혈관내벽에 홀뮴-166 방사선 분할 조사시 흡수선량 분포

  • 경피적관상동맥성형술(CPTCA)이나 스텐트삽입술 후에 발생하는 재협착을 방지하기 위한 방사선을 조사하는 방법 중에 베타 입자를 방출하는 액체 선원을 catheter풍선 내에 넣어 일정 시간 방사선 조사 시키는 방법이 있다. 조사시킬 혈관의 길이가 길어 한번의 방사선 조사가 어려울 경우 영역을 분할하여 두 번에 나누어 조사할 경우가 있다. 조사영역의 겹치는 부근의 흡수선량이 고선량이나 저선량이 되는가를 알기 위하여 두 풍선간의 접근 거리에 따른 혈관내벽의 흡수선량 분포를 알아보았다. 풍선내의 액체 선원은 Ho-l66을 이용하였고 Ho-l66의 물리적 반감기는 26.8시간이고 최대에너지 1.85 MeV, 평균에너지 0.69 MeV와 최대에너지 1.77 MeV, 평균에너지 0.65 MeV를 갖는 베타 입자를 방출한다. Ho-l66 의 방사선 흡수선량을 측정하기 위하여 GafChromic 필름(Nuclear Associates, Carle Place, NY, USA)을 이용하였고, 방사선이 조사된 필름의 optical density는 videodensitometer(Wellhofer, Schwarzen-bruck, Germany)를 이용하여 값을 읽었다. Catheter 풍선은 직경이 3 mm 이고 길이가 20 mm인 것을 이용하였다. 혈관 내벽의 최대 흡수선량을 표준화하여 겹치는 부분의 흡수선량 분포를 접근 거리에 따라 구하였다. 또한 몬테카를로 시abf레이션으로 확인하였다. 두 풍선의 겹치는 부근의 선량 분포는 풍선 중앙에서 중앙사이의 거리가 21 mm 일 때 중앙에서 20% 증가하였고, 거리가 22 mm일 때와 23 mm일 때 각각 10%와 40%의 감소를 보였다. 풍선 도자의 풍선 안에 베타입자 방출 액체 선원을 넣어 혈관내벽에 방사선 조사하는 방법은 비정거리가 짧아 혈관 내벽 부근에만 방사선을 조사시키고 그 외 중요 장기에는 영향을 덜 미치는 장점이 있다. 그러나 혈관 내벽 표면으로 부터의 거리에 따라 흡수선량이 급격히 떨어지는 분포를 이루기 때문에 두 개의 풍선이 겹치는 부근의 흡수선량은 아주 작은 접근 거리에서도 급격한 변화를 보였다. 따라서 시술 중에 겹치는 부분을 아주 적게 분할하여 정확하게 차례차례로 조사시키기 위해서는 신중한 거리 조정을 하여야 한다.

The Effect of Low Dose Radiation on the Neuronal Cell Proliferation in Diabetic Rats (당뇨병 유발 쥐에서 신경 세포 증식에 대한 저선량 방사선의 효과)

  • Purpose: To investigate the effect of low dose radiation on neuronal cell proliferation In diabetic rats. Materials and Methods: A group of rats (first group) were divided into three subgroups (nondiabetic control, nondiabetic 0.1 Gy and nondiabetic 10 Gy groups) to determine the effect of radiation on normal hippocampal neuronal ceil proliferation. A further group of rats (second group) were divided into six subgroups (nondiabetic control, diabetic control, diabetic 0.01 Gy, diabetic 0.1 Gy, diabetic 1 Gy and diabetic 10 Gy groups) to determine the effect of radiation on hippocampal neuronal cell proliferation under diabetic conditions. Using immuno-histochemistry for 5-bromo-2'-deoxyuridine (BrdU), the number of neuronal cells in the dentate gyrus of all the groups was counted. Results: The number of BrdU-positive cells in the dentate Gyrus of the nondiabetic control, nondiabetic 0.1 Gy and nondiabetic 10 Gy subgroups of the first group were $45.95{\pm}3.42,\;59.34{\pm}5.20\;and\;19.26{\pm}2.98/mm^2$, respectively. The number of BrdU-positive cells in the dentate gyrus of the diabetic control, diabetic 0.01 Gy, diabetic 0,1 Gy, diabetic 1 Gy and diabetic 10 Gy subgroups of the second group were $55.44{\pm}8.57,\;33.33{\pm}6.46,\;67.75{\pm}10.54,\;66.63{\pm}10.05,\;23.59{\pm}6.37\;and\;14.34{\pm}7.22/mm^2$, respectively. Conclusion: Low dose radiation enhances cell proliferation in the dentate gyrus of STZ-induced diabetic rats.

