In order to investigate the method for extension of shelf-life of Kimchi, the effect of low temperature heating and addition of mustard oil on pH and total acidity of Kimchi during storage at $15^{\circ}C$ were studied. Mustard oil was found to have the antimicrobial effect on the major lactic acid bacteria of Kimchi such as Lactobacillus plantarum, Lactobacillus brevis, Leuconostoc mesenteroides and Pediococcus cerevisiae, Addition of 200p.p.m. mustard oil, 0.1% mustard powder and 0.01% $H_2O_2$ to Kimchi effectively reduced the fermentation rate of Kimchi. Low temperature heating of salted cabbage and addition of 200p·p.m. mustard oil and 0.01% $H_2O_2$ to seasonings extented the time reaching optimum ripening of Kimchi about 2.5 times longer than control. Combination of low temperature heating, addition of mustard oil and $H_2O_2$ to seasonings and post low temperature heating delayed fermentation time Kimchi about 5 times longer than control after 15 days storage at $15^{\circ}C$.

This study was conducted to investigate the content of sulfur dioxide, carotenoids and the degree of contamination of Bacillus cereus in 33 kinds of dried sweet potato from domestic mainly dried agricultural products in Korea. According to the characteristics of dried sweet potato samples, it was classified into four clusters and as a result of analyzing the contents of sulfur dioxide, carotenoids and the degree of contamination B. cereus was no significant difference among the clusters. The detection ranges of residual sulfur dioxide from 33 dried sweet potatoes ranged from 0.38 to 28.16 mg/kg, three cases (9.09%) were detected at the reference level of 10 mg/kg or more. But no samples exceeding 30 mg/kg, the tolerance level of sulfur dioxide in dried sweet potatoes were detected. Since dried sweet potato does not have a standard for carotenoids, when comparing the national and international standards of carotenoids, the range of detection of carotenoids in dried sweet potato was $46{\sim}2,663{\mu}g$/100 g, which was within the reference range of $0{\sim}9,826{\mu}g$/100 g. In principle colonies suspected to be B. cereus in dried sweet potato were not detected. In 7 cases (21.21%), there were detected in the range of 0.05~1.59 log CFU/g but not more than 3 log CFU/g as the reference value. The results of this study are expected to be used as basic data to establish quality standard for dried sweet potatoes. In order to control the quality of dried sweet potatoes in domestic market, raw materials, drying method and packaging after distribution, it is necessary to maintain and maintain the process steadily.

In vitro fermentation was conducted to figure out alternative fiber sources for horse feed. For the development of value-added products as a horse feed resource, the pomaces from apple, carrot, grape, and citrus were fermented under solid-state conditions in the presence of 60% soybean meal with 40% of each fruit pomace at 60% of moisture content. Lactobacillus plantarum SK3873, Lactobacillus plantarum SK3893, Weissella cibaria SK3880, and Bacillus subtilis SK3889 were isolated from the fermented fruit pomace by inoculation of horse feces. For the growth of Bacillus subtilis, Saccharomyces cerevisiae, and Lactobacillus plantarum, they were inoculated in 3-step order at 0, 12, and 24 h, respectively. The fruit pomace was fermented for 48 h at $35^{\circ}C$. The pH of the apple, carrot, grape, citrus and all mixed pomaces decreased from 5.45~6.25 to 4.40~4.77. Microbial growth was maintained at $10^8{\sim}10^9cfu/g$. After 12 and 24 h incubation, dry matter of carrot pomace were highest at 54.84 and 56.66%, respectively (P<0.05) and that of grape pomace was lower than others during fermentation (P<0.05). Dry matter was generally reduced by about 20%. NDF decreased gradually or maintained after 24 h, indicating the fiber degradation. Ash content tended to decrease during fermentation. After 48 hours fermentation, Bacillus, yeast and Lactobacillus showed an excellent growth by using juice by-products. These results suggest that fermented juice pomace has a potential as horse feedstuff with probiotics to maintain beneficial microflora in horse gut.
