The Effect of Alcohol Fermented Feedstuff Made of Byproducts on In vitro Fermentation Characteristics and NDF Disappearance in the Rumen (조사료 대용 산업부산물을 이용한 알코올 발효사료가 In vitro 반추위내 발효특성 및 NDF 분해율에 미치는 영향)

  • An in vitro study was conducted to examine the influence of alcohol-fermented feedstuff formulated with byproducts on the fermentation characteristics and NDF disappearance in the rumen. Dietary treatments were either a soybean curd-based alcohol-fermented feedstuff (AFS) or brewery grain-based alcohol-fermented feedstuff (AFB). The AFS and AFB are composed of 50% commercial beef cattle feed, 50% soybean curd dreg, 5% molasses and 0.5% yeast and 25% commercial beef cattle feed, 25% brewery grain, 25% soybean curd dreg, 25% corn grit, 5% molasses and 0.5% yeast, respectively. The change of ammonia, pH alcohol, volatile fatty acids, and NDF disappearance were measured at 0, 2, 4, 8 and 12 hr after in vitro incubation in the rumen. After 2 hr incubation, higher ammonia concentrations were resulted in AFS (12.47 mg/dl) and AFB (12.85 mg/dl) compared to control (11.84 mg/dl) (p<0.05). Ruminal pH of AFS and AFB were significantly higher than control during 1 to 6 hr fermentation, but the pH of AFS and AFB were decreased after 6 hr. At 12 hr fermentation, the alcohol concentration of AFS and AFB was significantly increased by 43.9% and 48.0%, respectively. The acetate concentration was rapidly decreased in control, while the concentration was slowly decreased in AFS and AFB with increasing the fermentation time. Lower concentrations propionate and butyrate were observed in AFS and AFB during every fermentation time (p<0.05). The NDF disappearance was significantly lower in AFS and AFB after 4 hr fermentation. These results suggest that alcohol fermented feedstuff can control the fermentation pattern in the rumen.

The physicochemical characteristics of silk-tofu added with medicinal herb powder preserved in kochujang and deonjang (Tofujang) (약초첨가 비단두부의 장담금에 따른 이화학적 특성)

  • The effect of medicinal herb Powder addition on the Physicochemical characteristics of tofujang preserved in kochujang and deonjang was investigated. The moisture contents of tofujang decreased rapidly till 4 weeks of storage, after which they showed a slight tendency to increase. The PH values of cochujang tofujang and deonjang tofujangs showed a tendency to decrease, but then showed a tendency to increase a little. The saltiness of tofujang increased rapidly till 2 weeks of storage, and then increased gently. The chromaticity of L and a values decreased, and that of b value significantly increased, during a storage. The total microbial counts of kochujang tofujang decreased till 4 weeks of storage, and then increased a little (103 CFU/ml). The total microbial counts of deonjang tofujang were decreased during storage. The total free amino acids content of silk-tofu was extremely small $(0.16\~0.92 mg/ml)$, but after 4 weekn of preservation, that of tofujang preserved in kochujang and deonjang increased to 10mg/ml and 40mg/ml, respectively In the kochujang tofujang, the textural characteristics of hardness, gumminess and chewiness increased rapidly at initial preservation and then showed a tendency to decrease significantly. The springiness and cohesiveness values showed no specific tendency. In the deonjang toking, the textural characteristics of hardness and gumminess increased till 2 weeks of preservation, and then tended to decrease. The springiness, chewiness and cohesiveness value had a tendency to decrease significantly with preservation. In the sensory evaluation, color, aftertaste and overall acceptability of mok-k tofujang showed significantly high score, but there was no significance in nutty taste, sweet, salty and hot. Color, savory, sweet and overall acceptability of mok-d and con-d tofujang were high, and the saltiness and aftertaste of sin-d tofujang got high score.

