Taste Characteristics and Functionality of Two Stage Enzyme Hydrolysate from Low-Utilized Longfinned Squid (창오징어 2단 효소분해엑스분의 정미특성 및 기능성)

  • The taste characteristics and functionality of low-utilized small longfinned squid as affected by two stage enzyme hydrolysis were examined. In taste active-components, total free amino acid contents in hot-water and autolytic extract, two stage enzyme hydrolysate (TSEH) of longfinned squid were 2,792.5 mg%, 8,393.8 mg% and 9,186.1 mg%, respectively. The major free amino acids were Pro, Leu, Glu, Tau, Lys, Arg, Phe, Val and Ile. As for quarternary ammonium bases, betaine was the principal component (593.8 mg%) and also contents of TMAO, AMP in longfinned squid TSEH were 234.8% mg% and 51.0 mg%, respectively. The major inorganic ions in TSEH were Na(874.0 mg%), K (398.2 mg%), Cl (1,213.1 mg%) and PO$_4$(995.9 mg%). From the results in sensory tests, TSEH was superior to other extracts on the aspects of taste characteristics such as umami intensity, sweetness, taste harmony and transparency of extract. Also TSEH of longfinned squid revealed very higher Angiotensin-I converting enzyme inhibition ratio (92.1%) than those of hot-water and autolytic extract.

Effect of the Salt Concentration in Seafood Processing Wastewater on the Anaerobic Ultimate Biodegradability and Multiple Decay Rate of Organic Matter (해산물 가공폐수내 염분농도가 혐기성 최종생분해도와 유기물 다중분해속도에 미치는 영향)

  • In this study, the anaerobic ultimate biodegradability and multiple decay rate of organic matter were evaluated according to various salt concentrations in seafood processing wastewater. The evaluation was also performed with various types of anaerobic bacteria and S/I (substrate/inoculum) ratios. After the S/I ratio was fixed at 0.9, the ultimate biodegradability values of the anaerobic digested sludge and granular sludge were became 72.0% and 92.0%, respectively. The multiple decay rate coefficients ($k_1$) coefficients of the anaerobic digested sludge and granular sludge were $0.0478{\sim}0.1252\;day^{-1}$ and $0.0667{\sim}0.1709\;day^{-1}$, respectively. The optimum S/I ratio of the seafood wastewater, which was determined based on the ultimate anaerobic biodegradability and gas production, was 0.9. The organic matter removal rate never became less than 85.0% under a 3,000 mg/L chloride concentration. The multiple decay rate coefficients ($k_1$) were $0.1603{\sim}0.1709\;day^{-1}$ under $3,000\;mgCl^-/L$, and $0.0492{\sim}0.0760\;day^{-1}$ in more than $6,000\;mgCl^-/L$. The multiple decay rate coefficients ($k_2$) were $0.0183{\sim}0.0348\;day^{-1}$ under $6,000\;mgCl^-/L$, and $0.0154\;day^{-1}$ at $9,000\;mgCl^-/L$. With increasing chloride concentrations, the reaction rate ($k_1$, $k_2$) and ratio of the rapidly degraded organic matter ($S_1$) decreased.

Decay rate and Nutrient Dynamics during Litter Decomposition of Pinus rigida and Pinus koraiensis (리기다소나무와 잣나무 낙엽의 분해율 및 분해과정에 따른 영양염류 함량 변화)

  • We examined the nutrient dynamics during the leaf litter decomposition rate and process of Pinus rigida and Pinus koraiensis in Gongju for 21 months from December 2014 to September 2016 as a part of National Long-Term Ecological Research Program in Korea. The remaining weight rate of P. rigida and P. koraiensis leaf litter was $58.27{\pm}4.13$ and $54.08{\pm}4.32%$, respectively, indicating that the P. koraiensis leaf litter decomposed faster than P. rigida leaf litter. The decay constant (k) of P. rigida leaf litter and P.koraiensis leaf litter after 21 months was 0.95 and 1.08, respectively, indicating that P. koraiensis leaf litter decayed faster than P. rigida leaf litter probably due to the difference of nitrogen concentration between the two. The C/N ratio of P. rigida and P. koraiensis leaf litter was 64.4 and 40.6, respectively, initially, and then decreased to 41.0 and 18.9, respectively, after 21 months. The C/P ratio of P. rigida and P. koraiensis leaf litter was 529.8 and 236.5, respectively, and then decreased to 384.1, 205.2, respectively, after 21 months. The contents of N, P, K, Ca, and Mg were 6.78, 0.83, 2.84, 0.99, and 2.59 mg/g, respectively, in the P. rigida leaf litter and 10.90, 1.87, 5.82, 4.79, and 2.00 mg/g, respectively, in the P. koraiensis leaf litter, indicating that the elements except the magnesium showed higher contents in P. koraiensis. After 21 months elapsed, remaining N, P, K, Ca, and Mg was 88.4, 77.6, 26.7, 50.5 and 44.5%, respectively, in decomposing P. rigida, and 114.4, 61.3, 7.6, 115.2 and 72.0%, respectively, decomposing P. koraiensis leaf litter.

