This experiment was performed to improve the stability of Lactobacillus fermentum YL-3 as a poultry probiotics. The culture conditions that improve acid tolerance of L. fermentum YL-3 were investigated by changing several factors such as medium composition, temperature, anaerobic incubation and culture time. Also, L. fermentum YL-3 was encapsulated with alginate, calcium chloride and chitosan. The stable culture conditions of L. fermentum YL-3 were obtained in anaerobic incubation using MRS media without tween 80 for 20 hour at $42^{\circ}C$. The capsule after treatment with 1% chitosan was formed close membrane by a bridge bond. Immobilization of L. fermentum YL-3 in capsule was observed by confocal laser scanning microscopy, and cell viability was $2.0{\times}10^9\;CFU/g$ above the average. L. fermentum YL-3 capsule after acid treated at pH 2.0 for 3 hour survived about 40%, but those encapsulated with 1% chitosan survived about 65%. Survival rate of capsule stored at room temperature decreased about $2{\sim}3$ log cycle during 3 weeks, but viability of capsule stored at $4^{\circ}C$ during 3 weeks maintained almost $10^8\;CFU/g$ levels.

This study was carried out to investigate the optimal conditions for meat paste production with sardine. To improve the gel forming ability of meat paste, washing time and condition with alkaline solution, setting time and temperature, and heating temperature before pasteurization were controlled, and the influences of the freshness of raw sardine and the mixing ratios of ordinary and dark muscles on the duality of the meat paste product were discussed. The frozen storage showed a predominant effect on keeping freshness of raw sardine at different storage conditions and gel forming ability was maintained for 1 day at ice storage, for 3 days at $-3^{\circ}C$ and for 4 days at frozen condition, but there was no effect on keeping freshness of raw sardine in the storage at $25^{\circ}C$. Gel strength of meat paste product tended to decrease with washing time of raw meat, and in case of washing 3 times the meat appeared excellent in gel strength, but in case of seven and nine times the meat showed lower water holding capacity and decreased organoleptic test score in the quality of meat paste prtoduct. Raw meat washed with alkaline solution showed a desirable effect on gel forming ability compared with that washed with tap water, and in the case of washed with $0.5\%$ sodium bicarbonate solution exhibited the most favorable effect on gel forming. The gel strength of the meat paste product decreased with the increase of mixing ratios of dark muscle in the raw meat. Setting time and temperature for the gel forming ability of meat paste were good at $5^{\circ}C$ for 20 hours and at $20^{\circ}C$ for 2 hours. In the heating temperature of meat paste, heating treatment at $90^{\circ}C$ was desirable for gel forming.
Distribution of marine algae is diverse in Korea and the resource of edible algae is abundant marking 239,037 tons of yearly production in 1976. They have been known as a protein source and used as a supplement in Korean diet. It is necessary to estimate the potentiality and properties of usable algal proteins especially as food resources and studies of extraction and separation of the proteins, therefore, are basically required for this purpose. In this study, the influence of various factors including the sample treatment, extraction time and temperature, sample us extraction solvent ratio and pH upon the extractability of the water soluble protein was determined. And the effect of precipitation treatment for isolation of the algal protein from the extracts was also tested. Nine species of algae, the major ones in consumption as food namely Porphyra suborbiculata, Undaria pinnatifida, Hizikia fusiforme, Sargassum fulvellu, Enteromorpha linza, Codium fragile, Sargassum kjellmanianum and Ulva pertusa were collected as fresh from Kijang, Yangsan Gun, in the vicinity of Busan city. The content of crude protein $(N\times6.25)$ of the algae ranged from $9.46\%\;to\;24.14\% showing the highest value in Porphyra suborbiculata and the minimum in Hizikia fusiforme. In the effort of maceration of blending methods on the extractability, immersion freezing in dry ice-methanol solution appeared most effective yielding 1.5 to 2.5 times extractability than that of the mortar grinding method. The effect of the ratio of sample vs solvent on extractability differed from species. It was enhanced at the ratio of 1:20 (w/v) in Ulva pertusa and Enteromorpha linza while the ratio was 1:30 (w/v) for Cedium fragile, Undaria pinnatifida, Hizikia fusiferme, Sargassum fulvellum and Porphyra suborbiculata and 1:40 for Sargassum kjellmanianum respectively. The effect of extraction time and temperature was revealed differently from species which might be caused by differences in the constitution of algal tissues resulting in that the extraction for 1 hour at $50^{\circ}C$ gave the maximum extractabilily in Ulva pertusa and Enteromorpha linza, 2 hours in Porphyra suborbiculata, Hikikia fusiforme, Undaria pinnatifida, Sargassum kjellmanianum and 3 hours in Codium fragile. And the extractability was higher at $50^{\circ}C$ to $60^{\circ}C$ for the most of the tested samples except Hizikia fusiforme. The optimum pH for the extraction was 9 to 12. The recovery of extractable nitrogen to the total nitrogen was $63\%$ in average with the first extracts and $8.6\%$ with the second extracts respectively. Both extracts were prepared by 2 hour extraction at $50{\pm}1^{\circ}C$ with dry ice-methanol frozen and seasand macerated materials. And these conditions assumed to be an optimum for the extraction of water soluble algal proteins since the nitrogen content after the first extraction covered $90\%$ of the total water extractable nitrogen. In the precipitation of the extracted proteins, Barnstein method and methanol treatment seemed to be more efficient than other precipitation methods.

