  • For the elucidation of action mechanism on anti-HIV of natural resources, the extracts of $Campsis$ $grandiflora$ were tested for their inhibitory effects on HIV-1 replication and its essential enzymes as the reverse transcriptase (RT), protease and ${\alpha}$-glucosidase. In the assay of HIV-1-infected human T-cell line, water extracts of stem inhibited the HIV-1-induced cytopathic effects with IC (inhibitory concentration) of 100 ${\mu}g$/ml. Moreover water extracts (100 ${\mu}g$/ml) of stem showed strong activity of 37.9% on anti-HIV-1 RT using Enzyme Linked Oligonucleotide Sorbent Assay (ELOSA) method. In the HIV-1 protease inhibition assay, methanol extracts of stem and leaf extract showed 33.6% and 31.5% inhibition of the enzyme activity to cleave an oligopeptide resembling one of the cleavage sites in the viral polyprotein which can only be processed by HIV-1 protease, but did not exhibited glucosidase inhibitory activities. From these results, it is suggested that the inhibition of the viral replication $in$ $vitro$ is due to the inhibition of reverse transcriptase by water extracts of stem of $Campsis$ $grandiflora$.

  • As a specific and effective therapeutic genetic material against hepatitis C virus (HCV) multiplication, HCV internal ribosome entry site (IRES)-targeting hammerhead ribozyme which activity is allosterically regulated by HCV regulatory protein, NS5B RNA replicase, was constructed. The allosteric ribozyme was composed of sequence of RNA aptamer to HCV NS5B, communication module sequence which can transfer structural transition for inducing ribozyme activity upon binding NS5B to the aptamer, and sequence of ribozyme targeting +382 nucleotide of HCV IRES. With real-time PCR analysis, the ribozyme was found to efficiently inhibit HCV replicon replication in cells. Of note, the allosteric ribozyme was shown to inhibit HCV replicon replication more efficiently than either HCV genome-targeting ribozyme or NS5B aptamer only. This allosteric ribozyme can be used as a lead genetic agent for the specific and effective suppression of HCV replication.

  • To study the mechanism of syncytium formation, novel syncytia-inducing ecotropic murine retrovirus was used. Our previous result showed that amino acid substitutions at the RBD (receptor binding domain) of envelope glycoprotein contribute to syncytium formation. In this study, we have investigated if this fusion phenomenon could occur with retroviral vectors pseudotyped with the novel syncytia-inducing ecotropic murine leukemia virus Env. We have found that these vectors were not able to mediate virus-to-cell fusion in M. dunni murine cell lines. These findings indicate that syncytia-inducing ecotropic murine leukemia virus is capable of generating syncytia during its replication. There was also no correlation between the level of ecotropic murine leukemia virus receptor (mCAT-1) and the fusogenic effect.

  • The foot-and-mouth disease virus (FMDV), a member of the Aphthovirus genus in the Picornaviridae family, affects wild and domesticated ruminants and pigs. During replication of the FMDV RNA (ribonucleic acid) genome, FMDV-encoding RNA polymerase 3D acts in a highly location-specific manner. This suggests that specific RNA structures recognized by 3D polymerase within non-coding regions of the FMDV genome assist with binding during replication. One such region is the cis-acting replication element (CRE), which functions as a template for RNA replication. The FMDV CRE adopts a stem-loop conformation with an extended duplex stem, supporting a novel 15-17 nucleotide loop that derives stability from base-stacking interactions, with the exact RNA nucleotide sequence of the CRE producing different RNA secondary structures. Here, we show that CRE sequences of FMDVs isolated in Korea from 2010 to 2017 exhibit A and O genotypes. Interestingly, variations in the RNA secondary structure of the Korean FMDVs are consistent with the phylogenetic relationships between these viruses and reveal the specificity of FMDV infections for particular host species. Therefore, we conclude that each genetic clade of Korean FMDV is characterized by a unique functional CRE and that the evolutionary success of new genetic lineages may be associated with the invention of a novel CRE motif. Therefore, we propose that the specific RNA structure of a CRE is an additional criterion for FMDV classification dependent on the host species. These findings will help correctly analyze CRE sequences and indicate the specificity of host species for future FMDV epidemics.