Effects of Gibberellic Acid and Abscisic Acid on Proteolysis of Senescing Leaves from Rice Seedlings (노화 수도유묘엽의 단백질분해에 미치는 GA$_3$과 ABA의 영향)

  • The effect of gibberellic acid ($GA_3) and abscisic acid (ABA) on KCl-enhanced proteolysis of senescing leaves of rice(Oryza sativa L. cv. Chilsung) was studied. Emphasis was given to their effects on KCI-enhanced efflux of amino acids and proteinase activity. When treated singly, $GA_3 affected leaf proteolysis little, while ABA increased proteolysis, the rate of amino acid efflux, and ribulose -1,5 -bisphosphate carboxylase / oxygenase (Rubisco)-degrading endoproteinase activity. An additive increase in all three parameters mentioned above was observed when leaves were treated with ABA and KCl. No such an additive effect was found when $GA_3 was treated with KCl. Both $GA_3 and ABA helped to alleviate the KCI-suppressed activity of Rubisco-degrading exoproteinases. The additive increase in proteolysis of rice leaves in the presence of both ABA and KCl could thus be ascribed to a further increase in the efflux of protein hydrolyzates and Rubisco-degrading endoproteinase activity. An increase in proteolysis was accompanied by a decrease in water absorption, and the combined treatment of ABA with KCl resulted in a further reduction of water absorption.

Pesticide Degradation Activity of Several Isolates of Soil Bacteria and Their Identification (토양에서 분리한 수종 세균의 농약분해력 검정 및 동정)

  • Two bacteria were isolated from the continuously pesticide-used soil under plastic film house and upland condition. The degradation test of several pesticides by the selected bacteria, B59 and B71, were conducted. The degradation rates for 6 pesticides, procymidone, chlorothalonil, ethoprophos parathior, alachlor and pendimethalin, in medium by the isolates were 21.1% to 53.2% higher than non-inoculated medium. Under shaking culture condition, 90% to 95% of procymidone was degraded after 21 days treatment. Parathion was degraded in the range of 60% to 100% by B71 and B59, respectively. Otherwise 70% of alachlor was degraded by the two isolated bacteria during same period. The pH was not significantly affected for degradation of pesticides. The bacterial strains, B59 and B71 was identified as Acinetobacter sp. and as Pseudomonas sp. based on morphological, biochemical and physiological characteristics, and identity and similarity of automatic identification system, Biolog and MIDI.

preparation of Sauce from Enzymatic Hydrolysates of Cod Frame Protein (대구 Frame 단백질 가수분해물을 이용한 효소분해간장의 제조)

  • In order to utilize the protein source from a fish proessing by-product, cod was hydrolyzed with various enzymes such as tuna pyloric caeca crude enzyme (TPCCE), a-chymotrypsin, trypsin, papain and pronase E. The TPCCE hydrolysate acquired the highest sensory properties on taste, odor and color. The resultant cod rfame protein hydrolysate (CFPH) which was hydrolyzed with TPCCE, was separated through a series of ultrafiltration membranes with molecular weight cut-off (MWCO) of 30, 10, 5 and 1 kDa, and four types of permeates in cluding 30 K (permeate from 30 kDa membrane), 10 K (permeate from 10 kDa membrane), 5 K (permeate from 5 kDa membrane) and 1 K (permeate from 1 kDa membrane) were obtained. The natural sauces were prepared with 30 K, 10 K, 5 K and 1 K hydrolysate, and the sauce prepared with 1 K hydrolysate was the best score in sensory evaluations. In addition the mixed sauce prepared with 1 K hydrolysate and commercial soy sauce was similar to commercial sauce in sensory properties. These results suggest that the mixed sauce would be utilized as the substitute of acid-hydrolysis sauce.

