Ultrastructural Studies on Mercury Poisoning in the Liver, Kidney and Gills of Carassius carassius L. (수은중독에 의한 붕어(Carassius carassiusr L.) 장기의 미세구조 변화)

  • Electron microscopic studies were made to investigate changes in the fine structure of the liver, kidney and gills of Carassius carassius L. following exposure to 1 and 2.5 ppm of $HgCl_2$. The following results were obtained: 1. In the mercury-treated liver cells, an increase in the number of lysosomes were noticed. These lysosomes appeared to be of two types; round ones containing some crystalline structures and others with phagocytosed glycogen granules and mitochondria. Also observed were mitochondrial swelling where the matrix appeared less electrondense, and segregation of the nucleoli in the nucleus. 2. In the kidney, mercury treatment resulted in thickening of the basement membrane of the glomerulus, and appearance of vacuoles and cytoplasmic bodies in the proximal convoluted tubule. The vacuoles seemed to be formed from mitochondria. Nuclear shrinkage was also noticed at 2.5 ppm of $HgCl_2$. 3. Many large and small lysosomes appeared in response to mercury in the epithelial cells of the gill lamella. Also the lamellar membrane became fuzzy in appearance. 4. It can be concluded from these results that mercury-induced changes in the fine structure are associated with activation of detoxication processes and impairment of energy metabolism.

  • This study described the spermatozoa of the discoglossidae Bombina orientalis using light, scanning and transmission electron microscopes. Sperm head possess a crescent or leaf shape, with a moderately flexible head, and with a sharp anterior and posterior tips. The nucleus are a thick cone shaped in the widest middle part of nucleus, and a slender anterior and posterior of nuclear tips. The chromatin is not completely compact, but irregularly imbricated such as roof. Some nuclear lacunae, irregular in shape, are scattered within the nucleus. No neck and middle piece were developed. The flagellum is composed of 9+2 axoneme, axial rod and undulating membrane. The mitochondria were distributed only in cytoplasmic membrane around the nucleus. In particular, the nuclear rod contains bundles of fibers, the rod penetrating from anterior portion to the middle of the nucleus, is extended roughly two-thirds of the nucleus such as eyelashes shaped.

Electron Microscopic Studies on the Interpeduncular Nucleus and the Habenulo-Interpeduncular Projection in the Rat (흰쥐 뇌각간핵 및 고삐-각간로에 관한 전자현미경적 연구)

  • Electron microscopic degeneration studies were carried out on the rat's interpeduncular nucleus following lesion on both habenular nuclei to clarify the morphological nature of their presynaptic endings involved in the habenulo-interpeduncular pathway. Two distinct types of interpeduncular neurons were observed: 30% of large neurons and 70% of small ones. Four types of boutons with many characteristics could be differentiated; small spherical vesicle-containing boutons (40%), large spherical vesicle-containing boutons (18%), pleomorphic vesicle-containing boutons (33%), and elongated vesicle-containg boutons (9%). Vesicle containing dendrites were observed as dendro-dendritic synapses and axo-axonal synapses were also found, though rarely. Two to four days after the lesion on both habenular nuclei, the small spherical vesicle-containing boutons were found almost exclusively in the process of degeneration, while the other types were entirely free of degeneration. Therefore, it is inferred that habenural neurons terminate in the interpeduncular nucleus as small spherical vesicle-containing boutons. The neuronal origin of the other bouton types is discussed.

Studies on nuclear transplantation in mouse embryos II. Developmental potential of nuclei from embryos of different developmental stages (생쥐 수정란의 핵이식에 관한 연구 II. 발달단계별 수정란 핵의 이식후 생존성)

  • Single nuclei from two-, four- and eight-cell mouse embryos were transplanted into enucleated two-cell embryos by micromanipulation and Sendai virus mediated fusion. The developmental potential of these reconstituted embryos in vitro and in vivo was examined. It was found that the single nuclei which were transplanted to enucleated two-cell embryos were not only able to develop to the blastocyst stage in vitro(two-cell nuclei, 76.5%; four-cell nuclei, 68.4%; eight-cell nuclei, 48.3%), but also able to develop to full term in vivo after transfer to recipient mice(two-cell nuclei, 37.1%; four-cell nuclei, 29.6%; eight-cell nuclei, 16.3%). Although the proportion of live young produced after transfer of nucler of nuclear transplant embryos which received eight-cell nuclei was significantly (p<0.05) reduced, it would be suggested that the overall efficiency in producing identical offspring is greater when eight-cell embryos were selected for nuclear donor than two- or four-cell embryos were selected.

  • Paraganglioma is a benign tumor arising in the paraganglion system scattered throughout the body, but its cytopathologic findings arenot well known. We experienced a case of paraganglioma of carotid body diagnosed by fine needle aspiration. The patient was a 30 year-old female who suffered from the left neck mass for 3 years. The mass was $3\times3cm$ in size without pulsation or bruit. Cytologically, the smear revealed aggregated and singly scattered tumor cells haying abundant pale cytoplasm and indistinct cell borders. Their nuclei were round to oval, but enlarged nuclei were occasionally observed. The nuclear membrane was smooth with fine clumping of chromatin. Differentiation from metastatic follicular carcinoma of the thyroid gland was difficult.

