Effect of Fucoidan on Angiogenesis and Gene Expression in Human Umbilical Vein Endothelial Cells (후코이단이 혈관 내피세포의 신생혈관 생성 효과 및 관련 유전자의 발현에 미치는 영향)

  • Angiogenesis is a process including members of the angiogenic factors. In particular, fibroblast growth factor 2 (FGF2) is considered the most potent angiogenic factor because it promotes cell proliferation and tube formation. A recent study reported that fucoidan derived from marine plant potentiated FGF-2 induced tube formation in human endothelial cells. On the other hand, the molecular mechanisms involved in the angiogenic activity of fucoidan and FGF2 are unknown. In this study, a fucoidan treatment promoted angiogenesis induced by FGF2. The effects of fucoidan on FGF2-induced angiogenesis were confirmed by a proliferation assay using a CellTiter96 Aqueous One solution after a treatment with fucoidan and FGF2. The tube formation and wound healing assay for the angiogenic activity were also confirmed. Reverse transcription PCR showed a change in the mRNA of vascular endothelial growth factor-A (VEGF-A), intercellular adhesion molecule-1 (ICAM-1), matrix metallopeptidase9 (MMP9), and the signal transducer and activator of transcription3 (STAT3). In summary, the Fucoidan/FGF2 treatment induced an increase in cell proliferation, improved the tube formation and wound healing activity, and altered the STAT3, VEGF-A, ICAM-1, and MMP9 mRNA expression levels. Further research will be needed to provide a scientific explanation in terms of cell-signaling and confirm the present findings.

  • 장시간 허혈상태의 토끼 이개를 실험대상으로 하여 유로키나제와 헤파린을 병용 또는 단독으로 사용시와 또한 약물을 사용하지 않았을 때, 이들이 모세혈관의 개존성과 아울러 미세수술후의 조직 생존율에 미칠 수 있는 효과를 보기 위하여 허혈상태의 토끼 이개를 미세수술로 접합한 후 모세혈관으로의 혈류를 측정하기 위하여 레이저 초음파 혈류측정기(Laser doppler flowmetry)를 이용하였으며 방사선 구슬들(Cobalt-57 with plastic material with average diameter 15 micron)을 주입한 결과 유로키나제 조합에서 통계적으로 유효한 성적을 얻었다. 광학현미경 소견은 유로키나제와 헤파린을 병용한 조합에서 모세혈관내 내피세포의 배열이 유지되어 있었으며 헤파린을 사용한 조합에서도 유사한 소견을 보였으나 약물을 사용하지 않은 조합에서는 국소적인 내피세포의 배열이 결핍되어 있었다. 전자현미경 소견에서 유로키나제와 헤파린을 병용한 조합에서 내피세포가 혈관내벽에 배열되어 있었고 또 불규칙한 세포질이 돌출되어 있었다.

Stress Distribution in Microvascular Anastomotic Coupler (AnaFix®) Micropins with Respect to the Fillet Radius (필렛효과에 따른 미세혈관 문합커플러(AnaFix®) 마이크로핀의 응력분포)

  • An automated anastomotic ring-pin system consisting of both the anastomotic ring-pin system and the coupler device has eliminated the drawbacks of the suture method. High density polyethylene (HDPE), a material with outstanding biocompatibility and injection molding capability, was used in the ring. SUS316 stainless steel, Ti-6Al-4Nb, Ti-6Al-4V, and unalloyed titanium were used in FEM simulations of the micropin. The authors categorized the microvascular anastomotic ring micropins into short neck (SN) and long neck (LN) groups in order to evaluate the effect of the micropin's fillet radius and neck length on the von Mises stress. The micropins were further divided into those with and without fillet. On the basis of the fillet radius rate (FRR), which represents the rate of change in the von Mises stress with respect to the availability and shape of the fillet, and the neck length rate (NLR), which represents the rate of change in the von Mises stress with respect to changes in the length of the neck within the fillet shape, it can be concluded that the SN-3 neck design is the most stable.
