Biological Control of Plant Pathogens by Bacillus sp. AB02. (Bacillus sp. AB02를 이용한 식물 병원균에 대한 생물검정)

  • In the greenhouse fields for fruits and vegetables during the winter in Korea, there are serious damages by the sclerotium diseases due to the low temperature and humidity. This study was carried out to select an antagonic agent for the biological control of the sclerotium diseases. The 55 antagonic agents were selected from the rhizosphere in soil where the fruits and vegetables were cultivated in the green house fields, and strain AB02 among the tested isolates was estimated to be the strongest antagonist against the sclerotium disease. Using strain AB02, the antifungal spectrum was tested against 5 different plant pathogens. According to the results of the test, strain AB02 . showed the high antagonistic effect against Botrytis cinerea and Sclerotinia sclerotiorum. For the experiment of biological control against the sclerotium disease, it was estimated the suppression effect and the control effect by the strain in the pot experiment using the green perilla. According to the result of the pot experiments, the suppression effect was 40% and the control effect was 62%, respectively. For the stimulation effect of the tested plant growth by strain AB02 compared to the control, it was improved as 120% for the total length, 141% for the liveweight, 121% for the total number of leaves, 185% for the leaf area, and 327% for the liveweight of the root, respectively. Strain AB02 showing the antagonistic effect against the sclerotium disease and the stimulation effect for the plant growth was identified as Bacillus sp.

Cloning of Geranylgeranyl Pyrophosphate Synthase (CrtE) Gene from Kocuria gwangalliensis and Its Functional Co-expression in Escherichia coli (코쿠리아 광안리엔시스의 제라닐제라닐 피로인산염 합성 효소의 클로닝과 대장균에서 공발현을 통한 효소 활성에 관한 연구)

  • A gene encoding a novel geranylgeranyl pyrophosphate (GGPP) synthase from Kocuria gwangalliensis has been cloned and expressed in Escherichia coli. The deduced amino acid sequence showed 59.6% identity with a putative GGPP synthase (CrtE) from K. rhizophila. An expression plasmid containing the crtE gene was constructed, and E. coli cells containing this plasmid produced a recombinant protein with a theoretical molecular mass of 41 kDa, corresponding to the molecular weight of GGPP synthase. Due to the lack of crtE, crtB, and crtI in E. coli, the biosynthesis of lycopene was only obtained when the plasmid pCcrtE was co-transformed into E. coli expressing the pRScrtBI-carrying carotenoid biosynthesis crtB and crtI genes, which were sub-cloned from Paracoccus haeundaensis. The biochemical studies on the expressed proteins were performed via HPLC. The results obtained from this study will provide a wider base of knowledge regarding the primary structure of CrtE cloned from K. gwangalliensis at the molecular level.

Antioxidative Activity and Produced Condition of Antioxidative Substance by Bacillus sp. FF-7 (Bacillus sp. FF-7에 의한 항산화물질 생산조건과 항산화 활성)

  • The antioxidative activity of antioxidative substances produced from several bacterial strains isolated from fermented foods were tested by $DPPH\;({\alpha},{\alpha}'-diphenyl-{\beta}-picrylhydrazyl)$ free radical scavenging activity. One of the strains showing the highest antioxidative activity was identified as Bacillus sp. based on the morphological, biochemical, physiological characteristics, and 16S rRNA sequence, and named FF-7. The most optimal medium condition for the production of antioxidative substance from Bacillus sp. FF-7 was 2% galactose as carbon source and l% tryptone as nitrogen source. The antioxidative substance produced from FF-7 in these cultural medium was also tested by in vitro experimental models, the peruxidation of linoleic acid and the peroxidation of rat tissues microsomes by using thiobarbituric acid (TBA) for assay of free malondialdehyde production. The antioxidative activity against lipid peroxidation of rat tissues microsomes was shown in the following order; brain 97.50% > heart 79.95% > kidney 77.84% > spleen 77.47% > testis 69.96% > liver 62.45%. The antioxidative substance produced from FF-7 on linoleic acid peroxidation by IBA method was effectively inhibited during four days, and 0.05% BHT (butylated hydroxytoluene) used comparative control was also effectively inhibited. Results showed that the highest antioxidative activity by DPPH method of antioxidative substance produced from Bacillus sp. FF-7 was obtained by supplementing 2% galactose as carton source and l% tryptone as nitrogen source in cultured medium, this substance effectively inhibited the formation of TBARS in brain microsome in vit개 system and in linoleic acid peroxidation.

