To investigate occurrence of root-knot nematode (RKN) in plastic house of central area of Korea, 132 soil samples were collected in cucumber, water melon, tomato, red pepper and strawberry fields from 2013 to 2015. Among 132 soil samples, 65 soil samples (49%) were infested with RKN and mean density of RKN was 178 second-stage juveniles per $100cm^3$ soil (min. 1 ~ max. 3,947). The frequency of RKN by regional was the highest in Chuncheon with 80%, followed by Cheonan (68%), Nonsan (36%), Buyeo (33%) and Yesan (30%). The frequency of RKN by crops was the highest in tomato with 83%, followed by cucumber (61%), strawberry (41%), red pepper (30%), watermelon (26%). To identify the species of RKN, fifteen populations were selected for representative populations. As a phylogenetic analysis of 15 populations, southern root-knot nematode (Meloidogyne incognita), peanut root-knot nematode (M. arenaria) and northern root-knot nematode (M. hapla) were identified with 47%, 20% and 33% ratio, respectively. In crops, M. incognita, M. arenaria and M. hapla were detected in tomato, M. incognita and M. arenaria were detected in cucumber and watermelon, and M. hapla was detected in strawberry and lettuce. Thus, there should be a continuous management to major species of each crops to prevent dispersal of RKN damages.
This study was conducted to analyse the distribution of root-knot nematodes (Meloidogyne spp.) in fruit vegetable production areas in Korea. Soil samples were collected from greenhouses in Sungju (Kyungpook), Yeoju (Kyungki), Haman (Kyungnam), and Chungwon (Choongpook) provinces in 1997-1999. Plant parasitic nematodes were separated for density counting and some of the root-knot nematodes were identified using enzyme phenotypes of malate dehydrogenase (MDH) and esterase (EST). Among the 185 farms in Sungju province, Meloiciogyne spp. were detected from 99 farms (53.5%). Other plant parasitic nematodes detected were; Helic~otylmchuss pp. from 7 farms, Aphelenchus spp. from 43 farms, and Criconematids from 26 farms. Using the female enzyme phenotypes of MDH and EST. the four major root-knot nematodes in Korea, M. incognitc~(M I), M. uretznri~(M~ A), M. huplu (MH), and M. juvunica, could be identified. In the enzyme phenotype identification of 13 populations collected from Sunnam in Sungju province, 6 populations were identified as MA, 5 populations were identified as MI, and 2 populations were mixed with MI and MA. Among the 6 populations from Chojun in Sungju province. 4 populations were MA, one population was MI, and one population showed enzyme phenotypes of unknown species. Among the 14 populations of Yeoju province, l I populations were MH and 3 populations were MA.
Effects of application method of eight nematicides on reproduction of iweloidogyne arenaria were tested in pot. In pre-treatment, cadusafos , dazomet, fosthiazate reduced over 90% in number of egg masses of M. arenaria at recommended rate . Carbofuran showed similar reduction only at three times of recommended rate. Benfuracarb was the least effective. Post-treatments, in general, were less effective than pre-treatment except fostiazate which reduced root egg mass of iW. arenaria as much as pre-treatment at recommended rate . Systematic nematicide, fosthiazate has potential as post-treatment nematicide.
Cadusafos and fosthiazate were applied at the rate of 1.8 g a.i./6$\m^2$ as pre-plant or pre-plant+post-plant treatments on winter-grown oriental melon in a greenhouse soil infested with Meloidogyne arenaria. Nematicides reduced root-knot nematode population from 35 to 90% compared with control; fosthiazate was better than cadusafos (P = 0.003) and fosthiazate pre-plant+post-plant application reduced nematode population densities as much as 90%. Nematicides increased yield in an average of 23% (11-38%) in May, 39% (2-65%) in June, and 31% (12-46%) for the total (P = 0.085). The residue in the fruit of oriental melon by post-plant treatment of fosthiazate exceeded maximum residue limit of 0.2ppm, while the Cadusafos residue was below the limit. It is concluded that fosthiazate cannot be used as a post-plant treatment in winer-grown oriental melon with overall consideration, i. e., danger of residue, value of melon, costs of nematicide, consumer demand on safer agricultural products, and effects of fallow.
This study was investigated to compare the suppression of Meloidogyne arenaria by different treatments of Pasteuria penetrans which is known for biological control agent against Meloidogyne spp.. In order to select proper number of P. penetrans showing good suppression effect, P. penetrans were mixed with M, arenaria for attachment using three different concentration such as $3{\times}10^4$, $3{\times}10^5$ and $3{\times}10^6$ endospores/5 g medium, followed by treating them onto the roots of tomato. After 14 weeks incubation, P. penetrans at $3{\times}10^6$ endospores showed highest activity against the formation of gall caused by M, arenaria. At a dose of $3{\times}10^5$ endospores/5 g medium, P. penetrans was treated into soil either mixing with soil or spray onto soil surface for comparing of suppressive efficacy. When the antagonistic bacterium was treated by the former method, it suppressed more effectively Using P. penetrans at $3{\times}10^6$ endospores and mixing with soil method, suppression was compared among P. penetrans, $PASTORIA^{(R)}$(Japan) and $Fosthiazate^{(R)}$(Korea). P. penetrans was more potent than $PASTORIA^{(R)}$(Japan) and as similar as $Fosthiazate^{(R)}$(Korea). Therefore, these results suggested that P. penetrans can be used for controling of M. arenaria as biological control agent. Furthermore, thess results can be provided to develop environmentally-friendly nematicide.

