Genotoxicological Safety of Hot Water Extracts of the γ-Irradiated Astragali Radix, Atractylodes Rhizoma, and Cimicifugae Rhizoma in Vitro (감마선 조사 황기, 백출 및 승마 열수 추출물의 in vitro 유전독성학적 안전성 평가)

  • As the utilization of medicinal herbs in food and bio-industry increases, safe hygienic technologies for them are demanded. To consider the possibility of application of radiation technology for this purpose, the genotoxi-cological safety of three r -irradiated medicinal herbs were studied. Astragali Radix, Atractylodes Rhizoma and Cimicifugae Rhizoma were irradiated at 10 kGy, and then were extracted with hot water. The genotoxicity of the extracts was examined in two short-term in vitro tests: (1) Salmonella reversion assay (Ames test) in strains of TA98 and TA100; (2) Micronucleus test in cultured Chinese hamster ovary (CHO) cells. The extract was treated at maximum doses of 5 mg/plate in Salmonella reversion assay, and 1 mg/mL in micronucleus test where growth of CHO cells was inhibited by 50%. In Salmonella reversion assay with or without metabolic activation, both ex-tracts of irradiated and non-irradiated herbs showed no significant differences in formation of revertant colonies compared with the negative control. And also in micronucleus test, the incidences of micronucleus in CHO cells cultured with extracts of irradiated herbs were almost same as negative control in less than 3%. These results of two in vitro tests suggest that ${\gamma}$-irradiated herbs do not show mutagenicity and cytogenetic toxicity. Further tests of in vivo genotoxicity and chronic toxicity are needed to ascertain the safety of ${\gamma}$-irradiated herbs.

Effects of Mulching Materials on Growth and Yield of Atractylodes macrocephala Koidz (피복재료(被覆材料)가 백출(白朮)의 생육(生育) 및 수량(收量)에 미치는 영향(影響))

  • This study was conducted to investigate the soil mulching effect on growth and yield of Atractylodes macrocephala Koidz. The number of stem, stem diameter, and fresh weight of aerial part in black poly-ethylene film and transparent poly-ethylene film mulched condition were higher than those of non-mulching and rice-straw mulching. Number of flowers were high in the order of non-mulching, rice-straw, trans-P. E. film, and black P. E. film. Yield of underground part was higher in mulching condition than that of non-mulching. Dry rhizome yield in black P. E. film mulching was significantly high to compare with the other treatments. There was lower disease incidence of Phytophthora blight in trans-P. E and black P. E. film compare with the other treatments but any difference was not observed in essential oil content of rhizome. Significant positive correlation was found between rhizome weight and fresh weight of aerial part. It was also shown that the fresh weight of aerial part was greatly dependent on the number of stems and stem diameter.

Growth and Yield by the Different Seeding Methods and Cultivating Root Weight in Atractylodes macrocephala Koidz (백출(Atractylodes macrocephala Koidz)의 파종방법과 종근중에 따른 생육과 수량)

  • This study was conducted to investigate the effects on the growth and yield by the different seeding methods and cultivating root weight in Atractylodes macrocephata Koidz. Seeding distances were different such as 5${\times}$5 cm, 5${\times}$10 cm, 10${\times}$10 cm, 10${\times}$15 cm by the hand seeder and 15 cm seeding in drill. Emergence date, plant height were not significantly changed with seeding space, but the number of leaf, fresh weight of above-ground part and fresh weight of root were increased in the sparse seeding compared with the dense seeding. The highest fresh root yield was 1,012 kg/10 a at the 5${\times}$5 cm. Roots yield was increased in the sparse seeding compared with the dense seeding, but the highest number of roots above 16 g yield was observed at the 10${\times}$10 cm seeding distance. Emergence date was faster 1∼3 day root weight above 6 g than that root weight 5 g. The plant height, number of stem and fresh weight of above-ground part were more increased as the root weight was heavier. The growth of underground part were more increased as the root weight was heavier, yield was increased about 27% to 112% compared with root weight above 6 g than that 134.6 kg/10 a with root 5 g.

