Expression of Inhibin in the Whole-body γ-irradiated Mouse Ovary (감마선이 조사된 미성숙 생쥐 난소 내 인히빈의 발현 변화)

  • Objective: The purposes of the present study were to investigate the effect of ${\gamma}$-radiation on the expression of inhibin-${\alpha}$ proteins and genes for inhibin ${\alpha}$, ${\beta}A$, and ${\beta}B$ in the ovary. Methods: Immature mice were whole-body ${\gamma}$-irradiated with 25% of a lethal dose. At time 0, 3, 6, 12, and 24 hours after the irradiation, the ovaries were collected and used for immunohistochemistry for inhibin-${\alpha}$, and RT_PCR for inhibin-${\alpha}$, ${\beta}A$, and ${\beta}B$. Results: The expression of the immunoreactive inhibins-${\alpha}$ was maintained at 12 hours post-irradiation and reduced thereafter. The expression of inhibin-${\alpha}$ mRNA was significantly increased with the time after the irradiation. However there were no significant changes in the expression of ${\beta}A$ and ${\beta}B$ mRNAs. Conclusion: It might be thought that inhibin acts as one of the regulatory factors in the ${\gamma}$-radiation-induced follicular atresia in mice

Effect of Gamma Ray on Growth of Juvenile Abalone, Haliotis discus hannai (참전복(Haliotis discus hannai)의 성장에 대한 감마선 영향)

  • Radiation at very low doses frequently has a stimulating or hermetic effect on growth of organism. Effects of growth and survival rate on various dose of gamma irradiation in the farm culture of juvenile abalone (Haliotis discus hannai) were determined in Hallim (Jeju) from February 2004 to January 2005. The initial shell length of abalone juveniles in this study was average $3.45{\pm}0.4cm$. A change of growth after irradiation $(0{\sim}20Gy)$ was observed for 48 weeks. The highest growth rate was observed in 4 Gy-irradiated group and the lowest growth rate was observed in 20 Gy-irradiated group. The additional research about biochemical changes on juvenile abalone after irradiation should be accomplished. Continuous study for gamma radiation-induced hormesis on growth and metabolism of juvenile abalone will further induce the creation of value in ocean industry.

An Experimental Study of Radioprotective Effect of Ginseng Alkaloid Fraction on Cellular Damage (방사선 세포 손상에 대한 인삼 Alkaloid 분획의 보호 효과에 관한 실험적 연구)

  • This paper is to assess the effect of Adaptagen as a radioprotector in which main component is alkaloid fraction of ginseng. Evaluation was made in vitro and in vivo study with NIGP(S) mouse by the measurement of regeneration of jejunal crypt cell and micronucleus assay to analyze radioprotective effect of ginseng alkaloid fraction in comparison with that of water fraction after whole body irradiation. The results were as follows, 1. The degree of radiation damage of mouse jejunal crypt cell was diminished in both of alkaloid and water fraction groups compared to control group but more in alkaloid fraction group than water fraction group. Regeneration of mouse jejunal crypt cell was higher both in alkaloid and water fraction groups than control group. 3. In vitro study, frequency of micronucleus was diminished in tendency for the treated groups than control group but statistically insignificant. 4. In vitro study, frequency of micronucleus was diminished in both alkaloid and water fraction groups compared to control group but more in alkaloid fraction group than water fraction group.

Quality Properties of the Refrigerated or Frozen Irradiated Beef Patty (방사선조사된 패티용 분쇄우육의 가열전 품질특성)

