Microbial Control of the Tobacco Cutworm, Spodoptera litura (Fab.), Using S. litura Naclear Polyhedrosis Virus. I.   • A nuclear polyhedrosis virus (NPV) of the tobacco cutworm, Spodoptera litura would be a promisible agent for the control of the insect. To develop a viral insecticide using S. litura NPV, effect of spray on soybean leaves, temperature, storage, an sunlight on the pathogenicity of the virus were studies as follows: Median lethal concentration ($LC_{50}$) of the virus sprayed on the leaves against the third and the fifth instar larvae were $1.301\times10^{4 PIBS}/ml$ and $1.087\times10^{5 PIBS}/ml$, respectively. On the concentration of $1.0\times10^{5 PIBS}/ml$, median lethal times ($LT_{50}$) were 7.3 days for the 3rd and 8.9 days for the 5th instar larvae. Stability of S. litura NPV was quickly decreased at the higher temperate than $60^{\circ}C$ and at the longer exposure to the higher temperature. Storage of the virus at $-20^{\circ}C$ was kept higher pathogenicity than $4^{\circ}C$ and $25^{\circ}C$. Viral activity was maintained more than 10 days in the sprayed-under leaves, but decreased at 3 day after spray in th sprayed-on the leaf surface when exposed the virus to sunlight.

  • 본 연구는 이식 후 남은 잉여의 포배기 배아를 두 번의 냉동과 융해 과정을 반복적으로 실시한 후 이식한 결과에 관한 보고이다. 사람 포배기 배아의 동결보존에서 높은 생존율과 성공적인 임신율이 보고되고 있으나 미성숙 난자로부터 발달한 포배기 배아에 두 번의 초급속 냉동 방법을 실시한 후 이식한 보고는 되어 있지 않다. 이에 본 연구에서는 다낭성 난소 증후군 환자에게서 얻은 미성숙 난자로부터 발달한 포배기 배아를 artificial shrinkage 후 초급속 냉동함으로써 생존율을 높이는 방법을 이용하여 재냉동 이식하였을 때 임신에 성공한 증례를 보고하고자 한다. 29세의 환자로부터 채취한 55개의 미성숙 난자들(germinal vesicle stage oocytes)을 체외배양 하여 성숙한 37개의 난자들로부터 30개의 수정란을 얻을 수 있었다. 12개의 배아가 포배기 배아까지 발달하였으며 이 중 3개의 양질의 포배기 배아를 선별하여 이식하였고, 이식을 한 후에 남은 9개의 포배기 배아들은 artificial shrinkage의 과정을 마친 후에 초급속 냉동 방법을 이용하여 동결보존 하였다. 그 중, 4개의 포배기 배아들을 융해한 후 이식을 하지 않고 다시 재냉동을 하여 보관하였고 이 후 재냉동 되었던 4개의 포배기 배아들을 다시 융해 하여 이식을 한 결과 임신이 되어 건강한 남아를 분만하였다. 이로써 미성숙 난자로부터 얻은 포배기 배아가 두 번의 냉동과 융해의 과정을 통해 크게 손상을 입지 않고 생존할 수 있다는 것을 알 수 있었다. 그러므로 융해이식 후 남은 잉여의 포배기 배아를 다시 냉동 보관하여 다음 주기에 이용함으로써 축적된 임신율을 증가시킬 수 있을 것으로 사료된다.

Studies on the Culture Media and the Optimal Storage Conditions of Bioluminescent Bacteria Photobacterium phosphoreum (생체발광균주 Photobacterium phosphoreum의 배양배지 및 최적 저장조건에 관한 연구)

