Effect of Activated Protein C (APC) on Apoptosis of Cancer Cells (종양세포의 사멸에 있어서의 activated protein C의 효과)

  • Activated protein C (APC) has an anticoagulant effect and a non-hemostatic effect such as regulation of cell metastasis and modulation of inflammation. In this study, we investigated whether APC could modulate apoptosis in cancer cells. Tumor necrosis factor (TNF)-${\alpha}$, cyclohexamide, and FAS markedly induced apoptosis in human renal carcinoma Caki cells. When Caki cells were pretreated with APC, the percentage of death receptor-induced apoptosis did not change. Furthermore, we checked the effect of APC on tumor necrosis factor-related apoptosis-inducing ligand (TRAIL)-induced apoptosis in human glioma T98G and human breast carcinoma MDA231 cells. APC also had no effect on TRAIL-induced apoptosis in both cell lines. However, pretreatment with APC inhibited combination treatment (kahweol plus TRAIL and kahweol plus melatonin)-induced apoptosis and PARP cleavage in Caki cells. Taken together, our results suggest that APC can modulate anti-cancer therapeutic efficiency.

Determination of Bioconcentration Factor on BPMC and Carbaryl in Carassius auratus(goldfish) (Carassius auratus(goldfish)를 이용한 BPMC와 Carbaryl의 생물농축계수의 측정)

  • Bioconcentration Factor (BCF) is known as important criteria for ecotoxicology on hazardous chemicals. But there is no standard method for determining BCF and reported BCFs were slightly different in accordance with authors. This study was performed with aims to determine BCFs on BPMC and Carbaryl. Carassius auratus(goldfish) be chosen as test organism and test period were 3-day, 5-day and 10-day. Extract solvents were n-hexane and acetonitrile. GC-ECD was used to detecting carbamates. The obtained results were as follows: 1. It was possible to determine short term BCF$_s$ of Carbaryl or BPMC through relatively simple procedure. 2. BCF$_3$ of Carbaryl in concentration of 1, 2, 5, 10 ppm were 0.34 $\pm$ 0.06, 0.18 $\pm$ 0.02, 0.10 $\pm$ 0.01, 0.06 $\pm$ 0.01 respectively. BCF$_5$ of Carbaryl were 0.34 $\pm$ 0.05, 0.18 $\pm$ 0.02, 0.13 $\pm$ 0.01 and 0.07 $\pm$ 0.01, BCF$_{10}3$ of Carbaryl were 0.45 $\pm$ 0.05, 0.27 $\pm$ 0.02, 0.16 $\pm$ 0.02 and 0.09 $\pm$ 0.01. BCF$_3$ of BPMC in concentration of 1, 2, 5 ppm were 4.66 $\pm$ 0.17, 2.64 $\pm$ 0.49, 1.88 $\pm$ 0.24 respectively. BCF$_5$ of BPMC were 4.09 $\pm$ 0.50, 2.42 $\pm$ 0.37 and 1.83 $\pm$ 0.15. 3. BCF$_s$ of BPMC were decreased as increasing concentration. However, BPMC concentration in fish were increased in contrast to BCF. But more concentrated BPMC was found in fish 3-day test than found concentration in fish 5-day test. 4. Same trend appeared in Carbaryl. BCF$_s$ of Carbaryl were decreased as increasing concentration and prolonging test period. But found Carbaryl concentration in fish were increased. 5. BCF$_s$ of BPMC were higher than that of Carbaryl by 10 times, in spite of the physicochemical properties of the two carbamates were similar to each other. Further study is recommended to find out the reason of the difference.

A Study of the Histological Effects of Garlic Extract in a Liver-damage Rat Model (마늘 추출물이 간 손상 랫드의 조직학적 변화에 미치는 효과 연구)

  • In this study we examined the effects of water extract of garlic on carbon tetrachloride-induced liver injury, and demonstrated increased beneficial enzyme and anti-oxidant activity as well as histopathological changes (by Hematoxylin-Eosin (H&E) staining, Trichrome staining, and TEM examination), and showed that the treatment was dose-dependent and safe. A total of 42 male Sprague-Dawley rats were divided equally (n=7) into six groups. To induce hepatotoxicity in these subjects, carbon tetrachloride diluted in an equal volume of olive oil was intraperitoneally administrated at 0.5 ml/kg (0.20 g/kg/day) once a day for five days. Water extract of Korean-grown garlic was administered via a stomach sonde once a day, 5 days a week, for a total of 4 weeks. Groups received 0.35 g/kg (E1), 0.70 g/kg (E2), or 1.40 g/kg (E3), with the dose adjusted for body weight. Administration of garlic extract resulted in positive physiological effects in terms of reduced oxidative stress and toxicity, and induced functional changes in the liver. Comparing the subject groups (E1, E2, E3) administered different doses of garlic extract, the importance of morphological analysis in further studies is emphasized, because morphological changes indicating hepatotoxicity could occur, even though beneficial enzyme activities were found to be elevated.