Induction of Oocyte Maturation of Korean Frogs by OKadaic Acid in Vitro (Okadaic acid에 의한 한국산 개구리 난자의 성숙유도)

  • Phosphatase의 저해제로 알려진 okadaic acid(OA)가 한국산 개구리(북방산개구리 참개구리) 난자의 성숙에 미치는 효과를 조사하였다. 북방산개구리 난자에 약 50 nl의 okadaic acid(0 5-500 mM)를 미세주입한 후 18시간 배양한 결과 0.5 mM의 농도에서 부터 난자의 핵붕괴를 일으키기 시작하였다 동면초기에 처리한 progesterone에 반응하지 않는 난자들도 OA에 의하여 성숙을 일으켰으며. 이 성숙은 배양액에 첨가한 cycloheximide(10 mg/ml)에 영향을 받지 않았다 또한 OA의 처리를 받고 일정시간 배양한 난자의 세포질에는 미성숙 난자의 성숙을 유도하는 maturation promoting factor (MPF)의 활성이 생기었다 참개구리의 난자도 역시 OA에 의해 성숙이 유도되었으며, 주입 후 6시간에서부터 핵붕괴가 일어나기 시작하였다 참개구리에서도 OA의 처리가 MPF의 활성을 촉진하는 것을 세포질내에 Hl histone kinase의 활성도가 증가하는 것으로 확인할 수 있었다 참개구리에서도 OA에 의한 성숙은 cycloheximide나 CAMP에 의해 영향을 받지 않았다 이러한 결과들은 개구리 난자의 MPF 활성화와 성숙과정에 phosphatase가 관여함을 보여주는 것이다.

  • 동양달팽이(Nesiohelix samarangae)의 자성생식기 중 정자낭(spermatheca)의 미세구조를 투과전자현미경을 이용하여 관찰하였다. 성체의 정자낭의 내강에서는 정자와 이와 관련된 물질들 또는 상피로부터의 분비물질들이 관찰되었다. 정자낭의 내강을 둘러싸고 있는 벽의 상피는 단층원주상피로서 내강과 연한 세포의 원형질막은 미세융모를 가지고 있었으며 섬모는 가지고 있지 않았다. 이 미세융모들의 기부에서는 고배율 하에서 pinocytotic vesicle들이 관찰되었고 때로 세포의 분비물을 세포 밖으로 방출하는 모습도 관찰되었다. 세포들의 상부세포질에는 많은 수의 mitochondria가 있었고, 때로 분비과립과 lysosomd들이 존재하기도 하였다. 이들보다 아래에는 잘 발달한 골지체와 과립소포체(RER)들이 있었다. 이 부위에서 분비과립들이 많이 형성되어 있는 모습이 세포에 따라 관찰되었고, 그 아래에 즉, 각 세포의 하반부 세포질에 상하로 긴 핵이 관찰되었다. 세포의 기부 원형질막은 상부 세포질 내로 깊게 그리고 수없이 많이 주름을 이루고 있어 인접한 세포와의 경계를 구분할 수 없을 정도였다. 기저막의 아래에는 근육다발과 색소세포들이 결합조직 내에서 관찰되었고, 정자낭의 외막은 큰 액포로 인하여 세포질들이 원형질막 주변으로 밀려나 있는 특이한 세포들로 이루어져 있었다. 정자낭의 기능은 정자를 일시 저장하며 정자낭 상피에서 분비되는 소화액들로 정자를 일차적으로 세포외 소화하고 소화되지 않은 물질들을 흡수하여 이차적으로 세포내소화하는 것으로 생각된다.

A Study on the Properties of Hollow Silica Microspheres for Controlled-release Pesticide Formulation (농약 방출 조절제 소재로서 실리카 중공 미세구의 물성연구)

  • Pesticide formulations for controlled release were pepared with hollow silica microspheres. The hollow microsphere, which was obtained through calcination for the core removed after silica coating, showed maximum impregnation of benfuracarb up to 2.7 times of its mass in comparison with those obtained through the other core removal method. The release test of the pesticide formulation, when used with ESO(Epoxidized Soybean Oil) as a binder, showed ideal release pattern with steady release rate from the day 10 to 30 retaining the benfuracarb concentration in the water around 1.65 ppm.

  • Fine structure of spermatozoa of the bastard halibut (Paralichthys olivaceus) is described. Its structure is characterized by acrosomeless, having a spherical head region and a small mid piece containing five mitochondria disposed one row. The flagellum inserts centrally on the nucleus and has lateral ribbons. The present study supports the suggestion that spermatozoal ultrastructure is of taxonomic importance and may be a phylogenetic criterion in teleostei.

  • The ultrastructure of the gills of Oplegnathus fasciatus was examined by means of light and transmission electron microscopes. The gills have primary and secondary filaments (lamellae). The following cells are identified and described : epithelial cell, pillar cell, chloride cell and mucose cell. The simple epithelial layer consists of squamous epithelium containing a large nucleus and the surface is covered with some of microridges. The lamella pillar structures are characterized by axial microtubules and lateral membrane interdigitations. Chloride cells contain a lot of mitochondria and specifically developed tubular systems. The rough endoplasimic reticulum and golgi complex, and some of mucous granules were observed in immature mucous cells. The mature mucous cells were AB-PAS positive, globular in shape, and had mucous granules of similar size with various electron densities.