  • Vibrio, Photobacterium, Alteromonas and Xenorhabdus species are capable of emitting light, called bioluminescence. They exist in marine, freshwater and terrestrial environments. Bacterial bioluminescent reaction is that reduced riboflavin phosphates and a long-chain aldehyde are oxidized in the presence of molecular oxygen and enzyme luciferase. This experiment aims to develop the proper culture media and to optimize the storage condition for the recovery of bioluminescent activity in Photobacterium phosphoreum. The Luria broth (LB) medium was modified for cultivation of Photobacterium phophoreum, called as modified LB(mLB) medium. The mLB medium is LB fortified with 3% glycerol and 1.5% NaCl. In mLB medium. bacterial growth and bioluminescent activity are 25% higher than those in a Nutrient broth medium. When the cell stocks were stored at $-20^{\circ}C$, $-70^{\circ}C$ and LN2 for 3 months, cell growth and bioluminescent activity of culture after stored at $-20^{\circ}C$ were better than those of other treatments. The highest bioluminescent activity obtained at the late exponential phase in all treatments. When the cell stock was freeze-dried with 5% adonitol as a cryoprotectant, the recovery of cell was better than those of control and freeze-dried cell stock without addition of cryoprotectant.

Evaluation of Liquefaction Resistance of Sandy Soil Layer in Songlim-park, Pohang, Using Undisturbed Sample (불교란시료를 활용한 포항시 송림공원 사질토층의 액상화저항강도 평가)

  • In this study, a set of laboratory liquefaction test has been conducted using undisturbed samples collected from Songlim-park, Pohang where liquefaction phenomenon had been observed in 2017. Soil samples were frozen right after tube sampling to minimize the disturbances during transport, storage, and test preparation. Cyclic triaxial test has been carried out to evaluate the liquefaction resistance ratio of undisturbed soil samples. As a result, the liquefaction resistance ratio of samples collected from 8.0~8.8 m and 11.0~11.8 m were almost similar, and these values were approximately 0.04~0.07 larger than values estimated by simplified assessment method using field test results. It is expected that the application of undisturbed sample for the evaluation of liquefaction resistance could contribute to the economical design of geotechnical structures.

Conservation of Bamboo Dining Tables excavated from Taean Mado Shipwreck No. 1 (태안 마도1호선에서 출수된 대나무 소반의 보존처리)

  • The small dining tables which are made of bamboo were deteriorated condition when those were excavated from Mado shipwreck No.1. Therefore, preliminary test was conducted for consolidation (Sucrose, Dammar gum, Cetyl alcohol and Polyethylene glycol 4000) then impregnated and vacuum freeze dried. After the test, since the treatment with vacuum freeze drying using an aqueous solution of PEG 4000 keep the shape well the most and present bright colour, waterlogged archaeological bamboo conserved with vacuum freeze drying using an aqueous solution of PEG 4000. One bamboo table consolidated with soluble PEG 4000 from 10% to 40%, then vacuum freeze dried. Another impregnated with soluble PEG 4000 from 10% to 70%. After that, it was separated and held in polycarbonate sheet for preventing bending during dry. The small dining tables are the three dimensional shapes and have some loss. Therefore the mounting for support tables were necessary which are made of acrylic sheet. It keep the table shape and make it convenient for storage and handling.

Compressive Strength of Waterlogged Archaeological Wood after PEG Treatment with Concentration and Solvent (PEG 처리 수침고목재의 농도 및 용매에 따른 압축강도 변화)

  • The compressive strength of PEG along processing concentration and solvent is willing to be measured and proper processing condition for exhibition and storage is also willing to be measured by comparing with dimensional stability. In the advanced research of setting PEG-preprocessing concentration & solvent for freeze drying of waterlogged archaeological wood of high water content, vacuum freeze drying showed the highest dimension stability after 40% PEG-preprocessing of aqueous solution. In this study, the compressive strength increased in proportion of processing concentration and water showed the relatively-higher compressive strength than t-butanol regarding solvent. Especially, it showed that there is no big strength difference between PEG 40% and PEG 50% in aqueous solution by 6.6%(16kgf/$cm^2$). According to the above results, it was recognized that it is most effective to implement freeze drying after 40% PEG-preprocessing when want to dimensional stability and compressive strength simultaneously.

