  • Forty one samples from thirty three species of algae (19 from 15 species of Rhodophyceae, 18 from 14 species of Phaeophyceae, 3 from 3 species of Chlorophyceae, and 1 of marine Phanerogams) collected from several locations on the east, west and south coast of Korea, were analyzed for their contents of crude protein, fat, cellulose, ash, nitrogen free extract, amino nitrogen, and total amino acids. For the examination of extractability of algal protein with water, 4 species of algae, Sargassum thunberggi, Grateloupia filicina, Phyllospadix japonica, and Sargassum confusum, were analyzed. And the effect of some precipitation treatments for isolation of algal protein was also tested. As a matter of fact, Rhodophyceae showed high content in crude protein and low in crude fat while the case was opposite for Phaeophyceae and Chlorophyceae. Refering to the content of crude protein and total amino acids, the recommendable algae for protein sources were Sargassum thunbergii, Acrosorium flabellata, Phacelocarpus japonicus, Laurencia okamurai, Laurencia intermedia, Grateloupia filicina, Chondrus ocellatus, Gloiopeltis furcata, Gigartina tenella, Dictyota dichotoma, and Scytosiphon lomentaria. Methanol treatment appeared most effective in precipitation isolation of protein from water extracts whereas pH control method did not so beneficial. The precipitation rate of protein was particularity higher in the extract of Sargassum confusum ana the lowest was makted from the extract of Sargassum thunbergii.

Effect of Water Soluble Extract of Lichens on Weights of Various Organs, Blood Components and Activities of Transaminases and 5-Nucleotidase in Rat (지의류(地衣類)의 수용성 추출물이 흰쥐의 각 장기무게, 혈액성분 및 Transaminases와 5-Nucleotidase 활성에 미치는 영향(I))

  • In order to evaluate the effect of water soluble extract of lichens (Physcia, Parmelia and Clandonia species) on liver damage, activities transaminase(GPT) and 5'-nucleotidase in serum and liver were measured in rats fed lichens extract. DNA and RNA were measured in liver and spleen, as well as various organ weights and blood components. Control group was fed water to compare with the lichen group. Three sets of experiments were conducted: the first set was done with normal rats, the second one with rats with liver damage induced by $CCl_4$ injection was divided into three subgroups. i.e. no treatment group, pre-treatment group and post-treatment group, and the third one was with rats with acute and chronic liver damage. In normal rats, lichens extract feeding reduced serum GOT and GPT activities. In liver damaged rats, both pre-and post-treatment had suppressing effect against increase of serum enzymes. In rats with acute and chronic liver damage, lichens fed group had lower activities of serum GOT, GPT and 5'-nucleotidase but higher activities of liver enzymes than control group. This effect was more pronounced in rats with acute liver damage. Liver weight increased considerably with lichens intake. Hemoglobin and hematocrit levels were also higher in lichens fed group. Nucleic acid contents in spleen but not in liver were increased in lichens fed group. The latter increase was more significant with chronic liver damage. It is suggested from the present study that water soluble lichens extract play protective and therapeutic roles in organs against infection and atrophic disease.

Diurnal Changes of Leaf Water Potential in Cuttings (삽수(挿穗)의 Leaf Water Potential의 변화(變化))

  • The diurnal pattern of leaf water potential in cuttings by Dye Method was as follows: 1. Diurnal pattern of leaf water potential (${\psi}_l$) in Viburnum Awabuki K. Koch and Daphne odora Thunb. was shown the pattern of the curves without mutual relation with soil classes when soil water potential (${\psi}_s$) was 0 bar. When ${\psi}_s$ was above -0.01 bar, the cuttings in the loamy sand (L.S.) was shown by the maximum values than that in sandy clay (S.C.) by about -1 bar gap (Fig. 1). 2. The diurnal changes of ${\psi}_l$ was shown the most high from two to eight O'clock in the morning, the maximum value was -3 bars when ${\psi}_s$ was above -0.01 to -0.02 bar, and was -4 bars below -0.03 bar. The diurnal the lowest values of ${\psi}_l$ showed -20 to -22 bars from one to two O'clock in the afternoon. In the fifteenth day after cutting V.A., the staying time in the diurnal maximum values of ${\psi}_l$ is about half in comparison with it in the fifth day. The curves of recovery of water stress (Fig. 1), the former reached to the diurnal maximum values -1 to -2 bars lately every hours comparing with it of the latter. The general diurnal pattern of ${\psi}_l$ was most clearly related to change with air temperature and the relative humitidy. 3. Comparing the treatment block by IAA 50 ppm with controlled block in fifteenth day after V.A. cuttings, in case of treatment reached to maximum values -2 to -3 bars lately as shown Fig. 2., and also staying times was only half in comparison with controlled block. 4. The cuttings 4 leafs was much rootings than 2 leafs in V.A. (Table. 1), and the former reached maximum value -2 to -3 bars lately every hours comparing with the latter. 5. In case of Buxus microphylla var. Koreana as shown Fig. 3., comparing the pattern curves of in the cuttings 8 leafs with 4 leafs, the former reached to maximum values -2 to -3 bars lately in comparison with the latter, but reffering to the amount of rooting (Table. 2), the former is less than the latter.

