Isolation and Identification of Fibrinolytic Enzyme Producing Strain from Shrimp Jeot-Gal, a Tiny Salted Shrimps, and Medium Optimization for Enzyme Production (새우젓에서 혈전용해효소 생산균주의 분리, 동정 및 효소생산 배지의 최적화)

  • A strain of potential producer of fibrinolytic enzyme was isolated from shrimp Jeot-Gal, a tiny salted shrimps, and identified as Bacillus sp.. The preliminary experiment showed an enzyme yield of 18 U/mL in medium for screening. The carbon, nitrogen and salts significantly influenced the fibrinolytic enzyme production. An optimized medium containing 2% skim milk, 2% soluble starch and 3% NaCl (pH 7.5) after 72 hrs fermentation time at $37^{\circ}C$ yielded 3-fold increase in enzyme production, 62 U/mL.

Optimization of Medium Components using Response Surface Methodology for Cost-effective Mannitol Production by Leuconostoc mesenteroides SRCM201425 (반응표면분석법을 이용한 Leuconostoc mesenteroides SRCM201425의 만니톨 생산배지 최적화)

  • This study was undertaken to establish optimum medium compositions for cost-effective mannitol production by Leuconostoc mesenteroides SRCM201425 isolated from kimchi. L. mesenteroides SRCM21425 from kimchi was selected for efficient mannitol production based on fructose analysis and identified by its 16S rRNA gene sequence, as well as by carbohydrate fermentation pattern analysis. To enhance mannitol production by L. mesenteroides SRCM201425, the effects of carbon, nitrogen, and mineral sources on mannitol production were first determined using Plackett-Burman design (PBD). The effects of 11 variables on mannitol production were investigated of which three variables, fructose, sucrose, and peptone, were selected. In the second step, each concentration of fructose, sucrose, and peptone was optimized using a central composite design (CCD) and response surface analysis. The predicted concentrations of fructose, sucrose, and peptone were 38.68 g/l, 30 g/l, and 39.67 g/l, respectively. The mathematical response model was reliable, with a coefficient of determination of $R^2=0.9185$. Mannitol production increased 20-fold as compared with the MRS medium, corresponding to a mannitol yield 97.46% when compared to MRS supplemented with 100 g/l of fructose in flask system. Furthermore, the production in the optimized medium was cost-effective. The findings of this study can be expected to be useful in biological production for catalytic hydrogenation causing byproduct and additional production costs.

Effect of Medium Components on the Lipstatin Production by Streptomyces toxytricini (배지 성분이 Streptomyces toxytricini에서의 lipstatin 발효에 미치는 영향)

  • In order to increase the productivity of lipstatin by Steptomyces toxytricini, the effect of medium components on the lipstatin production was investigated. Using TSB medium as a basal medium, a variety of carbon sources, nitrogen sources, lipid and fatty acids was supplemented into a fermentation medium. The seed culture of S. toxytricini grown in 25 ml TSB medium at $28^{\circ}C$ for 3 days with agitation at 200 rpm was inoculated in the size of 2% in fermentation media containing different components and fermented at $28^{\circ}C$ for 60 more hrs. In the examination of the effect of carbon sources, the best cell growth was observed in fermentation media supplemented with glucose or glycerol, but the lipstatin productivity was the highest in media containing lactose or sucrose. Among complex nitrogen sources, yeast extract was the best one for cell growth, but the highest lipstatin production was found in TSB media composed of 1.7% casitone and 0.3% soytone. The increased concentration of triolein as a lipid caused the promotion of cell growth but the significant suppression of lipstatin production. When 0.5% fatty acids were supplemented to fermentation medium, unsaturated fatty acids like linoleic or oleic acid suppressed cell growth as well as lipstatin production, but 2 times higher lipstatin production was achieved by stearic acid, a saturated fatty acid, differently from expectation.

  • The medium optimization was investigated to maximize the production of fibrinolytic enzyme by Bacillus subtilis KCK-7 isolated from Chungkookjang, which could hydrolyze the fibrin produced through the blood coagulation mechanism in human body. The simultaneous addition of 5% soluble starch and 0.5% cellobiose to the medium as carbon sources resulted in the highest production of the fibrinolytic enzyme. Likewise, the optimized composition of medium appeared to be 0.5% peptone, 0.3% beef extract, 0.5% cellobiose, 5% soluble starch, 2% raw soybean meal and 0.02% Na$_2$HPO$_4$. In addition, the fibrinolytic enzyme production by Bacillus subtilis KCK-7 reached to the maximum level after the cultivation for 48 hr, using the optimized medium.

