Effect of Light on Growth of Chlorella sp. HA-1 in Large-scale Photobioreactors for Biological $CO_2$ Fixation (생물학적 $CO_2$ 고정화를 위한 스케일-업된 광생물반응기에서 Chlorella sp. HA-1의 성장에 미치는 빛의 영향)

  • Feasibility of large-scale photobioreactors for biological CO$_2$ fixation was investigated using Chlorella sp. HA-1. Generally, as the volume of photobioreactor increased, the CO$_2$ fixation rate decreased because of a lower illumination efficiency in large-scale than in small-scale photobioreactors. Though controlling the arrangement and the number of light source, the maximum CO$_2$ fixation rates that could be achieved were 530 and 357 gCO$_2$/㎡day for 40 L and 188 L photobioreactor, respectively, which was higher than the CO$_2$ fixation rate of lab-scale photobioreactor.

Removal of ${\alpha}$-Gal Epitopes in Aortic Valve and Pericardium of Pig Using Green Coffee Bean ${\alpha}$-Galactosidase (돼지의 대동맥 판막 및 심낭에서 녹색콩 알파-갈락토시다아제를 이용한 알파-갈 항원결정인자 제거)

  • Background: It is currently thought that tissue valve degeneration is related to an animal's immune response, which is mainly due to cell surface ${\alpha}$-Gal epitopes. Cell surface ${\alpha}$-Gal epitopes are known to be degraded by the enzyme called green coffee bean ${\alpha}$-Galactosidase. It is also well known that ${\alpha}$-Gal epitopes are immunologically stained by Griffonia Simplicifolia isolectin type B4. We know that many commercially available tissue valves are made of aortic valves and pericardial tissue of pig. So, we investigated whether ${\alpha}$-Gal epitopes of the aortic valve and pericardial tissue of a pig can be removed by green coffee bean ${\alpha}$-Galactosidase, and we did so by comparing immunologic staining of the tissues before and after the enzyme treatment. Material and method: After treating fresh porcine aortic valve and pericardial tissue with green coffee bean ${\alpha}$-Galactosidase at concentrations of 0.5 unit/mL, 1.0 unit/mL, 2.0 unit/mL, respectively, under the condition of pH 6.5, temperature. $4^{\circ}C$ and 24 hours of incubation, each sample was stained with Griffonia Simplicifolia isolectin type B4 immunpfluorescent labeling. We then examined whether the ${\alpha}$-Gal epitopes were reduced or abolished in each consecutive. concentration of green coffee bean ${\alpha}$-Galactosidase by comparing the degree of the Griffonia Simplicifolia isolectin B4 staining in each sample. Result: In the pig aortic valve tissue, a 1.0 unit/mL concentration of green coffee bean ${\alpha}$-Galactosidase at pH 6.5, $4^{\circ}C$ and reaction for 24 hours was enough for complete removal of ${\alpha}$-Gal epitopes from the cell sur face on the immunostaining with Griffonia Simplicifolia isolectin B4. On the other hand, more ${\alpha}$-Gal epitopes were present in the pig pericardial tissue on Griffonia Simplicifolia isolectin B4 staining before the enzyme treatment, and 1.0 unit/mL of galactosidase was not sufficient for complete removal of ${\alpha}$-Gal from the tissue. 2.0 units/mL of green coffee bean ${\alpha}$-Galactosidase was needed to completely remove the ${\alpha}$-Gal epitopes from the pericardial tissue on immunostaining. Conclusion: The ${\alpha}$-Gal epitopes of the pig's aortic valve and pericardial tissue were successfully stained with Griffonia Simplicifolia isolectin B4. We could remove nearly all the ${\alpha}$-Gal epitopes using green coffee bean ${\alpha}$-Galactosidase at the concentration of 1.0 unit/mL in the aortic valve. Of pig, and 2.0 unit/mL was need to nearly completely remove all the ${\alpha}$-Gal epitopes in the pericardial tissue of pig under the condition of pH 6.5, $4^{\circ}C$ and 24 hours of reaction time. In the near future, removal of ${\alpha}$-Gal epitapes in the pig's aortic valve and pericardial tissue will become a powerful tool for the improvement of the tissue valve durability. It needs to be determined if ${\alpha}$-galactosidase treated pig tissue is immune to human anti-Gal antibody or anit-Gal mooclonal antibodies.