Effects of Seeding Dates on the Growth Characteristics, Dry Matter Yield and Feed Value of Teosinte New Variety, "Geukdong 6" [Zea mays L. subsp. mexicana (Schrad.) H. H. Iltis] (파종시기가 테오신트 [Zea mays L. subsp. mexicana (Schrad.) H. H. Iltis] 신품종 "극동 6호"의 생육 특성, 수량 및 사료가치에 미치는 영향)

  • This study was carried out to investigate the effects of seeding dates and cultivated period on the growth characteristics, dry matter yield and feed value of Teosinte new variety "Geukdong 6"[Zea mays L. subsp. mexicana (Schrad.) H. H. Iltis]for feed. The experimental design was arranged in a randomized block design with three replications. Treatments consisted of five time seeding dates, 10 May(T1), 25 May(T2), 10 June(T3), 25 June(T4) and 10 July(T5), and same time harvesting, 22 October. Therefore, growing period were 164 days(T1), 149 days(T2), 134 days(T3), 119 days(T4) and 103 days(T5), respectively. In growth stage at harvest time, T1, T2, T3, T4 and T5 were early flowering, end heading, middle heading, early heading and early heading stage, respectively. Plant length and dead leaf were highest in T1, but leaf width and number of leaf were highest in T2 than others (p<0.05). Leaf length, stem diameter and number of tiller were not significantly different among the treatments (p<0.05). Stem hardness was higher in order of T1(2.0)> T2(1.9) > T3=T4(1.7) > T5($1.2kg/cm^2$). Fresh yield and dry matter yield showed significantly higher as the sowing time was faster and the cultivation period was longer (T1 > T2 > T3 > T4 > T5, p<0.05). Crude protein, crude fat and TDN content were highest in T5, but ADF and NDF content were highest in T1 than others (p<0.05). T1, T2 and T3 showed significantly higher crude protein yield compared to T4 and T5 (p<0.05). Total digestible nutrients yield were higher in order of T1 > T2 > T3 > T4 > T5 (p<0.05), and relative feed value were higher in order of T5 > T4 > T3 > T2 > T1 (p<0.05). Based on the above results, seeding dates could be recommended from the early May to the late May, and it is efficient that the cultivation period is over 150 days for dry matter yield, crude protein yield and total digestible nutrients yield.

Growth of Tomato and Pepper Grafted Plug Seedlings under Different Shading Condition During Acclimatization after Graft-taking (접목활착 후 순화시 차광조건에 따른 토마토와 고추 접목묘의 생육)

  • Acclimatization after grafting and graft-take that in order to produce plug seedlings of high-quality are important plug seedling stage which not reduce the plug seedlings quality before shipment. Appropriate acclimatization environment can not only increase seedling quality before secondary growth period, but also effective in promoting the growth of plug seedlings. This study was conducted to determine the optimal the shading period and shading level for acclimatization of tomato and pepper grafted plug seedlings after graft-take. The tomato and pepper seedlings were used in this experiment, and after graft-take, a tunnel was installed on a bed in glasshouse of venlo type to started shading treatment. The shading levels were 35, 55, 75 and 95%, and the shading periods were 1 and 2 weeks, and non-treatment was set as the control. In the case of tomato grafted plug seedling, plant height, stem diameter, dry weight of root, leaf area were significantly higher at the shading period of 1 week and the shading level was 55%. In the case of pepper grafted plug seedling, plant height, stem diameter, and leaf area were the highest in the shading period of 2 weeks and the shading level was 35%. However, dry weight of root, compactness, and T/R ratio, which seedling quality indicators, were lower than in the shading period of 1 week and the shading level 55%. Therefore, considering the quality of seedlings, it is suggested that shading period of 1 week with shading level of 55% treatment can be suitable to produce high quality grafted plug seedlings of tomato and pepper.

