Microstructure and Microdefects of Diamond Thin Films Deposited by MPECVD (마이크로웨이브 화학증착법에 의한 다이아몬드 박막의 미세구조오 미세결함)

  • Diamond thin films were deposited on p-type (100) Si wafers using MPECVD. Prior to deposition, ultrasonic striking was done to improve density of nucleation sites with dimond powder of 40~$60\mu$m size. Then diamond thin films were deposited at $^900{\circ}C$, 40Torr and 1000W microwave power using ${CH}_{4}$ and ${H}_{2}$ gases. The purity, the morphology and the microstructur'e and microdefects of diamond thin films were characterized by Raman spectroscopy, SEM and TEM, repectively. In Raman spectroscopy the peaks of non-diamond phase increased as ${CH}_{4}$, concentration increased. In SEM, the morphology of diamond thin films varied from crystalline to cauliflower as ${CH}_{4}$, concentration increased. As ${CH}_{4}$ con centration increased, the density of defects increased, with most defects being {III} twin. ${MTP}_{5}$, were formed with five (II]) planes. As these (Ill) Planes were twinned, ${MTP}_{5}$, represented five-fold symmetry. ]n the interfaces, defects in diamond thin films fanned out from small regions implying nucleation sites.

Study on evaluating the significance of 3D nuclear texture features for diagnosis of cervical cancer (자궁경부암 진단을 위한 3차원 세포핵 질감 특성값 유의성 평가에 관한 연구)

  • The aim of this study is to evaluate whether 3D nuclear chromatin texture features are significant in recognizing the progression of cervical cancer. In particular, we assessed that our method could detect subtle differences in the chromatin pattern of seemingly normal cells on specimens with malignancy. We extracted nuclear texture features based on 3D GLCM(Gray Level Co occurrence Matrix) and 3D Wavelet transform from 100 cell volume data for each group (Normal, LSIL and HSIL). To evaluate the feasibility of 3D chromatin texture analysis, we compared the correct classification rate for each of the classifiers using them. In addition to this, we compared the correct classification rates for the classifiers using the proposed 3D nuclear texture features and the 2D nuclear texture features which were extracted in the same way. The results showed that the classifier using the 3D nuclear texture features provided better results. This means our method could improve the accuracy and reproducibility of quantification of cervical cell.

The Ultrastructure of the Cutaneous Pigment Cells in the Amphibia (양서류 피부 색소세포의 미세구조)

  • The ultrastructures of the pigment cells in the Asiatic land salamander (Hynobius leechi) dorsal skin were obtained by means of electron microscope. The results were as follows; 1. The pigment cells of the epidermis consisted of the melanocytes in the germinal layer and of the melanophores distributing to the keratinocyte layer. The traits of these cells in the epidermis were as follows: A. The nuclei of the melanocytes were round or oval in shape and appeared as partly small or large infoldings of the nuclear envelope. B. Rough-surfaced endoplasmic reticulums and Golgi complexes were well developed in infranuclear cytoplasm. Many ribosomes were mainly distributed around the perinuclear portion. C. The melanosomes of the melanocytes were observed as a found or an oval shape and strong electron-dense or less electron-dense melanosomes were observed. D. The infoldings of the nuclear envelope in the melanophore were partly found deeper than those of the melanocyte. The cytoplasm of the melanophore filled with melanosomes caused organelles not to be observed in that. 2. The pigment cells in the dermis were composed of the xanthophores just beneath basement membrane and the melanophores in the connective tissue. The traits of these cells in the dermis were as follows: A. The xanthophores contained round or oval vesicles, and these vesicles were divided into 6 types (type I pterinosome, type II pterinosome, type III pterinosomes, type iv pterinosome, type V pterinosome, type VI pterinosome). B. Most of the nuclei of the melanophores in the dermis were elongate in shape, and a portion of the nuclear envelope was deep infolded. C. Becuase the cytoplasm was filled with the melanosomes of the same electron-density, organelles were not observed in the cytoplasm. D. Two processes of the melanophore in the dermis extended in parallel with a xanthophore and the cytoplasm in those processes were filled with the melanosomes.

Gill Ultrastructure of the Spiny Top Shell, Batillus cornutus (Gastropoda: Turbinidae) (소라, Batillus cornutus 아가미의 미세구조)

  • Gill morphology and ultrastructure of the spiny top shell, Batillus cornutus were described using light and electron microscopy (SEM and TEM). The spiny top shell, Batillus cornutus has bipectinate gill. The epithelial layer of gill filament was simple and composed of columnar epithelium, ciliated cell, mitochondria-rich cell and secretory cell. Microvilli were well-developed on the free surface of columnar epithelial cell. The epithelial cells are connected to the neighboring cells with intercelluar junctions at the apico-lateral surface. The cilia and microvilli were commonly observed on the free surface of ciliated cell. Tubular mitochondria appeared in the apical cytoplasm, and connected ciliary rootlet. Mitochondria-rich cells contained a oval-shaped nucleus in the basal area. And majority of cytoplasm was occupied by well-developed mitochondria. Result of AB-PAS (pH 2.5) and AF-AB reaction showed that secretory cells contained mainly acidic carboxylated mucosubstances. Secretory cells are unicellular glands and can be divided into four types (A, B, C and D) depending on the cell shape and ultrastructure of secretory granules.

