A Research on Superparamagnetic Iron Oxide Nanoparticles' Toxicity to U373MG Cell and its Effect on the Radiation Survival Curve (산화철 나노입자의 U373MG 세포 독성평가 및 방사선 세포생존 곡선에 미치는 영향에 대한 연구)

  • This research was performed to evaluate the superparamagnetic iron oxide nanoparticles'(SPIONs) cell toxicity and to measure the radiation cell survival curve changes of SPIONs-uptake glioblastoma multiforme cells. The results could be practically used as the fundamental data to ameliorate proton beam cancer therapy, for example, providing necessary GBM treatment dose in the proton beam therapy when the therapy takes advantage of SPIONs. The assessment of the toxicological evaluation of synthesized SPIONs was accomplished by MTT assay as an in vitro experiment. The results showed no meaningful differences in the cell survival rate at the $1-100{\mu}g/ml$ SPIONs concentrations, but the cell toxicity was shown as the cell survival rate decreased up to 74.2% at the $200{\mu}g/ml$ SPIONs concentration. Then, we measured each radiation cell survival curve for U373MG cells and SPIONs-uptake U373MG cells with 0~5 Gy of proton beam irradiations. It is learned from the analysis of the experimental results that the SPION-uptake cells' radiation survival rate was more rapidly decreased as the irradiation dose increased. In conclusion we confirmed that SPIONs-uptake in U373MG cells induces cell death at the much less dose than the lethal dose of SPION-non-uptake cell. This research shows that the therapeutic efficacy of glioblastoma multiforme treatment in proton beam therapy can be improved by SPIONs targeting to the GBM cells.

  • International Commission on Radiological Protection (ICRP) has revised its recommendations concerning the tissue reaction to ionizing radiation in accordance with consideration of the detriment arising from non-cancer effects of radiation on health based on recent epidemiological basis. Particularly, for the lens of the eye, the threshold in absorbed dose revised to be 0.5 Gy, for occupational exposure in planned exposure situation the commission recommended "An equivalent dose limit for the lens of the eye of 20 mSv in a year, averaged over defined periods of 5 years, with no single year exceeding 50 mSv." To monitor the radiation exposure of radiation worker, TLD is typically provided and the lens of eye dose can be assessed by run of dose calculation algorithm with TL element response data. This study is to assess equivalent dose of the lens of eye using the Harshaw TLD system and its two different dose calculation algorithms. The result provides the Harshaw TLD system showed the assessment of the lens of eye dose with 48.84% error range.

Effectiveness of Fentanyl Transdermal Patch (Fentanyl-TTS, $Durogegic^{(R)}$) for Radiotherapy Induced Pain and Cancer Pain: Multi-center Trial (방사선치료로 인한 통증 및 암성통증에 대한 듀로제식의 효과: 다기관연구)

  • $\underline{Purpose}$: To evaluate the effectiveness and safety of fentanyl-TTS in the management of radiotherapy induced acute pain and cancer pain treated with radiotherapy. $\underline{Materials\;and\;Methods}$: Our study was open labelled prospective phase IV multi-center study. the study population included patients with more 4 numeric rating scale(NRS) score pain although managed with other analgesics or more than 6 NRS score pain without analgesics. Patients divided into two groups; patients with radiotherapy induced pain (Group A) and patients with cancer pain treated with radiotherapy (Group B). All patients received 25 ug/hr of fentanyl transdermal patch. Primary end point was pain relief; second end points were change in patient quality of life, a degree of satisfaction for patients and clinician, side effects. $\underline{Results}$: Between March 2005 and June 2005, 312 patients from 26 participating institutes were registered, but 249 patients completed this study. Total number of patients in each group was 185 in Group A, 64 in Group B. Mean age was 60 years and male to female ratio was 76:24. Severe pain NRS score at 2 weeks after the application of fentanyl was decreased from 7.03 to 4.01, p=0.003. There was a significant improvement in insomnia, social functioning, and quality of life. A degree of satisfaction for patients and clinician was very high. The most common reasons of patients' satisfactions was good pain control. Ninety six patients reported side effect. Nausea was the most common side effect. There was no serious side effect. $\underline{Conclusion}$: Fentanyl-TTS was effective in both relieving pain with good tolerability and improving the quality of life for patients with radiotherapy induced acute pain and cancer pain treated with radiotherapy. The satisfaction of the patients and doctors was good. There was no major side effect.