Effects of Green Tea Seed Oil on the Cholesterol, TBARS and Inflammatory Responses in C57BL/6 Mice Fed High Cholesterol Diet (녹차씨기름이 고콜레스테롤식이를 섭취시킨 C57BL/6 Mice의 콜레스테롤, TBARS 및 염증반응에 미치는 영향)

  • Effects of green tea seed oil intake on the serum cholesterol, the thiobarbituric reactive substances (TBARS) formation of liver and inflammatory markers of plasma and macrophage in cholesterol fed mice were investigated, comparing to the intakes of corn oil and olive oil. C57BL/6 mice were divided into three groups and fed the experimental diets: supplemented corn oil, green tea seed oil and olive oil to cholesterol diet, respectively, at the level of 10% for 9 weeks. The increased levels of serum cholesterol of green tea seed oil group were significantly (p<0.05) lower than that of corn oil group until 6 weeks. The TBARS formation in liver of green tea seed oil and olive oil groups were significantly (p<0.05) reduced than those of corn oil group. The contents of inflammatory markers ($LTB4,\;TNF-{\alpha},\;PGE2,\;NO$) in plasma and macrophage did not show significant differences among the experimental groups. These results showed that green tea seed oil could exert the activity of decreasing serum cholesterol and the antioxidative activity in cholesterol fed mice liver. Therefore, we suggest that green tea seed oil might be developed as a high quality edible oil.

Gastric mucosal immune response of Helicobacter pylori-infected children (Helicobacter pylori 감염 소아에서 위점막 면역반응)

  • Purpose : Helicobacter pylori infection is one of the most common gastrointestinal infections worldwide; it almost invariably causes chronic gastritis. Pediatric studies may provide important insights into the mucosal immune response of H. pylori-infection, as children are not submitted to environmental factors such as alcohol, tobacco and anti-inflammatory medication. The aim of the present study was to investigate the mucosal immune response against H. pylori in clinically well-defined groups: H. pylori-positive (divided into peptic ulcer disease and gastritis) and H. pylori-negative control. Methods : Antral biopsies were obtained from 45 children undergoing an upper GI endoscopy for dyspeptic symptoms. T cells (CD3+, CD4+, CD8+) and B cells (CD20+) were analyzed by quantitative immunohistochemistry. The correlation of lymphocyte subsets of gastric mucosa with histology was evaluated. Results : T cells (CD3+, CD4+, CD8+) and B cells (CD20+) were significantly increased in the lamina propria of H. pylori-positive group (P<0.01). CD8+ T cells were significantly increased in the lamina propria of the H. pylori-positive peptic ulcer disease (P<0.01). Within the epithelium, only CD4+ T cells were significantly increased in the H. pylori-positive group (P<0.01). Gastric histological parameters had a closer correlation with lymphocytes in the lamina propria than intraepithelial lymphocytes. Conclusion : This study suggests that both T cells and B cells in the lamina propria play important roles in the local immune response of H. pylori-infected children. Furthermore, it remains to be elucidated whether CD8+ T cells in the lamina propria may contribute to peptic ulcer formation in H. pylori-infected children.

Optimization of TDA Recycling Process for TDI Residue using Near-critical Hydrolysis Process (근임계수 가수분해 공정을 이용한 TDI 공정 폐기물로부터 TDA 회수 공정 최적화)

  • The recycling of TDA from solid waste of TDI plant(TDI-R) by near-critical hydrolysis reaction had been studied by means of a statistical design of experiment. The main and interaction effects of process variables had been defined from the experiments in a batch reactor and the correlation equation with process variables for TDA yield had been obtained from the experiments in a continuous pilot plant. It was confirmed that the effects of reaction temperature, catalyst type and concentration, and the weight ratio of water to TDI-R(WR) on TDA yield were significant. TDA yield decreased with increases in reaction temperature and catalyst concentration, and increased with an increase in WR. As a catalyst, NaOH was more effective than $Na_2CO_3$ for TDA yield. The interaction effects between catalyst concentration and temperature, WR and temperature, catalyst type and reaction time on TDA yield had been defined as significant. Although the effect of catalyst concentration on TDA yield at $300^{\circ}C$ as subcritical water was insignificant, the TDA yield decreased with increasing catalyst concentration at $400^{\circ}C$ as supercritical water. On the other hand, the yield increased with an increase in WR at $300^{\circ}C$ but showed negligible effect with WR at $400^{\circ}C$. The optimization of process variables for TDA yield has been explored with a pilot plant for scale-up. The catalyst concentration and WR were selected as process variables with respect to economic feasibility and efficiency. The effects of process variables on TDA yield had been explored by means of central composite design. The TDA yield increased with an increase in catalyst concentration. It showed maximum value at below 2.5 of WR and then decreased with an increase in WR. However, the ratio at which the TDA yield showed a maximum value increased with increasing catalyst concentration. The correlation equation of a quadratic model with catalyst concentration and WR had been obtained by the regression analysis of experimental results in a pilot plant.

  • Increased base cation loss and Al mobilization, a consequence of soil acid neutralization responses, are common in air polluted areas showing forest decline. The prediction models of acid neutralization responses were developed by using indicators of soil acidification level(pH, and base saturation) in order to assess the forest soil sensitivity to acidification. The soil acidification level was greatest in Namsan followed by Kanghwa, Ulsan, and Hongcheon, being contrary to regional total $ANC_H$ pattern through soil columns leached with additional acid ($16.7mmol_c\;H^+/kg$), Both base exchange and Al dissolution were main acid neutralization processes in all study regions. There were low base exchange and high Al dissolution in the regions of the low total $ANC_H$. The $ANC_M$ by sulfate adsorption was greatest in Hongcheon compared with other regions even though the AN rate was very low as 6.4%. Coefficients of adjusted determination of simple and multiple regression models between soil acidification level indicators and the acid neutralization responses were more than 0.52(p<0.04) and 0.89(p<0.01), respectively. The result suggests that soil pH and base saturation are available indicators for predicting the acid neutralization responses. These prediction models could be used as an useful method to measure forest soil sensitivity to acidification.

