Effect of the supplementation of Coconopsis lanceolata extract on lipid metabolism amelioration in type 2 diabetes mouse model induced by high fat diet (울릉도 섬더덕 추출물의 급여가 제2형 당뇨 동물의 지질대사 개선에 미치는 영향)

  • The present study was designed to investigate the antihyperlipidemic effect of Coconopsis lanceolata extract in C57BL/6J mice. The mice were divided into four groups: normal diet group (ND), high fat diet group (HFD), positive control group with 0.05% metformin (PC), Coconopsis lanceolata extract group (UCL). After 5 weeks of feeding, average body weight of the UCL group mice was slightly decreased, while that of the HFD group significantly increased) Also, liver and adipose tissue weights in the UCL group significantly increased. The levels of trigliceride (TG) and total in the plasma of UCL-supplemented group were significantly lower than those of high fat diet group. On the other hand, HDL-cholesterol level was increased. Expression level of proteins related with adipogenesis such as SREBP-1c, ACC, and FAS in the liver of the UCL group mice was much lower comparing with the HFD group mice. In conclusion, the results showed that the Coconopsis lanceolata extract possesses significant antihyperlipidemic effects in C57BL/6J mice.

Influence of Houttuynia cordata Thnub Powder Mixtures on Plasma Lipid Composition in Pigs (어성초 분말 혼합 식이가 돼지의 혈장 지질성분에 미치는 영향)

  • This study was designed to investigate the influence of Eosungcho (Houttuynia cordata Thunb) mixture on the plasma lipids of pigs. Experimental groups were divided into 3 groups: control group(0%), 5% and 10% Eosungcho feeding groups. Plasma concentration of total cholesterol, atherosclerotic index, free cholesterol and cholesteryl ester were significantly lower in the Eosungcho powder 5% and 10% feeding groups than control group. Plasma contents of HDL-cholesterol, cholesteryl ester ratio, triglyceride and phospholipid were significantly different between 10% Eosungcho feeding group and control group. Plasma concentration of LDL-cholesterol was significantly love in 10% Eosungcho feeding group($76.5{\pm}5.4mg/dL$) than in control group($86.5{\pm}5.6mg/dL$). Plasma concentration of VLDL and chylomicron were showed similar tendency to LDL-cholesterol. Also, blood sugar, GOT and GPT concentration were significantly lower in 10% Eosungcho feeding group than in control group. In control group, the ratio of saturated fatty acid to unsaturated fatty acid was 58.1 : 51.9, but in 5% and 10% Eosungcho feeding groups, those were 42.1 : 57.9 and 41.0 : 59.0, respectively. The component of main fatty acid was different between control group and Eosungcho feeding groups. In Eosungcho feeding groups, main unsaturated fatty acids were oleic acid(18:1) and linoleic acid(18:2). From the results, addition of Eosungcho powder might be effective in improving the plasma lipid components in pigs.

Effects of Capsosiphon fulvescens Extract on Lipid Metabolism in Rats Fed High Cholesterol Diet (매생이 추출물이 고콜레스테롤 식이를 급여한 흰쥐의 지질대사에 미치는 영향)

  • This study was performed to investigate the effects of Capsosiphon fulvescens (CF) extract on serum and liver lipid metabolism in rats. Male Sprague-Dawley rats were administered 1% cholesterol and 0.25% sodium cholate to induce hypercholesterolemia and were divided into six groups. Experimental groups were administered with following diets; normal diet group (NC), high cholesterol diet group (HC), normal diet and CF extract (200 mg/kg) administered group (NC-CFL), normal diet and CF extract (400 mg/kg) administered group (NC-CFH), high cholesterol diet and CF extract (200 mg/kg) adwlinistered group (HC-CFL), and high cholesterol diet and CF extract (400 mg/kg) administered group (HC-CFH). The body weight gain and feed efficiency ratio were increased by high cholesterol diet, but gradually decreased to the corresponding level of the normal diet group by administered CF extract. The serum total cholesterol concentration of normal diet group was significantly higher than those of NC-CFL group and NC-CFH group. The concentrations of serum total cholesterol LDL-cholesterol and triglyceride and the atherogenic index .were tended to decrease in the CF extract administered groups compared with the high cholesterol diet group. However, HDL-cholesterol and phospholipid concentrations in serum decreased in the high cholesterol diet group and markedly increased by the CF extract administered groups. There were no differences in the concentrations of serum triglyceride, phopholipid, LDL-cholesterol, HDL-cholesterol and free cholesterol in normal diet groups. Concentrations of total cholesterol and triglyceride in liver and kidney were also markedly decreased in the CF administered groups. Total cholesterol and triglyceride concentrations of adipose tissue in the CF administered groups were also decreased compared with the high cholesterol diet group. In addition, there were no differences in the concentrations of liver, kidney and adipose tissue total lipid, triglyceride and total cholesterol in normal diet groups. The activities of heparin-releasable lipoprotein lipase (LPL) and total-extractable LPL in adipose tissue were increased in the high cholesterol diet group compared with the CF extract administered groups, but those of the NC -CFL group and NC-CFH group were similar to the normal diet group. Taken together, it is suggested that the extract of CF have an antiatherosclerotic effect by reducing cholesterol level of serum and liver. Furthermore, the extract of CF decreased LPL activity and triglyceride concentration, resulting in less lipid storage.