Nutritional and Sensory of Green Leafy Vegetables Cultivated from Medicinal Plant Seed (약용식물 종자로 재배된 쌈잎채소의 영양학 및 관능적 평가)

  • Ten green leafy vegetables were cultivated from medicinal plant seed. Their nutritional content, taste, appearance and texture were evaluated. Moisture content was $91.23{\pm}0.00%$ in $Saururus$ $chinensis$ (Sc), $90.39{\pm}0.05%$ in $Perilla$ $frutescens$ (Pa) and $66.78{\pm}0.05%$ in $Agastache$ $rugosa$ (Ar). The fat content was 8.89% in $Salvia$ $plebeia$ (Sp) and 0.13% in $Peucedanum$ $japonicum$ (Pj). The protein content was $11.1{\pm}0.00%$ in Ar and $0.54{\pm}0.03%$ in Pj. Total sugar content was $20.06{\pm}0.03%$ in $Geum$ $japonicum$ (Gj), $12.73{\pm}0.03%$ in $Coixlacry$ $majobi$ (Cm) and $1.04{\pm}0.03%$ in $Atractylodes$ $japonica$ (Aj). Medicinal leafy vegetables had higher glucose, sucrose and fructose, but lower maltose. Ascorbic acid content was $33.67{\pm}1.40$ mg% in Sc, $27.91{\pm}2.91$ mg% in Pa and $15.68{\pm}1.09$ mg% in At. Chlorophyll content was higher in $Perilla$ $frutescens$ var. $acuta$ Kudo than other leafy vegetables. The evaluation also showed that Cm, Pa and Aj tasted sweeter and had low acidity.

The Quality characteristics of sponge cake added with the ingredients of Sagoonja-Tang (사군자탕(四君子湯) 재료를 첨가한 스폰지 케이크의 품질특성)

  • This study was carried out to determine the functional works and basic ingredients of Sagoonja-Tang on sponge cake. The experimental groups consisted of 6% mixtures of five (Ed-confirm the number) kinds powders : S1 for Sagoonja-Tang powder, S2 for Panax ginseng, S3 for Poria cocos, Koidz, S4 for Atractylodes macrocephala, and S5 for Glycyrrhiza uralensis Fisch. The volume of sponge cake somewhat decreased after adding the ingredients, and the volume differences in order from highest to lowest, were control>S1>S3>S2>S4>S5. In the microbiological quality test, viable cell counts were high in control groups of sponge cake, and the numbers of viable cell for control sponge cake reached to $9{\times}10^7CFU/g$ on the 7th day of storage, and decreased to $2.5{\times}10^5CFU/g$ on the 10th day of storage. All sponge cakes added with ingredient's powder showed pretty low viable cell counts. Especially, the group S5 showed the lowest counts of $1.2{\times}10^2CFU/g$ on the 7th day of storing. In the texture analyses of sponge cake, All groups showed higher degree of hardness, gumminess and chewiness than SC. The antioxidative activity of the Sagoonja-Tang's ingredients was measured. The POV value measured was S5>S4>S3>S2 >S1>control in order of highest to lowest. In sensory test, the overall acceptability of sponge cake was from highest to lowest S3>SC S2>S1>S5>S4.

Antioxidative Properties and Whitening Effects of the Astragali Radix, Atractylodis Rhizoma Alba and Acanthopanacis Cortex (황기, 백출 및 오가피의 항산화성 및 미백효과에 관한 연구)

  • In an attempt to find natural sources of antioxidants and whitening agents, Comparisons of the antioxidative and tyrosinase inhibitory activities of various ethanol extracts of Astragali Radix, Atractylodis Rhizoma Alba and Acanthopanacis Cortex were carried out. Comparison of the four ethanol extracts revealed that, Astragali Radix had the highest electron-donating ability(72.5%) and the highest SOD-like ability(26.1%). The xanthine oxidase experiment exhibited a hindrance effect of 88.5% in Atractylodis Rhizoma Alba, 81.1% in Acanthopanacis Cortex, 75.8% in Astragali Radix. A tyrosinase inhibitory activity assay was conducted to evaluate the whitening effects of the extracts, The tyrosinase inhibitory activity was 42.1% in the Acanthopanacis Cortex, 37.2% in the Atractylodis Rhizoma Alba, 6.0% in the Astragali Radix. Based on these results, we suggest that the ethanol extracts of Astragali Radix, Atractylodis Rhizoma Alba and Acanthopanacis Cortex can be used as food and cosmetic ingredients.