Two dominant thrips in hot pepper (Capsicum annuum) cultivating in greenhouses are Frankliniella occidentalis and F. intonsa in Korea. This study investigated their overwintering physiology. These two thrips were freeze-susceptible and suppressed the body freezing temperature by lowering supercooling point (SCP) down to -15~-27℃. However, these SCPs varied among species and developmental stages. SCPs of F. occidentalis were -25.7±0.5℃ for adults, -17.2±0.3℃ for pupae, and -15.0±0.4℃ for larvae. SCPs of F. intonsa were -24.0±1.0℃ for adults, -27.0±0.5℃ for pupae, -17.2±0.8℃ for larvae. Cold injuries of both species occurred at low temperature treatments above SCPs. Thrips mortality increased as the treatment temperature decreased and its exposure period increased. F. occidentalis exhibited higher cold tolerance than F. intonsa. In both species, adults were more cold-tolerant than larvae. Two thrips species exhibited a rapid cold hardening because a pre-exposure to 0℃ for 2 h significantly enhanced the cold tolerance to a lethal cold temperature treatment at -10℃ for 2 h. In addition, a sequential exposure of the thrips to decreasing temperatures made them to be acclimated to low temperatures. To investigate the overwintering sites of the two species, winter monitoring of the thrips was performed at the greenhouses. During winter season (November~February), adults of the two species were not captured in outside of the greenhouses. However, F. occidentalis adults were captured to the traps and observed in weeds within the greenhouses. F. occidentalis adults were also emerged from soil samples obtained from the greenhouses during the winter season. F. intonsa adults did not come out from the soil samples at November and December, but emerged from the soil samples obtained after January. To determine the adult emergence due to diapause development, two thrips species were reared under different photoperiods. Adult development occurred in all photoperiod treatments in F. occidentalis, but did not in F. intonsa especially under short periods. Tomato spotted wilt virus, which is transmitted by these two species, was detected in the weeds infested by the thrips during the winter season. These results suggest that F. occidentalis develops on weeds in the greenhouses while F. intonsa undergoes a diapause in the soil during winter.

In this study, the extraction of Conger myriaster oil by using supercritical carbon dioxide (SC-CO2) and organic solvent was investigated. The extraction conditions conducted for SC-CO2 varied for pressure (25, 30 MPa) and temperature (45, 55 ℃), while the SC-CO2 flow rate was kept constant during the experiment (27 g min-1) and hexane was used as a conventional organic solvent. The extraction yield indicated that the best extraction condition would be SC-CO2 at 55 ℃ and 30 MPa, resulting in the highest yield of 37.73 ± 0.14%. The oils were characterized for their fatty acid (FAs) composition using gas chromatography, while it was revealed that the major FAs were mystric acid, palmitoleic acid, oleic acid, electroosapentaenoic acid (EPA), and docosahexaenoic acid (DHA). The oxidation stability of the extracted C. myriaster oil was evaluated by measuring the acid value, peroxide value, and free fatty acid. The best oxidative stability was obtained from SC-CO2 extracted oil at 30 MPa and 55 ℃. There was a significant difference in the color properties of the SC-CO2 and hexane extracted oils, with the SC-CO2 extracted oil showing better chromaticity than the oil extracted using hexane. Extracting oils from C. myriaster with SC-CO2 could bring better economic benefits than using organic solvents. When supercritical carbon dioxide was used, there was no post-treatment process; thus, it was confirmed that this is a more environmentally friendly oil extraction method.
