광합성 미세조류인 Chlorococcum littorale을 이용한 이산화탄소의 생물학적 고정화

  • Chlorococcum littorale has been grown in high $CO_2$ concentrations to utilize $CO_2$ gas in the polluted air. The effect of incident light intensity on the specific growth rate is expressed by a photoinhibition model, showing half- saturation constant, $K_0\;as\;8\;(W/m^2)$ and inhibition constant, Ki as 35 $(W/m^2)$. The maximum specific growth rate was also estimated as 0.095 (1/day) under this condition. This strain maintained the optimum growth rate in 20% of $CO_2$ gas but 50% of input $CO_2$ gas is the maximum concentration considering the economical efficiency. The maximum Specific $CO_2$ consumption rate, $qCO_2$ was measured as 17.48 (mg $CO_2/g$ dry wt./day) in batch cultivation, 11.2 (mg $CO_2/g$ dry wt./day) in fed-batch cultivation and 10.87 (mg $CO_2/g$ dry wt./day) at 0.065 (1/day) of dilution rate in continuous cultivation. The chemical composition of the biomass obtained from this process showed 32.5% of protein, 27.5% of lipid, 16.5% of carbohydrate and ash 11.7%.

Prospects of Insect Biodiesel Production in Korea: A review (곤충 유래 바이오디젤의 국내 생산 가능성에 관한 고찰)

  • Biodiesel is a renewable and environmentally friendly liquid biofuel for transportation. Insect is considered as a new valuable biomass to convert into biodiesel. In particular, BSF(Black Soldier Fly) containing high fat is a renewable source of biodiesel. Biodiesel drived BSF has high concentration of saturated fatty acid methyl ester and low concentration of polyunsaturted fatty acid methyl ester which makes it potentially an ideal substrate for producing excellent quality biodiesel. Most of the fuel properties of BSF biodiesel were met the requirements of standard EN 14214. BSF have a higher lipid yield and biodiesel productivity as compared to microalgae and vegetable oils. This review paper includes the overall summary and compilation of the insect research conducted on biodiesel production and includes the BSF biodiesel properties.

Toxicity Evaluation of Organochloride Pesticide, Endosulfan and its Metabolites Using Microalgae (미세조류를 이용한 유기염소계 농약 Endosulfan 및 Endosulfan 분해산물의 독성평가)

  • A growth inhibition assay using Chlorella sp. AG 10002 based on the OECD 201 standard test procedure was applied to the toxicity testing of endosulfan and its reported metabolites. Comparison of dry cell weight, optical density (OD) at 680 nm, and chlorophyll a concentration indicated that optical density at 680 nm of culture broth is convenient, rapid, and accurate method for cell growth. In this microalgae system, the $IC_{50}$ values of endosulfan, endosulfan sulfate, endosulfan lactone, and endosulfan ether were determined as 9.45, 18.8, 18.2 and 37.5 mg/L, respectively. In a while, endosulfan diol did not show a significant toxicity up to 50 mg/L. Since endosulfan is liable at acidic or alkaline conditions, treatment of endosulfan in pH 3, 4, and 11 for 3 days resulted in reduced toxicity, as expected. These results suggested that the microalgae system is useful to evaluate various toxic chemicals and provide a new notion for bioremediation of endosulfan in aqueous systems.

Characteristics of Microalgal Growth on Anaerobic Effluent of Animal Waste (축산분뇨 혐기성 처리수에서 미세조류의 성장특성)

  • Characteristics of microalgal growth was investigated using anaerobic effluent from two-phase animal waste digestor as substrate. Batch experiments were carried out to investigate the effect of the initial nitrogen and phosphorus concentrations on growth of Microcystis aeruginosa, Chlorella sp. and Euglena gracilis. In 400 times diluted anaerobic effluent (TN 3 mg/L), single cell growth of the Euglena gracilis population increased twice without delay, although Chlorella sp. and Microcystis aerugenos take over 144 hours. Similar appearance with single cell growth was observed in mixed cultures. However, microalgae population did not increase under condition of 10 times diluted influent (TP 3 mg/L) in both pure and mixed cultures, which was affected by high organic and nitrogen concentration. Logistic growth model successfully fitted to determine biokinetic parameters such as ${\lambda}$: lag time, ${\mu}m$: maximal specific growth rate, A: asymptote of growth.

Effects of nutrient and inorganic carbon on single cell formation of Pediastrum duplex (Pediastrum duplex의 single cell 형성에 미치는 영양염류 및 무기탄소의 영향)

  • In this study, effects of nutrient and inorganic carbon on single cell emergence during the cultivation of microalgae were observed using colonial green algae, Pediastrum duplex. The concentration of inorganic carbon had significant effect on single cell emergence and its growth, but nitrogen and phosphorus concentration showed minor effects. According to P. duplex cultivation experiment, single cell started to be emerged around 500~750 mg-C/L of inorganic carbon concentration and it was bloomed dramatically at the higher values. And growth of P. duplex was started to be surpressed at the single cell formation concentration. From the results, it could be said that when we operate the microalgae systems for cultivation/harvesting or wastewater treatment, in order to avoid single cell formation, inorganic carbon should be maintained to the proper level.

