  • This study was undertaken to observe the mutagenic occurrence in urine excreted after the ingestion of roast beef. Two healthy nonsmoker persons of both sex were selected for this test, employing two strains(TA98, TA100) of Salmonella typhimurium according to Ames' method. The mutagenic activity began to appear in urine of both sex three hours after ingestion of 300g of roast beef, gradually increasing until 6 hours and declining thereafter.

  • Serum albumin은 혈청 단백질 중 50-60%를 차지한다. 알부민은 순환계 내에서 삼투압을 유지시켜 세포내외의 체액을 유지 및 지용성 물질치 이동에 관여한다. 신사구체와 기저막의 투과성이 증가하거나 세뇨관의 흡수감소 등으로 단백질의 요배설량이 하루에 150mmg 이상되는 단백뇨는 신장내의 병변이 있음을 나타내는 중요한 지표이다. 이러한 단백뇨는 크게 사구체성 단백뇨, 세뇨관성 단백뇨, 혼합성 단백뇨로 나눌수 있는데 이중 사구체 기저막의 손상으로 발생되는 사구체성 단백뇨는 알부민과 같은 고분자량의 단백질이 소변 내로 배설되는 현상이다. 일반적으로 알부민이 정상치 이상으로 소변을 통하여 배설되는 것을 'microalbuminuria'라고 하며 당뇨병으로 인한 신장병변을 예견하는데 중요한 지표로 알려져 있다. Microalbuminuria는 방사 면역확산법, 방사성 면역측정법, 면역혼탁측정법, 비탁측정법, 효소결합면역측정법(ELISA) 등으로 검출할 수 있으나 최근에는 안전하면서도 민감도가 높고 측정이 용이한 방법인 ELISA를 이용한 면역분석방법이 많이 사용되고 있다. 면역특이성이 떨어지는 다세포군 항체를 사용한 방법의 문제점을 극복하기 위해서 세포융합기술을 이용한 단세포군항체를 생산하는 세포주를 개발하였고 그 특성을 규명하였다. 따라서 본 연구에서 세포융합기술을 도입하여 개발된 인간 혈청 알부민에 대한 단세포군항체는 glycohemoglobin에 대한 단세포군항체와 더불어 당뇨병의 진행 정도를 진단할 수 있는 키트의 원료로 사용될 수 있다.

  • This study has been described the metabolism and excretion in a healthy male urine collected for 24 hr after oral administration of a complex (pseudoephedrine and dextromethorphan). To detect the trace amounts of parent drugs and their metabolites, acid-hydrolyzed urine was extracted and derivatized with MSTFA and MBTFA followed by gas chromatography/mass spectrometric analysis. Two parent drugs and their metabolites were tentatively identified as their derivatives based on the mass spectral interpretation and compared with previous reports. In addition, the time profile of urinary excretion rate for parent drugs and metabolites was studied. On the basis of metabolites identified and excretion rate, the metabolic pathways of both drugs are suggested.

Effects of Cecal Ligation and Colostomy on Food and Water Intake and loafer Excretion in Chickens Fed Restrictedly and Freely (닭에 있어서 사료섭취의 자유 및 제한급여시킬 때의 사료섭취량, 음수량 및 수분 배설량에 미치는 맹장결찰 및 인공항문 수술의 효과)

  • It was examined whether the ceca and the back-flow of urine into the lower intestine are involved in water intake and excretion in chickens and food intake affects those. Colostomy significantly increased water intake, total water excretion and the ratio of the water intake to food intake in the ceca-ligated chickens under restrict and ad libitum feeding conditions (P<0.05), but the increases were much larger in chickens fed ad libitum than in those fed restrictedly. Cecal ligation increased water intake, total water excretion and the ratio of water intake to food intake in the colostomised chickens which were fed freely (P<0.05). but not in those fed restrictedly, None of colostomy and cecal ligation affected the resultant water balances in chickens under both feeding conditions. Colostomy increased food intake in the ceca-ligated chickens (P<0.05), but no increase by cecal ligation was observed in colostomised chickens. It is concluded that the lower intestine takes a very important role in water recovery from urine to maintain water balance in chickens.