  • Microbial reduction, physicochemical property, and sensory evaluation of irradiated beef patty were investigated. The microbial counts of refrigerated beef patty were reduced to below the number of 3 logs after irradiation at 3 kGy. But no viable microorganism was detected in frozen beef patty irradiated at 3 kGy. Food additives such as nitrite, salt, phosphate and ascorbic acid did not affect on the inactivation of microorganism by irradiation. The irradiation effect on the water holding capacity was not significant, but frozen irradiated beef patty showed higher water holding capacity than refrigerated beef patty. The drip loss of irradiated beef patty did not show significant differences according to irradiation doses. Considering the influence of food additives, the irradiated beef patty mixed with salt and phosphate showed lower drip loss than that without food additives. In refrigerated beef patty, TBARS values were increased with increase of irradiation doses and showed lower values in the beef patty mixed with food additives than that without food additives. The redness of refrigerated beef patty showed highest values at 3 kGy of irradiation and then decreased with increasing irradiation doses, while in the frozen beef patty did not show distinct tendency according to the irradiation doses or food additives. In sensory evaluation. the irradiated beef patty showed unpleasant smell as compared with the non irradiated beef patty, but showed somewhat higher score in smell at the sample contained ascorbic acid regardless of irradiation doses.

  • Effect of various dosages of gamma ray ranging from 0 to 16 krad on wound periderm formation was investigated with aging potato tuber slices $(1cm{\times}2mm)$ under aseptic condition. Cell division was gradually inhibited with increasing dosage, and completely prevented with 16 krad treatment. Solanine content was not significantly different due to dosages. Gas chromatographic separation of 2,4-dinitrophenylhydrazone precipitate from radiation-induced carbonyl compounds in potato tubers showed that formaldehyde and acetone tended to be increased only with high dosages.

Comparison of the Effects of Gamma Ray and Electron Beam Irradiation to Improve Safety of Spices for Meat Processing (육가공용 향신료의 위생화를 위한 감마선 및 전자선 조사 효과 비교)

  • This study evaluated the effects of gamma ray and electron beam (E-beam) to improve the safety of spices for meat processing. The spices (garlic powder, curry powder, turmeric powder, black pepper, white pepper, oregano, parsley, laurel leaf powder, basil, and rosemary) were irradiated by gamma ray and E-beam at 0, 2, 4, 6, 8, and 10 kGy. Total bacterial populations were then enumerated on total plate count agar, and bacteria isolated from the samples were identified by polymerase chain reaction (PCR). In addition, $D_{10}$ values for Bacillus cereus and Staphylococcus aureus inoculated in spices was determined, and the Ames test was conducted for genotoxicity analysis. The contaminated total bacterial populations in spices ranged from 1.5 to 3.8 Log CFU/g, and most of identified bacteria were Bacillus spp., and Staphylococcus spp. However, the bacterial populations decreased below the detection limit (2 Log CFU/g) after irradiation at 4 kGy except for parsley, which required 6 kGy in gamma ray and 8 kGy in E-beam to decrease total bacterial populations below detection limit. $D_{10}$ values were also higher (p<0.05) in E-beam treated samples than gamma-ray treated samples. No genotoxicity was observed in both conditions with and without metabolic activation. These results indicate that gamma ray (>4 kGy and <6 kGy) could be more useful to improve food safety of meat processing spices compared to E-beam.

  • The most remarkable aspect in the hormesis law is that dose of harmful agents can produce effect that are diametrically opposite to the effect found with high doses of the same agent. Minute quantities of a harmful agent bring about very small change in the organism and control mechanisms appear to subjugate normal processes to place the organism in a state of albert and repair. The stimulated organism in more responsive to changes in environmental factors than it did before being alerted. Routine functions, including repair and defense, have priority for available energy and matetial. The alerted organism utilizes nutrients more efficiently, grows faster, shows improved defense, and lives longer. Accelerated germination, sprouting, growth, development, blooming and ripening, and increased crop yield and resistance to disease are found in plants. Another concept supported by the data in that low doses of ionizing radiation provide increased resistance to subsequent high doses of radiation. The hormesis varies with subject plant, variety, state of seed, environmental and cultural conditions, physiologic function measured, dose rate and total exposure. The results of hormesis are less consistently found, probably due to the great number of uncontrolled variables in the experiments. The general dosage for radiation homlesis in about 100 (10 to 1,000) times ambient or 100 (10 to 1,000) times less than a definitely harmful dose, but these must be modified to the occasion. Although little is known about most mechanisms of homzesis reaction, overcompensation of repair mechanism is offered as one mechanism.

