Immunomodulatory Activities of Ethanol Extract of Cordyceps militaris in Immunocompromised Mice (밀리타리스 동충하초(Cordyceps militaris) 에탄올 추출물의 면역억제 마우스 면역활성에 미치는 영향)

  • In order to determine the functional benefits of $Cordyceps$ $militaris$ in the immune system, we examined the immunomodulatory activities of $Cordyceps$ $militaris$ in an immunocompromised C57BL/6 mice model. Mice were injected intraperitoneally with an immunosuppressive drug, cyclophosphamide, and then administered orally with 3% hydroxypropylmethylcellulose or 30, 100, and 300 mg/kg of 50% ethanol extract of $Cordyceps$ $militaris$ (CM 30, CM 100, and CM 300, respectively) for 12 days. Mice treated with CM displayed significantly increased splenocyte proliferation and natural killer cell activity compared to immunosuppressed control mice (p<0.05). The spleen cells isolated from mice treated with CM also displayed increased production of Th1 cytokines, including IL-2, IL-12, IFN-${\gamma}$ and TNF-${\alpha}$, suggesting enhanced cellular immunity in response to CM. However, CM had no significant effect on the production of IL-4 and IL-10. These results indicate that $Cordyceps$ $militaris$ enhances immune function by promoting immune cell proliferation and Th1 cytokine production.

Gelidium amansii Extract, a Potent α-glucosidase and α-amylase Inhibitor, Alleviates Postprandial Hyperglycemia in Diabetic Mice (당뇨 마우스에서 우뭇가사리(Gelidium amansii)의 식후 고혈당 완화 효과)

  • Gelidium amansii shows antioxidant and anti-obesity effects; however, the effect on postprandial blood glucose levels is not known. The objective of the present study was to investigate the inhibitory effect of Gelidium amansii extract (GAE) on carbohydrate-digesting enzymes and its ability to alleviate postprandial hyperglycemia in streptozotocin (STZ)-induced diabetic mice. Gelidium amansii was extracted with 80% ethanol and concentrated for use in this study. The ${\alpha}-glucosidase$ and ${\alpha}-amylase$ inhibition assays were performed using the colorimetric method. ICR normal and STZ-induced diabetic mice were orally administered GAE (300 mg/kg body weight) or acarbose (100 mg/kg body weight) alone or soluble starch (2 g/kg body weight). Blood samples were taken from the tail vein at 0, 30, 60 and 120 min. Our results indicated that GAE markedly inhibited ${\alpha}-glucosidase$ and ${\alpha}-amylase$ activities with $IC_{50}$ values of $0.099{\pm}0.009mg/ml$ and $0.178{\pm}0.038mg/ml$, respectively, and was a more effective inhibitor than acarbose, the positive control. Further, the postprandial blood glucose levels of STZ-induced diabetic mice in the GAE-administered group were significantly lower than those of control group mice (p<0.05). Moreover, the area under the curves (AUC) significantly decreased with GAE administration in STZ-induced diabetic mice (p<0.05). These results indicate that GAE may be effective in decreasing postprandial blood glucose levels by inhibiting carbohydrate-digesting enzymes such as ${\alpha}-amylase$ and ${\alpha}-glucosidase$. Therefore, GAE could be used as a potential functional food for alleviating postprandial hyperglycemia.

Effects of Oral Administering Kefir on Blood Glucose Levels in Diabetic Mice (Kefir의 투여가 당뇨병 마우스의 혈당치에 미치는 영향)

  • This study was conducted to investigate the effect of oral administering kefir on blood glucose levels in diabetic mice. Kefir($10\;m\ell/kg$ body weight) was orally administered to diabetic mice twice daily for 45 days. Diabetic mice fed saline or 10% skim milk were used as a control. No significant difference in feed intake and weight gain were observed among the treatments. Compared to the saline-fed or skim milk-fed group, blood glucose in the kefir-fed group decreased significantly at 45 days(p<0.05). At 45 days, serum triglyceride and non-esterified fatty acid concentrations in diabetic mice fed kefir decreased significantly compared to those of the saline and skim milk groups(p<0.05). Both high density lipoprotein and low density lipoprotein concentrations were not significantly different among the three treatment groups. In conclusion, the symptoms of diabetic mice improved when kefir was fed for >45 days.

Ulmus macrocarpa Hance Reduces Cyclophosphamide-induced Toxicity in Mouse Liver (Cyclophosphamide가 유발한 간 조직변화에 대한 느릅나무 열수추출물의 완화 효과)

  • Cyclophosphamide (CP) is widely used in cancer and lymphoma treatments and as an immunosuppressant drug. CP is a DNA alkylating agent that metabolizes into 4-hydrocyclophosphamide (4H-CYP) and aldophosphamide in hepatocytes. However, its metabolites cause DNA synthesis disorder, leading to apoptosis and toxic side effects. The development of technology to minimize this side effect is essential to improve CP's clinical application. Various bioactive compounds have been reported to have anti-cancer and antioxidant functions and preventive or therapeutic roles in metabolic diseases. Many researchers have attempted to minimize the side effects and improve the efficacy of these drugs together with the use of bioactive compounds. Ulmus macrocarpa Hance has been used for the treatment of edema, mastitis, stomach pain, tumors, cystitis, and other inflammatory diseases. The aim of this study was to investigate at the histological level the protective function of U. macrocarpa Hance against CP's side effects and any potential toxic effect of U. macrocarpa Hance in the liver and kidney. Water extracts of U. macrocarpa Hance reduced CP-induced toxicity and did not induce any histological damage in the liver and kidney. Therefore, U. macrocarpa Hance would be applicable in the pharmaceutical industry.

