Color change of Zambian amethyst by heat treatment (잠비아산 천연 자수정의 열처리에 따른 색상변화)

  • It is known that the natural amethyst is changed to citrine after heat treatment. However, when all amethyst samples from Zambia were heat-treated in the temperature range of $350{\sim}380^{\circ}C$ for 1 hour, the result was that five out of eight samples were changed to citrine and all the rest of samples became rock crystal quartz. These differences in the color appearance seem to be influenced by the original colors contained in the amethyst before the heat treatment. The amethyst containing yellow color changed to citrine and the amethyst without containing yellow color changed to rock crystal quartz after the heat treatment. The results compared after the instrumental analysis on the difference of color change, it showed the differences of peak intensity in 3,400 $cm^{-1}$ and the existence and non-existence of peak at the range of 5,200${\sim}$5,400 $cm^{-1}$ in FTIR. It revealed the difference in the quantity of Cr which is a trace element in the WD-XRF analysis. The identical result in the FTIR spectra before and after the heat treatment reveals that the heat treatment did not cause any change in the main composing elements or crystal structure.

  • DNA sequence information on small-subunit rRNA gene (16S rDNA) obtained from food-poisoning bacterial culture was used to investigate the presence of bacterial pathogens in food. By reverse dot blot detection method, presence of food-poisoning bacteria could be confirmed on hybridization of digoxigenin-labeled 16S rDNA Polymerase Chain Reaction (PCR) primer product and biotin-labeled specific oligonucleotide probe. Escherichia coli, Bacillus cereus. and Salmonella sp. were used as the representative food-poisoning bacterial microorganisms. An oligonucleotide probe, based on the variable region of 16S rRNA gene, was used as the specific probe. These tools may be more useful than classic biochemical method for rapid identification of contaminated food.

  • The significance of soil environment is gradually increased due to the soil and underwater contamination caused by petroleum leak accidents. It requires a high cost and long period for the purification of soil compared with other environmental matrix such as water and air. For this reason, it has been embroiled in a legal conflict to find the pollution source and charge of cleanup. In this study, we analyzed the physical properties and typical additives of jet fuel to search a method that can distinguish kerosene and jet fuel contamination. In particular, the chemical marker in kerosene was visualized by the developer and the additives in jet fuel, such as antioxidant and metal deactivator were detected by GC-MS. This study could be used to judge petroleum source at soil contaminant accident sites.

파이렌 이중체 기반 2 킬레이트 결합자리 형광분자에 대한 금속 양이온의 형광 소광 원리 탐색

  • 형광화학센서는 형광분자의 화학적으로 유도된 형광의 소광 또는 증광을 통해 생체 등에서 특정 물질을 관찰할 수 있기에 그 활용도가 높다. 본 연구에서는 두 개의 서로 다른 킬레이트 결합자리를 가지는, 파이렌 이중체를 발색단의 형광분자(Pyex)를 이용하여, PET (photoinduced electron transition)와 AID (absorbance intensity decreasing)의 형광 소광 원리에 집중하여, 전자구조계산과 TD (time-dependent) 계산을 근거로 금속 양이온의 형광 소광 원리를 분류하고, 더불어 그에 관여하는 금속 양이온의 원자오비탈까지 탐색하였다. 그 결과 Pyex와 그 칼륨이온 복합체에서는 실험값과 일치하는 형광이, 납과 은 이온 복합체에서는 소광이 나타났다. 구체적으로는 납 이온의 경우 PET를 주된 원인으로 AID와 함께 작용하여 소광을 발생시키고, 은 이온의 경우는 AID에 의해 소광이 일으키는 것으로 밝혀졌다. 또한 납 이온의 p 오비탈이 소광에 관여하는 것으로 볼 수 있는 결과도 나타났다.

Factors Affecting Reactivity of Various Phenolic Compounds with the Folin-Ciocalteu Reagent (다양한 페놀성 물질과 Folin-Ciocalteu 시약의 반응성에 미치는 영향 요인 평가)

  • The Folin-Ciocalteu (F-C) reagent has been extensively used for quantifying total phenolic contents in many different types of food materials. Since several different procedures of the assay methods using the F-C reagent have been applied, we investigated changes in reactivity of various phenolic compounds with the F-C reagent under three different assay conditions and factors affecting reactivity. Among 10 standard compounds tested, compounds with high hydroxyl density (number of -OH/molecular weight) showed a largely different response according to addition sequence of the F-C reagent or $Na_2CO_3$. Preincubation in $Na_2CO_3$ significantly reduced the reactivity of the phenolic compounds bearing galloyl moiety (e.g. gallic acid, tannic acid, and epigallocatechin-3-gallate) with the F-C reagent, while monophenol compounds including ferulic acid and sinapinic acid were more stable as compared to diphenols. There was little change in response to the F-C reagent of all phenolic compounds incubated in acidic pH; their reactivity except ferulic acid was reduced significantly when incubated in neutral or alkaline pH. Changes in reactivity of gallic acid incubated in $Na_2CO_3$ or neutral/alkaline pH conditions were the most prominent. $H_2O_2$ generated from phenolic compounds did not affect the reaction with the F-C reagents. The present results suggest that reactivity of different phenolic compounds with F-C reagent was affected considerably by different procedures of the assay, and the total phenolic contents could be fluctuated according to standard compounds and assay scheme.