Effect of Fermented Water Extracts from Ligularia fischeri on Hepatotoxicity Induced by D-Galactosamine in Rats (D-Galactosamine 투여 랫트에서 곰취 열수 추출 발효물이 간 독성 저하에 미치는 영향)

  • This study was conducted to determine the effect of fermented water extracts from Ligularia fischeri (LAF) on reduction of hepatotoxicity induced by D-galactosamine (D-GalN) in rats. In this experiment, male Sprague-Dawley rats were used as experimental animals, which were divided into eight groups: normal group, D-GalN-treated group (control), D-GalN and non-fermented water extracts from Ligularia fischeri (LA)-treated groups [100, 200, and 400 mg/kg BW (body weight)], and D-GalN and LAF-treated groups (100, 200, and 400 mg/kg BW). ${\gamma}$-Glutamyl transferase, aspartate aminotransferase, alanine aminotransferase, and lactate dehydrogenase activities in serum of the D-GalN and LAF-treated groups decreased significantly compared to those of the control group (P<0.05). The high density lipoprotein-cholesterol levels of the D-GalN and LAF-treated groups increased significantly compared to those of the control group (P<0.05). The low density lipoprotein-cholesterol and triglyceride levels of the D-GalN and LAF-treated groups decreased significantly compared to those of the control group (P<0.05). The atherogenic index values of the D-GalN and LAF-treated groups decreased significantly compared to those of the control group (P<0.05), and their high density lipoprotein cholesterol by total cholesterol ratio increased significantly in these groups (P<0.05). Superoxide dismutase activity of liver tissues were enhanced significantly (P<0.05) in the D-GalN and LAF-treated groups compared to that of the control group (P<0.05), whereas their malondialdehyde content decreased significantly in these groups (P<0.05). The histopathological observations revealed apoptotic cells and mild portal inflammation in liver tissues of the D-GalN and LAF-treated groups. Taken together, these results demonstrate that LAF may improve plasma lipid profile and alleviate hepatic damage.

Relationship between HsCRP and Pulse Transit Time (HsCRP와 맥파전달시간에 대한 연구)

  • The purpose of this study is to evaluate the relationship between high sensitive C-reactive protein (hsCRP) and pulse transit time (PPT). Apparently healthy 233 subjects had been enrolled in the health promotion center of the Pusan National University Hospital from Jan. 29 to Feb. 26, 2004. They had no previous history of diabetes, hypertension and hyperlipidemia. Subjects were categorized according to tertiles of hsCRP level [Group 1: first tertile $(0.01\;{\sim}\;0.02\;mg/dl)$, Group 2: second tertile $(0.03\;{\sim}\;0.05\;mg/dl)$, Group 3: third tertile $(0.06\;{\sim}\;0.12\;mg/dl)$, and Group 4: Fourth tertile $(0.13\;{\sim}\;16.8\;mg/dl)$]. PTT body mass index (BMI), total cholesterol (T-C), LDL-cholesterol(LDL-C), blood sugar (BS), systolic blood pressure (sBP) and diastolic blood pressure (dBP) were significantly different among hsCRP groups (p<0.05). HsCRP is positively related with BMI, tryglyceride (TG), LDL, sBP and dBP (p<0.05), and negatively related with PTT and HDL-cholesterol (HDL-C) (p<0.05). PTT is significantly negatively related with hsCRP, T-C, TG, LDL-C, BS, dBP and sBP (p<0.05). The hsCRP and PTT were related before controlling BMI, T-C, LDL-C, sBP, and dBP, but not related after conkolling. The relationship between hsCRP and PTT depends on cardiovascular disease risk factors.