Effects of Atractylodis Rhizoma Alba extract on collagen-induced arthritis in mice (백출(白朮)의 콜라겐 유도 관절염 마우스에서의 관절염 개선 효과 연구)

  • Objectives : The present study was undertaken to evaluate whether Atractylodis Rhizoma Alba ethanol (ARA-E) extract, which is the pericarp of $Atractylodes$ $japonica$ Koidz. has an effect on collagen-induced arthritis (CIA) in mice. Methods : Male DBA/1J mice were induced by intradermal injection of bovine collagen-II in Freund's incomplete adjuvant (IFA). The CIA mice in the onset of arthritis were treated daily with oral administration of ARA-E extract at dose of 50 mg/kg/bw for 28 days. Arthritis index, histopathological changes and the levels of anti-type II collagen (CII) IgG and inflammatory cytokine, TNF-${\alpha}$ in sera of mice were measured to evaluate the antiarthritic effect of ARA-E. Results : ARA-E extract significantly decreased the arthritic scores and inhibited pathological changes of knee joint tissues in CIA mice. ARA-E extract also significantly decreased the serum levels of anti-CII IgG and TNF-${\alpha}$ in CIA mice. These results indicate that ARA-E extract may effectively prevent arthritic damages in CIA mice, at least partially, by inhibiting the production of autoantibodies and inflammatory cytokine. Conclusions : This studies suggest that ARA-E has a therapeutic potential in inflammatory joint diseases such as rheumatoid arthritis.

  • A simple and reliable reverse phase HPLC method was developed to determine pharmacologically active marker compounds of Epimedii Herba, Atractylodis Rhizoma Alba, and Polygalae Radix. The stability test of water-extracts of the three natural medicines has been evaluated for six months. However, no significant changes in the content of the marker compounds of each extract were observed during the time of investigation.

  • An oriental medicinal database was used from medicinal plants for screening of anticancer agents. The prescription frequency and dosage of medicinal plats in 170 anticancer prescriptions were analyzed. From the total prescription score of each medical plant, it was found that high-scored medicinal plants have been widely studied in the previous research on the cancer treatment. Consequently, dangui, hwangui, banha, bachul, jinpy, insam, bacjakyak, deahwang, chungoong, jimo, chunnamsung, omija, hyunggae, huekchook, banggi, boclyung, osooyou were selected as raw materials for the screening of anticancer agents.

  • 1. In the rabbit and the dog, the blood pressure response to water extract and methanol extract obtained from Atractylodes rhizoma alb'a was investigated. 2. Water extract and methanol extract, when administered into the rabbit and the dog by the route of vein, produced fall of the blood pressure. 3. The depressor response of the rabbit to water extract and methanol extract was not affected by $Avicel{\circledR}$, propranolol and atropine. 4. The depressor response by water extract and methanol extract in the rabbit was not affected by guanethidine, but water extract and methanol extract produced elevation of blood pressure in this rabbit. 5. Pretreatment of rabbit with chlorisendamine or phenoxybenzamine weakened the depressor response to water extract and methanol extract, and the both extracts produced secondary elevation of blood pressure in this rabbit. 6. The pressor response of the chlorisondamine-treated rabbit to water extract and methanol extract was not affected by atropine. 7. Water extract decreased the pressor action of tyramine and depressor action of pilocarpine and isoproterenol, but did not affect the blood pressure response of nor einephrine, angiotensin and dimethylpehnyl piperazinium iodide(DMPP).