The thickness of anti-freezing layer has been empirically determined using the frost depth obtained from the freezing index and has not been generally considered as a structural layer in pavement design procedure. In fact, the anti-freezing layer makes a role in structural layer and enables to reduce the total thickness of pavement system. The objective of this study is to develop the statistical regression model for evaluating the structural capacity of anti-freezing layer using Falling Weight Deflectormeter(FWD) test data in asphalt pavements. The FWD testing was conducted at the embankment, cutting, and boundary area of various test sections to estimate the structural capacity of anti-freezing layer in different foundation condition. It is observed from this testing that the center deflections of pavement structure with anti-freezing layer are smaller than those without anti-freezing layer ranging from 0.4 to 82.6%. To determine the variables of statistical model, the correlation study has been conducted between various FWD deflection indexes and the anti-freezing layer thickness. It is found that the ${\Delta}BDI$(%)(${\Delta}Basin$ Damage Index(%)) is highly correlated with anti-freezing layer thickness. The ${\Delta}BDI$(%) model were developed for evaluating structural capacity of anti-freezing layer using linear mixed-effect models.
Anchovy oil was extracted using supercritical carbon dioxide ($SCO_2$) and organic solvents. Extraction was carried out at temperature range from 40 to $60^{\circ}C$, and pressure range from 15 to 25 MPa. The flow rate of $CO_2$ (22 $gmin^{-1}$) was constant entire the extraction period of 1.5 h. The fatty acid composition of anchovy oil was analyzed by gas chromatography (GC). The main fatty acids of anchovy oil were myristic acid, palmitic acid, stearic acid, palmitoleic acid, EPA (eicosapentaenoic acid), and DHA (docosahexaenoic acid). In addition, the oil obtained by $SCO_2$ extraction contained a higher percentage of polyunsaturated fatty acids especially EPA and DHA comparing to the organic solvent extracted oil. The oxidative stability of oils extracted from Anchovy by $SCO_2$ extraction was compared to those extracted by organic solvents. Results showed that the storage periods of oils obtained by $SCO_2$ extraction were longer than those of organic solvents extraction.
This study was performed to investigate the inhibitory effect of the water-extract from fermented compositions containing Inonotus obliquus with added Houttuynia cordata on the growth of either human AGS gastric and HCT-15 colon cancer cells or NIH3T3 normal mouse fibroblast cells. Cytotoxic activity on cancer cells was investigated by viable cell count, MTT assay and morphological observation. Mixtures of Inonotus obliquus with added Houttuynia cordata were fermented at $30{\sim}37^{\circ}C$, $50{\sim}60%$ humidity for 30 days, extracted with water, freeze dried, powered, and then dissolved in water for the experiment. In MTT assay, the fermented compositions exhibited inhibitory effects of 13, 25, 40, 67 and 78% for AGS and 22, 40, 50, 69 and 76% for HCT-15 at 0.16, 0.4, 0.8, 1.6 and 4.0 mg/ml, respectively. However, normal NIH3T3 cells were exhibited 86% survival under the same experimental condition. Fermented compositions showed highly inhibitory effect against human cancer cell line HCT-15 and AGS, but not on normal cell line NIH3T3.

An investigation was carried out to study the effects of extraction, heat treatment, and storage temperature on functional ingredients of onion. Extracts of onion paste and freeze dried onion were prepared by treatments with several solvents including hot water, methanol, and ethanol, and then contents of functional ingredients such as total phenol, flavonoid, quercetin, and thiosulfinate were determined. The highest levels of functional ingredients were observed in samples extracted using water at 100$^{\circ}C$ or ethanol. The skin extract contained 60-fold more quercetin than onion paste extract, whereas no thiosulfinate was detected in onion skin extract. Heat treatment of onion extracts at 60, 80, 105, and 121$^{\circ}C$ did not effect on the levels of all functional ingredients tested. During four-week storage, the levels of total phenol, flavonoid, and quercetin were not significantly changed regardless of storage temperature. However, the total thiosulfinate was rapidly reduced as the storage temperature increased.
Cordyceps and its allies fungi has been described as a secret medicine that gives eternal youth and a long life. Some species of Cordyceps are sources of biochemicals, such ascordycepin, with interesting biological and pharmacological properties. Hence, it has been studied to uncover its pharmacological effect. We attempted to study the formation of fruiting bodies and to develop means of mass production Korean isolate of Paecilomyces tenuipes has been inoculated into silkworms, where it reproduced using culture container, mini-kit successfully. Culture container, mini-kit is composed of a cylinder-shaped body and lid. The container is made of translucent polyethylene terephthalate. The size of the container is $82{\times}75mm$, reduced by 10 times as compared with the conventional culture kit. The mini kit has many advantages - high culture amount, ability of maintaining optimal humidity, parasite-free cultivation and high-end appearance. With the kit, the optimal cultivation condition is under $22^{\circ}C$, culture period of 53 days. And synnemata of P. tenuipes could be kept fresh for 14 days at the temperature of under $10^{\circ}C$. Therefore, the Min-kit can be used in both ways as a culture container and a packing kit for end-user customers.