  • As a preliminary study, effects of environmetal pollutants on propagation of freshwater microalgae, Cryptomonas ovata and feeding rate of Corbicula leana were investigated at 20${\pm}$1$^{\circ}C$, over 20 days after treatment of pollutants, glucose, complex fertilizer and NH4Cl. Number of C. ovata in control group was increased from 38${\times}$104 cell/ml to 1.910${\times}$104 cell/ml after 20 days cultivation in Sorokin-Krauss medium. Increments of cell number in experimental groups treated with glucose, complex fertilizer and NH4Cl were higher than that of control group. The higher propagation rate of C.ovata was observed when 30 mg/l of glucose treated, 120 mg/l of complex fertilizer treated, and 4 mg/l of NH4Cl treated, compared with other concentrations in each pollutant treated group. The feeding rates of large size group of C. leana which fed with a living organism, C. ovata in each experimental group were higher than small size group, and slightly reduced with the increase of pollutant concentrations. The feeding rates were not significantly different between any concentrations of the pollutant, and among experimental groups treated with glucose, complex fertilizer and NH4Cl.

  • The Green Lamp Fixture (GLF) of LED prepared with thin panel structure was investigated for illumination of street lamps and other lighting system uses, which was also very useful to aquaculture and aquafarm lighting uses, or fish luring lights and marine aquanautics of aquamarinautics (aquamarine+aquanautics) uses, etc. In the case of fish luring lights, it was verified that the fish luring of Green Lamp Fixture of LED was very effective for phototaxis movement and ecological community promotion to the micro-living things of organisms and the small fries and fishes, like as 'crowding together'. For the aquaculture lightings, it was also very excellent in waterproof and heat-sink properties, photosynthetic growing of algae and micro-organisms, water-weeds and seaweeds living underwater.

On-off Control for Continuous Culture of Microalgae in Flat Panel Photobioreactor (평판형 광생물반응기의 미세조류 연속배양을 위한 On-off 제어)

  • Recently, technologies that produce biofuels from microalgae are being studied worldwide. It is necessary to significantly reduce the production costs of biofuels from microalgae for economic reasons. In this study, the growth curve of the microalgae was obtained using the batch-culture method, and the specific growth rate was predicted using the regression method. Based on the culture conditions of the estimated specific growth rate, the turbidity of the microalgae in the flat panel photobioreactor (PBR) was measured. Furthermore, an on-off control scheme was applied to the flat panel PBR in order to culture the microalgae continuously on the basis of turbidity. The parameters of the on-off control system were displayed by LabView. The on-off scheme of peristaltic pump was controlled based on the turbidity in the PBR. In addition, the turbidity values of growth curves were compared and analyzed in the continuous culture process using the on-off controller.

Treatment of Acid Mine Drainage using Eggshells and Microalgae (폐난각과 미세조류를 이용한 산성광산배수처리)

  • The aim of this study was to investigate the heavy metal removal and biomass productivity in the Acid Mine Drainage (AMD) using eggshell and microalgae. The experiment was operated 6 days in the eggshell and microalgae hybrid system, and using eggshell powder and microalgae as Chlorella vulgaris. The obtained result indicated that the biomass productivity of 2.82 g/L/d from 1.12 g/L initial concentration in 6 days was reached with light transmittance of 97% at a 305 mm depth in the optical panel photobioreactor (OPPBR). The total removal efficiency of Fe, Cu, Zn, Mn and Cd was found to be 98.92%, 99.91%, 98.78%, 88.99% and 98.00% in the AMD using eggshell and Chlorella vulgaris hybrid system, respectively. Additionally, there were significant relationships between biomass and concentration of each heavy metal ($R^2$ = 0.8771, 0.8643, 0.8669, 0.9134 and 0.6277 for Fe, Cu, Zn, Mn and Cd). These results indicated that the eggshell and microalgae hybrid system was highly effective for heavy metal removal when compared to the conventional biological process in the AMD. Therefore, the eggshell and microalgae hybrid system was effective for heavy metal removal and biomass productivity and can be applied to treat AMD in treatment plant.

미세조류의 Methane 발효특성

  • This study was carried out to examine degradation characteristics of microalgae Chlorella vulgaris in methane fermentation. We measured COD and VS reduction, gas and methane productivity, VFA (volatile fatty acid), respectively. Then we calculated material balance and hydrolysis rates in soluble and solid material. The substrate concentration was controlled from 14 gCOD$_{cr}$/l to 64 gCOD$_{cr}$/l in batch cultures, and HRT (hydraulic retention time) controlled from 2 days to 30 days in continuous experi- ments. The results were as follows. In batch culture, accumulated gas productivity increased with the increase of the substrate concentration. The SS and VSS was removed all about 30% increase of substrate concentration and the most of the degradable material removed during the first 10 days. The curve of gas and methane production rate straightly increased until substrate concentration is 26 gCOD$_{cr}$/l. In continuous culture experiments, the removal rates at HRT 10days were 20% for total COD and TOC, respectively. At longer HRT, there was no increase in the removal efficiency. At HRT 15 days, the removal rates were 30% for SS and VSS, respectively. Soluble organic materials were rapidly degraded, and so there was no accumulated. Soluble COD concentration was not increase regardless of HRT-increasing. That meaned the hydrolysis was one of the rate-limiting stage of methane fermentation. The first-order rate constants of hydrolysis were 0.23-0.28 day$^{-1}$ for VSS, and 0.07-0.08 day$^{-1}$ for COD.
