  • To investigate the antialopecic effect of N-4-salicylomide-2-amino-6-piperidinopyrimidine 3-oxide(SALMI) and its salt on the hair of black mouse (C57BL/6), this study was carrie d out. When salmi and its salt in ethanol solution were administered to the black mouse (C57BL/6) by route of skin, both of them promoted the growth of hair.

Studies on the effect of Betula platyphylla extract on human dermal papilla cell proliferation and its mechanism of action (자작나무 추출물에서 보이는 모유두(HDP) 세포 성장 촉진 효과와 작용 메커니즘 연구)

  • Betula platyphylla extract includes various materials which showed biological activity such as terpenoids. For this reason, Betula platyphylla extract has been used to alleviate inflammation. In this study, extract of Betula platyphylla was obtained and purified using several solvents and evaluated whether they showed effect on prevention of hair loss. Cell cytotoxicity assay was performed to investigate the effect of extracts on cell proliferation. Western blotting was performed to observe the changes in expression of several related growth factors such as β-catenin, VEGF, IGF1, and cyclin D. Also, 5-α-reductase activity was measured. The ethyl acetate extract was divided into four partial extracts and named as H3-1, H3-2, H3-3, and H3-4. The H3-2 extract showed proliferation activity of human derma papilla cell and increased the protein expression of several related growth factors such as β-catenin, VEGF, IGF1, and cyclin D, comparable to the effect of Ethyl 3,4,5-Trimethoxy Benzoate (ETB)and Lupeol (LPO). Moreover, we found that the fraction H3 was shown to decrease 5-α-reductase activity while ETB and LPO had no significant effect on 5-α-reductase activity.

Hair-growth Promoting Effect of Microneedle Roller Therapy (미세침요법의 모발성장효과)

  • Micro needle roller therapy has been used for cosmetic purposes, such as reducing skin winkles and improving elasticity of skin. It is claimed that micro needle roller therapy has potentials for connective tissue regeneration by facilitating collagen synthesis. Therefore, there seems to be a possibility that connective tissue regenerating potential of micro needle roller therapy could influence the hair growth cycle. This study, we investigated the hair growth-promoting effects of micro needle roller therapy. C57BL/6 mice were devided into three groups as follows: normal saline-treated, minoxidil-treated, and micro needle roller therapy-received group. Hair growth activity was evaluated by handscopic and microscopic observations. Sections of dorsal skin were stained with hematoxylin and eosin. Expression of BrdU, FGF, and VEGF was detected by immunohistochemical staining. Micro needle roller therapy enhanced the development of hair follicle during anagen. Immunohistochemical analysis revealed that micro neeld roller therapy incresed the expression of BrdU and FGF in the hair follicles of C57BL/6 mice. Furthermore, micro needle roller therapy upregulated mRNA expression of VEGFR-2, FGF-2, EGF - growth factors that play a central role in hair follicle development during anagen. These results suggest that Micro needle roller therapy can potentially be used for the treatment of alopecia.

Cutaneous Adverse Reactions Induced by Gefitinib (Iressa) in Lung Cancer Patients (폐암 환자들에서 Gefitinib (Iressa)에 의한 피부 부작용)

  • Background: Gefitinib (ZD 1839, Iressa) is a new anticancer agent; more specifically, it is a selective epidermal growth factor receptor tyrosine kinase inhibitor that is, widely used for various solid cancers, including lung cancer. Cutaneous adverse reactions induced by gefitinib have recently been reported; however, not much on this topic has been reported in the Korean literature. Method: We studied cutaneous adverse reactions of gefitinib in 23 patients who suffered with non-small cell lung cancer at Chonnam National University Hwasun Hospital from October 2004 to September 2005. Result: The patients ranged from 23-72 years old, and there were 17 patients with adenocarcinoma, 5 with squamous cell carcinoma and 1 with bronchioloalveolar carcinoma. The most common adverse reaction was acneiform eruptions in 15 patients (65.2%). This reaction appeared within 2 months after medication, and it didn't correlate with the therapeutic response and tumor type. Pruritus was the second most common reaction (39.1%), which was mild and generalized, especially around eyelid area. Xerosis (26.1%), exfoliation on palm and sole (21.7%), and paronychia (21.7%) followed. Hair breakage and intertrigo were rare adverse reactions. Conclusion: Various cutaneous adverse reactions were observed in patients with non-small cell lung carcinoma after gefitinib treatment. The skin complications could be alleviated with dermatologic consultations and treatments, skin complications could be alleviated.