Effects of Different Levels of CP Intake on Protein Utilization and N Excretion in Varying Growth Stages of Hanwoo Steers (한우 거세우의 성장단계별 CP 급여수준이 단백질 체내 이용성 및 N 배설량에 미치는 효과)

  • Three experiments with different stage of Hanwoo steers (average BW of 167kg, 355kg and 449kg, respectively) were conducted to examine the effect of different quantities of CP intake with similar DM intake on protein balance and N excretion. Twelve Hanwoo steers in each experiment were offered three diets containing three levels of protein (9, 14 and 18% on DM basis) in the concentrate formulated with using different amount of ground corn grain or corn gluten meal. There were significant effects of increasing CP intake on CP digestibility but not on DM digestibility. Despite different CP intake, apparent fecal N outputs were not significantly affected. However, amounts of N excreted into urine increased (p<0.05) with increasing CP intake. There was an obvious effect of live body weight on efficiency of body protein conversion of CP consumed in excess of animal requirement for maintenance, with slopes of 70% and 46~39% for growing and fattening stage, respectively. Nitrogen excretion into feces and urine in this experiment was 70% of the daily N consumption for fattening stage, compared with 60% for growing stage.

Influence of Intracerebroventricular Ketanserin on Rabbit Renal Function (가토신장기능에 미치는 뇌실내 Ketanserin의 영향)

  • 5-Hydroxytryptamine (5-HT) was reported to elicit natriuresis and diuresis when given intracerebroventricularly (icv) and these effects were shown to be abolished by icv methysergide, $5-HT{_1}$ antagonist, thus suggesting that central tryptaminergic system may also participate in the regulation of renal function. We tried in this study to elucidate the role of $5-HT_2$ receptors in the central tryptaminergic regulation of renal function, observing the effects of icv ketanserin, a specific $5-HT_2$ antagonist. Ketanserin (KET) icv in doses of $120{\mu}g$ $(=0.3\;{\mu}moles)/kg$ produced significant natriuresis without affecting renal hemodynamics, indicating that it resulted from decreased tubular Na reabsorption. Systemic blood pressure decreased slightly but significantly. When given iv, no significant effect was observed. 5-HT, $200{\mu}g/kg$ icv, produced mild but significant natriuresis and diuresis. However, after KET, $40{\mu}\;g/kg$ icv, a dose which minimally affects renal function, the natriuresis and diuresis by 5-HT was greatly augmented, with the fractional excretion of filtered sodium reaching 9.3%. The renal effects of other biogenic amines administered icv, such as norepinephrine, dopamine and histamine, were not significantly affected by the KET pretreatment. These observations suggest that central tryptaminergic system influences renal function in dual ways, i.e., natriuretic and diuretic influence via $5-HT_1$ receptors, whereas $5-HT_2$ subtypes mediate the antinatriuretic and antidiuretic effects, and that the central tryptaminergic system plays a role in the regulation of rabbit renal function.

A Study on the Amount and Major Compositions of Excreta from Broilers (육계의 분뇨 배설량 및 분뇨 성분 조사 결과)

  • This work was conducted to investigate the amount and major compositions of excreta from broilers. One hundred fifty 1-d-old male and female Ross broilers were used in this work and ten broilers were selected to measure excreta. Average amount of excreta per broiler was 85.4 g, and average moisture content was 76.9%. N, $P_2O_5$ and $K_2O$ in excreta were 1.19, 0.29 and 0.49%, respectively. pH, $BOD_5$, $COD_{mn}$, and SS were 7.80, 42,134 mg/L, 52,668 mg/L, and 106,669 mg/L, respectively. Pb in poultry excreta was 0.803 mg/kg and highly lowed compared with the standard contents (150 mg/kg). Cd was not founded in excreta. Hg was founded in excreta as 0.002 mg/kg, but it was low compared with the standard level (2 mg/kg).

