Cytotoxic Effect of Extracts from Styela clava against Human Cancer Cell Lines (암세포주에 대한 미더덕 추출물의 세포독성 효과)

  • The present study describes the preliminary evaluation of the cytotoxic effect from Styela clava (Korean name : miduduk) extracts. S. clava was treated with methanol, ethanol, acetone, and water, then cytotoxic effect of the extracts were evaluated by the MTT reduction assay. The ethanol extracts from S. clava showed the cytotoxic activity on the HT-29 human colon cancer cells. The ethanol extracts was further fractionated with n-hexane, diethyl ether, ethyl acetate, and water according to the degree of polarity. The diethyl ether fraction showed high cytotoxic activity of HT-29 cells. however, the other fractions showed low cytotoxicity. The diethyl ether layer also showed the cytotoxic activity against SW620, HeLa, and MCF-7 cells. These studies support that extracts from S. clava may be a potential candidate as a possible chemotherapeutic agent against human cancer cells.

  • 'Mideuduck', Styele clava, has been esteemed as one of the most tasty sea foods in the south coast of Korea. The free amino acids in Mideuduck were analysed by amino acid auto-analyser. In Mideuduck extract, taurine, proline, glutamic acid, glycine, alanine and serine were abundant, holding $32.8\%,\;17.6\%,\;11.9\%,\;11.2\%,\;8.9\%,\;7.1\%$ respectively of total amino acid content and followed by threonine, lysine, methionine in order. While valine, leucine, phenylalanine, isoleucine and histidine were poor ana aspartic acid, arginine and tyrosine were trace in content.

  • Glycosaminoglycans(GAGs) from sea squirt, Styela clava was extracted with sodium phosphate at $105^{\circ}C$ for 2 hr and deprotein with trichloroacetic acid or hydrochloride. This GAGs was mainly constituted of galactose, glucosamine, glucose, mannose and galactosamine, and was phenylalanine, threonine, glutamic acid and aspartic acid. Mineral contents was mainly constituted 3.0mg% sodium, 1.6mg% potassium and 1.2mg% phosphorus and heavy metal was not detected. At pharmaceutical and cosmetic code of GAGs, protein and sulfate contents should included each range $14.0{\sim}22.0%$, $35.0 {\sim}45.0%$. After 5.0% trichloroacetic acid(w/v) and 10.0% HCl(v/v) treatment, protein and sulfate contents of GAGs was contained each 35.1%, 35.4% and each 22.0%, 18.5%.

Antioxidative Activity of Mustard Leaf Kimchi with Optional Ingredients (부재료 첨가에 따른 갓김치의 항산화성)

  • Antioxidative activities (AA) of mustard leaf kimchi (MLK) by the addition of optional ingredients among selected minor materials were studied. In order to determine AA of MLK with different spices, the model systems of ground cooked beef with green onion, garlic, and red pepper powder were prepared and stored for 4 weeks at 4$^{\circ}C$. AA of red pepper added group was stronger than those of others. AA of red MLK was relatively higher than that of (green) MLK. For the enhancement of AA of MLK, another model systems were prepared with the selected antioxidative optional ingredients, which were bonnet bellflower root, leek, burdock, sea tangle, sea mustard, seastaghorn at the level of 2% or 4%. The extracts of water, 75% methanol and hexane of MLK, bonnet bellflower root added MLK, and seastaghorn added MLK had a considerable AA with the inhibition of peroxide formation during the autioxidation of linoleic acid mixtures in aqueous model systems at 37$^{\circ}C$. Therefore, AA was more effective in MLK containing specific optional ingredients than that of MLK alone significantly.

  • This study was performed to evaluate the effect of alkaline treatment on Styela clava tunic (SCT). Considerable damage to the surface of alkali-treated SCT was observed by scanning electron microscopy (SEM) in a concentration-dependent manner upon alkaline treatment. The amount of crystalline region in SCT gradually increased with increasing NaOH concentration, which was analyzed by X-ray diffraction and thermogravimetric analysis. The initial enzymatic reaction of Celluclast toward SCT was elevated by treatment with NaOH up to 1.0 N concentration due to disruption of the SCT surface by promoting binding of enzymes with SCT. However, in the late stage of the enzyme reaction, hydrolysis rate decreased with elevation of NaOH concentration, thereby increasing the amount of non-reacted residuals. This result was due to the increase in the crystalline regions in SCT.