Gasification and Methanation Characteristics for SNG(Substitute Natural Gas) from Coal Char (석탄촤로부터 대체천연가스(SNG)를 얻기 위한 가스화 및 메탄화 반응 특성)

  • 본 연구에서는 가스화 반응, 수성가스 전환 반응, 메탄화 반응 등으로 구성된 SNG제조 공정에 대한 해석을 통해, 석탄 촤의 가스화 반응에 의해 생성된 합성가스를 이용한SNG제조 공정 특성을 파악하고자 하였고, SNG제조 공정 중 가스화 공정에 대한 실험을 통해 가스화 공정의 조건에 따른 합성가스 발생 특성 및 메탄화 반응의 특성을 살펴보았다. 석탄 촤를 대상으로 하여 가스화 공정의 $O_2$/feed ratio와 steam/feed ratio 조건 변화에 따른 합성가스 발생 특성을 살펴본 결과 steam을 투입하지 않은 경우 발생되는 합성가스 중 CO의 농도는 55$\sim$65%, $H_2$ 9$\sim$11%, $CO_2$ 24$\sim$29% 범위였고, $O_2$/feed ratio가 증가할수록 CO의 농도는 증가하고, $H_2$$CO_2$의 농도는 감소하는 경향을 나타내었다. 또한,steam을 투입하는 경우 합성가스 중 CO의 농도는 20$\sim$37%, $H_2$ 16$\sim$18%, $CO_2$ 42$\sim$55% 범위였다. 메탄화 공정 해석 결과 메탄의 농도를 최대로 얻을 수 있는 조건은 $H_2$/CO 비가 3인 조건이었고 온도가 낮을 수록 생성농도가 높아짐을 알 수 있었다. 가스화 특성 실험 결과 및 공정해석 결과, 메탄화 반응에 대한 실험 및 공정해석 결과는 고체시료의 가스화 반응을 통해 발생한 합성가스를 이용한 SNG 제조 공정 특성 파악 및 SNG를 제조하기 위해 필요한 단위 공정에 대한 설계 자료 및 운전조건을 결정할 수 있는 주요 인자로 활용될 수 있을 것으로 판단된다.

  • Whole cell penicillin amidase of Escherichia coli was immobilized by entrapment in gelatin followed by extrusion and crosslinking with glutaraldehyde. The immobilized engyme preparation demonstrated the recovery yield of activity up to 70% and good stability during storage and operation. The half life of activity decay during the operation was estimated to be about 50 days. The optimum pH and temperature for both of immobilized and soluble enzyme are 8.5 and 5$0^{\circ}C$, respectively. No significant change was demonstrated in the effect of pH and temperature, but the increase in heat stability at high temperature was observed in the case of the immobilized enzyme. It was found that the plug flow reactor could be operated favorably since the pH drop along the column path due to tile reaction product was minimized by employing substrate solution with moderate buffer strength. The optimal condition of reactor operation was discussed with regard to the effect of substrate concentration and the residence time on the conversion efficiency and productivity.

An Experimental Study on the Determination of Minimum Response Concentration of Inorganic Pollutants in Tap Water (수돗물에서 무기 오염물질 최소 반응 농도 결정의 실험적 고찰)

  • In this study, four inorganic pollutants (cadmium, chromium, manganese, lead), that could cause contamination events in drinking water distribution system, were selected and batch tests were carried out to determine the "minimum response concentration (MRC)", a part of Korean Contamination Warning system establishment. As the results, the minimum response concentration of cadmium was found to be 0.05 to 0.08 mg/L (0.005 mg/L : water quality standard) and that of chrome was 0.03 mg/L (0.05 mg/L). And the minimum reaction concentration was 0.005 mg/L for manganese (0.05 mg / L for water quality) and 0.02~0.08 mg/L for lead (0.01 mg/L).

  • The optimum conditions for the removal of hydrogen sulfide by Fe-EDTA complex in the bubble column reactor were investigated. As the concentrations of the complex increased, the conversion rate of hydrogen sulfide increased, while Fe concentration and pH were stably decreased and the amount of elemental sulfur produced was also increased. Hydrogen sulfide was removed efficiently when the concentration of Fe-EDTA complex was maintained more than 0.05M. pH acts as an important factor for the stability of complex in the oxidation of hydrogen sulfide and optimum pH range was 8.5~9.5. As the molar ratio of EDTA : Fe was increased, the conversion rate of hydrogen sulfide became stable. However, the rate was decreased due to the precipitation of FeS when the concentration of EDTA was decreased. As the concentration of EDTA increased, the conversion rate of hydrogen sulfide increased due to the high stability of Fe-EDTA complex.