Characteristics of Watermelon Mosaic Virus Transmission Occurring in Korean Ginseng (인삼에서 발생하는 수박모자이크바이러스의 감염 특성)

  • Korean ginseng (Panax ginseng) is the most popular herb for medical purpose in Korea. Recently, viral diseases from Korean ginseng showing various degrees of severe mottling, variegation and mosaic symptoms have caused quantity losses of Korean ginseng in a large number of farms. Watermelon mosaic virus (named WMV-gin) was identified as a causal agent for the disease of Korean ginseng. Interestingly, WMV-gin failed to infect both Korean ginseng plant and susceptible host species including cucurbitaceous plants by mechanical inoculation. However, WMV-gin could successfully infect Korean ginseng by transmission of two aphid species (Myzus persicae and Aphis gossypii). It is likely that transmission of WMV-gin was done by both the aphid species during feeding behavior of the two aphid species on Korean ginseng, though the aphids dislike feeding in Korea ginseng. Similarly, a strain of WMV (WMV-wm) isolated from watermelon was transmitted successfully to Korean ginseng plant by the two aphid species, but not by mechanical inoculations. Transmission assays using M. persicae and A. gossypii clearly showed both WMV-gin and WMV-wm were not transmitted from infected Korean ginseng plant to cucurbit species that are good host species for WMV. These results suggest WMV disease occurring in Korean ginseng plant can be controlled by ecological approaches.

Physiological Characterization of BTEX Degrading Bacteria Microbacterium sp. EMB-1 and Rhodococ-cus sp. EMB-2 Isolated from Reed Rhizosphere of Sunchon Bay (순천만 갈대의 근권으로부터 분리한 BTEX 분해세균 Microbacterium sp. EMB-1과 Rhodococcus sp. EMB-2의 생리학적 특성 분석)

  • This study focuses on investigating roles of microorganisms in decontamination of reed rhizosphere in Sunchon Bay, Korea, which is considered one of the marsh and mud environment severely affected by human activities such as agriculture and fisheries. In general, the bay is known to play the role of the buffering zone to reduce the sudden impact or change by environmental stresses. In our initial efforts to elucidate the microbial functions in decontamination process in reed rhizosphere, pure bacteria capable of degrading aromatic hydrocarbons were isolated from reed (Phragmites communis) rhizosphere of Sunchon bay by enrichment culture using either benzene, toluene, ethylbenzene, or xylene (BTEX) as a sole source of carbon and energy. Measurement of the rates of BTEX degradation and cell growth during the incubation in BTEX media under several temperature conditions demonstrated maximized degradation of BTEX at $37^{\circ}C$ in both strains. Both strains were also resistant to all the heavy metals and antibiotics tested in this study, as well as they grew well at $42^{\circ}C$. Identification of the isolates based on 16S rRNA gene sequences, and a variety of phenotypic and morphologic properties revealed that the two strains capable of BTEX catabolism were among Microbacterium sp., and Rhodococcus sp. with over $95{\%}$ confidence, designated Microbacterium sp. EMB-1 and Rhodococcus sp. EMB-2, respectively This result suggested that in the rhizosphere of reed, one of major salt marsh plants they might play an important roles in decontamination process of reed rhizosphere contaminated with petroleum such as BTEX.

An Accurate and Efficient Analysis of Composite Plates Based on Enhanced First-order Shear Deformation Theory (개선된 일차전단변형이론을 이용한 복합재료 적층평판의 고정밀 해석)

  • In this paper, an efficient yet accurate stress analysis based on the first-order shear deformation theory (FSDT) is presented. The transverse shear strain energy is modified via the mixed variational theorem, so that the shear correction factors are automatically involved in the formulation. In the mixed variational formulation, the transverse stresses are taken to be functions subject to variations. The transverse shear stresses based on an efficient higher order plate theory (EHOPT, Cho and Parmerter, 1993) are utilized and modified, while the transverse normal stress is assumed to be the third-order polynomial of thickness coordinates, which satisfies both zero transverse shear stresses and prescribed surface fractions in top and bottom surfaces. On the other hand, the displacements are assumed to be those of the FSDT Resulting strain energy expressions are referred to as an EFSDTM3D that stands for an enhanced first-order shear deformation theory based on the mixed formulation for three dimensional elasticity, The developed EFSDTM3D preserves the computational advantage of the classical FSDT while allowing for important local through-the-thickness variations of displacements and stresses through the recovery procedure that is based on the least square minimization of in-plane stresses. Comparisons of displacements and stresses of both laminated and sandwich plates using the present theory are made with the classical FSDT, three-dimensional exact solutions, and available data in the literature.