Flavor Modification of Mideoduck (Styela clava) Drips by Maillard Reaction (Maillard 반응에 의한 미더덕 체액의 풍미개선)

  • Mideoduck drips were mixed with amino acids (Met, Tau, Gly, Ala, Thr, Cys), thiamine and sugars (Glucose, Ribose) for flavor modification and evaluation using the Maillard reaction. To mask the seafood flavor, onions, spring onions, garlic, ginger, citric orange and green tea were mixed with Mideoduck drips at $160^{\circ}C$ for 2.5 hr in a stainless still reaction bomb. The glucose/thiamine model reaction system was estimated to be lower than the ribose/thiamine model system, and an extreme case is the ribose/Met model system. Mixed system of glucose, ribose and taurine containing sulfur compounds showed fair results. Among the Mideoduck drips mixed with sugars and amino groups, only thiamine model systems were estimated to be normal. The flavor composition of Mideoduck drips/sugars model system, and long chain fatty acids were composed of 31.32~62.71% total flavor content. The 1,2-benzenedicarboxylic acid dibutylester contents made up more than 20% of the model system in groups A, B and C. From the model system in this study, drip/glucose, drip/ribose, drip/glucose/citric orange, and drip/glucose/glycine/cystine groups showed most intense good flavor.

  • 우리는 살아가면서 흔히 개인적으로 극심한 스트레스를 겪거나 불행히도 어떤 뜻밖의 재난이나 큰 사고를 경험할 수 있다. 이런 외상적 사건들을 겪고 대부분의 사람들은 잘 극복할 수 있으나 일부 사람들은 심각한 의상 후 스트레스 반응을 보여 그런 외상이 지나감에도 불구하고 괴로운 기억 속에서 그것을 회상시키는 활동과 상황을 피하며 늘 불안한 마음으로 몸은 현재에 살고 있지만 마음은 실상 과거에 매달려 있고 과거의 고통 속에서 헤어 나오지 못하고 있다. 외상 후 스트레스 장애는 다른 불안 장애, 우울증처럼 흔한 질환으로 사회생활에 큰 지장을 주며 삶의 질을 손상시킨다. 그렇지만 외상을 당한 당사자들이 이런 이야기를 하기 꺼려하고, 이런 사람들을 진료실에서 종종 만날 수 있는 임상가들도 외상의 심각성과 외상 후 스트레스 장애에 대한 인식이 부족하여, 우리 주위에 많은 사람이 외상 후 스트레스 장애로 고생하고 있지만 외상 후 스트레스 장애로 진단되는 경우가 실제보다 훨씬 적다. 따라서 외상 후 스트레스 장애에 대한 평가를 배움으로써 더 많은 환자들을 정확히 진단하며 다른 질환과의 감별을 쉽게 할 수 있고, 환자들의 고통과 어려움이 과연 무엇인지 잘 알 수 있으며, 치료 표적과 치료 전략을 정하고, 치료 계획을 세우는 데 도움을 얻을 수 있다. 평가 방법으로는 구조화된 면담, 평가 척도, 정신 생리적 측정법, 치료자의 임상적인 결정 등이 있다. 향후에는 외상 후 스트레스 장애의 증상 정도뿐만 아니라 일상생활, 사회적 관계, 직업적, 사회적 적응 및 기능에 미치는 영향도 같이 평가하는 도구들이 개발되고, 현재 개발된 많은 평가 척도들이 새로이 수정되고 표준화되어, 이런 평가 척도들을 통해 외상 후 스트레스 장애의 경과와 외상 후 스트레스 장애로 인한 정신 사회적 영향을 더 잘 알 수 있는 자료들이 많이 수집되길 바란다. 대칭이, 작은대칭이, 펄조개는 저질이 니질인 곳에서 주로 서식하고 있었고 귀이빨대칭이는 사니질과 사질에 주로 서식하고 있었다. 특히 삽교호 수계 채집된 귀이빨대칭이는 사니질과 사질의 저질에서 출현하였다.및 내장낭 조직들이 중요한 에너지 저장소이며, 영양 공급 기관임을 알 수 있었으며, 폐각근 조직과 내장낭 조직들의 영양물질 함량 변화는 생식소의 에너지 요구에 따라 변하고 있음을 알 수 있었다.. 따른 신장 기능에 대한 영향을 평가해 보아야 한다.하를 조사한 결과, 연간 진동해역으로 유입되는 오염부하량은 부유물질 기준으로 전체 37,316 ton/yr로 추정되었고, 이중 육지기원 부하량이 9,809 ton/yr (26.3%) 였고, 해역의 침강량 23,576 ton/yr (63.2%), 양식생물의 배설량이 3,932 ton/yr (10.5%) 로 대부분이 해역의 침강량에 의한 것으로 조사되었다. 육지오염부하량이 변동이 없다는 전제 조건 아래, 진동만의 피조개 채묘장을 미더덕 양식장으로 품종전환시킬 경우, 전체 부하량은 산술적으로 약 196%가 증가한 72,998 ton/yr 가 되고, 양식생물의 배설량은 10배 정도 증가하게 되어 진동만의 오염부하를 가중하게 된다. 진동만은 여름철 빈산소수괴 및 적조가 빈번하게 발생하고 있으므로 미더덕 양식장을 새로이 시설할 경우 오염부하가 가중될 것으로 판단되어, 앞으로 진동만의 양식장 수용능력을 파악하여 적정량의 양식물량이 시설되어야 할 것이다.25 psu에서 가장 높게 나타났다.인 단백뇨는 진행성 경과의 위험성을 동시에 암시해 주는 매우 유용한 지표이다. 본 연구 결과 학교 신체 검사상에서 발견된 단독 단백뇨의 주된 원인은 기립성 단백뇨로 81.8%를 차지하였다. 그러나 지속성 단백뇨의 경우 비록 9.1%의 빈도를 보였지만, 신장 조직검사를 시행한 결과 진행성 경과를 취할 수 있는

