Changes in Rheological Properties of Culture Broth During the Biopolymer Production by Bacillus sp (Bacillus sp.에 의한 생물 고분자의 발효 중 배양액의 유변학적 특성 변화)

  • Variations of rheological properties of culture broth during the production of biopolymer by an alkali tolerant Bacillus sp. were investigated. Correlations among the rheological characteristics of culture broth, cell growth and biopolymer production were examined. Rheology of the culture broth changed in the course of fermentation. The culture broth showed a non-Newtonian flow behavior, as the viscosity and pseudoplasticsity increased during the cultivation. The rheological parameters such as flow index, consistency index, yield stress and apparent viscosity during the cultivation were not influenced by the cell growth, but significantly related to the biopolymer synthesis. Changes in the rheological parameters of the broth were affected not only by the biopolymer concentration, but also by the progress of fermentation. Some rheological parameters showed maximum values just before the completion of biopolymer production and substrate consumption. Hence, it was shown that the rheological characteristics of the culture broth could be used as a good indicator for the detection of the progress or completion of fermentation.

Quality Properties of Fermented Gingers (발효생강의 품질 특성)

  • Applying ginger to processed foods to improve health is limited due to its pungent odor. In this study, fermented gingers were prepared using lactic acid bacteria from dairy products or kimchi and their chemical and sensory properties were examined. Adding dried ginger or fermented gingers (GSt, GLa, GLm) to plain yogurt made them less tasty and lowered the overall acceptability of yogurt except GLp. For all fermented gingers including GLp, the characteristic flavors of the ginger decreased by fermentation so adding fermented ginger into yogurt did not affect its flavor significantly. Thus GLp would be expected to be a food ingredient instead of dried ginger without lowering organoleptic qualities. The content of 6-gingerol, a bioactive component in ginger, decreased in all fermented gingers. However, 6-shogaol, which increased in GSt, GLa, and GLm, was not detected in GLp.

Component Analysis and Digestive Enzyme Activities of Fermented Crataegi Fructus Extracts (산사 발효액의 함유 성분 분석 및 소화 활성)

  • Currently many studies aimed at enhancing efficacy of medicinal food on biological activity using bioconversion technology including fermentation process. In this study, the quality characteristics and antioxidative activity of fermented Crataegi fructus was investigated. The antioxidant activity of fermented Crataegi Fructus was assessed by various radical scavenging assays using DPPH (2,2-Diphenyl-1-picrylhydrazyl), FRAP (Ferric ion reducing antioxidant power), Reducing power and ABTS (2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid)). Moisture content of fermented Crataegi Fructus was $39.3{\pm}0.06%$. Contents of crude ash, crude protein, and crude fat were $0.20{\pm}0.01$, $1.77{\pm}0.04$, and $1.40{\pm}0.59%$, respectively. Moreover, the hunter's color values of fermented Crataegi Fructus were 79.24 (lightnees), 1.58 (redness), and 31.25 (yellowness), respectively. Total phenolic contents of fermented Crataegi Fructus were $3,015{\pm}250$ GAE ${\mu}g/g$. The antioxidative activities of fermented Crataegi Fructus significantly increased in a dose dependent manner. In addition, fermented Crataegi Fructus slightly (10.4%) inhibited ${\alpha}$-glucosidase activity; however, there was no inhibitory activity against ${\alpha}$-amylase. In terms of proteolytic activity, fermented Crataegi Fructus showed a strong activity than pancreatin (used as a positive control). These results indicate that fermented Crataegi Fructus can be used as a natural resource for material aiding digestion.

전기투석과 확산투석을 이용한 공정개발

  • 전기투석은 역삼투압, 한외여과와 함께 가장 많이 이용되고 막공정 중의 하나이다. 전기투석은 다른 막공정과 같이 막의 선택성에 의한 분리조작이며 병렬식 배열에 의한 막의 이용이 가능하고 막오염 현상이 있으며 따라서 막-유체간의 접촉에 대한 제어가 필요하다. 전기투석은 운전목적에 따라 desalting electrodialysis(ED)와 water-splitting electrodiaiysis(WSED)로 구분할 수 있다. Desalting electrodialysis는 고전적 의미의 탈염을 위한 전기투석공정이며 WSED는 bipolar membrane을 이용하여 염을 산과 염기로 분리시키는 기능을 갖는 전기투석 공정을 말한다. WSED는 전기적으로 물을 분리한다는 의미로서 Electrohydrolysis로 불리기도 한다. WSED의 기본원리는 bipolar membrane의 양쪽면에서 수소이옹과 수산이온을 발생시켜 산 또는 염기용액으로 전달하고 bipolar membrane에 접하고 있는 양이온 또는 음이온 교환막에서는 각 용액의 전기적 중성을 유지하기 위해 대응하는 이온을 투과시키는 것이다. WSED는 염으로부터 산 염기제조 뿐만아니라 염의 형태로 생성되는 유기산, 아미노산 등 발효생성물의 회수 또는 acidification에 이용되고 있다.

  • An excellent foam stability in the freshly fermented green beer of a commercial brewery was found to be destroyed substantially by the time it was finished. A further survey proved that the processes of aging and pasteurization accompanied by enzymatic chillproofing were responsible for the foam deterioration.