  • Purpose : Macrovascular complications are the main cause of mortality in type 1 diabetes mellitus (T1DM). The purpose of this study was to clarify the presence of early vascular changes and to assess the risk factors of macrovascular complications in young adults with T1DM diagnosed in childhood and adolescence. Methods : Seventy-two patients ($23.9{\pm}2.4$ years) with T1DM diagnosed before 18 years of age and twenty normal controls were included. The incidence of hypertension, dyslipidemia, and other risk factors of macrovascular complication were reviewed. Flow-mediated vasodilation (FMD) and mean intima-media thickness (IMT) measured by ultrasound were compared between patients and control subjects, and their correlations with macrovascular risk factors were analyzed. Results : Of the 72 patients, 32 (44.4%) had hypertension. The proportions of maleness (P=0.03) and mean body mass index (P=0.04) were higher in the hypertensive patients than in normotensive patients. Thirty-one (N=69, 44.9%) patients had dyslipidemia and LDL-cholesterol was positively correlated with mean HbA1c (r=0.32, P=0.008) and total daily insulin dose (r=0.27, P=0.02). The mean IMT was significantly higher in patients than in control subjects ($0.43{\pm}0.06$ mm vs $0.39{\pm}0.06$ mm, P=0.03). There was no difference in the value of FMD between patients and controls, but the duration of the disease after pubertal onset was negatively correlated with FMD (r=-0.34, P=0.01). Conclusion : Hypertension, dyslipidemia and atherosclerotic vascular change were observed in young adults with T1DM diagnosed during childhood and adolescence; this strongly suggests that meticulous screening of macrovascular complications and control of their risk factors should be conducted.

Effect of Monascus purpureus-Fermented Korean Red Ginseng Powder on the Serum Lipid Levels and Antioxidative Activity in Rats (홍국발효홍삼 분말 급여에 의한 흰쥐의 혈청 지질 농도 및 항산화 활성에 미치는 영향)

  • Monascus-fermented Korean red ginseng (MFRG), fermented by Monascus purpureus KCCM 12002, may be an ideal candidate for the hepatoprotectic, hypolipidemic and antioxidative activities. Effect of MFRG powder on these parameters in rats was investigated. Body weight gain, food intake, and water consumption were not significantly different among the groups. Total and relative weights of liver were significantly higher in MFRG group than that in other groups. The activities of AST and $\gamma$-GTP were highly lowered in MFRG group compared to control group. Contents of serum total lipid and triglyceride were significantly lowered in silymarin group and were significantly increased in MFRG group compared to control group, but tended to be lowered in RG group. Serum content of total cholesterol tended to be lowered in silymarin, RG, and MFRG groups compared to control group. HDL-cholesterol contents was only significantly increased in MFRG group compared to control group. At the same time, atherogenic index (AI) was also significantly lowered in silymarin, RG and MFRG groups compared to control group, and this effect was more pronounced in MFRG group. Content of thiobarbituric acid reactive substances (TBARS) in the liver was significantly lowered in MFRG group and tended to lowered in silymarin and RG groups compared to control group. The hepatic glutathione concentration was significantly higher in silymarin and MFRG groups. Hepatic morphology in all experimental groups revealed clear-cut hepatic lobules with the uniform pattern of the polyhedral hepatocytes radiating towards the periphery from the central vein. These results suggested that MFRG may have anti-atherogenic index (AI) and antioxidative activity in normal dietary feeding rats.

Effects of Sasa coreana Nakai on the Lipid Compositions of Serum in High Cholesterol Diet Rats (신의대(Sasa coreana Nakai) 추출물이 고콜레스테롤 식이 투여 흰쥐의 혈청 지질에 미치는 영향)

  • This study was performed to investigate the effects of Sineui-Dae (Sasa coreana Nakai, a kind of bamboo) extracts on the improvement of serum lipid composition by using rats fed a hypercholesterol diet for 4 weeks. The experiment animals were administered with the following diets; high cholesterol diet group (HC diet) as a control and three supplemented groups with high cholesterol diets (HCW, HCM or HCH diet). Three kinds of extracts were prepared by orderly extraction with hexane, methanol, and water. We measured free cholesterol, cholesteryl ester, total cholesterol LDL- and HDL-cholesterol triglyceride and phospholipid in the serum of rats in three experimental and control groups. There were no significant differences in body weights and feed intakes between the HC control group and the extract supplemented groups. The levels of total cholesterol and LDL-cholesterol in the serum of extract supplemented groups were lower than that of the group fed with HC diet only. Serum HDL-cholesterol levet which is known as an antiatherosclerosis factor, was higher in all groups supplemented with bamboo extract by 17.2-21.9% compared to the HC control group. All groups supplemented with bamboo extracts showed the lowering effect of atherogenic index compared to the HC control group (HC group: 2.96${\pm}$0.08, HCM group: 1.48${\pm}$0.02, HCW group: 1.69${\pm}$0.04, HCH group: 1.84${\pm}$0.01). Furthermore, serum triglyceride and phospholipid decreased significantly in the HCM diet compared to the HC control diet. These results suggest that Sineui-Dae bamboo extract, especially methanol extract, has improving effects on hyperlipidemia of rats fed a high cholesterol diet.