Acute High-Altitude Cerebral Edema Presenting as Extensive Microbleeds along the Corpus Callosum without T2 Hyperintensity: A Case Report and Literature Review (T2 고신호강도가 동반되지 않은 뇌량의 광범위한 미세출혈의 형태로 나타난 고산병: 증례 보고 및 문헌 고찰)

  • High-altitude cerebral edema (HACE) is a potentially fatal neurological syndrome that develops in persons traveling to a high altitude. We report the case of a 49-year-old male who had traveled to a high altitude, and lost consciousness for a few hours. Susceptibility-weighted images revealed multiple, fine black pepper like microbleeds along the corpus callosum with several microbleeds in the left frontal and parietal subcortical white matter. The T2-weighted images did not show any abnormal signal intensities along the corpus callosum. The diffusion-weighted images revealed small nodular high signal intensities in the basal ganglia. This report describes the atypical radiologic findings of HACE showing multiple microbleeds along the corpus callosum, without abnormal high-signal intensity on T2-weighted images.

Ultrastructure of Germ Cells, Cyst Epithelial Cells and Interstitial Cells during Spermatogenesis of the Stone Flounder, Kareius bicoloratus (돌가자미 Kareius bicoloratus의 정자형성과정 중 생식세포, Cyst 상피세포 및 간질세포의 미세구조)

  • Ultrastructure of germ cells, the cyst epithelial cells and interstitial cells during spermatogenesis of the stone flounder, Kareius bicoloratus (Pleuronectidae) sampled on the west coast of Korea were investigated by electron microscopic observations. In the primary spermatocyte, the synaptonemal complexes appear in the zygotene stage of the prophase during maturation division. In the growing testis, especially, the interstitial cells (Leydig cells) appear near the primary, secondary spermatocytes and spermatids. Well-developed interstitial cells (steroid hormone secreting cells) which are located in the interlobular space in growing testis have three morphological characteristics of a vesicular nucleus, mitochondria with tubular cristae and smooth endoplasmic reticulum. During spermatogenesis, the primary and secondary spermatocytes attach to the cyst epithelial cell (Sertoli cell) having an elongated ovoid or triangular nucleus and several mitochondria in the cytoplasm. In the growing testis, lipid droplets, the mitochondrial rosettes and glycogen particles appear in the cytoplasm of the cyst epithelial cells near the secondary spermatocytes and spermatids. Particularly, the mitochondria, endoplasmic reticulum, little lipid droplets and the large amount of glycogen particles are present in the cytoplasm of the cyst epithelial cell in the late growing testis. In the late stage of spermiogenesis, the proximal centriole is joined to the nuclear envelope, the distal centriole forms the basal body of the flagellum and gives rise to the axial filament of the flagellum. No acrosome of the sperm is formed as seen in other teleost fish. The head of the spermatozoon is approximately $3{\mu}m$ in length and its tail is about $30{\mu}m$ in length. The axoneme of the tail flagellum of the spermatozoon consists of nine outer doublet microtubules at the periphery and two centrial singlet microtubules at the center. The spermatozoon of this species has two axonemal lateral fins. Especially, the cyst epithelial cells which located near groups of gametes in the various stages, show three functions: nutrition, phagocytosis and steroidogenesis. Especially, the nuclei of cyst epithelial cells in the recovery stage of the testicular developmental stages appear to be irregular in shape after spermiation. Of three functions of the cyst epithelial cell, several characteristics of phagocytosis are showed in the cytoplasm of the cyst epithelial cells in the recovery stage of the testicular developmental stages. At this stage, therefore, it is assumed that the cyst epithelial cells are involved in degeneration and resorption of undischarged germ cells after spermiation.