Effect of Corticosteroids on Renal Excretion of Lithium (Lithium 이온의 배설에 미치는 Corticosteroid의 영향)

  • Lithium salts are being used increasingly to treat patient with affective disorders, especially acute mania, or bipolar manic-depressive illness. For therapeutic effect the lithium content must be maintained at or above a particular level. Lithium poisoning due to overdosage may be seen occasionally, and its course is determined primarily by the rate of renal lithium elimination. A search is therefore indicated for procedures that could raise the lithium clearance. In a number of reports renal lithium excretion has been studied in relation to the excretion of water, sodium, potassium and hydrogen, but effects of sodium or water on the lithium excretion has not yet been clarified. Hence the present study was undertaken to investigate the effects of corticosteroid on the excretion of lithium ion. The female rat(Sprague-Dowley), weighing from 200 to 300g, was injected with 50mg/kg of lithium chloride intraperitoneally, and then injected with graded dosage of fludrocortisone and dexamethasone in each group. During the injected rats were incubated in metabolic cage, 24 hour urine of rats were collected. At 24 hours after injection, the rats were sacrificed with guillotin, the blood were collected. And then the concentratios of $Na^+$, $K^+$, $Li^+$ of collected urine and serum were checked by Flame photometer. The results are summarized as follows; 1. Fludrocortisone decreased the serum concentration of lithium and increased the urinary excretion of lithium. 2. In the group treated with low dose of dexamethasone(0.1mg/kg), the serum concentration of lithium was decreased and high dose of dexamethasone (1mg/kg) increased the urinary excretion of lithium. 3. Fludrocortisone increased the urinary $[Na^+]/[K^+]$ in serum and decreased $[Na^+]/[K^+]$ in urine, but opposite effects were occurred in dexamethasone. By above results, it may be concluded that corticosteroid increased the urinary excretion of lithium and decreased the serum concentration of lithium, but it seems to be there is no relationship between these effects of corticosteroid and of the renal $Na^+$ or $K^+$ transport.

Oxygen Consumption, Operculum Movement Number and Ammonia Excretion of Cultured Tilapia (Oreochromis mossambicus) by Salinity Changes (염분 변화에 따른 자바틸라피아(Oreochromis mossambicus)의 산소소비, 아가미 호흡수 및 암모니아 배설)

  • The effects of salinity on oxygen consumption, operculum movement number and ammonia nitrogen excretion were examined in cultured tilapia, Oreochromis mossambicus (total length 10$\pm$1.0 cm, total weight 17.3$\pm$4.2 g). The fish were exposured to certain salinities (0, 3, 9, 15, 21, 27 and 32 psu) step by step with 3 psu$\cdot$d$^{-1}$ and changed rapidly from certain salinity to another salinity. The oxygen consumption of the fish which was transferred step by step showed increasing tendency in the range of 3 $\sim$ 27 psu, but markedly appeared very low value at 32 psu. The tolerance limit in tilapia by salinity change without acclimation was shown 23.3 psu from 96-h TL$_{50}$. The oxygen consumption, operculum movement and ammonia nitrogen excretion of fish which was transferred rapidly from certain salinity to each salinity (0, 9, 15, 21 and 32 psu) showed a changing point at 15 psu; they showed increasing and/or decreasing pattern before 15 psu, and showed decreasing pattern after 15 psu. From these results, it was concluded that the appropriate salinity without physiological change for Java tilapia was below 15 psu.

Effects of Dietary Zizyphus jujuba Seed Meal on Broiler Performance (대추씨(Zizyphus jujuba Seed) 급여가 육계의 생산성에 미치는 영향)

  • The study was conducted to investigate the effect of dietary Zizyphus jujuba seed meal on broiler performance, small intestine and cecal length, gizzard weight and fecal gas emission. A total of 180 day-old broiler chicks (Ross) were randomly divided into 4 groups, fed the commercial diet containing 0, 0.3 (T1), 0.6 (T2) and 0.9 % (T3) of Zizyphus jujuba seed meal from two to 5 weeks of age. Body weight gain and feed efficiency (feed/gain) were significantly increase in T2 than other groups at 3 weeks age (P<0.05). There was no statistical difference in body weight gain and feed efficiency (feed/gain) by feeding Zizyphus jujuba seed meal, although they tended to be improved by treatment during whole experimental period (5 weeks age). The gizzard weights of three treatment groups were higher than control. But no difference among three treatment groups was observed. The small intestine and cecal length was no change by feeding Zizyphus jujuba seed meal. Emission of $NH_3$ and VFA gas from excreta were significantly decreased from 5 to 10 days of storage period (P<0.05). In conclusion, the results of this study showed that the addition of Zizyphus jujuba seed meal at 0.3 to 0.6 % to the diet has increased digestibility of feed and the reduction of $NH_3$ emissions from feces, there was a possibility of improvement in body weight gain of broiler chickens.