  • The present study was aimed for the fetor in the livestock facilities. Simple detection systems for ammonia and hydrogen sulfide were examined to see the mechanism making the discoloration length of malodorous substance concentration. Detector tube for ammonia showed a good linearity of 99.6% while hydrogen sulfide detector tube gave 99.7% linearity with reproductivity of 95%.

전자빔 조사를 통한 핑크 투어멀린의 특성 변화

  • 투어멀린는 $XY_3Z_6(Si_6O_{18})(BO_3)_3$ 화학 방정식을 가진 광물로써 조성에 따라 alkali 투어멀린, calcic 투어멀린, X-site vacant 투어멀린 등으로 불린다. 투어멀린 특성과 화학성분에 따라 종을 구분하는데, 주된 종들은 엘바이트, 리디코타이트, 드라바이트, 우바이트, 숄이 있다. 일반적으로 적색 투어멀린의 발색 원인은 Mn, Fe 그리고 Cu의 함량에 따라 색상의 차이를 나타낸다. 본 연구에서 우리는 10MeV 에너지와 $1{\times}10^{17}cm^2$ 조건에서 전자빔을 수행 한 후 투어멀린의 컬러 변화를 관찰하였다. 자외선-가시광선 분광분석결과 모든 시료는 전자빔 조사 후 530 nm의 $Mn^{3+}$부근의 흡수 peak들이 증가하는 것이 관찰되었다. 이는 $Mn^{2+}$에서 $Mn^{3+}$ 이동 때문이여, $Mn^{3+}$는 Y-site에서 O(1)H-O(3)H 축에 따라 Jann-taller 변형으로 안정된 구조를 가지게 된다. 따라서 전자빔 조사 후 적색으로 변하게 되는 것이다. 또한 전자빔 조사 후 컬러가 모두 변했지만 상온에 뒀을 때 변화 된 컬러가 원래의 색으로 되돌아가는 향상을 보였다. 이는 전자빔 조사 후 전자가 튕겨져 나가서 불안정한 상태로 존재하고 있다가 상온의 열에 의한 에너지에 통해 다시 안정된 상태로 되돌아오는 결과로 볼 수 있다. 또한 우리는 WD-XRF를 통해 미량의 Mn 원소함량 차이에 따라 전자빔 조사 시 컬러 변화에 미치는 영향에 대해 확인할 수 있었다. 그리고 적외선 분광분석에서는 4,300-4,600 $cm^{-1}$사이에 특징적인 밴드들이 관찰되었다.

Ozone Application for the Bleaching of Unsaturated Coloring Material (오존을 이용한 불포화 색소물질의 탈색반응)

  • Bleaching effect of ozone on unsaturated coloring material was compared quantitatively with the change of composition of reactants through the ozone oxidation of ${\beta}$-carotene. Beeswax colorized by carotenoids was reacted with ozone in water, 0.5 % and 1.0 % sodium hydroxide solution, and 20 % and 95 % acetic acid solution and bleaching effect of each reaction condition was compared with each other. And then the additive effect of hydrogen peroxide was investigated. Decrease of unsaturated double bond of ${\beta}$-carotene was proportioned quantitatively to the bleaching effect of ozone on ${\beta}$-carotene and was in accordance with the bleaching effect on beeswax. The bleaching effect of ozone on coloring material in 1.0 % sodium hydroxide solution was better than any other condition and the absorbance of beeswax at 390 nm was improved to 0.045 with the addition of hydrogen peroxide(2 % of wax). For the bleaching of beeswax, it was concluded that the ozone oxidation was more effective than physical adsorption.

Sapphire Enhancement with BeO Powder Source (BeO를 확산원으로 한 사파이어의 향상처리)

  • We heat treated pure sapphire samples at $1800^{\circ}C{\sim}100$ hours in BeO-${Al_2}{O_3}$ powder by varying BeO contents of $5{\sim}50\;wt%$ in order to quantify the BeO diffusion enhancement for corundum gem stones. We investigated the heat treated samples with visual evaluation, color coordination analysis, and surface roughness measurement. We confirmed that $Be^{2+}$ ions did not lead to orange color but to dark gray. The lightness in CIE Lab index decreased while surface roughness increased rapidly as BeO contents increased. We propose that BeO yellow color enhancement may be feasible only for $Fe^{3+}$ rich blue sapphires and sub-5% BeO content be appropriate to shorten post re- cutting process.

  • Pigments were extracted by aqueous acetone from the boiled canned oyster colored during storage and then the components were separated by thin layer chromatography. Totally eleven pigments could be isolated including one yellow, one red, five orange or reddish orange and four green components, and their UV-visible spectra were measured. It can be envisioned from the electronic spectral study and color reaction on the indivisual pigments isolated from the brown acetone extracts that the green pigments as well as most of the yellow orange ones may be porphyrin derivatives originated probably from the chlorophyll and some of the orange pigments contains ketocarotenoids. In particular, the pigment of band 8 which is expected to be pheophytin a or its derivatives and the carotenoid band 7 seem to be the major pigment. The close resemblance of the chromatogram of the colored muscle extract to that of the viscera suggests that the brown coloring material is probably originated from the viscera pigments.