This study intends to evaluate the efficiency of non-point source reduction technique by using the porous lightweight and recycled aggregate which microorganism is seeded. In case of infiltration velocity 30~70 mm/hr in high concentration of influent, it is indicated that SS was 40~94%, COD 44~91%, BOD 4~91%, TN 1.2~66%, TP 7~70% of removal efficiency. Removal efficiency is good in infiltration velocity 30 > 50 > 70㎜/hr order. Therefore, the non-point source treatment facility filled with lightweight and recycled aggregate using microbial seeding shows higher removal efficiency than a conventional sand and gravel. We confirm that the function and efficiency are improved significantly and applied to treat non-point sources.
A series of field and laboratory experimentes were conducted to find out the effects of kinds of crops residues and the different amount of fertilizer to the microbial interaction, chemical properties, plant growth and their interaction under continous cultivation of hot pepper, soybean, sesame and upland rice. The results obtained were summerized follows : 1. Total number of bacteria and actinomycetes were enhanced by cultivation of upland rice and soybean while no appreciable effects were obtained by the cultivation of hot pepper and sesame. 2. The number of bacteria, actinomycetes and fungi were increased by return of crop residues when the cultivation of hot pepper, soybean, sesame and upland rice. Specially, actinomycetes was remarkably increased by upland rice cultivation. 3. Increased amount of fertilizer were remarkably affected to decrease of number of soil microorganisms. Specially, actinomycetes succession was appearently affects while plant growing time. 4. The number of identified soil bacterial species were obtained high in order of Bacillus sp.>Rhizobium sp.>Agrobacterium sp.>Pseudomonas sp. The number of Gram positive bacteria were superior than that of Gram negative bateria. 5. Interaction between microbial succession and crops cultivation, the number of Bacillus sp. was increased in hot pepper, Rhizobium sp. was in soybean, and sesame, and Agrobacterium sp. were increased in soybean, respectively. 6. Survival and habitate of soil microorganisms were differ with kinds of crop, application of residue and fertilizers. Most high number of Bacillus sp. Rhizobium sp. and Pseudomonas sp. were obtained on the rhizoplane and rhizosphere while Agrobacterium sp. and Fusarium sp. were high in rhizosphere. 7. Factors in relation to change of soil microbial population was obtained high in order of climates>crops>organic>matter>fertilizer.
This study was performed to determine the ranking foods and related foodborne pathogens for the impact of climate change, e.g., increasing temperature, on microbiological food safety. To do this, we developed an impact-ranking model comprising an Excel spreadsheet by using Risk Ranger. Because of a lack of data, input values in this model were determined on the basis of an expert's opinion. These values also were converted to normal distribution, and the developed model was simulated using @RISK. In conclusion, the 5 superior ranking foods and related foodborne pathogens for climate change impact were as follows: ready-to-eat foods (RTE) (Staphylococcus aureus, Salmonella spp., and Escherichia coli O157:H7); bread and rice cakes (S. aureus and Bacillus cereus); meat and egg products (Salmonella spp., E. coli O157:H7, and S. aureus); tofu (bean curds) and jellies (B. cereus, E. coli O157:H7, and S. aureus); and fish products (S. aureus, Vibrio spp., and E. coli O157:H7).
The evaluation of organic matters and nutrients removal was investigated for the synthetic wastewater and the high strength of dairy wastewater. Two different systems were performed for this research. System A composing of a single RBC with tapered aeration was fed with the synthetic wastewater for 74 days with 173L/day of influent, 200% of internal return and 100% of sludge return for the period 1 and 2. The feed conditions were maintained 346L/day of influent, 50% of internal return and sludge return for the period 3. The dairy wastewater was introduced to evaluate treatment efficiency for system B composing of dual RBCs and tapered aeration tanks for 50 days of experimental run time, in which hydraulic rates were maintained at the constant ratios of 346L/day, 50% of internal return and 50% of sludge return. The spiral string media made of nylon fibre was attached by Bacillus sp. in RBC for both systems. The specific area of string media was $1.4m^2$/m and biomass was maintained at the concentrations of 23g/m. The synthetic wastewater was supplied by 1,800mg/L of glucose, 500mg/L of $NH_4Cl$, and 500mg/L of $KH_2PO_4$ to system A. The dairy wastewater was supplied to system B from dairy production plant. The average influent concentrations were 1,334mg/L of BOD, 2,014mg/L of CODcr, 160mg/L of T-N, and 12mg/L of T-P in system A. The average influent concentrations of parameters were 1,006 mg/L for BOD, 1,875mg/L for $COD_{cr}$, 51.6mg/L for T-N and 8.9mg/L for T-P in system B. Results indicated that removal efficiencies of BOD and $COD_{cr}$ were more than 90% however, the removal efficiency of T-N was 87%, and that of T-P was 82% for system A. Removal efficiencies were 98.5% of BOD, 91.3% of nitrogen and 89% of phosphorus for system B. The removal efficiencies of organic matters, T-N and T-P were higher in system B than in system A. The effluent quality issued by the stringent national legislations for the discharge of the high strength of dairy products wastewater can be improved using sequential RBCsand tapered aeration reactors rather than a single RBC and tapered aeration reactors with Bacillus sp.