Development of simultaneous analytical method for investigation of ketamine and dexmedetomidine in feed (사료 내 케타민과 덱스메데토미딘의 잔류조사를 위한 동시분석법 개발)

  • According to media reports, the carcasses of euthanized abandoned dogs were processed at high temperature and pressure to make powder, and then used as feed materials (meat and bone meal), raising the possibility of residuals in the feed of the anesthetic ketamine and dexmedetomidine used for euthanasia. Therefore, a simultaneous analysis method using QuEChERS combined with high-performance liquid chromatography coupled with electrospray ionization tandem mass spectrometry was developed for rapid residue analysis. The method developed in this study exhibited linearity of 0.999 and higher. Selectivity was evaluated by analyzing blank and spiked samples at the limit of quantification. The MRM chromatograms of blank samples were compared with those of spiked samples with the analyte, and there were no interferences at the respective retention times of ketamine and dexmedetomidine. The detection and quantitation limits of the instrument were 0.6 ㎍/L and 2 ㎍/L, respectively. The limit of quantitation for the method was 10 ㎍/kg. The results of the recovery test on meat and bone meal, meat meal, and pet food showed ketamine in the range of 80.48-98.63 % with less than 5.00 % RSD, and dexmedetomidine in the range of 72.75-93.00 % with less than 4.83 % RSD. As a result of collecting and analyzing six feeds, such as meat and bone meal, prepared at the time the raw material was distributed, 10.8 ㎍/kg of ketamine was detected in one sample of meat and bone meal, while dexmedetomidine was found to have a concentration below the limit of quantitation. It was confirmed that the detected sample was distributed before the safety issue was known, and thereafter, all the meat and bone meal made with the carcasses of euthanized abandoned dogs was recalled and completely discarded. To ensure the safety of the meat and bone meal, 32 samples of the meat and bone meal as well as compound feed were collected, and additional residue investigations were conducted for ketamine and dexmedetomidine. As a result of the analysis, no component was detected. However, through this investigation, it was confirmed that some animal drugs, such as anesthetics, can remain without decomposition even at high temperature and pressure; therefore, there is a need for further investigation of other potentially hazardous substances not controlled in the feed.

A Study on the Effect of Water Soluble Extractive upon Physical Properties of Wood (수용성(水溶性) 추출물(抽出物)이 목재(木材)의 물리적(物理的) 성질(性質)에 미치는 영향(影響))