Utilization of Probiotic Starter Cultures for the Manufacture of Low-fat Functional Fermented Sausages (저지방 기능성 발효소시지의 제조를 위한 복합 유산균주의 이용)

  • This study was performed to evaluate the physico-chemical properties of fermented sausages containing probiotic starter cultures (LK-30 plus, Lactobacillus plantarum 155 and 167, and Pediococcus damnosus L12) with reduced fat levels, and to determine the optimum condition for the manufacture of these products. Although low-fat fermented sausages were reduced fat content at the amount of 90% and the ripening time by 1-2 weeks, as compared to regular-fat counterpart, they became harder and had many winkles outside due to the extreme drying. In addition, fat level in fermented sausages affected the composition and shear force values. During ripening, pH, lightness and yellowness values tended to decrease, however, microbial counts of inoculated lactic acid bacteria were increased up to $10^8-10^9cfu/g$ within 3 days and remained constant thereafter. Low-fat fermented sausages had higher microbial counts than regular-fat ones. Although the inoculated probiotic starter cultures alone had the functional properties, such as cholesterol reduction, anti-high blood pressure and antimicrobial activity, they did not have distinctive characteristics in the fermented sausages. Based on these results, the low-fat fermented sausages were successfully manufactured, but a little bit increased fat level and improved functional properties in the fermented sausages would be required to have better quality as compared to regular-fat counterparts.

An Investigation of Microbial Contamination of Side Dishes sold at Traditional Market and Super Market in Ulsan (울산지역 재래시장 및 대형 할인점 유통 반찬류의 미생물 오염도 조사)

  • This study was investigated to determine the contamination levels of total aerobic bacteria, coliform group, $E.$ $coli$ and food-borne pathogens of side dishes from 2 traditional markets (100 samples) and 2 super markets (100 samples) located on Ulsan. The levels (range) of total aerobic bacteria was 4.75 log CFU/g (1.60~6.92 log CFU/g) in traditional market and 4.62 log CFU/g (2.00~6.46 log CFU/g) in super market, respectively. Coliform was detected in 64 and 66 samples sold at traditional markets and super markets, respectively. $E.$ $coli$ was detected in 4 and 6 samples sold at traditional markets and super markets, respectively. The food-borne pathogens, namely $Bacillus$ $cereus$ and $Listeria$ $monocytogenes$ were detected in 1 sample sold at traditional markets, respectively, and $Bacillus$ $cereus$ was detected in 4 samples sold at super markets. However, other pathogens such as $Salmonella$ spp., $Shigella$ spp., $Vibrio$ $parahaemolyticus$, $Yersinia$ $enterocolitica$, $Clostridium$ $perfringens$, $Camphylobacter$ $jejuni$ and Pathogenic $E.$ $coli$ were not detected. The $Saengchae$ and $Seasoned$ $Jeotgal$ were relatively vulnerable compared to the others in the food-borne pathogens.

Product Quality and Extension of Shelf-life of Low-fat Functional Sausages Manufactured with Sodium Lactate and Chitosans during Refrigerated Storage (젖산나트륨과 키토산을 첨가한 저지방 기능성 소시지의 냉장 저장 중의 품질 및 저장성 증진 효과)

  • This study was peformed to evaluate physico-chemical and textural properties, and shelf-life effect of low-fat functional sausages(LFFS) manufactured with sodium lactate(SL), lac color and various molecular weights of chitosans(low=1.5 kDa, medium=30∼40 kDa and high=200 kDa) during storage at 4$^{\circ}C$ for 8 weeks. LFFS had a pH range of 6.39∼6.50, 76∼78% moisture, <2% fat, 14∼15% protein. The combination of SL and low molecular weight(MW) of chitosan improved water holding capacity(WHC), however those of SL and medium MW of chitosan reduced WHC. Vacuum purge(VP) reduced with increased MW of chitosans during refrigerated storage. The addition of chitosans reduced the lightness and yellowness, but increased the redness values, which was comparable to the sodium nitrite concentration between 75 and 150 ppm. LFFS containing SL and medium MW of chitosan increased most texture profile analysis(TPA) values, as compared to controls with 75 and 150 ppm. The addition of SL in LFFS retarded the microbial growth for Listeria monocytogenes, however no synergistic effect with the addition of chitosans were observed. E coli O157:H7 and Salmonella typhimurium reduced during refrigerated storage, regardless of SL and chitosan treatments. Increased storage time increased values for VP, yellowness and textural properties. These results indicated that the combination of SL and various MW of chitosans affected the functional and textural properties, and inhibited the microbial growth for LM effectively. In addition, 0.5% lac color as a replacer for sodium nitrite improved the color development, resulting in similar hunter color values, which was comparable to the sodium nitrite concentration between 75 and 150 ppm.