To select resistant vegetables against two species of root-knot nematodes, M. incognita and M. arenaria, 39 vegetables belongs to 7 families, 13 genera, 25 species were screened in greenhouse pot test. Susceptible vegetables to both nematodes were amarath and leaf beet in Amaranthaceae, Malabar spinach in Basellaceae, Moroheiya in Tiliaceae, and Water-convolvulus in Convolvulaceae, Pak-choi in Brassica campestris var. chinensis, Tah tasai in B. campestris var. narinosa, B. campestris var. chinensis x narinosa, Leaf mustard, Mustard green in B. juncea, Kyona in B. juncea var. laciniate, Choy sum in B. rapa subsp. arachinenesis, Kairan in B. oleracea var. alboglabra, Arugula in Eruca sativa, Garland chrysanthemum in Chrysanthemum coronarium, Endive in Cichorium endivia, Artichoke in Cynara cardunculus var. scolymus, Lettuce in Lactuca sativa. Resistant to M. arenaria but susceptible to M. incognita were B. oleracea cv. Matjjang kale, B. oleracea var. gongyloides cv. Jeok kohlrabi, and C. intybus cv. Radicchio. Resistant vegetables to both nematodes were C. intybus cv. Sugar loaf, Grumoro, Radichio treviso, B. oleracea cv. Manchu collard, Super matjjang, B. oleracea italica, B. oleracea var. botrytis italiana, and Perilla in Lamiaceae. Vegetables resistant to both species of root-knot nematodes could be used as high-valued rotation crops in greenhouses where root-knot nematodes are problem.
The effect of cultural, physical and chemical control methods on the population density of Meloidogyne arenaria second-stage juveniles (J2) and on fruit yields of oriental melon was investigated at Seongju Fruit Vegetable Experiments Station, Kyungpook province, Korea, for two years from 1999 to 2000. Crops used in a rotation prior to Oriental melon were rice, corn, sesame, and green onion. The physical methods used were either solarization, soil addition or soil drying, and a nematicide, fosthiazate of granular formula, was used as the chemical method, applying at a rate of 300g a. i./10 a. Growing rice in the rotation, solarization, and soil addition controlled the nematode disease most effectively, reducing the number of J2 by 90% and increasing fruit yields two times. However, the effects of these control methods on the J2 population were limited to the early growing season; the J2 population increased later, suggesting that additional control practices may be needed in the following season. The next effective control methods were use of corn in the rotation, the nematicide application, and soil drying. The nematicide application was effective only for the early fruit yield, but neither for the late nor for the total yields. Use of sesame or green onion in the rotation was not effective in controlling the nematode.
In this study, we analyzed the phylogenetic characterization of root-knot nematodes (Meloidogyne spp.) in soils from fruits and vegetables greenhouses in Korea. Soil samples were collected from 12 greenhouse fields in which tomato, cucumber, watermelon, and Oriental melon were being cultivated. Meloidogyne spp. were detected in all the soil samples at an average number of $72{\pm}6$ nematodes/300 g of soil to $2,898{\pm}468$ nematodes/300 g of soil. Phylogenetic analysis using polymerase chain reaction-restriction fragment length polymorphism was attempted for the second-stage juveniles (J2) of Meloidogyne spp. collected from the greenhouse soils. Twelve Meloidogyne spp. from the greenhouse soils were classified into two groups by using HinfI digestion of mitochondrial DNA, resulting in 900, 410, 290, and 170 bp fragments (group A) and 900, 700, and 170 bp fragments (group B). Phylogenetic analysis based on mitochondrial DNA sequences (1,483-1,521 bp) showed that nine group A isolates were identified as Meloidogyne incognita (99.73-99.93%) and three group B isolates showed 99.54-99.73% similarity to Meloidogyne arenaria.
Total of 36 sweet potato field soils were sampled to survey the occurrence of the root-knot nematodes (RKN). The 61% of sweet potato fields in Haenam, 40% in Iksan and 31% in Yeju were infested with RKN, respectively. Average population density of RKN was 324 juveniles per 300 g soil. The resistance screening of sweet potato cultivars against RKN was carried out by using clay pots in a greenhouse. Average temperature under ground 10 cm in pot was 21.5$^{\circ}C$ during the test. There was no difference in number of egg sacs among different inoculation methods, however the egg-inoculation method was easy for treatlnent and had stable far results. The multiplication ratio of Melioidogyne incognita differed from 6.3 times (Jeungmi) to 63.2 times (Yulmi) by sweet potato cultivas. There were no cultivars showing resistance to M. incognita, but Jinmi, Jeungmi and Borami had resistance to if arenaria, M. hapla and M. javanica.
Nineteen pepper (Capsicum annuum L.) cultivars were screened for the resistance to two common species of root-knot nematodes, Meloidogyne incognita and M. arenaria. The tested pepper cultivars showed different degrees of resistance to M. incognita but all were resistant to M. arenaria. The cultivars cv. Gonggongchil, Green No. 500 Pimento, Dahonggeon, Manitta, Shinbaram, Perfecto, and Hanmaeum were resistant to the both species of root-knot nematodes. Therefore, seven selected resistant pepper cultivars are recommended as resistant rotation crops in greenhouses where root-knot nematodes are problematic.