  • Maximization of productivity of recombinant cell fermentations requires consideration of the inverse relationship between the host cell growth rate and product formation rate. The problem of maximizing a weighted performance index was solved by using optimal control theory for recombinant E. coli fermentation. Concentration of a growth inhibitor was used as a control variable to manipulate the specific growth rate, and consequently the cloned-gene expression rate. Using a simple unstructured model to describe the main characteristics of this system, theoretical analysis showed that the optimal control profile results in an initial high growth rate phase followed by a low growth rate and high product formation rate phase. Numerical calculations were done to determine optimal switching times from the growth to the production stage for two representative cases corresponding to different dependency of the product formation rate on the growth rate. For the case when product formation rate is sensitive to the specific growth rate, the optimized operation yields about 60% increase in the final product concentration compared with a simple batch fermentation.

Fermentation Characteristics of Extruded Tissue Cultured Mountain Ginseng (압출성형 산삼배양근의 발효 특성)

  • The aim of this study was to compare the fermentation characteristics of tissue cultured mountain ginseng, extruded tissue cultured mountain ginseng, and root hair of red ginseng. Also, pH, acidity, brix, reducing sugar, total sugar, and alcohol were analyzed. The extrusion conditions were barrel temperature of 110 and $140^{\circ}C$ and moisture content of 25 and 35%. Fermentation temperature was $27^{\circ}C$ for 15 days and the cultivation was fixed with Saccharomyces cerevisiae, Aspergillus usamii, and Rhizopus japonicus. The results showed that pH, brix, reducing sugar content, and total sugar content of fermented broths were decreased after 5 days and then maintained steadily for the following 10 days. Acidity of final fermented broths were 1.12% (root hair of red ginseng), 1.19% (tissue cultured mountain ginseng), and $0.97{\sim}1.02%$ (extruded tissue cultured mountain ginseng), respectively. Alcohol content of final fermented broths were 3.82% (root hair of red ginseng), 0.91% (tissue cultured mountain ginseng), and $1.86{\sim}2.18%$ (extruded tissue cultured mountain ginseng). The fermentation efficiency of extruded tissue cultured mountain ginseng (barrel temperature $140^{\circ}C$, moisture content 25%) were the highest. In conclusion, the fermentation efficiency was increased by extrusion process.

Development of the Dynamic Model for the Metabolic Network of Clostridium acetobutylicum (Clostridium acetobutylicum의 대사망의 동적모델 개발)

  • To produce biobutanol, fermentation processes using clostridia that mainly produce acetone, butanol and ethanol are used. In this work, a dynamic model describing the metabolic reactions in an acetone-butanol-ethanol (ABE)-producing clostridium, Clostridium acetobutylicum ATCC824, was proposed. To estimate the 58 kinetic parameters of the metabolic network model with experimental data obtained from a batch fermentor, we used an efficient optimization method combining a genetic algorithm and the Levenberg-Marquardt method because of the complexity of the metabolism of the clostridium. For the verification of the determined parameters, the developed metabolic model was evaluated by experiments where genetically modified clostridium was used and the initial concentration of glucose was changed. Consequently, we found that the developed kinetic model for the metabolic network was considered to describe the dynamic metabolic state of the clostridium sufficiently. Thus, this dynamic model for the metabolic reactions will contribute to designing the clostridium as well as the fermentor for higher productivity.

Changes in the Viability of Lactic Acid Bacteria during Storage of Freeze-Dried Yogurt Snacks (저장기간에 따른 동결건조 농후 발효유 내 유산균 생균수 변화)

  • The majority of food drying processes are based on the use of thermal energy. However, such methods may deteriorate the quality of the final product. Freeze-drying is one of the most useful processes for drying thermosensitive substances. Food that contains beneficial bacteria, for example, is susceptible to heat treatment, but during freeze-drying beneficial bacteria are preserved in these food items. The primary goals of this study were to develop yogurt snacks and to compare the viability of lactic acid bacteria (LAB) in yogurt snacks under different freeze-drying temperatures. In addition, the survival of LAB during storage was investigated. Survival of LAB in freeze-dried yogurt snacks gradually decreased over 16 weeks of storage. LAB had a residual viability of 25.5% after 16 weeks of storage at room temperature. LAB survived better in freeze-dried plain yogurt snacks than in freeze-dried strawberry yogurt snacks during storage. Freeze-dried yogurt snacks contained 11.9% fat, 57.1% carbohydrate, and 18.7% protein. In conclusion, the viability of LAB in freeze-dried yogurt snacks depends on the temperature during freeze-drying: the higher the freeze-drying temperature, the lower the viability of LAB in yogurt snacks. The viability of LAB in yogurt snacks was also dependent on the moisture content and nutritional value.

Change in amygdalin contents of maesil (Prunus mume) wine according to preparation steps and its characteristics (매실 와인의 제조공정에 따른 아미그달린(amygdalin) 함량의 변화와 품질 특성)

  • The purpose of this study was to compare the amygdalin content, alcohol content, pH, titratable acidity, and color of maesil wine prepared with two different manufacturing processes. Maesil wine was made from maesil chung, and maesil was preserved (MW1) or removed (MW2) before fermentation. During aging, amygdalin content in all the wines was gradually reduced, and the content of MW2 was less than that of the other. The alcohol content, pH, and titratable acidity ranged from around 9.87-10.94, 3.57-3.80, and 8.89-10.68%, respectively. The difference between the samples was not significant. For color, MW1 had lower L and higher a and b values than MW2. In this study, the difference in physicochemical properties according to the presence of maesil was not significant, indicating no degradation of the quality according to the manufacturing processes. However, the MW2 showed a reduction in amygdalin contents.