The Promoting Effect of Cornus officinalis fermented with Lactobacillus rhamnosus on Hair Growth (산수유 유산균 발효액의 모발성장 효과)

  • The bacterial growth and pH of Cornus officinalis fermented with Lactobacillus rhamnosus during fermentation were evaluated. As the results, the number of the fermentation after fermentation always remained higher than 6 log CFU/mL and the pH of those ranged from 4 to 6. To evaluate the effect of Cornus officinalis fermented with Lactobacillus rhamnosus on hair growth promotion in C57BL/6 mice, Six weeks old male mice were divided into four groups including normal group (saline), negative control group (essence base), positive control group (minoxidil) and experimental group (Cornus officinalis and animal milk fermented with Lactobacillus rhamnosus mixed in negative control). And they were applied topically with test materials for 8 days. Hair regrowth effect in experimental group using gross and histological examination was higher than that in positive control group. Body weight and food intake of four groups didn't show significant difference. These results indicated that the Cornus officinalis fermented with Lactobacillus rhamnosus can be used practically for hair growth or prevention of hair loss.

Effect of Origanum vulgare Extracts on Hair Regeneration (오레가노 추출물의 모발성장 촉진효과)

  • This study was carried out to investigate the effect of Origanum vulgare extracts on cell proliferation of human hair dermal papilla cell (HHDPC) using sulforhodamine B (SRB) assay, antioxidant activity by 1,1-diphenyl-2-picryl hydrazyl (DPPH) method, expression of insulin-like growth factor-1 (IGF-1) by analyzing reverse transcriptase polymerase chain reaction (RT-PCR) and hair growth in a shaving animal model of C57BL/6 mice topically applying with an amount of 0.1 mL once a day for 3 weeks. The mice were divided into 4 groups including normal group (saline, N), negative control group (dimethyl sulfoxide, NC), positive control group (5 mg/mL minoxidil, PC), and experimental group (Origanum vulgare extracts, OV). Treatment of OV didn't show cytotoxicity in HHDPC up to 10 ${\mu}g/mL$ and exhibited antioxidant activity with $IC_{50}$ of 31.0 ${\mu}g/mL$. IGF-1 expression in the skin was significantly (p<0.05) increased in the PC and OV compared to the N or NC. PC and OV also showed a prominently promoted hair regrowth compared to the N or NC in hair growth observation. The hair regrowth of OV was significantly higher than that of PC (p<0.05). Therefore, these results indicate that O. vulgare extracts effectively stimulated hair growth in an animal model.

Medicinal Herbal Complex Extract with Potential for Hair Growth-Promoting Activity (발모효과를 가지는 한방복합처방단)

  • To develop new therapeutic materials to prevent hair loss and enhance hair growth, we developed a medicinal herbal complex extract (MHCE) using 23 herbs traditionally used in oriental medicine. Medicinal Herbal complex extract was consist of Angelica gigas Nakai, Psoralea corylifolia Linne, Biota orientalis Endlicher, and Eclipta prostrata Linne, Rehmannia glutinosa Liboschitz var. purpurea Makino, Ligustrum lucidum Aiton, Polygonum multiflorum Thunberg, and Sesamum indicum Linne, Sophora angustifolia Sieboldet Zuccarini, Angelica dahurica Benthamet Hooker, and Leonurus sibiricus Linne, Salvia miltiorrhiza Bunge, Prunus persica Batsch, Commiphora molmol Engler, Chrysanthemum indicum Linne, Boswellia carterii Birdwood, Panax ginseng C. A. Meyer, Cnidium officinale Makino, Albizia julibrissin Durazzini, and Corydalis ternata Nakai that have traditionally been used for treating hair loss, preventing gray hair, anti-inflammation, and blood circulation in oriental medicine. In addition, we examined the hair growth effect of MHCE in vitro and in vivo. In vitro, we evaluated the effects of MHCE on cultured HFDPC, HaCaT cells, and murine embryonal fibroblasts (NIH3T3 cells). Also, we evaluated the ability of MHCE to prevent gray hair on murine melanoma cells (B16F1 cells). The hair growth-promoting effect of MHCE in vitro was also observed in vivo using C57BL/6 mice. Our results showed that MHCE significantly increased the proliferation of HFDPC (175 % proliferation at $50{\mu}g/mL$), HaCaT cells (133 % proliferation at $20{\mu}g/mL$), and NIH3T3 cells (120 % proliferation at $50{\mu}g/mL$). MHCE also showed consistent melanogenesis in B16F1 cells (154 % melanin synthesis at $50{\mu}g/mL$). Moreover, MHCE showed potential for hair growth stimulation in C57BL/6 mice experiments (98 % hair growth area on 4 weeks). These results indicate that MHCE may be a good candidate for promotion of hair growth.