${\gamma}$-Glutamyltranspeptidase (GGT, EC 2.3.2.2) transfers ${\gamma}$-glutamyl moiety from glutamine to amino acids or peptides and hydrolyzes glutamine to glutamate and ammonia. In order to overproduce ${\gamma}$-glutamyltranspeptidase from Bacillus amyloliquefaciens (BAGGT), the encoding gene was cloned and expressed in Bacillus subtilis. The productivity of BAGGT in Bacillus subtilis was improved by 42-fold by using a dual-promoter system that was generated by combining promoters from B. subtilis ${\alpha}$-amylase and BAGGT genes. Through optimization of medium composition by Plackett-Burman design and central composition design, BAGGT was produced at $18.3{\times}10^7U/L$ of culture in the optimized medium. Compared to previously used Luria-Bertani medium, the optimized culture medium (15 g/L molasses, 60 g/L corn steep liquor, 6 g/L yeast extract, 4 g/L NaCl, 6 g/L $K_2HPO_4$, and 2 g/L $KH_2PO_4$), resulted in a 4.3-fold increase in production of BAGGT.

This study assessed the feasibility of odor removal by the application of Bacillus sp. that has many advantages in sewage treatment to sewage sludge. The NH3 removal rates in the treatment of primary sludge using only aeration were measured at 24, 48, and 72 hours of treatment and the results were 12.5 %, 12 %, and 42.1 %, respectively. The NH3 removal rates of a reactor injected with BIO-CLOD made by solidifying Bacillus sp. concentrated 10 % together with other substances were measured after 24, 48, and 72 hours of treatment and the results were 43 %, 70 %, and 81 % respectively. In the cases where the Bacillus sp. cultured in NB medium was injected into the primary sludge reactor to reach injection rates of 0 %, 1.7 %, 3.3 %, and 6.7(v/v%), the TVOC removal rates measured when 72 hours had passed after the injection were 59 %, 71 %, 88 %, and 98 % respectively, which were higher than the NH3 removal rates as the NH3 removal rates measured at the same time were shown to be 29 %, 25 %, 31 %, and 48 %, respectively. In the sludge dewaterability conducted with various Bacillus sp. injection concentrations, a Bacillus sp. concentration of 4(v/v%) was considered to be suitable. The Bacillus sp. concentrations and reduction in the bad odor substances were correlated with each other. The results showed that aeration and Bacillus sp. injection will assist biological oxidation so that the bad odor substances can be removed. Based on the SRF values of the primary sludge and digested sludge, in which Alum and PAC were used, the appropriate amount of Alum aggregate reagent was judged to be 500 mg/L, and when PAC was used, 6 mg/L was judged to be appropriate.

Lab scale experimental study was carried out for SBR process, to investigate the effects of influent ship sewage organic compound removal and Bacillus sp. state on design parameters. This process was able to remove nitrogen and phosphorus as well as organic matter efficiently. More than 95% of chemical oxygen demand(COD) were removed. In addition, about 97% of total nitrogen (T-N) was reduced. The total phosphorus(T-P) reduction averaged 93%. The performance load of SBR process was shown to be 0.095kg ${\cdot}$ TOC/$m^{3}$${\cdot}$ day. The pH was decreased from 8.1 to 7.0 within 30 min and increased to 7.3 at the end of anoxic stage, and these phenomena were explained. The sluge produced in the SBR process is characterized by low generation rate (about 0.36kg ${\cdot}$ MLSS/kg ${\cdot}$ TOC) and excellent settleability. The number of Bacillus sp. in the SBR was 24.2%, indicating that Bacillus sp. was a predominant species in the reactor.