NUI/NUX of the Virtual Monitor Concept using the Concentration Indicator and the User's Physical Features (사용자의 신체적 특징과 뇌파 집중 지수를 이용한 가상 모니터 개념의 NUI/NUX)

  • As growing interest in Human-Computer Interaction(HCI), research on HCI has been actively conducted. Also with that, research on Natural User Interface/Natural User eXperience(NUI/NUX) that uses user's gesture and voice has been actively conducted. In case of NUI/NUX, it needs recognition algorithm such as gesture recognition or voice recognition. However these recognition algorithms have weakness because their implementation is complex and a lot of time are needed in training because they have to go through steps including preprocessing, normalization, feature extraction. Recently, Kinect is launched by Microsoft as NUI/NUX development tool which attracts people's attention, and studies using Kinect has been conducted. The authors of this paper implemented hand-mouse interface with outstanding intuitiveness using the physical features of a user in a previous study. However, there are weaknesses such as unnatural movement of mouse and low accuracy of mouse functions. In this study, we designed and implemented a hand mouse interface which introduce a new concept called 'Virtual monitor' extracting user's physical features through Kinect in real-time. Virtual monitor means virtual space that can be controlled by hand mouse. It is possible that the coordinate on virtual monitor is accurately mapped onto the coordinate on real monitor. Hand-mouse interface based on virtual monitor concept maintains outstanding intuitiveness that is strength of the previous study and enhance accuracy of mouse functions. Further, we increased accuracy of the interface by recognizing user's unnecessary actions using his concentration indicator from his encephalogram(EEG) data. In order to evaluate intuitiveness and accuracy of the interface, we experimented it for 50 people from 10s to 50s. As the result of intuitiveness experiment, 84% of subjects learned how to use it within 1 minute. Also, as the result of accuracy experiment, accuracy of mouse functions (drag(80.4%), click(80%), double-click(76.7%)) is shown. The intuitiveness and accuracy of the proposed hand-mouse interface is checked through experiment, this is expected to be a good example of the interface for controlling the system by hand in the future.

The Functional Effects for the Prevention and Treatment on Hair Loss from Astringent Persimmon Fruit Extracts (떫은감 추출물의 발모 및 탈모의 기능성 효과)

  • Astrigent persimmon fruit resources have been used traditionally to treat different systemic diseases and acclaimed for various biological activities including hair growth. This study investigates the hair restoration efficacy of astrigent persimmon fruit extracts on genetically predisposed to balding pattern Black mice model. Water extract of astrigent persimmon fruit(10 mg/mouse/day) with standardized vehicle formulation, only vehicle and positive control minoxidil (2%) were applied daily until completion of two full hair growth generations. The changing pattern of hair growth were observed through two hair growth generations of C57BL/6 Black mice. The hair existing area and hair length was increased significantly (P > 0.001) in astrigent persimmon fruit treated mice than vehicle-treated control mice. Furthermore, histological assessment revealed that the number of hair follicles did not remarkably increase after astrigent persimmon fruit treatment in compare to control mice. Thus, our data revealed that the topical application of astrigent persimmon fruit may promote hair growth in nude mice by extend the hair existing area and increase hair length which is an indicator of prolong anagen phase.

The Effect of Pumpkin and Medical Herb Extract Supplement on Blood Composition of Mouse (호박 및 한방생약재 추출액이 마우스 혈액성상에 미치는 영향)

  • 본 연구는 한방에서 여러 가지 기능성을 주목받고 있는 호박을 주원료로 하고 8가지 한방 생약재를 부재료로 첨가해 이것이 마우스의 혈액 성상 중 적혈구수와 hemoglobin에 어떤 영향을 미치는지 알아보기 위해 진행되었다. 실험은 부재료로 첨가되는 한방생 약재 의 종류와 1회 복용량에 따라9가지 그룹으로 나누어 진행되었으며 먼저 마우스에게서 출혈을 유도한 후 호박 및 한방생 약재 추출물을 먹인 대조군과 비교해 출혈 전으로 회복되는 정도를 살펴보았다. 그 결과 적혈구의 경우9가지 실험구 모두가 대조구에 비해서 유의적으로 적혈구 수가 증가하였다 첨가된 부재료의 수를 줄인 A와 B그룹보다 8가지 한방생약재를 모두 부재료로 첨가한 C그룹에서 가장 높은 증가효과를 보여 주었다. Hemoglobin의 경우에도 9가지 실험구 모두가 대조군에 비해 유의성있는 증가효과를 보여주었다 Hemoglobin도 적혈구와 마찬가지로 A와 B그룹보다 8가지 한방생약재를 모두 첨가한 C그룹에서 회복효과가 가장 우수하였다 적혈구와 hemoglobin모두 1회 복용량보다 첨가되는 한방생약재의 종류에 따라 회복효과가 더 큰 것으로 생각된다.

