolates of Sclerotinia sp. were obtained from diseased leaves of crisphead lettuce. Among them, the fungi YR-1 was isolated, which showed highly virulent on the whole plant. the YR-1 was identified as Sclerotinia sclerotiorum based on the formation, color, shape and size of sclerotium and apothecium. For the pathogenicity test, the most suitable inoculum quantity of YR-1 strain was selected as the triturated mycelial suspension of $A_{550}$=0.8, 40 ml showing disease incidence of 94%, and the symptom showed as same as at the fields, the leaves and stem had rotten and developed white downy mycelial at the diseased lesion on the leaves and stems, and produced black and irregular sclerotinia. This is the first report on the pathogenicity test using by triturated mycelial suspension-inoculum of the pathogen for the sclerotinia rot of crisphead lettuce.
Incidence of Fusarium wilt was surveyed in fields of summer radish in Gangwon province in Korea in 2018 and 2019. The disease started in early July and spread rapidly in hot summer of late July and August and in severe case, reached up to 80% in a field in Gangneung area. Symptoms in the seedling stage include poor growth and browning of internal tissue of root. During mid-growth, the leaves of diseased plant turned yellow over time, the surface of the roots changed from white to blackish, and the vascular tissues turned brown. A total of 23 isolates was obtained from the diseased plants and identified as Fusarium oxysporum f. sp. raphani by elongation factor-1α and intergenic spacer sequence analysis. Pathogenicity of the isolates was tested by artificial inoculation to the radish and other plants. All the isolates tested were pathogenic to radish plant, although there were differences in virulence on radish 11 cultivars. However, the isolates were not virulent to other plants except some cruciferous vegetables including Brussels sprouts, rocket, stock, and turnip. The results of pathogenicity test showed that it is necessary to rotate with crops other than cruciferous vegetables in order to prevent Fusarium wilt from radish fields.
This study was investigated the occurrence of sclerotinia rot caused by Sclerotinia sclerotiorum at the major perilla cultivating area, Gangdong-dong, Gangseo-gu, Busan in 1998. The incidence of this disease ranged from 8.1 to 28.3% at Gangdong-dong area during the growing seasons. Symptoms of the disease initially appeared damping-off of infected stems and soft-rot on the leaves of perilla. Under the relatively high humidity, abundant white mycelia of the pathogen formed on the lesion developed into black sclerotia later and the infected leaves were finally fell down. Sixteen isolates, Sl-S16, isolated from diseased lesions showing typical symptoms, and pathogenicity was tested using mycerlial disks. Among them, S2 isolate showing the most strong pathogenicity was selected and identified as Sclerotinia sclerotiorum on the basis of morphological and cultural characteristics. For biological control, an antagonistic bacteria, N4 isolate which effectively inhibited not only mycelial growth of S2 isolate but also suppress sclerotinia rot on the pot assay, was selected and identified as Bacillus megaterium according to Bergey's manual and API system., Wettable powder type, N4 formulation using B. megaterium N4 isolate was developed and estimated its control effect on perilla crops in a plastic house. As a results, N4 formulation which applied before 3 days inoculation of pathogen was effectually controlled Sclerotinia rot as the control value of 98.0%, was more effective than chemical fungicide, benomyl showing the control value of 78.0%. This is the first report of wettable powder formulation as a biocontrol agent using B. megaterium N4 against Sclerotinia rot caused by S. sclerotiorum on perilla.
A total of 29 Botrytis were isolated from ginseng gray mold at 8 locations in Korea from June to July, 2004 and 2005. The causative agent of them was identified as Botrytis cinerea based on morphological characteristics of conidia and RPB2 gene sequence analysis. In inoculation experiments with sclerotia, one isolate was pathogenic to ginseng plant whereas two isolates were non-pathogenic. Scanning electron microscopy revealed that the sclerotia of pathogenic isolate has rough surface and that of non-pathogenic isolate very smooth surface. Optimum temperature and culture conditions for sclerotia production were $5{\sim}20^{\circ}C$ and darkness, respectively. The number of sclerotia was increased tenfold on media added with 0.5% old stem fragment of ginseng.
In early 2000's, about six Phytophthora species have been newly described leading mortality on coniferous and broad-leaved trees in forests. Also, some species of Phytophthora are responsible for ink disease in chestnut plantation near or within forests. Similar symptoms of ink disease were appeared in some areas of Kyungnam and Jeonnam providences in 2005, and the pathogen was isolated using Phytophthora- selective medium in 2006. Morphological and genetic analysis were performed to identify the isolate. Also, the pathogenicity was conducted to complete $K\ddot{o}ch^{\prime}s$ postulate and compare susceptibility among chestnut cultivars. The molecular analysis between P. katsurae and P. hevae were performed with the isolates obtained from different countries including Korea or the sequences downloaded from Phytophthora webpage. The result showed that the isolated pathogen from chestnut was P. katsurae. There is no report of P. katsurae in Korea until now. P. katsurae was re-isolated from inoculated chestnut cultivars. Also, there was a slight difference in susceptibility among chestnut cultivars. The rDNA sequence of our isolate showed 100% similarity with sequence of the isolate cultured from Japan and New Zealand.
Effects of tomato-juice and KH$_2$PO$_4$ as exogenous nutrients on the infection of Botrytis cinerea LVF12 and pathogenicity to tomato were investigated. B. cinerea LVF12, which was previously reported as a casual agent of the gray mold rot of perilla, was used for pathogenesis on tomato leaves. No infection was induced, and no lesion developed on tomato leaves by the conidial suspension of LVF12 when the inoculum was prepared in sterilized water. However, when the conidial suspensions of LVF12 added with various concentrations and conditions of tomato-juice were inoculated on whole tomato plants, the disease was induced readily, Among them, 20% tomato juice with 0.1M KH$_2$PO$_4$ appeared to be the most suitable nutrient to promote high disease incidence on tomato. For the pathogenicity test according to the growing stage of tomato, the mature leaves were more susceptible than seedlings. Symptoms on the infected plants were initial small gray spots at the inoculated area. Later the whole leaves, petioles and stems became gray and eventually fell off, Under high humidity conditions, the diseased leaves and stems were covered with gray hyphae and conidia. All symptoms of infected plants were identical to those in the field conditions.