Identification of Germination Properties for the Screening of Gamma-irradiated Beans (방사선 조사된 콩류의 선별 검지를 위한 발아특성 확인)

  • Germination method was used to screen the biological changes in soybean, kidney bean, and red bean caused by gamma irradiation. Beans were irradiated at 0.1, 0.3, 0.5, 0.7, and 1.0 kGy. Ten beans of each sample were placed on moistened cotton and germinated at $30{\circ}C$. The root lengths were measured daily for 5 days. Root lengths of all beans grew continuously for 5 days, but the growth rate of irradiated beans decreased significantly from fourth day. Unirradiated beans showed the highest growth rate during 5 days of germination. Gamma-irradiated beans could be screened by measuring the daily growth rate and root length during germination.

Effect of the Paclitaxel and Radiation on the Large Bowel Mucosa of the Rat (횐쥐의 대장점막에 Paclitaxel(Taxol)과 방사선조사의 효과)

  • Purpose : Paclitaxel is a chemotherapeutic agent with a potent microtubule stabilizing activity that arrests mitosis at G2-M phase of cell cycle which is the most radiosensitive period. Therefore paclitaxel is considered as a cell cycle-specific radiosensitizer. This study investigates the effect of paclitaxel on the radiation response of the normal large bowel mucosa of the rat. Materials and Methods: The rats were divided into the three groups i.e., single intraperitoneal infusion of paclitaxel (10 mg/kg), a single fraction of irradiation (8 Gy, x-ray) to the whole abdomen, and a combination of irradiation (8 Gy, x-ray) given 24 hours after paclitaxel infusion. The histological changes as well as kinetics of mitotic arrest and apoptosis were evaluated on the large bowel mucosa at 6 hours, 1 day, 3 days and 5 days after treatment with paclitaxel alone, radiation alone and combination of paclitaxel and radiation. Results : The incidence of the mitotic arrest was not increased by paclitaxel infusion. The apoptosis appeared in 24 hours after paclitaxel infusion, and the histopathologic changes such as vesiculation, atypia and reduction of the goblet cell of the mucosa of the large bowel were demonstrated during the period from 6 hours to 3 days after, and returned to normal in 5 days after paclitaxel infusion. In irradiated group, the apoptosis was increased in 6 and 24 hours after irradiation, and the histopathologic changes of the mucosa were appeared in 24 hours and markedly increased in 3 days and returned to normal in 5 days. In combined group of irradiation and paclitaxel infusion, the apoptosis was appeared in 3 days and the histopathologic changes appeared during the period from 6 hours to 3 days after infusion. On the basis of the incidence of apoptosis and the degree of the histopathologic changes of the large bowel mucosa, there seemed to be additive effect by paclitaxel on radiation rather than sensitizing effect. Conclusions: The histopathological changes of large bowel mucosa in combined group compared to radiation alone group suggested an additive effect of paclitaxel on radiation response in the large bowel of rat.

Effect of High Protein Diet and Resveratrol Supplementation on the Nutritional Status and Immunoreactivity in the Irradiation-induced Inflammatory Rats (방사선 조사된 흰쥐에서 고단백식이와 레스베라트롤 첨가가 영양상태 및 면역기능 증진에 미치는 효과)

  • Most cancer patients are treated with surgery, chemotherapy or radiation as anticancer therapies. Especially in the case of radiation, these treatments produce adverse effects such as vomiting, weight loss, anorexia, normal cell damage and malabsorption. The major goal of this study was to determine the effect of irradiation on the nutritional and immune status in irradiated rats. A secondary goal was to determine the effectiveness of high protein diet (HP) and resveratrol (Res) in minimizing the adverse effects of radiation. Rats were divided into four groups: normal diet (NP), HP, NP + Res and HP + Res groups. Each group was further divided into subgroups that received radiation (RT group) and one that did not (non-RT group). Each diet was supplied from $12^{th}$ day prior to irradiation treatment with irradiation dose of 17.5 Gy. The diets were continued until 10th day after radiation treatment and animals were sacrificed. The radiation treatment showed decreased body weight, serum protein and HDL levels and increased TG and LDL levels in nutritional status. HP, NP + Res and HP + Res groups reduced the level of serum LDL and TG in irradiated rats. NP + Res and HP + Res groups increased reduced albumin level of serum in RT group. In case of immune status, the radiation treat-ment showed decreased WBC, lymphocytes and increased neutrophil and eosinophil levels. The levels of serum IL-2 and IL-6 were significantly increased by radiation, however the cytokine levels decreased in all dietary treatment groups. These results showed that high protein diet and resveratrol supplementation seem to minimize the adverse effects of radiation on lipid nutritional status and inflammation response in the rat model.