Effect of Brassica rapa Sprouts on Lipid Metabolism in Rats Fed High Fat Diet (순무 싹이 고지방식이를 급여한 흰쥐의 지질대사에 미치는 영향)

  • This study was performed to investigate the effect of Brassica rapa (BR) sprouts on weight reduction and cholesterol-lowering action in rats fed high fat diet for 4 weeks. Weight-matched male Sprague-Dawley rats were assigned to four groups according to dietary fat levels (10% or 20% of diet wt.). Experimental groups were normal diet group (N), high fat diet group (HF), high fat diet with 5% BR sprouts powder group (HF-BRL), and high fat diet with 10% BR sprouts powder group (HF-BRH). The body weight gain was increased in HF group, but gradually decreased to the corresponding level of the N group fed BR sprouts powder. The concentrations of serum LDL-cholesterol, atherogenic index and cardiac risk factor tended to decrease in the BR sprouts powder fed groups compared with the HF group. However, HDL-cholesterol concentration in serum decreased in the HF group and markedly increased in the BR sprouts powder fed groups. Concentrations of triglyceride and total cholesterol in liver were also markedly decreased in the BR sprouts powder fed groups. Triglyceride concentrations of epididymal and mesenteric adipose tissues in the BR sprouts powder fed groups were also decreased compared with the HF group. These results indicate that BR sprouts powder may reduce fat accumulation and body weight, and have cholesterol-lowering effect.

Effects of Ramie Leaves on Improvement of Lipid Metabolism and Antiobesity Effect in Rats Fed a High Fat/High Cholesterol Diet (모시잎이 고지방-고콜레스테롤 식이 흰쥐의 지질대사 개선 및 항비만 효과에 미치는 영향)

  • This study was performed to investigate the effects of ramie (Boehmeria nivea) leaf powder on improvements in lipid metabolism in serum, liver and adipose tissue, and the anti-obesity effect in rats fed a high fat/high cholesterol diet for 4 weeks to induce a hyperlipidemic and obese model rat. Male Sprague-Dawley rats weighting 210 g were divided into four groups; a normal diet group (N), a high fat/high cholesterol diet group (HFC), a high fat/high cholesterol diet with 5% ramie leaf powder group (HFC-RL), and a high fat/high cholesterol diet with 10% ramie leaf powder group (HFC-RH). The body weight gain increased with a high fat/high cholesterol diet, but gradually decreased in the ramie leaf powder fed groups compared with the HFC group. The liver index in the HFC group was highest among the four groups, although the difference was not significant compared with the ramie leaf powder fed groups. The adipose tissues weight in the HFC group was heavier than that of the N group, whereas those of groups fed ramie leaf powder decreased gradually. Alkaline phosphatase activity was not different between the HFC groups, but serum alkaline aminotransferase and lactate dehydrogenase activities decreased significantly after ramie leaf powder feeding. Levels of serum triglyceride, total cholesterol, LDL-cholesterol, atherogenic index and cardiac risk factors tended to decrease in the ramie leaf powder fed groups compared with the HFC group, whereas serum HDL-cholesterol level decreased in the HFC group and markedly increased in the ramie leaf powder fed groups. Levels of triglyceride and total cholesterol in the liver were significantly lower in the ramie leaf powder fed groups than in the HFC group. Levels of triglyceride and total cholesterol in adipose tissues were also lower in the ramie leaves powder fed groups than in the HFC group. The activities of heparinreleasable lipoprotein lipase (LPL) and total-extractable LPL in adipose tissues increased in the HFC group compared to that of the N group, but those of the ramie leaf powder fed groups decreased significantly. These results suggest that ramie leaf powder may improve lipid metabolism in serum, liver and adipose tissue and potentially reduce lipid storage.