Hygienic process of recycling salting solution of salted chinese cabbage (총설 - 절인배추의 위생적 절임수 재활용 처리기술)

  • 배추 절임 공정에서 위해 미생물로부터의 안전성 확보를 위한 위생적 세척 처리기술개발로 효율적인 재활용 시스템을 개발하고자 전기분해수를 활용하여 실험하였다. 전기 분해수의 농도는 100 ppm 으로 염수와 전해수의 비율을 1:9, 2:8, 5:5로 20회 반복 실험을 하였다. 그 결과 기존의 절임수 재활용 횟수인 3~5회를 훨씬 초과하여 10회 이상 반복하여도 총균수가 초기에 비해 2 log CFU/g 이상 감소된 결과를 나타내었다. 또한 재활용 처리된 염수의 pH나 산도 또한 초기에 비해 큰 차이를 보이지 않아 기존의 재활용 방법에 비해 효율적인 결과를 나타내었다. 조사되어진 바에 따르면 절임 공정에서 사용되어진 염수의 회수율은 70~80%에 달했으며, 절임공정에서 본 연구에서와 같이 전해수와 사용된 절임수가 1:9 또는 2:8 비율이 효율적일 것으로 판단된다. 재활용 처리되어진 염수로 절인 배추로 김치를 제조하여 관능평가를 한 결과, 기존의 방법으로 만들어진 김치와 큰 차이를 보이지 않아 본 재활용 시스템 방법은 기존의 시스템의 문제점으로 제기되었던 절인 배추의 성상 변화와 김치의 맛의 변화의 문제점을 해결할 수 있을 것으로 판단된다.

핵산분해효소에 관 연구 제 3보 새로운 Carrier에 의한 Phosphodiesterare의 고정화

  • 고분자 기질을 분해하는 효소의 고정화 후의 활성 yield을 높히고자 Polysaccharide의 표면에 acrylamide 및 N-hydroxysuccinimidyl acrylate을 graft 공중합 시킴으로써 긴 측쇄에 활성 ester을 가지는 새로운 carrier에 phosphiesterase을 고정화 시켰으며 이 고정화된 phosphodiesterase의 몇가지 성질을 조사하여 본래의 phosphiesterase의 성질과 비교하였다. 1) 이 Carrier에 phosphodiesterase을 직접 고정화시킨 결과 52%가 고정화되었으며 고정화된 효소는 38%의 비활성을 나타내 주었다. 2) phosphodiesterase는 이상과 같이 고정화됨으로써 최적작용 pH는 8.0부근에서 9.0부근으로 최적작용온도는 $50^{\circ}C부근에서$ $60^{\circ}C부근으로$ KM치는 1.11mg에서 2.1mg/ml로 변하였으며 열안정성은 훨씬 증가하여 본래의 phosphodiesterase의 열불활성속도상수가 0.4인데 고정화된 효소는 0.03 이었다. 또 $5^{\circ}C에서$ 수용액상태(pH 8.0 Tris buffer)로 6개월 보관후의 관존 활성은 본래의 효소가 0.2%인데 고정화된 효소는 42%로 높은 치를 나타내주었다.

  • III족 원료 가스로 triethylgallium(TEGa)과 trimethylindium(TMIn)을 사용하고 V족 원료 가스로 사전 열 분해하지 않은 arsine(AsH3)과 monoethylarsine(MEAs)을 사용하여 ultrahigh vacuum chemical vapor deposition(UHVCVD)법으로 Si3N4로 패턴된 GaAs(100)기판 위에 GaAs와 InGaAsqkr막을 선택적으로 에피택시 성장을 하였다. V족 원료 가스를 사전 열 분해하지 않으므로 넓은 성장 온도 구간과 V/lll 비율에서도 선택적으로 박막이 성장되었다. 또한 선택 에피택시의 성장 메카니즘을 규명하기 위하여 다양한 filling factor(전체면적중 opening된 면적의 비율)를 가지는 기판을 제작하여 성장에 사용하였다. UHVCVD법에서는 마스크에 면적중 opening된 면적의 비율)를 가지는 기판을 제작하여 성장에 사용하였다. UHVCVD법에서는 마스크에 입사된 분자 상태의 원료 기체가 탈착된 후 표면 이동이나 가스 상태의 확산과정 없이 마스크로부터 제거되므로 패턴의 크기와 모양에 따른 성장 속도의 변화나 조성의 변화가 없을 뿐만 아니라 chemical beam epitaxy(CBE)/metalorganic molecular beam epitaxy(MOMBE)법에서 알려진 한계 성장온도 이하에서 선택 에피택시 성장이 이루어졌다.

  • 본 논문에서는 다항식 H(x)로부터 H(x) = f(g(x))를 만족하는 두 개의 다항식 f(x), g(x)로 분해하는 알고리즘을 제시한다. 간단한 구조의 비선형 방정식 들을 풀면 이 문제를 해결할 수 있다. 일반적으로 비선형 방정식을 푸는 문제는 상당히 어렵다고 여겨지고 있지만, 이 문제에서는 변수들간의 점화식을 이용하여 정확하게 해를 구할 수 있다. 논문에서는 몇 가지 예제를 통해서 제시한 알고리즘의 효율성을 보이고, 기하 모델링 분야에서 중요한 응용에 대해 논하고자 한다.