Comparison of antioxidant activities and polyphenolic compounds of extracts from artificially cultivated Sanghwang mushroom species, Phellinus linteus and P. baumii (인공재배 상황버섯 재배종 Phellinus linteus 와 P. baumii 자실체 추출물의 항산화활성과 폴리페놀 성분 비교)

  • P. linteus (Korea Sanghwang, PLKS) and P. baumii (Jangsoo Sangwhang, PBJS) were artificially cultivated under the same conditions. Fruiting bodies were extracted using 70% methanol, 60% ethanol, and hot water. Phenol and flavonoid contents were optimally extracted using 60% ethanol. Antioxidant activities of 60% ethanol extracts from fruiting bodies and mycelia from each Sanghwang mushroom species were measured using DPPH (2,2-diphenyl-1-picrylhydrazyl), ABTS (2,2'-azino-bis (3-ethylbisthiazoline-6-sulfonic acid) radical scavenging activities, and FRAP (ferric reducing antioxidant power). The antioxidant activities of fruiting bodies were relatively higher in comparison to those of mycelial samples. In high performance liquid chromatography (HPLC) analysis, styrylpyrone-class poly phenolic compounds, davallialactone, hispidin, hypholomine B, and inoscavin A were detected in fruiting body samples of two Sanghwang mushroom species, but not in their mycelial samples.

Performance Evaluation between Alternating Type Process and Recirculating Type Process by using a Mathematical Model (수학적 모델을 활용한 alternating 형태 공정과 recirculating 형태 공정의 성능 평가)

  • In this research, the performance evaluation between an alternating type process and a recirculating type process was investigated by using mathematical models. The Advanced Phase Isolation Ditch (APID) process and the $A^2/O$ process were selected the target processes of the alternating type and recirculating type, respectively. For more quantitative evaluation, 5 performance indexes which included economy and energy efficiency as well as effluent quality were used, and various disturbance conditions of influent were given to the process models. As simulation results, the APID process which had the specific operation modes to use the organic matter in influent effectively showed higher efficiency of denitrification than the $A^2/O$ process. In the case of effluent TSS, the $A^2/O$ process that the retention time in reactors could be maintained stably was more effective than the APID process. In the cases of various disturbance condition, although it was identified that both two processes had similar effluent quality, the sludge production of the $A^2/O$ process showed lower than that of the APID process while the APID process showed higher energy efficiency.

Determination of Bioconcentration Factor in Some Pesticides (일부 농약의 생물농축계수의 측정)

  • The present study was performed to investigate the bioconcentration of BPMC, chlorothalonil, dichlorvos and methidathion. The BCFs(bioconcentration factors) and depuration rate constants for four pesticides in zebrafish(brachydanio rerio) were measured under semi-static conditions(OECD guideline 305-B) in a concentration of one-hundredth of the 96 hours LC50 of each pesticide at the equilibrium condition. The results obtained are summarized as follows : The BCFs of BPMC, chlorothalonil, dichlorvos and methidathion were 1.44$\pm$0.09, 2.223$\pm$0.063, 0.81$\pm$0.08 and 5.53$\pm$0.13, respectively. Depuration rate constants of BPMC, chlorothalonil, dichlorvos and methidathion were 0.028, 0.015, 0.220 and 0.152, respectively. The concentrations of BPMC, dichlorovs and methidathion in zebrafish reached an equilibrium in 3 days, and the equilibrium of chlorothalonil was reached after 14 days. Depuration rate of dichlorvos was the fastest followed by methidathion, BPMC and chlorothalonil. The lower BCF of BPMC was due to its relatively high KOW, slow KDEP, and low SW and VP, compared to chlorothalonil and methidathion. The BCF of chlorothalonil was much lower than that excepted on the basis of high KOW, slow KDEP, SW and VP. The reason is that the experimental concentration for chlorothalonil is 1/100~1/1000 lower than that of BPMC, dichlorvos and methidathion. The BCF of dichlorvos was lower than that of other pesticides due to its very rapid KDEP, very high VP and SW, and very low KOW. The BCF of methidathion was higher than that of other pesticides due to its very low VP and SW. Therefore, these data suggest that physicochemical properties of pesticides may be important in the bioconcentration.

