The Study of Factor Concerning Ovum Pick-up and Conception Rate by Ultrasound-Guided Follicular Aspiration in Hanwoo Heifers (한우에 있어서 초음파기기를 이용한 생체내 난자 채취시 채란조건 및 수태율에 관한 연구)

  • This study was carried out to investigate of factors concerning to ultrasound-guided follicular aspiration; level of vacuum pressure, diameter of use needle, effect of FSH hormone and conception rate after embryos transfer. 1. Oocytes collection number were 4.2$\pm$2.9 e.a to luteuml phase and follicular phase were 4.4$\pm$3.5 e.a to ovaries of Hanwoo. 2. We taked proper level of aspiration vacuum pressure was 40~120 mmHg to oocytes collection. Oocytes collected number were 4.2$\pm$3.2, 4.3$\pm$3.4, 4.5$\pm$3.4 e.a. to 40, 80, 120mmHg, respectively, follicles aspiration rate were 49, 47, 45%. 3. Effect of collection needle diameter was not difference significantly(P<0.05), oocytes collected number were 4.4$\pm$3.5 e.a to 170 and 3.0$\pm$1.8 e.a to 18G needle, collected oocytes quality were no difference significantly (P<0.05). 4. Follicles increase number to FSH hormone injection were 6.2$\pm$2.3 e.a to intramuscle and 1.1$\pm$2.7 e.a. to epithelial injection method. 5. Conception rate derived from E.T. was 11.1% to freezing embryos and 46.2% to fresh E.T., difference significantly(P<0.05).

Effects of Growth Factors and Hexoses on In Vitro Development in Porcine Embryos (돼지수정란의 체외발육에 있어서 Growth Factors와 Hexoses의 영향)

  • The aim of this study was to investigate the effects of different hexoses in medium with IGF-I and/or EGF on in vitro development of porcine embryos. In first experiment, when the embryos were cultured in medium with concentrations of 5 ng/ml IGF-I and 10 ng/ml EGF, the morula and blastocyst rates were higher than in another culture conditions (P<0.05). In second experiment, the effect of IGF-I in medium with different hexoses during the embryo culture was examined. The higher cleavage rates were obtained in medium with glucose and IGF-I (P<0.05). However, the higher proportion of embryos developed to morula and blastocyst stages in medium with IGF-I were obtained than in medium without IGF-I in medium with glucose, mannose and galactose. In third experiment, the effect of EGF on in vitro development of porcine embryos in medium with different hexoses were examined. In the culture medium was supplemented with glucose, the higher proportions (P<0.05) of embryos developed to molura and blastocyst stages were obtained in medium with that than without EGF. However, the proportions of embryos developed to blastocyst stage were not significantly different between with and without of EGF in medium with different hexoses. In firth experiment, the effects of presence of IGF-I and EGF in medium with different hexoses on in vitro development of porcine embryos were examined. When culture medium was supplemented with IGF-I and EGF, the higher proportions of oocytes cleaved and developed to 8-cell stage were obtained in medium with glucose than mannose, galactose and fructose (P<0.05). These results show that glucose and growth factors, supported the development in vitro of porcine embryos, especially with greater embryo cleavage and development in medium with glucose added to media with IGF-I and/or EGF.

Effects of Embryo Density on Development of In Vitro Produced Bovine Embryos (수정란의 밀도가 소 체외수정란의 체외발달에 미치는 효과)

  • This study was carried out to improve of effective culture system on development of IVM/IVF/IVC bovine embryos. The cumulus-oocyte-complexes (COCs) collected from Korean cattle ovaries harvested at a local abattoir were matured in 50 ${mu}ell$ of TCM199 supplemented with 10% fetal bovine serum (FBS) and hormones (35 $\mu\textrm{g}$/$m\ell$ FSH, 10 $\mu\textrm{g}$/$m\ell$ LH, 1$\mu\textrm{g}$/$m\ell$ estradiol 17 $\beta$ under paraffin oil at 39$^{\circ}C$ in a humidified atmosphere of 5% $CO_2$in air. At 24 hrs after culture, matured oocytes were fertilized in vitro for 22~24 hrs with motile semen in which obtained by centrifugation of a frozen thawed semen on Percoll-density gradients (45% vs. 90%) at 500 g for 20 min. The presumptive zygotes were divided into three experimental groups. Single egg (Group 1), 25 (Group 2) or 50 eggs (Group 3) were cultured on cumulus cell in 50 ${mu}ell$ TCM199 supplement with 10% FBS for 6~9 days after fertilization. In vitro developmental rates into the blastocysts in the groups 2 and 3 were significantly (P<0.05) higher than those of group 1 (37,27 vs. 6%, respectively). Cell number of blastocysts obtained in groups 2 and 3 at day 8 were significantly (P${mu}ell$) resulted in higher developmental competence and cell number of bovine blastocysts produced in vitro than those the culture of single embryos with cumulus cells.