Change in tooth length and angulation on panoramic radiographs taken at different labiolingual and buccolingual inclinations (치아의 순(협)설 경사도 변화에 따라 파노라마 방사선 사진에 나타난 치아 길이 및 각도 변화)

  • Objective: The purpose of this study was to examine how the mesio-distal angulation and the length of each tooth changes on panoramic radiograph at different bucco-lingual inclinations. Methods: After constructing an acrylic model based on the mean arch of 30 adults with normal occlusion, the wire was placed in the center of the teeth on the acrylic model. First, the wire was implanted in normal angulation and inclination and a panoramic radiograph taken. After changing the inclination from $I-5^{\circ}\;to\;I+15^{\circ}\;by\;5^{\circ}$, a panoramic radiograph was taken again and the mesio-distal angle and wire length on the panoramic radiograph were assessed. Results: When the wire was implanted at the normal angulation and inclination, the length measured in the panoramic radiograph was magnified $111{\sim}117%$ from the original length in the anterior region and $121{\sim}125%$ in the posterior region. Only the central and lateral incisors showed significant length differences when the inclination was changed from $l-15^{\circ}\;to\;I+15^{\circ}$ at fixed angulation. When the inclination was changed from $l-15^{\circ}\;to\;I+15^{\circ}$, the angulation of most teeth on panoramic radiograph appeared to be more disto-angulated than in reality, and the lateral incisor and canine showed the largest difference. Only $l-15^{\circ}\;to\;I+15^{\circ}$ groups of premolars and $I+15^{\circ}$ group of molars showed more mesio-angulation than in reality. As the labio(bucco)lingual inclination of all teeth were decreased, tooth angulation in the panoramic radiograph appeared to be more disto-angulated. Conclusion: The labio-liugual inclination of teeth should be considered because it affects panoramic image of teeth, such as length of incisors and angulation of other teeth.

Effect of Tumor Hypoxia on Efficacy of Tirapazamine Combined with Fractionated Irradiation in Mouse Tumor (마우스종양에서 분할방사선조사와 병용된 Tirapazamine의 효과에 미치는 종양 저산소상태의 영향)

  • Purpose : Tumor hypoxia can be overcome with hypoxic cytotoxin. In mouse tumor, tirapazamine's efficacy of the potentiating radiation effect was tested by the tumor oxygenation status combined with hype facti on ated rad iotherapy .:The control and hypoxic mouse tumors we established by inoculation of RIF-1 tumor cells into the normal or previously irradiated back and thigh of C3H mice. When the tumors reached a proper size, both the control and hypoxic tumors were given hypefractionated treatments (8fractions/4 days) with saline (0.02 ml/g), tirapazamin (0.08 mM/0.02 ml/kg), irradiation (2.5 Gy), irradiation combined with tirapazamine given 30 minutes prior to each irradiation. The response was evaluated by the growth delay assay by measuring tumor size from day 0 (12 hrs prior to the first fractionation) to the day when the volume had 4-fold increase or cross sectional area had 2-fold increase. Results : Overall growth pattern showed that tirapazamine Potentiated radiation effect in back and thigh tumors grew in the normal and preirradiated tumor bed. With growth delay assay using reference point of initial tumor volume or cross sectional area, tirapazamine potentiated radiation effect 1.9 times for the control and 2.4 times for the hypoxic tumors in back, and 1.85 times for the control and 1.6 times for the hypoxic tumors. With reference of 4-fold increase of the initial volume or 2-fold increase of the cross sectional area, tirapazamine potentiated radiation effect 1.48 times for the control and 2.02 times for the hypxic tumors in back, and 1.85 times for the control and 1.6 times for the hypoxic tumors. Conclusions : Present result indicated that radiation response of hypoxic tumors was potentiated by tirapazamine in the back or thigh tumors grew in the control or preirradiated tumor bed, and potentiation of the hypoxic tumors was eDual to or greater than that of the control tumors in the back or thigh.

  • Placenta accrete patients whose mother mortality rates are rather high due to massive bleeding during childbirth need to have Prophylactic placement of Internal Iliac Artery Balloon Occlusion Catheters procedure to reduce amount of blood loss and inoperative transfusion. Nevertheless, studies for mothers inevitably exposed to dose during PIIABOCs procedure have not been published many yet. Therefore, this study is to investigate exact information on radiation dose exposed to fetus during PIIABOCs procedure. Average effective dose of fetus per organ is 2.38~8.83 mGy, measured highest at beam center and followed by eyeball, stomach and bladder. The result showed that the longer fluoroscopy time is used, the closer beam center is and the thicker abdominal thickness is, the more effective dose on fetus is increasing. When using the collimator and protection shown to decrease the effective dose and when using higher the patient table shown to decrease the effective dose. It has been reported that the threshold of deterministic effect is about 100mGy. Deterministic effect was regarded as a factor that would influence on fetus exposed by medical radiation than stochastic effect. Consequently, it concluded that dose exposed on fetus in PIIABOCs procedure was approximately 10% of threshold of deterministic effect with effective dose of 0.49~18.27 mGy.