In Vitro Anti-aging and Hair Follicle Dermal Papilla Cells Activation Effects of Usnea diffracta Vain Extract (송라 추출물의 세포 수준에서 항노화 및 모유두세포 활성화 효과)

  • Songla (Usnea diffracta Vain.) is one of the lichens belonging to the genus Usnea, and pharmacological activities such as antioxidant, antimicrobial, anti-inflammatory, anti-tumor and cardiovascular protection have been reported in previous studies, but its efficacy in skin and hair is not well known. In this study, the effect of Usnea diffracta extract (UDE) on anti-aging and dermal papilla cell proliferation was verified in vitro. As a result of the experiment, it was confirmed that the UDE significantly reduced the expression of MMP-1 and the activity of MAPKs (ERK, p38, JNK) and AP-1 (c-Fos, c-Jun), which were increased by UVA in HDFn. In addition, the UDE significantly increased the proliferation of HFDPC and significantly increased the mRNA expression of VEGF and KGF, which are hair growth factors. Accordingly, the phosphorylation of ERK/CREB involved in hair proliferation and expression of growth factors was increased in a concentration-dependent manner. The main component represented by the main peak was separated and purified using Prep LC by concentrating the UDE, which was confirmed as diffractaic acid through NMR and Mess analysis. Isolated diffractaic acid significantly reduced the expression of MMP-1 increased by UVA in HDFn and increased the proliferation of HFDPC in a concentration-dependent manner. The result suggest that UDE proved its usability as a natural cosmetic material with anti-aging and dermal papilla cell activation effects.

Collagen Increasing and Hair Growth Effect of Supramolecular Cosmetic Materials Containing Phytochemicals (식물성 유용물질을 함유하는 초분자체 화장제재의 콜라겐 증가 및 육모효과)

  • Medicinal herbs have been shown to have protective functions for skin and hair. We investigated the effects of complex of soluble ${\beta}$-cyclodextrin and phytochemicals on the functions of skin and hair. In previous report, we evaluated the safety of supramolecules and found their anti-microbial effects and anti-fungal effect against Gram (+) and Malassezia furfur which is known to cause dandruff. Here we present that functional supramolecules-containing cream promotes the biological skin activity by inducing the collagen formation. And treatment of supramolecules-containing hair tonic increased the rate of hair growth of mouse. Taken together, supramolecular cosmetic compounds containing water insoluble phytochemicals and water soluble ${\beta}$-cyclodextrin exhibit the potential ability for hair growth promotion and delaying the aging of skin.

Synergistic Effects of the Combination of Oral Herbal Composition and Topical Hair Tonic on Hair Growth in C57BL6 Mice (경구용 생약 복합 조성물과 경피용 육모 조성물의 복합 사용에 따른 육모 상승 효과)

  • Currently, various treatments available for alleviating hair loss and combination treatments are commonly used, which are frequently questionable and in effective. We aimed to investigate the synergistic effect regarding the combination of oral herbal composition and topical hair tonic on anagen induction and hair restoration in a shaving model of C57BL/6 mice. Seven-week old mice were trimmed by electric clippers and treated with oral herbal composition and topical hair tonic either alone or in combination. The combination treatment showed the highest hair growth promoting effect. Moreover, it significantly improved total blood antioxidant capacity and reduced lipid peroxidation and triglyceride level, which was not observed in the topical hair tonic treated group. These results suggest that the combination of oral herbal composition and topical hair tonic has a synergistic hair growth promoting effect and such synergism may be the result of the differing hair regrowth mechanisms concerning treatments.