  • The process of the formation of polyhedra occlusion bodies and occlusion of viral nucleocapsids of Autographa californica Nuclear Polyhedrosis Virus in Spodoptera frugiperda cell were photomicrographed and described. Progeny viral nucleocapsids were observed in the nuclei of the host cells, bundled and then enveloped. The nucleoapsids were mainly accumulated near the membrane-like profiles. The nuclear membrane were hypertophied up to the cytoplasmic membrane. Prepolyhedral bodies were observed and they were growing with the accumulations of thread-like materials(polypeptides) produced by viral genes. The bundled and enveloped nucleocapsids were occluded into the growing polyhedra.

  • ${\beta}$-Glucan is a natural compound contained in cell walls of yeast or fungi, and cereal's fiber. It is also known to boost the immune system in human. Aureobasidium is a producer of water-soluble ${\beta}$-1,3/1,6-glucan. In this study, natural killer (NK) cell and macrophage activity were tested to investigate the effects of ${\beta}$-1,3/1,6-glucan isolated from A. pullulans on immune activity. Activation of NK cell was increased about 63-39% by the treatment of $10-200{\mu}g/mL$ ${\beta}$-1,3/1,6-glucan than control. Besides, only $10{\mu}g/mL$ of ${\beta}$-1,3/1,6-glucan was enough to boost activation of NK cell. Phagocytosis of macrophage was increased to 15~21% by the treatment of $10{\sim}200{\mu}g/mL$ of ${\beta}$-1,3/1,6-glucan than zymosan-treatment. In LP-BM5 proliferating inhibition test, relative mRNA level of LP-BM5 virus was decreased in ${\beta}$-1,3/1,6-glucan-treated cell about 36~74% than control. The decline of LP-BM5 mRNA level appeared to depend on the concentration of ${\beta}$-1,3/1,6-glucan. These results suggest that pure ${\beta}$-1,3/1,6-glucan from A. pullulans might be contributing to enhancement of immune activity through the activation of NK cell and phagocytosis of macrophage. Moreover, treatment of the ${\beta}$-1,3/1,6-glucan could increase the resistance to virus infection such as LP-BM5 through the restraining of the multiplication.

  • The chemical composition, anti-porcine epidemic diarrhea virus (PEDV) activity and antimicrobial activity of Artemisia dubia essential oil were evaluated in this study. Fifty eight compounds from A. dubia essential oil were identified through analysis by gas chromatography-mass spectrometry (GC-MS). The major constituents of the oil were camphor (17.18 %), germacrene-D (15.70%), trans (${\beta}-$) racaryophyllene (6.79%), ene thujones (6.57%), 1, 8-cineole (5.94%) and camphene (5.08%). The essential oil was evaluated for antiviral activity against PEDV in Vero cells using a cytopathic effect (CPE) reduction method. The oils actively inhibited PEDV replication with a 50% inhibitory concentration ($IC_{50}$) of 43.7 ${\mu}^3/mL$. The 50% cytotoxicity concentration ($CC_{50}$) of the oils was over 100 ${\mu}/mL$ and the derived therapeutic index was >2.3. Similar analysis of the ribavirin revealed that they have a relatively weaker efficacy when compared to the oils. The antimicrobial activity of the essential oil against 5 microorganisms was evaluated by the disc diffusion method. The essential oil exhibited antimicrobial activity against 5 tested microorganisms with a clear zone of 8-22 mm. Among the tested microorganisms, Streptococcus pyogenes was the most sensitive and Candida albicans the least. Therefore, in can be concluded that essential oils of A. dubia may have interesting applications for microbial control or the control of PEDV-derived diseases.