Educational Effects of Self-directed Learning Method Using 3D Printing Products on Radiological Science Students (3D 프린팅 구조물을 이용한 자기주도 학습방법이 방사선학과 학생들에 미치는 교육 효과)

  • The purpose of this study was to analyze the educational effect of self-directed learning method using 3D printed anatomy on radiological science students. The subjects were 32 students (20 males and 12 females) in the second year of radiological science at university. They were divided two groups as a non-active student group and an active student group. A learning method was self-directed learning using 3D printed anatomical structures, and the effects of quantitative learning improvement were evaluated before and after the learning. The qualitative evaluation of the students was analyzed on the Likert's 5-point scale for the interest, satisfaction, and learning effects (memorization convenience of anatomy name, radiography Interpret ability, understanding on bones structure, and X-ray projection technique). As a result, the enhancement of learning improved 65.4% on average, and all students got scored high on all variables. Especially non-active student groups showed higher correlation coefficients in all variables except interest and radiography interpret than active student groups. These results might suggest that self-directed learning using 3D printed anatomical structures could have a positive educational effect on radiological science students.

Comparison of Setup Deviations for Two Thermoplastic Immobilization Masks in Glottis Cancer (성문암 세기변조방사선치료에서 두 가지 열가소성 마스크에 대한 환자위치잡이 오차 평가)

  • The purpose of this study was compare to the patient setup deviation of two different type thermoplastic immobilization masks for glottis cancer in the intensity-modulated radiation therapy (IMRT). A total of 16 glottis cancer cases were divided into two groups based on applied mask type: standard or alternative group. The mean error (M), three-dimensional setup displacement error (3D-error), systematic error (${\Sigma}$), random error (${\sigma}$) were calculated for each group, and also analyzed setup margin (mm). The 3D-errors were $5.2{\pm}1.3mm$ and $5.9{\pm}0.7mm$ for the standard and alternative groups, respectively; the alternative group was 13.6% higher than the standard group. The systematic errors in the roll angle and the x, y, z directions were $0.8^{\circ}$, 1.7 mm, 1.0 mm, and 1.5 mm in the alternative group and $0.8^{\circ}$, 1.1 mm, 1.8 mm, and 2.0 mm in the alternative group. The random errors in the x, y, z directions were 10.9%, 1.7%, and 23.1% lower in the alternative group than in the standard group. However, absolute rotational angle (i.e., roll) in the alternative group was 12.4% higher than in the standard group. For calculated setup margin, the alternative group in x direction was 31.8% lower than in standard group. In contrast, the y and z direction were 52.6% and 21.6% higher than in the standard group. Although using a modified thermoplastic immobilization mask could be affect patient setup deviation in terms of numerical results, various point of view for an immobilization masks has need to research in terms of clinic issue.

Comparative Study on Human Risk by Ionizing Radiation and Pesticide as Biological Information about Environmental Disaster (환경재해에 관한 생물정보로서의 이온화 방사선과 살충제의 인체 위해성 비교 연구)

  • Environmental risk factors such as ionizing radiations, heavy metals, and pesticides can cause environmental disasters when they exist in excess. The increases in use of ionizing radiation and agricultural pesticide are somewhat related to the possibility of the disaster. The risk of radiation and pesticide was evaluated by means of the single cell gel electrophoresis (SCGE) assay on the human blood lymphocytes. The lymphocytes were irradiated with $0{\sim}2.0Gy$ of $^{60}Co$ gamma ray. Another groups of lymphocytes were exposed to various concentrations of parathion. Significantly increased tail moment, which was a marker of DNA strand breaks in SCGE assay, showed a clear dose- or concentration-response relationship. Parathion of a recommended concentration for agricultural use ($1mg {\ell}^{-1}$ ) has a strong cytotoxic effect on lymphocytes, which is equivalent to damage induced by 0.1 Gy of ${\gamma}$-ray. Furthermore, $2mg{\ell}^{-1}$ of parathion can give rise to DNA damage equivalent to that induced by 0.25 Gy at which the radiation-induced damage can start to develop into clinical symptoms. The comparative results of this study can provide an experimental basis and biological information for the prevention of environmental disaster.

  • We tried to assessment about characteristics of image through quantitative evaluation method and qualitative evaluation method in Full Field Digital Mammography. It satisfied an approval standard of ten score regardless of compression ratio measuring detection score after compressing and appling an algorithm of JPEG2000 orJPEG compression targeting ACR accreditation phantom. Also, it was apparent that when we selected and compressed the image of real fine lesion and measured a change of diagnosis ability magnifing over 50 percent after compressing over 20:1 ratio, it had a strong influence on diagnosis ability. We realized that the difference between the original image according to compression ratio measuring a quantitative evaluation which is PSNR,RMSE,MAE and SSIM was relatively allowable.