Properties of Regenerated Cellulose Films Prepared from the Tunicate Styela clava (미더덕 껍질을 이용한 셀룰로오스 필름의 제조 및 특성)

  • The tunic of Styela clava(SCT) consists of a proteoglycan network. Regenerated cellulose films were prepared by solution casting and coagulation of SCT in N-methylmorpholine-N-oxide(NMMO)/$H_2O$(87/13 wt%). The crystalline structure of powdered SCT was primarily that of cellulose I. The crystalline structure of SCT films exhibited a cellulose II structure, similar to that of viscose rayon. Physical characterization of SCT films and fibers revealed an intrinsic viscosity($\eta$) of 6.35 dL/g, average molecular weight($M_w$) of 423,000 g/M, and fiber density of 1.50 $g/cm^3$ with a moisture regain and water absorption of 10.20% and 365%, respectively. The results were similar to those of cellulose films regenerated from wood pulp. Films prepared with 6 wt% SCT exhibited strong tensile strength, high water absorption, and a greater degree of elongation. Scanning electron micrographs(SEM) of film cross-sections showed a layered, sponge-like structure.

The Change in Hematological Factors of Ascidian (Styela clava) Extract Diets Fed Rainbow Trout (Oncorhynchus mykiss). (미더덕 추출액을 섭취한 무지개 송어 혈액성상의 변화)

  • Effects of muscle fluid concentrates from Styela clava on fish diets were investigated in rainbow trout(Oncorhynchus mykiss). Fish were fed on one of the isonitrogenous(48%) and isolipic(20%) feed containing 5 to 20% of muscle fluid concentrates for 8 weeks. Hematological parameters such as hemoglobin, hematocrit, albumin, glucose, total bililubin, triglyceride and glutamic pyruvate transaminase of the rainbow trout which were fed on the diets varied, but no specific trend became apparent. However, glutamic oxaloacetic transaminase value was significantly higher than that of normal fishes. Therefore, concentrated ascidian fluid which is normally discarded, can be combined with commercial diets for the normal growth of rainbow trout.

Purification and Characterization of a Carotenoprotein from Style clava (미더덕(Styela clava)으로부터 분리.정제한 carotenoprotein의 특성에 과한 연구)

  • In this study Carotenoprotein from Styela clava were extracted, and purified by ammonium sulfate fraction, Sephadex G- 200 gel chromatography and DEAE-cellulose ion exchange chromarography. The purified carotenoprotein from styela clava had absorption maxima of 487nm, 463nm and 280nm, and the carotenoid liberated from carotenoprotein had 478nm and 452nm with inflexion. One miligram of caroteno-protein contained 0.35 $\mu$g of carotenoid. The carotenoprotein had an approximate molecular weight of 398 kDa (gel filtration). SDS-PAGE showed only a single polypeptide chain with a molecular weight of 62.4 kDa. The amino acid composition of the carotenoprotein were mainly glutamic acid(11.48%), valine(10.75%), leucine (10.45%), aspartic acid(9.94%), while cysteine and tryptophan were not found. The carotenoprotein contained lipid as structure units. In the carotenoprotein, the major fatty acids were oleic acid, plamitoleic acid and palmiunsaturated fatty acids 33.6%, saturated fatty acids 18.6%. In addition, the levels of higher unsaturated fatty acids were high as much as 30.8% of the total fatty acids. Carotenoid was extracted from the carotenoprotein by acetone. Thin layer chromatography showed only carotenoid to be present. Its chemical reactivities and spectroscopic properties were studied and elucidated as astaxanthin.