Application of Predictive Microbiology for Microbiological Shelf Life Estimation of Fresh-cut Salad with Short-term Temperature Abuse (PMP 모델을 활용한 시판 Salad의 Short-term Temperature Abuse 시 미생물학적 유통기한 예측에의 적용성 검토)

  • The aim of this study was to investigate the growth of aerobic bacteria in fresh-cut salad during short-term temperature abuse ($4{\sim}30^{\circ}C$temperature for 1, 2, and 3 h) for 72 h and to develop predictive models for the growth of total viable cells (TVC) based on Predictive food microbiology (PFM). The tool that was used, Pathogen Modeling program (PMP 7.0), predicts the growth of Aeromonas hydrophila (broth Culture, aerobic) at pH 5.6, NaCl 2.5%, and sodium nitrite 150 ppm for 72 h. Linear models through linear regression analysis; DMFit program were created based on the results obtained at 5, 10, 20, and $30^{\circ}C$ for 72 h ($r^2$ >0.9). Secondary models for the growth rate and lag time, as a function of storage temperature, were developed using the polynomial model. The initial contamination level of fresh-cut salad was 5.6 log CFU/mL of TVC during 72 h storage, and the growth rate of TVC was shown to be 0.020~1.083 CFU/mL/h ($r^2$ >0.9). Also, the growth tendency of TVC was similar to that of PMP (grow rate: 0.017~0.235 CFU/mL/h; $r^2=0.994{\sim}1.000$). The predicted shelf life with PMP was 24.1~626.5 h, and the estimated shelf life of the fresh-cut salads with short-term temperature abuse was 15.6~31.1 h. The predicted shelf life was more than two times the observed one. This result indicates a 'fail safe' model. It can be taken to a ludicrous extreme by adopting a model that always predicts that a pathogenic microorganism will grow even under conditions so strict as to be actually impossible.

Physico-chemical and Microbial Properties of Sausages Affected by Plant Scale and Cooking Treatments during Refrigerated Storage (가열조건 및 공장 규모에 따른 소시지의 냉장저장 중 이화학적 및 미생물적 품질특성)

  • Purpose: The objective of this study was to examine the effect of plant scale and cooking conditions on the quality characteristics of sausages during refrigerated storage. Methods: Sausages used in this study were classified into two groups: those submitted to $1^{st}$ cooked treatments and those submitted to $2^{nd}$ cooked treatments. The pH, volatile basic nitrogen (VBN), gas production ratio, and microorganisms were measured in triplicate. Results: The change of quality in the products was assessed every 7 days by measuring pH, VBN levels, total microbes, coliform bacteria, Escherichia coli, and pathogenic bacteria in the products. Pathogenic bacteria such as Staphylococcus aureus, Listeria monocytogenes, Clostridium perfringens, and E. coli were not detected in the sausages with $1^{st}$ cooked treatments. The results showed that the pH of the sausages decline as storage time increased. The pH value of the sausages with $2^{nd}$ cooked treatments changed gradually. VBN levels were generally lower in products with $2^{nd}$ cooked treatments than in those with $1^{st}$ cooked treatments, but they varied with the type of products. On the $35^{th}$ day, the number of total microbes ranged between 6.13-7.12 log CFU/g in products with $1^{st}$ cooked treatments and 3.44-6.92 log CFU/g in products with $2^{nd}$ cooked treatments, showing fewer bacteria in the latter products. Conclusions: $1^{st}$ cooked treatments were effective in microbial control, but $2^{nd}$ cooked treatments could prolong the shelf life of the sausages, indicating a need for differential management of each product.

Removal of Arsenite by Nanocrystalline Mackinawite(FeS)-Coated Alumina (나노크기 매킨나와이트로 코팅된 알루미나에 의한 아비산염의 제거)