Research about Relationship between the Quality of life, Oral Health and Total Health of adults (성인들의 구강보건과 건강.삶의 질의 관련성 연구 - 영남지역 일부 주민들을 대상으로 -)

  • The purpose of this research is to increase adults' quality of life by improving oral health. Total 572 people participated in this study in Young-Nam area. Survey research method was used to measure various tools of participants' general index, subjective view of oral health status, oral health knowledge, oral health practice, dietary pattern, OHIP-14, THI, QOL for analyzing the effect on quality of life. The result of the study is summarized as follows; 1. Oral health knowledge has positive effects on oral health practice (0.442), dietary pattern(0.343). Diet has positive effects on OHIP-14(0.187) and OHIP-14 has significant positive effects on THI(0.564). THI also has positive effects on QOL(0.380). But oral health practice index has slight effects on OHIP-14(0.056). 2. Indirect effect indexes on QOL are OHIP-14(0.214), diet(0.040), oral health knowledge(0.019), oral health practice(0.012) listed in order of effect size, and the biggest direct effect on QOL is from THI(0.308). The result of this study shows oral health and total health are important to improve the quality of life. And the knowledge about oral health dietary pattern should be important to improve oral health. But, even with high level of oral health knowledge, the oral health practice is at low level. Health education developing program should be needed. It is required to present basic data which represent adults with national view by extracting groups using sample probability methods as of next task of this study.

Intranasal Phototherapy for Allergic Rhinitis : a systematic review (알레르기 비염의 비강 내 광 치료 : 체계적 문헌고찰)

  • Objectives : We investigated the effectiveness and safety of intranasal phototherapy for allergic rhinitis (AR). Methods : We searched 8 electronic databases (PubMed, Cochrane Library, CNKI, CiNii Articles, OASIS, NDSL, KISS, KMbase) to identify randomized controlled trials (RCTs) that reported the use of intranasal phototherapy for AR from their inception until May 30, 2020. Two investigators independently searched, collected, and screened the RCTs. We performed data extraction and evaluation for risk of bias using the Cochrane risk-of-bias tool. Results : This study included 12 RCTs; six studies compared intranasal and sham phototherapy, of which four studies reported a significant inter group difference and two studies reported a significant difference partially. No significant changes in symptoms were observed between the phototherapy and conventional therapy groups. The phototherapy and concurrent acupuncture treatment group showed a significantly higher effectiveness rate compared with the group that received only acupuncture. Both the phototherapy and laser acupuncture group showed significant improvement in the symptom severity scale scores. Six studies reported mild adverse effects, such as dryness and nasal pain in the intranasal phototherapy group; however, no severe adverse effects were reported. Conclusions : This study confirmed the safety and effectiveness of intranasal phototherapy for symptom relief and improved quality of life in patients with AR. However, further studies are needed on this topic in order to demonstrate it clearly.

A study on Smart Water Grid through IT Convergence (IT 융합을 통한 스마트 워터그리드 추진방안에 대한 연구)

  • Due to global population growth and urbanization, water problems are expected to increase in severity. In response to this expectation, Smart Water Management that refers to implement intelligent water information systems by IT convergence is considered a new paradigm. In this paper, we try to study the Smart Water policy, especially Smart Water Grid, and technology development trends in major countries. On the basis of results from the comparative analysis with Smart Grid in power sector, we evaluate political. legal, economic and technological feasibility related with the Smart Water Grid. In the conclusion, we suggest some strategic recommendations for the promotion of the Smart Water Grid.

Culture Characteristics and Optimal Conditions for Mycelial Growth of Calocybe indica (Calocybe indica의 배양적 특성과 균사 배양 적합 조건 설정)

  • Calocybe indica is an edible mushroom commercially cultivated in India and other tropical countries. In this study, the culture characteristics and optimal conditions of milky mushroom strains were determined. The growth temperature and pH range of milky mushrooms was extensively investigated between 15-35 ℃ and pH 3-11. For efficient cultivation, 20 types of nutrient sources were selected that consisted of one of 21 types of carbon sources, 6 organic nitrogen sources, 6 inorganic nitrogen sources, 13 amino acids, 6 organic acids and 12 inorganic salts. The impact of each of the selected nutrition sources and their concentration on growth was investigated. The optimum pH and temperature were determined to be pH 6.0 and 15 ℃, respectively. The optimum concentration of medium elements for the mycelial growth of C. indica was determined to be as follows: carbon source, 2% maltose; organic nitrogen source, 1% yeast extract; inorganic nitrogen source, 0.1% NaNO3; amino acid, 0.7% asparagine; organic acid, 0.07% acetic acid; inorganic salt, 0.07 mM MnSO4.