고밀도 알루미늄 박막이 코팅된 강판의 부식 특성

  • 알루미늄은 경량 금속으로 부식 저항력이 높아 철을 부식으로부터 보호하기 위한 표면처리 소재로 사용되고 있다. 철의 부식을 방지하기 위해서 알루미늄을 코팅하는 경우, 코팅 방법은 용융도금법이 주로 사용되고 있으며, 알루미늄을 빛의 반사막으로 활용하는 경우 진공 중에서 물리기상증착(physical vapor deposition; PVD)법을 사용하기도 한다. 알루미늄 박막을 물리기상 증착으로 코팅하면 박막성장 초기에 핵(nucleus)을 형성하고, 형성된 핵을 중심으로 주상정(column)으로 박막이 성장하는 것이 일반적이다. 알루미늄 박막의 주상정과 주상정 사이에 공극(pore)이 존재하기 때문에 알루미늄 박막을 부식방지 막으로 이용하기 위해서는 두께를 증가시켜야 한다. 본 연구에서는 스퍼터링(unbalanced magnetron sputtering)을 이용하여 치밀한 조직을 갖는 알루미늄 박막을 코팅할 수 있는 공정변수를 도출하고, 치밀한 알루미늄 조직이 철의 부식에 미치는 영향을 평가하였다. 기판은 냉연강판(cold rolled steel sheet)이 사용되었으며, 알루미늄 타겟의 크기는 직경 4 inch이었다. 알루미늄 박막의 미세조직과 밀도에 영향을 주는 공정변수를 확인하기 위해서 스퍼터링 파워, 공정 압력, 외부 자기장 세기 등의 조건을 변화시켜 코팅을 실시하였다. 알루미늄 박막의 밀도 변화에 가장 큰 영향을 준 공정변수는 외부 자기장의 세기와 방향이었다. 알루미늄 박막이 약 3 ${\mu}m$의 두께로 코팅된 냉연강판을 염수분무시험(salt spray test, 5% NaCl)으로 부식특성을 평가한 결과, 시험을 시작한 후 120시간 후에도 적청이 발생하지 않았다. 이러한 결과는 기존의 동일한 두께를 갖는 알루미늄이 코팅된 강판의 내부식 특성의 2배의 성능을 보여준다.

Ultrastructural Localization of GABAergic Neuronal Components in the Dog Basilar Pons (개의 교핵내 GABA성 신경세포 성분의 미세구조적 위치관찰)

  • An immunocytochemical study of GABA-positive neuronal elements was performed at the electron microscopic level to examine subcellular distribution of the inhibitory neurotransmitter in the dog basilar pons. Electron-dense reaction product was observed in neuronal somata and dendritic processes. One or more unlabeled axon terminals made asymmetric synaptic contacts with these GABAergic somatic and dendritic profiles. A large number of GABA-positive axon terminals were also observed. They made symmetric as well as asymmetric synaptic contacts with unlabeled dendritic profiles. In axo-axonic synapses, postsynaptic axon-like processes were consistently GABA-immunoreactive. These observations suggest that the inhibitory local circuit neurons in the dog basilar pons play a major role in cerebro-ponto-cerebellar circuitry by integrating various afferent inputs and conveying them into the cerebellar cortex and the deep cerebellar nuclei.

  • Urechis unicinctus spermatogenic cells, sperm and spermatids, prepared from testis are investigated to identify nuclear actin using amoeba monorlonal anti-actin as the first Ab and gold particles (10 nm) conjugated mouse IgG (immunogold) as the Ab marker. The Ag-Ab reactions analyzed the localization of nuclear actin of the spermatogenic cells and the immunogold particles incorporated mainly with nuclear matrices. A few immunogold particles are merged into the acrosomes and the other architectures of spermatogenic cells, such as mitochondrion and centrioles. It is often observed and there is a tendency in which the incorporated immunogold particles are increased in number in the nuclear matrices of sperm compared with that of spermatids The increments and decrements of the incorporated immunogold particles according to developmental stages and the spermatogenic architec-tures are interpreted and discussed in aspect of acrosomal function and of nuclear condensation of spermatids.

Textural, Isotopic, and Chemical Investigation of Cultured Pearls (양식진주의 조직적, 동위원소적 및 화학적 연구)

  • Cultivated pearls, composed of aragonite crystals, show two distinctive layers: nacreous and conchiolin organic layers. Each aragonite crystal is surrounded by organic matrix, which probably consists of amino acids. Nucleus, surrounded by pearl layer, also consists of nacreous crystals, suggesting that there is a close mineralogical and ultrasturctural relationship between pearl and nucleus. Carbon isotopic values of cultivated pearls are within the range of marine carbonate carbon. Oxygen isotopic composition indicates that the temperature for the growth of pearl and pearl oyster ranges from 16.4 to $21.4^{\circ}C$ and from 15.5 to $24.8^{\circ}C$, corresponding to the summer temperature range of the cultivating area. Elemental composition of pearl, pearl oyster, and nucleus shows that there is a difference in chemical composition depending upon the original mineralogy and the chemical composition of water in which shells grow. Especially, a strong relationship exists between pearl and the inner layer of pearl oyster because both are composed of nacreous aragonite and formed in a shallow marine environment.