Gamma-ray-induced skin injury in the mini-pig: Effects of irradiation exposure on cyclooxygenase-2 expression in the skin (감마선조사에 의한 돼지 피부장애에 cyclooxygenase-2의 발현변화)

  • The basic concepts of radiation-induced skin damage have been established, the biological mechanism has not been studied. In this study, we have examined the effects of gamma rays on skin injury and cyclooxygenase(COX)-2 expression. Gamma irradiation induced clinicopathological changes in a dose- and time-dependent manner in mini-pig skin. The histological changes were consistent with the changes in gross appearance at 12 weeks after irradiation. After three days' irradiation, apoptotic cells in the basal layer were found more frequently in irradiated skin than in normal skin, with the magnitude of the effect being dose-dependent. The thickness of the epidermis transiently increased 3 days after irradiation, and then gradually decreased, although changes in the epithelial thickness of the irradiated field were not observed with irradiation doses over 50 Gy. In the epithelium, there was an initial degenerative phase, during which the rate of basal cell depletion was dependent on the radiation dose (20-70 Gy). One week after irradiation, COX-2 expression was mostly limited to the basal cell layer and was scattered across these cells. High COX-2 expression was detected throughout the full depth of the skin after irradiation. The COX-2 protein is upregulated after irradiation in mini-pig skin. These histological changes associated with radiation exposure dose cause the increased COX-2 expression in a dose-dependent fashion.

  • 양어용 사료로 사용되는 생어사료(고등어, 전어, 새우) 및 배합사료에 대한 방사선 조사(5 kGy) 후 조성분의 변화와, 사료의 방사선 조사에 의한 산패정도 및 BHA (200 ppm)의 효과를 저장온도 (2$0^{\circ}C$, 4$^{\circ}C$, -2$0^{\circ}C$) 및 저장기간 (0, 3, 7, 14일)에 따라 측정하였다. TBA가 및 과산화물가를 산패도 측정에 적용하였다. 5 kGy의 방사선 조사에서 사료의 조성분은 변화되지 않았다. 방사선 조사에 의한 초기 산패는 증가되었으며, BHA첨가에 따라 양어용 생어사료의 자연산패 및 방사선에 의한 초기산패는 억제되었으며 (p<0.05) 농림부에서 고시된 사료의 공정규격의 미량첨가 사료의 사용제한 범위 (300 ppm)내 에서도 충분한 산패억제 효과를 나타냈다.

  • Microbial reduction, physicochemical property, and sensory evaluation of irradiated beef patty were investigated. The microbial counts of refrigerated beef patty were reduced to below the number of 3 logs after irradiation at 3 kGy. But no viable microorganism was detected in frozen beef patty irradiated at 3 kGy. Food additives such as nitrite, salt, phosphate and ascorbic acid did not affect on the inactivation of microorganism by irradiation. The irradiation effect on the water holding capacity was not significant, but frozen irradiated beef patty showed higher water holding capacity than refrigerated beef patty. The drip loss of irradiated beef patty did not show significant differences according to irradiation doses. Considering the influence of food additives, the irradiated beef patty mixed with salt and phosphate showed lower drip loss than that without food additives. In refrigerated beef patty, TBARS values were increased with increase of irradiation doses and showed lower values in the beef patty mixed with food additives than that without food additives. The redness of refrigerated beef patty showed highest values at 3 kGy of irradiation and then decreased with increasing irradiation doses, while in the frozen beef patty did not show distinct tendency according to the irradiation doses or food additives. In sensory evaluation. the irradiated beef patty showed unpleasant smell as compared with the non irradiated beef patty, but showed somewhat higher score in smell at the sample contained ascorbic acid regardless of irradiation doses.