  • Due to the large specific surface area and great reactivity toward environmental contaminants, nanocrystalline mackinawite (FeS) has been widely applied for the remediation of contaminated groundwater and soil. Furthermore, nanocrystalline FeS is rather thermodynamically stable against anoxic corrosion, and its reactivity can be regenerated continuously by the activity of sulfate-reducing bacteria. However, nanocrystalline mackinawite is prone to either spread out along the groundwater flow or cause pore clogging in aquifers by particle aggregation. Accordingly, this mineral should be modified for the application of permeable reactive barriers (PRBs). In this study, coating methods were investigated by which mackinawite nanoparticles were deposited on the surface of alumina or activated alumina. The amount of FeS coating was found to significantly vary with pH, with the highest amount occurring at pH ~6.9 for both minerals. At this pH, the surfaces of mackinawite and alumina (or activated alumina) were oppositely charged, with the resultant electrostatic attraction making the coating highly effective. At this pH, the coating amounts by alumina and activated alumina were 0.038 and 0.114 $mmol{\cdot}FeS/g$, respectively. Under anoxic conditions, arsenite sorption experiments were conducted with uncoated alumina, uncoated activated alumina, and both minerals coated with FeS at the optimal pH for comparison of their reactivity. Uncoated activated alumina showed the higher arsenite removal compared to uncoated alumina. Notably, the arsenite sorption capacity of activated alumina was little changed by the coating with FeS. This might be attributed to the abundance of highly reactive hydroxyl functional groups (${\equiv}$AlOH) on the surface of activated alumina, making the arsenite sorption by the coated FeS unnoticeable. In contrast, the arsenite sorption capacity of alumina was found to increase substantially by the FeS coating. This was due to the consumption of the surface hydroxyl functional groups on the alumina surface and the subsequent occurrence of As(III) sorption by the coated FeS. Alumina, on the surface area basis, has about 8 times higher FeS coating amount and higher As(III) sorption capacity than silica. This study indicates that alumina is a better candidate than silica for the coating of nanocrystalline mackinawite.

Selection and Mechanisms of Indigenous Antagonistic Microorganisms against Sheath Rot and Dry Rot Disease of Garlic (마늘 잎집썩음병과 마른썩음병을 길항하는 토착길항미생물의 선발 및 기작)

  • Sheath rot and dry rot disease caused by Pseudomonas marginalis and Fusarium oxysporum were serious problems in garlic farmland. In this study, total of 160 indigenous antagonistic bacteria were isolated from 16 farmlands in Yeongcheon, Korea. Among these, 15 strains were able to inhibited P. marginalis and F. oxysporum. The 16s rDNA genes of the selected 15 strains were amplified and sequenced. The strains has strong antagonistic ability against garlic pathogens was achieved Bacillus subtilis YC82, B. vallismortis YC84, B. amyloliquefaciens YC240. The selected 3 strains tested for investigation of antifungal mechanisms further analyses; 3 strains of these validated for production of siderophore, ${\beta}$-glucanase and chitinase using CAS (chrome azurol S) blue agar, CMC-congo red agar and DNS method. The 3 strains were able to utilized insoluble phosphate as dertermined by vanado-molybdate method. The 3 strains verified for production of auxin and gibberellic acid using Salkowski test and holdbrook test. Also, 3 strains showed stimulation germination, stem growth promoting activity on the in vivo test. The 3 strains were able to effectively suppress P. marginalis and F. oxysporum causing sheath rot and dry rot diseases on the in vivo pot test.

Growth Patterns of Lactic Acid Bacteria during Fermentation of Radish with Rice Water and Rice Bran (쌀뜨물과 쌀겨를 이용한 무 발효과정 중 젖산균의 증식 양상)

  • Changes in total aerobic bacteria (TAB), lactic acid bacteria (LAB), rod- and coccal-shaped lactic acid bacteria (R- and C-LAB), pH, and total acidity were investigated during fermentation of radish with new fermentation base at $20^{\circ}C$ for up to 16 days. New fermentation base was prepared by pre-fermenting a mixture of rice bran and rice water (1 : 0.1, w/v) at $20^{\circ}C$ for 7 days. Initially, radish showed 5.41, 4.23, 4.57, and 3.1 log CFU/g, and base showed 6.68, 6.60, 5.95, and 5.6 log CFU/mL for TAB, LAB, R-LAB, and C-LAB, respectively. Initial pH and total acidity of radish were $6.6^{\circ}C$ and 0.09%, and those of base were 5.76 and 0.36%, respectively, Counts of LAB (4.23 to 8.33 Bog Cpu/g, 6.6 to 9.7 log CFU/mL), R-LAB (4.57 to 7.15 log CFU/g, 5.95 to 8.5 log CFU/mL), and C-LAB (3.1 to 7.5 log CFU/g, 5.6 to 8.6 log CFU/mL) of radish and base respectively increased during initial fermentation period, then remained constant during late fermentation period, and far 4 days after fermentation, pH values (6.6 to 4.19, 5.76 to 4.57) drastically decreased and thereafter slowly decreased. Total acidities of radish and base (0.09 to 0.63%, 0.36 to 0.63%, respectively) drastically increased far 7 days after fermentation and increased slightly thereafter.