Purification of Two Novel Antimicrobial Peptides from Pyloric Caeca of the Starfish Asterina pectinifera (별불가사리 Asterina pectinifera의 유문맹낭 추출물로부터 새로운 2종류의 항균활성 펩타이드의 정제)

  • PAP-1, a novel antimicrobial peptide isolated from pyloric caeca extract of the starfish Asterina pectinifera was purified and characterized. First, the acidified pyloric caeca extract was put through Sep-Pak C18 solid phase extraction cartridge using a stepwise gradient. Among the eluents, RM 60 (retained materials at 60% methanol) showed good antimicrobial activity against Bacillus subtilis and Escherichia coli D31 and was purified in C18 reversed-phase and ion-exchange high-performance liquid chromatography columns. The purification steps yielded two novel peptides showing strong antimicrobial activities. These peptides were named pyloric caeca A. pectinifera peptide 1 and 2 (PAP-1 and PAP-2). For the characterization of the purified peptides, the molecular weights and amino acid sequences were determined by MALDI-TOF MS and Edman degradation. The molecular weights of PAP-1 and PAP-2 were about 2951.54 Da and 2980.15 Da respectively. The amino acid sequences of PAP-1 and PAP-2 were partially determined: AIQNAGES and AIQNAAES, respectively. PAP-2 is an isoform of PAP-1, differing merely by a single residue at position 6 (glycine or alanine). The comparison of the N-terminal amino acid sequences and molecular weights of the peptides with those of other known antimicrobial peptides revealed that PAP-1 and PAP-2 have no homology with any known peptides. These findings suggest that PAP-1 and PAP-2 play a significant role in the innate defense system of starfish pyloric caeca.

Isolation and Identification of Cellulase-producing Microorganism, and Determination of Optimal Culture Condition (토양으로부터 Cellulose 분해효소를 생산하는 미생물의 분리, 동정 및 최적배양조건의 결정)

  • The strain No. 33, which produces cellulose-degrading enzyme, was isolated from soil. Yellow halo was identified when the culture supernatant of the strain was loaded onto agar plate containing 2.0% CMC using paper disc method. From scanning electron microscopic observation, the morphology of the stain was rod-shaped. For identification, several biochemical characteristics were tested, and this strain was identified to Bacillus sp. So, we named this strain as Bacillus sp. No. 33. The maximal growth was observed when the stain was cultured in the medium containing 1.0% glucose, 3.0% yeast extract, 0.5% $KH_2PO_4$, 0.02% $MgSO_4{\cdot}7H_2O$, pH 7.0 at $30^{\circ}C$ for 39 hours with shaking. The maximal enzyme production was accomplished using the medium containing 4.0% CMC, 2.0% yeast extract, 0.5% $KH_2PO_4$, 0.04% $MgSO_4{\cdot}7H_2O$, pH 7.0 at $30^{\circ}C$ for 42 hours with shaking.

Studies on Audiological Significance of the Bing Test (Being test의 임상청각학적 의의에 대한 고찰)

  • In normal auditory systems, the difference between the bone conduction thresholds obtained with the test ear opened and occluded in low frequency signals below 1, 000Hz is about 10~25 dB. But no difference in these thresholds suggests the presence of a lesion in the conductive mechanism. Sullivan added the differences in these thresholds at 250Hz, 500Hz and 1,000Hz and called "Occlusion Index". In oder to study the audiological significance, we have measured the occlusion index in 40 ears of normal persons, 20 ears of chronic otitis media patients and 20 ears of sensori-neural hearing impairment patients. We also measured the static compliance in 40 normal ears and observed the correlation with the occlusion index. The results are as follows: 1. Occlusion index was 33. $10\pm10.63dB$ in normal group, $3.10\pm3.03dB$ in chronic otitis media; group, $28.10\pm15.17dB$ in sensori-neural group. 2. Static compliance in normal group was $0.61\pm0.31cc$ (0.22~1.75cc) 3. Occlusion index showed inverse proportion to static compliance.

A Study on the 'fragmentation' trend of modern film montage (현대영화 몽타주의 '파편화(fragmentation)' 경향 연구)

  • The film scholar Vincent Amiel divides into three types of montage through his book The Aesthetics of Montage ; Montage narratif, Montage discursif, and Montage decorrespondances. These three categories are the concept that encompasses the aesthetic class to which most movies belong. Early films pursued the essential and basic functions of editing, which tend to be modified in the direction of enhancing the director's goals over time. In this way, "Expressive Montage" is one of most important concepts of montage, not as a 'methodology' that combines narrative but as a 'purpose'. In the montage stage, the expressive montage work is done through three steps of decision. The process of 'combining' to combine the selected films in a certain order, after the process of 'selection' which selects only necessary parts of the rush film, and 'connection' to determine the scene connection considering the duration of the shot. The connection is the final stage of the montage. There are exceptions, of course. When fiction films of classical narratives use close-ups, or when using models or objects of neutered animals, the film induces the tendency of a "montage decorrespondances" rather than a "montage narratif" or "montage discursif". This study attempts to analyze the tendency of montage of works with 'uncertain connection' through 'collage' used by close-ups and montage decorrespondances as 'fragmentation tendency of modern films'. The fragmentation of the montage in contemporary film breaks the continuous and structural nature of the film, and confuses the narration structure that is visible on the surface of the film. The tendency of the fragmentation of the montage, which started from this close-up, seems to give an answer to the extensibility of the modern image.