Untouched Camera Mouse System for The Disabled Person's Assistant Device (뇌병변 장애인 보조기기용 노터치 카메라 마우스 시스템)

  • In this paper, we propose assistant device system based on camera that can perform general mouse function for the physically disabled. Most disabled people use special hardware and rely largely on foreign-made assistance device and the using rate of assistance device was very low because of high price. To solve this problem, we propose effective system based on web camera that can perform general mouse function and can raise accessibility for the physically disabled. In the proposed method, we firstly attach color marker on arbitrary device or body parts and then input the information of color using camera and extract coordinate information of color marker and finally, match processing information(coordinate) to special event(keyboard, mouse) and control various applications. The proposed method shows good performance in price and performance compared with other techniques by real field test.

Effects of Inhaling Essential Oils from Lavender, Silver Fir, Grapefruit on the Restraint Stress in Mice (라벤더, 실버퍼, 그레이프후룻 정유 흡입이 마우스의 구속 스트레스에 미치는 효과)

  • The effects of inhaling three essential oils (lavender [Lavendular angustifolia], silver fir [Abies alba], and grapefruit [Citrus paradise]) against restraint stress were investigated in mice. Serum cortisol levels were significantly increased in the 1h-restraint group. Also, silver fir and grapefruit oils prevented the elevation of cortisol levels under experimental conditions. Inhalation of 10% lavender oil for 30 min before inducing stress significantly inhibited restraint-induced hypothermia. We also investigated the effects of restraint stress on triglycerides, total cholesterol, and high-density lipoprotein cholesterol. It was found that the concentration of triglycerides in the serum samples increased after restraint stress. Silver fir essential oil inhibited elevation of triglyceride levels. Taken all together, the correlation of herb essential oil inhalation against restraint stress in animals can partly be explained.

The role of p38 MAP kinase on RANKL regulation in mouse periodontal ligament fibroblasts (마우스 치주인대 섬유모세포에서 RANKL 조절에 대한 p38 MAP kinase의 역할)

  • Receptor activation of nuclear factor ${\kappa}$ B ligand (RANKL)은 파골세포의 분화와 기능에 중요한 역할을 하는 단백질로 이들 물질의 조절에는 p38 MAP kinase가 관여한다. 그러나 치주인대 섬유모세포에서 RANKL 발현 시 p38 MAP kinase의 역할은 잘 알려져 있지 않다. 이에 이번 연구는 마우스 치주인대 섬유모세포의 $IL-1{\beta}-induced$ RANKL 발현과정에서 p38의 역할을 규명하고자 하여 다음과 같은 결과를 얻었다. 마우스 치주인대 섬유모세포에 $IL-1{\beta}$ (1ng/ml)의 자극은 수용성 RANKL의 합성을 증가시켰다. 수용성 RANKL의 합성은 p38 MAP kinase 억제제인 SB203580에 의해 농도 의존적으로 억제되었으나 다른 MAP kinase 억제제인 SP600125, JNK 억제제와 PD98059, ERK 억제제에 의해서는 수용성 RANKL의 합성이 조절되지 않았다. NF-kB 억제제에 의해서도 수용성 RANKL의 합성이 억제되지 않았다. RANKL 유전자의 발현은 $IL-1{\beta}$로 자극 시에는 대조군에 비해 약 5배의 발현 증가를 보였으나 SB203580으로 전처치 시 $IL-1{\beta}$ (1ng/ml)로 자극시보다 약 1.5배의 감소를 보였다. 그러나 SP600125, PD98059, 및 NF-kB 억제제로 전처치한 경우에는 $IL-1{\beta}$로 자극한 경우와 비슷한 수준을 보였다. $IL-1{\beta}$로 자극 시 RANKL 유전자의 반감기가 90분 이었으나 SB203580로 전처치 후 $IL-1{\beta}$로 자극 시 RANKL 유전자의 반감기는 60분으로 감소하였다. Cycloheximide 전처리 시 SB203580에 의한 RANKL 유전자 발현 억제가 관찰되지 않았다. 단백질 분석결과 p38 MAP kinase의 인산화 활성은 30분까지 증가하였으나 그 이후 감소하여 2시간째에는 그 발현이 미약하였다. SB203580로 전처치 후 $IL-1{\beta}$로 자극 시 p38 MAP kinase의 인산화 활성이 감소하였다. 이상의 결과는 p38 MAP kinase가 RANKL 유전자 조절에 중요한 역할을 담당하고 있음을 시사한다.