  • In this paper, the electrical values of resistance, current, and voltage condition of reactivity is analyzed by applying the direct current (DC) voltage in medium for stem cell regeneration treatment. The voltage response by medium is related to electrical stimulation in the process of induction of differentiation for stem cell and the differentiation condition can be checked depending on the response of voltage condition. If the voltage level is lower in reacting response of a medium, the stem cell stimulation condition is stable, and if the voltage changing level is higher, the stem cell stimulation condition is unstable and a considerable loss will be resulted in the differentiation process. In this research, the optimization of electrical stimulation condition is expected for possible stem cell regeneration treatment.

  • Media optimization for the production of MR-387A and B, novel aminopeptidase M inhibitors by Streptomyces sp. SL-387 isolated from a soil was studied. Optimized medium was consisted of 1% glucose, 3% soybean meal, 0.2% yeast extract, 0.1% beef extract, 0.3% NaCl, 0.01% $K_2HPO_4$, 0.3% $CaCO_3$, 0.001% $MnCl_2{\cdot}4H_2O$, 0.001% $ZnCl_2{\cdot}7H_2O$, and 0.0005% $MgSO_4{\cdot}7H_2O$, and adjusted to pH 7.0 before autoclaving. When the optimized medium was used as a fermentation medium, maximum productivity of MR-387 was reached at 120 hours of fermentation, and total productivity was 909.1 U/ml.

Optimization of Culture Conditions for the [+]-Eudesmin Production in Magnolia Sieboldii Cells (함박꽃나무의 현탁배양세포로부터 [+]-Eudesmin의 생산을 위한 최적화)

  • In order to product the furofuranoid lignans, (+)-eudesmin which is one of the secondary products from Magnolia sieboldii. through cell suspension cultures; various culture media, initial sucrose concentration, elicitations, shaking speeds, and inoculum sizes. Among the culture media tested, MS medium had a pronounced effect on suspension cell growth and (+)-eudesmin contents. The maximum dry cell weight (DCW) of 3.71 g per flask was obtained at inoculum size of 0.5 g and in MS medium supplemented with $3\%$ sucrose plus 0.5 mg/L 2,4-D after 8 weeks. (+)-Eudesmin biosynthesis was stimulated with high initial sucrose concentration ,and the maximum (+)-eudesmin production of $3.2{\mu}g/g$ DCW was achieved at 200mg/L chitosan and $5\%$ initial medium sucrose. The optimal shaking speeds for dry biomass accumulation and (+)-eudesmin contents was 130 rpm. This work is considered to be helpful for large-scale bioprocessing of Magnolia sieboldii suspension cell cultures in bioreactor.

  • In this study, media optimization by statistically designed experiments stimulated an increase in cell growth of Bacillus sp. during batch cultivation. Plackett-Surman design method selected 3 components among 7 components of production medium. Box-Behnken design method calculated the optimum concentration of selected components by Plackett-Surman design. In the optimized medium, viable cell number increased 2 times.   • Bacterial strains showing the firinolytic and $\beta$-glucosidase activity were screened from Doeniang. The strain of KH-15 revealed a high level of fibrinolytic and $\beta$-gluocosidase activity. The isolated bacterium was identified and desingnated as Bacillus sp. KH-15. The carbon, nitrogen and salts sgnificantly influenced te fibrinolytic enzyme and $\beta$-glucosidase production. The optimized composition of medium appeared to be 2% glucose, 0.5% yeast extract and 0.1% calcium chloride. The optimum pH and temperature for fibrinolytic enzyme and $\beta$-glucosidase activities were pH 7∼8, 4$0^{\circ}C$ and pH 6∼8, 30∼4$0^{\circ}C$, respectively.

Production Medium Optimization for Monascus Biomass Containing High Content of Monacolin-K by Using Soybean Flour Substrates (기능성 원료를 기질로 이용하는 Monacolin-K 고함유 모나스커스 균주의 생산배지 최적화)

  • During the last decade, monacolin-K biosynthesized by fermentation of red yeast rice (Monascus strains) was proved to have an efficient cholesterol lowering capability, leading to rapid increase in the market demand for the functional red yeast rice. In this study, the production medium composition and components were optimized on a shake flask scale for monacolin-K production by Monascus pilosus (KCCM 60160). The effect of three different soybean flours on the monacolin-K production were studied in order to replace the nitrogen sources of basic production medium (yeast extract, malt extract and beef extract). Among the several experiments, the production medium with dietary soybean flour to replace a half of yeast extract was very good for monacolin-K production. Plackett-Burman experimental design was used to determine the key factors which are critical to produce the biological products in the fermentation. According to the result of Plackett-Burman experimental design, a second order response surface design was applied using yeast extract, beef extract and $(NH_4)_2SO_4$ as factors. Applying this model, the optimum concentration of the three variables was obtained. The maximum monacolin-K production (369.6 mg/L) predicted by model agrees well with the experimental value (418 mg/L) obtained from the experimental verification at the optimal medium. The yield of monacolin-K was increased by 67% as compared to that obtained with basic production medium in shake flasks.