Since long time ago, it has been talked about that soaking wood into water for a long time would be profitable for the decreasing of defects such as checking, cupping and bow due to the undue-shrinking and swelling. There are, however, no any actual data providing this fact definitly, although there are some guesses that water soluble extractives might effect on this problem. On the other hand, this is a few work which has been done about the effect of water soluble extractives upon the some physical properties of wood and that it might be related to the above mentioned problem. If man does account for that whether soaking wood into water for a long time would be profitable for the decreasing of defects due to the undue-shrinking and swelling in comparison with unsoaking wood or not, it may bring a great contribution on the reasonable uses of wood. To account for the effect of water soluble extractives upon physical properties of wood, this study has been made at the wood technology laboratory, School of Forestry, Yale university, under competent guidance of Dr. F. F. Wangaard, with the following three different species which had been provided at the same laboratory. 1. Pinus strobus 2. Quercus borealis 3. Hymenaea courbaril 2. The physical properties investigated in this study are as follows. a. Equilibrium moisture content at different relative humidity conditions. b. Shrinkage value from gre condition to different relative humidity conditions and oven dry condition. c. Swelling value from oven dry condition to different relative humidity conditions. d. Specific gravity 3. In order to investigate the effect of water soluble extractives upon physical properties of wood, the experiment has been carried out with two differently treated specimens, that is, one has been treated into water and the other into sugar solution, and with controlled specimens. 4. The quantity of water soluble extractives of each species and the group of chemical compounds in the extracted liquid from each species have shown in Table 36. Between species, there is some difference in quantity of extractives and group of chemical compounds. 5. In the case of equilibrium moisture contents at different relative humidity condition, (a) Except the desorption case at 80% R. H. C. (Relative Humidity Condition), there is a definite line between untreated specimens and treated specimens that is, untreated specimens hold water more than treated specimens at the same R.H.C. (b) The specimens treated into sugar solution have shown almost the same tendency in results compared with the untreated specimens. (c) Between species, there is no any definite relation in equilibrium moisture content each other, however E. M. C. in heartwood of pine is lesser than in sapwood. This might cause from the difference of wood anatomical structure. 6. In the case of shrinkage, (a) The shrinkage value of the treated specimen into water is more than that of the untreated specimens, except anyone case of heartwood of pine at 80% R. H. C. (b) The shrinkage value of treated specimens in the sugar solution is less than that of the others and has almost the same tendency to the untreated specimens. It would mean that the penetration of some sugar into the wood can decrease the shrinkage value of wood. (c) Between species, the shrinkage value of heartwood of pine is less than sapwood of the same, shrinkage value of oak is the largest, Hymenaea is lesser than oak and more than pine. (d) Directional difference of shrinkage value through all species can also see as other all kind of species previously tested. (e) There is a definite relation in between the difference of shrinkage value of treated and untreated specimens and amount of extractives, that is, increasing extractives gives increasing the difference of shrinkage value between treated and untreated specimens. 7. In the case of swelling, (a) The swelling value of treated specimens is greater than that of the untreated specimens through all cases. (b) In comparison with the tangential direction and radial direction, the swelling value of tangential direction is larger than that of radial direction in the same species. (c) Between species, the largest one in swelling values is oak and the smallest pine heartwood, there are also a tendency that species which shrink more swell also more and, on the contrary, species which shrink lesser swell also lesser than the others. 8. In the case of specific gravity, (a) The specific gravity of the treated specimens is larger than that of untreated specimens. This reversed value between treated and untreated specimens has been resulted from the volume of specimen of oven dry condition. (b) Between species, there are differences, that is, the specific gravity of Hymenaea is the largest one and the sapwood of pine is the smallest. 9. Through this investigation, it has been concluded that soaking wood into plain water before use without any special consideration may bring more hastful results than unsoaking for use of wood. However soaking wood into the some specially provided solutions such as salt water or inorganic matter may be dissolved in it, can be profitable for the decreasing shrinkage and swelling, checking, shaking and bow etc. if soaking wood into plain water might bring the decreasing defects, it might come from even shrinking and swelling through all dimension.

Relationship between the Economical Defects of Broiler Meat Carcass and Quality Grade A with a Meat Grader (경제성 관련 계육 도체 결함과 축산물등급사에 의한 품질 A등급의 상관관계)

  • To investigate the relationship between the carcass defects considered by a meat grader and quality grade A, a total of 59,700 broiler carcasses, which were reared from September 1, 2007 to August 31, 2008 and randomly selected from 597 broiler farms with 100 broilers in each farm, were graded by a meat grader using the Korean meat grading system. The average slaughter weight of the broilers increased at the 31- to 33-d age but decreased after the 37-d age. The average daily gain to grade A ratio of the broilers was highest in autumn, and the bruises, flesh blemishes, and abnormalities of the carcasses were found to have been most frequent in winter (p<0.05). The number of economical defects (EDs) increased with the increase in the slaughter age, and the correlation coefficients of the EDs and grade A were -0.88, -0.48, and -0.12 in terms of the conformation, exposed flesh/cuts/tears, and skin discolorations of the carcasses, respectively. The correlation coefficients of the non-economical defects (NEDs) and the grade A of the broiler carcasses were -0.16, -0.22, and -0.32 in terms of the feathers, alien substances, and carcass treatment, respectively. The total number of defects (EDs + NEDs) and the grade A were highly correlated (-0.93), and in particular, the correlation between the grade A and the EDs was -0.91. It can be deemed that the grade A from a meat grader is highly correlated with the EDs. Therefore, the results suggest that broiler carcass grade evaluation using a meat grading system can provide direct information on profits in the market. The resulting carcass grade of broilers can also provide the information needed to decide on a proper incentive for a farm that has attained a good quality grade.