Conservation for Wooden Lacquer Ware Excavated from Isong Sansung Fortress (이성산성 출토 목제칠기 보존)

  • The wooden lacquer wares, which had excavated from the water reservoir in the Isong Sansung Fortress, were applied with vacuum freeze drying method and treated, after they had been substituted with tbutanol, and treated with PEG-pretreatment. The manufacturing technique, which had not been investigated in the past, was examined through the investigation of the wood and the lacquer used for wooden lacquer wares, and the pieces of the lacquer wares, which had been stored for a long time, were restored for the exhibition and conservation. As the result, the wooden lacquer wares were manufactured, using black alder belonging to the group of Alnus, and the lacquer was painted on the bottom-layer by mixing the powdered soil and lacquer, and it was confirmed that the lacquer mixed with the black pigment and the pure lacquer had been painted alternatively over 2 times, and that the wooden lacquer ware had been manufactured through total 5 times of being painted.

동결건조를 이용한 레트로바이러스의 저장 시 안정성에 미치는 인자에 관한 연구

  • We studied to improve the stability of retrovirus during freeze-drying. Through selection of the additives, we chose the trehalose, trehalose-PVP colyophilized mixture which were most effective additive on retrovirus storage. Through thermal analysis by DSC. in case of adding trehalose, thermal change shift to $15^{\circ}C$ from $10^{\circ}C$. As a result, we found it that Tg have important role to improve the stability of retrovirus. When retrovirus was storaged at the different temperature, the activity of freeze-dried retrovirus with trehalose sustained for 8 week below $4^{\circ}C$, But freeze-dried retrovirus without additives showed the activity less than 40% at all temperature.

Cryopreservation of Primordial Germ Cells(PGCs) from Korean Native Chicken(Ogye) Embryos using Commercial Cryoprotectants (상업용 동결보호제를 이용한 한국재래닭(오계) 원시생식세포의 동결 보존)

  • Cryopreservation of poultry semen has been reported, but preservation of female genetic material has not been possible because of the unique anatomical and physiological characteristics of the avian egg. Thus an alternative strategy for conservation of oviparous species of animals must be developed. Recent technological developments for producing germline chimeras by the transfer of primordial germ cells (PGCs) into recipient embryos has enabled the conservation and retrieval of chicken genetic resources in their complete form. In the present study, fertilized eggs were incubated for about 5.5 days to obtain embryos at stage 28. The whole embryo was collected from the germinal gonad using a fine glass micro pipette under a microscope. The PGCs were then purified using MACS method. Two commercially available cryoprotectants (A and B) were used to preserve the PGCs, and EG were used as a control. The average recovery rate of PGCs after thawing was 35.5% and 60.5% with the A and B treatments, respectively. There was no significant difference between B treatments and control, which showed an average recovery rate of 52.8%. However, the recovery rate obtained using A cryoprotectant (35.5%) was significantly lower than using treatment control and B. The average viability of the PGCs after thawing were 77.9% and 77.4% for cryoprotectants A and B, respectively, and the control were was 81.6%. There was no statistically significant difference between the two treatments and control. It was concluded that all of the available cryoprotectants examined in this study could be used for preservation of PGCs from embryos. Further experiments to produce germline chimera from PGCs preserved using this techniques are strongly recommended.

Quality Characteristics of Frozen Welsh Onion (Allium fistulosum L.) according to Various Blanching Treatment Conditions (Blanching 처리 조건에 따른 동결 대파의 품질 특성)

  • In this study, we set an optimum blanching condition as a pretreatment condition to freeze welsh onion (Allium fistulosum L.) for the purpose of preserving the cooking material for long-term usage. With higher blanching temperature and longer processing time, the Hunter color values and the hardness of welsh onion tended to decrease, which confirmed softening of the tissue. Compared to before blanching, total bacterial counts decreased to near 2 log scale, whereas coliforms were not detected or decreased to a 1-2 log scale. Blanching by heating at 100$^{\circ}C$ for 5 min decreased peroxidase inactivity to more than 94%, whereas only 47% of the inactivity was achieved at 80$^{\circ}C$ for 1 min. The sensory evaluation for the blanched and defrosted welsh onion showed that the group blanched at 80$^{\circ}C$ for 1 min was preferred significantly (p<0.05).