The Combined Effect of Heat Treatment and Irradiation on the Inactivation of Major Lactic Acid Bacteria Associated with Kimchi Fermentation (김치의 숙성관련 주요 젖산균 살균에 대한 가열처리와 방사선 조사의 병용효과)

  • The combined effects of heat treatment and ${\gamma}-irradiation$ on the inactivation of major lactic acid bacteria associated with Kimchi fermentation were investigated. The radiosensitivities $(D_{10}\;values)$ of lactic acid bacteria in case of a single treatment of irradiation were 0.61 kGy in Lactobacillus brevis, 0.60 kGy in Lactobacillus plantarum, 0.50 kGy in Leuconostoc mesenteroides, 0.4 kGy in Pediococcus cerevisiae, 0.39 kGy in Streptococcus faecalis. The heat sensitization $(D_{min}\;values)$ by a single treatment of heat ranged 9.2-15.6 at $50^{\circ}C$ and 3.7-5.5 at $60^{\circ}C$. Synegistic effects were shown in the radiosensitivities of Streptococcus faecalis, Pediococcus cerevisiae, Lactobacillus plantarum, and Lactobacillus brevis by the combined treatment(Dose multiplying factors ranged $1.20{\sim}1.56$). It seems, therefore, that the combined treatment can be applied to the radiation preservation of Kimchi, minimizing the side-effects like physical changes induced by the high dose irradiation or heat treatment.

Consciousness and Behavior of the Radiation Safety Management by Clinical Pathologists (임상병리사의 방사선안전관리에 대한 고찰)

  • 이 연구는 동위원소실에 근무하는 임상병리사들의 방사선 방어에 대한 지식, 태도 및 행위수준을 조사하여 상관관계를 파악하고 방사선 안전관리에 대한 행위에 영향을 미치는 요인을 분석하기 위해 시행하였다. 설문지는 2010년 10월부터 11월까지 대전 충청권 및 서울 경기권 동위원소실 임상병리사를 연구 대상자로 하여 연구자자 임의로 선정 직접설문, 또는 우편방식으로 조사하였고, 수집된 자료는 SPSS12.0 프로그램을 이용하여 빈도와 백분율, T-검정, ANOVA, 다중회귀분석법을 이용하여 분석하였다. 방사선 안전관리에 대한 지식 점수는 평균은 11.5점이었으며 표준편차는 ${\pm}$1.9 분포를 보였다. 방사선 안전관리에 대한 태도 점수는 평균 69.0점, 표준편차는 ${\pm}$5.4이었다. 방사선 방어에 대한 행위점수는 평균은 57.1점, 표준편차 ${\pm}$3.5이었다. 방사선 안전관리 행위에 있어 이론적으로 알고 있는 내용에 비해 수행이 현저히 떨어진 결과가 나왔으며, 방사선 안전관리에 대한 지식, 태도, 행위와의 상관성은 통계적으로 유의한 상관관계를 보였다(p<0.01). 행위점수에 영향을 미치는 요인으로는 종사자법정교육, RI 면허취득, 지식, 태도로 나타났다. 결론적으로 동위원소실에 근무하는 임상병리사의 방사성동위원소 안전관리 행위는 태도에 비하여 수행이 현저히 떨어진 결과가 나왔고, 안전관리 행위에 영향을 주는 인자로 종사자법정교육, RI 면허취득, 지식점수, 태도점수를 알 수 있었다. 업무 진행에 있어 잘못된 지식의 습득 및 태도를 취할 수 있는 가능성이 충분히 있으며 이를 바로 잡을 수 있도록 안전관리에 대한 지속적인 교육을 실시함과 더불어 RI면허 취득, 방사선 종사자에게 교육의 효과를 더욱 높이기 위한 적절한 교육 자료의 개발이 필요하다고 할 수 있다.