Effects of Nitric Oxide Scavenger and Inhibitor on the Development of Bovine IVM/IVF Embryos (Nitric Oxide 화합물 첨가가 소 체외수정란의 체외발육에 미치는 효과)

  • This study was designed to evaluate the effects of nitric oxide scavenger (hemoglobin) and inhibitor (L-nitro-L-arginine methyl ester; L-NAME) with or without cumulus cell on the development of bovine IVM/IVF embryos. When CR/sub 1aa/ medium were supplemented with different dosage (lug/m, 5ug/m and 10ug/ml) of hemoglobin at 48hrs for in vitro culture, the proportion of embryos developing beyond morulae stage in 0, 1ug/ml and 5ug/ml with or without cumulus cell were 23.8%, 33.3 % and 26.8%, and 39.5%, 54.8% and 48.8%, respectively. There was a significantly difference the developmental rate of 1ug/ml hemoglobin intact cumulus cells to any other groups (P<0.05). On the other hand, when added to hemoglobin at 96 hrs, 1 ug/ml hemoglobin with cumulus cell group was significantly increased the percentage of developing into morulae and blastocysts to any other groups (P<0.05), and similar trend that of added at 48hrs. The overall means of the percentage of developing into morulae and blstocysts in 1ug/ml hemoglobin group was significantly increased than those of any other groups (P<0.05) and cumulus co-culture with hemoglobin was increased the in vitro developing rate of IVM/IVF embryos. In CR/sub 1aa/ medium treated with L-NAME 0, 10, 50 and 100mM, the developmental rate of morula plus blastocysts were 55.6%, 64.9%, 58.8% and 66.7%, respectively. The L-NAME did not affect the developmental rate and the cell numbers of blastocysts in all treated groups. These results indicate that hemoglobin and cumulus co-culture can increase the proportion of embryos that developed into morulae and blastocysts, but cell numbers of blastocysts were not affect in all groups.

Change of Nutrition Loss of Long-term Application with Different Organic Material Sources in Upland Soil (유기물원이 다른 퇴비연용 밭토양에서 양분유실량 변화)

  • The objective of this study was to determine the effects of various kinds of composts on the change of nutrition loss in upland soils. Field experiments were conducted in the loam and sandy loam soils, while the clay loam and sandy loam soils were used for laboratory experiments. Various kinds of composts such as poultry manure compost(PMC), cow manure compost(CMC), human excrement sludge(HES), and food industrial sludge compost(FISC) were applied annually at rates of 0, 40, and $80mg\;ha^{-1}$ to soils grown with soybean and maize plants for 4 years during 1994 to 1997. The results of this study were as follows : The loss of nutrients in the form of cation and anion by run-off water increased with the increase of compost application rate. Compared with bare soils, maize cultivation decreased the nutrient loss by run-off from soils by 43% in anionic form and 32% in cationic form. Amount of cation loss were ordered $K^+$ > $Ca^{2+}$ > $Na^+$ > $Mg^{2+}$ > $NH_4{^+} $ and that of anion loss were ordered $SO_4{^{2-}}$ > $NO_3{^-}$ > $Cl^-$ > $PO_4{^{3-}}$. Nutrient loss of sand loam soil in the cation and anion by percolation water increased 1.7 times compared with loam soil. $NO_3{^-}-N$ contents in percolated water were high at the initial stage after compost application, and the amounts were higher in sandy loam soil than loam soil. The maize cultivation also decreased the $NO_3{^-}-N$ contents in percolated water by 82% in loam soil, and 58% in sand loam soil. Soil pH of composts determined by laboratory incubation test increased pH 6.1~6.8 application with poultry and cow manure compost but application with human excrement sludge decreased pH 4.5~4.7. Soil EC were increased initially composts application and decreased up to 2 weeks, thereafter kept a certain level. Nitrogen mineralization rates of composts determined by laboratory incubation test at $25^{\circ}C$ were 39~76% in sandy loam soil, and 16~48% in clay loam soil.
