Antioxidant Effects of Berchemia berchemiaefolia in Nerve Pain Models

  • Berchemia berchemiaefolia (BB) are climbing plants or small to medium-sized trees that live in Africa, Asia and America. We performed the present study to investigate whether oral administration of Berchemia berchemiaefolia extract (BBE) protects SD rats from pain. The SD rat experimental groups were divided into four groups. Two of the animal model groups were fed on BBE (200 mg/kg or 100 mg/kg). We performed oral acute toxicity test to determine the optimal oral dose of BBE. To explore if BBE alleviated pain in the SD rat, we undertook the tail flick latency test and formalin test. Additionally, we conducted the anti-oxidative test. The findings of the present study suggest that Berchemia berchemiaefolia extract exhibits strong antioxidant and analgesic activities.

Comparative Proteome Analysis of Celastrol-Treated Helicobacter pylori

  • Various preclinical and clinical trials have been conducted the efficacy of celastrol. In data presented in the current manuscript is the first trial to inhibit Helicobacter pylori with celastrol. In this study, the quantitative change of various H. pylori proteins including CagA and VacA by the anti-bacterial effect of celastrol was determined. The anti-H. pylori effects of celastrol was investigated by performing 2-dimensional electrophoresis and additional supporting experiments. After 2-dimensional electrophoresis analysis, spot intensities were analyzed and then each spot was identified using matrix assisted laser desorption ionization-time of flight mass spectrometry (MALDI-TOF-MS) or peptide sequencing using Finnigan LCQ ion trap mass spectrometer (LC-MS/MS). The results show that celastrol has multiple effects on protein expression in H. pylori.

유전자 조작된 대장균이 생산한 알파 인터페론(rHu IFN-$\alpha$A)의 변이원성에 대한 연구

  • Ames test와 포유동물 배양세포를 이용한 염색체 이상시험을 통하여 유전자 조작된 대장균이 생산한 알파 인터페론의 변이원성 유무를 조사하였다. rHu IFN-$\alpha$ A는 S. typhimurium (TA 1535, TA 1537, TA 1538, TA 98, TA 100) 에 30, 3000, 30,000 IU/plate의 농도에서 변이원성을 나타내지 않았으며, rat(Sprague-Dawley)의 골수세포를 이용한 생체내 염색체 이상시험을 실시하여 양성대조군인 nitroso-guanidine과 rHu IFN-$\alpha$A 각각에 대한 염색체 구조 이상의 출현율을 조사하였다.

Bacteriophage Sigma FA1의 치사활성과 구조특성 (Killing Activity and Molecular Properties of Bacteriophage Sigma FA1 of Bacillus circulans)

  • Bacillus circulans로부터 mitomycin C처리에 의해 유발되는 Bacteriophage Sigma FA1을 서당밀도균배 원심법 (sucrose gradient centrifugation)에 의해 분리, 정제하였다. Sigma FA1의 감수성숙주에 대한 치사작용은 single-hit 방식이었다. 한편, 세포에 감여된 후 phage DNA는 천천히 분해되기 시작했으며, 본 연구의 결과는 Sigma FA1의 세포에 대한 치사작용이 기존의 것과는 다른 것임을 입증시켰으며, 이는 단 백질분해기능에서 유래되는 듯 했다.

  • 제주도 서귀포의 해변에서 채집한 해양퇴적물로부터 Myxococcus속의 점액세균 JW154를 분리하여 그 대사산물로부터 강한 항균활성을 나타내는 두 개의 bithiazole계 화합물을 분리하고 각종 기기분석을 통하여 구조를 확인한 바, 이들은 각각 KR025와 melithiazole F로 보고된 화합물임을 밝혔다. 유묘 검정법으로 조사된 이 화합물의 주요 식물병원균에 대한 in vivo 항균활성은 토마토 역병과 밀 녹병 및 보리 흰가루병에 대해 높게 나타났는데, 20$\mu\textrm{g}$/m1 수준으로 처리하였을 때 80% 이상의 높은 방제가를 보였다.

  • 단백질-단백질 상호작용(PPI :Protein-Protein Interaction) 데이터는 생물체가 어떠한 메커니즘으로 생명을 유지하는지에 대한 정보를 담고 있다. 최근에는 생물학자들의 실험에 의해 많은 데이터가 축적되어 있으며, 데이터베이스로 구축되어 인터넷에 공개되어 있다. PPI 데이터는 단백질를 노드(node)로, 상호작용은 에지(edge)로 갖는 그래프(Graph) 구조로 표현 가능하다. 본 논문에서는 사용자가 PPI 데이터를 쉽게 가공하고 분석할 수 있도록 그래프 이론 기반에 기반하여 구현한 Proteinca(PROTEin INteraction CAbaret) 시스템에 대해 소개한다. The Effect of Alpha Subunit of Go on Cell Growth

  • Heterotrimeric GTP binding proteins (G proteins) mediate signals generated by neurotransmitters and hormones Among G proteins, Go is found in a large quantity in brain and growth cone membranes of neurons. In spite of its abundance in neurons, the role of Go is not fully understood. In our previous study, we identified promyelocytic leukemia zinc finger protein (PLZF) as an interacting partner of alpha subunit of Go ($Go{\alpha}$) and confirmed their interaction employing several biochemical assays. To date, it is reported that PLZF functioned as a cell growth suppressor and a transcription repressor. To determine effect of $Go{\alpha}$ and PLZF interaction on the cellular function of PLZF, we performed luciferase reporter gene assay and BrdU incorporation assay. Co-expression of $Go{\alpha}$ and PLZF synergistically increased the effect of PLZF alone. These results suggest that $Go{\alpha}$ may act as cellular activator of PLZF. This novel feature of Go may provide insights into understanding diverse role of Go-coupled receptor as well as its cellular actions.

Effect of Propolis on the Activity of Antioxidant Enzymes in Rat Liver Irradiated by X-ray

  • We investigated the effect of propolis on the activity of antioxidant enzymes in rat liver exposed by X-ray irradiation. The dosage of propolis showed the effect of lowering the concentration of superoxide anion in irradiated rat liver, suggesting that propolis has a significant role to remove superoxide anion as an antioxidant and/or by activating the antioxidant enzyme. The activities of superoxide dismutase (SOD) and glutathione reductase (GR), disturbed by X-ray irradiation, were restored in 30 days to normal status in the group which dosed propolis before X-ray irradiation. Interestingly, catalase (CAT) and glutathione peroxidase (GPOX) activities were highly increased with feeding propolis to rat compared to untreated group, whereas glutathione s-transferase (GST) activity was little affected. Taken together, it suggests that the propolis has a protective role in the rat liver cells against X-ray irradiation by increasing and recovering the activities of antioxidant enzymes.

Estimation of Death Time by Changes of Postmortem Xanthine Oxidase Activity in Rats

  • To evaluate the postmortem changes in activities of oxygen free radical metabolizing enzymes, the rats were sacrificed with cervical dislocation and were kept in an incubator at $25^{\circ}C$, 70% of humidity for 12 hours. The activities of aniline hydroxylase, catalase, glutathione-S-transferase and superoxlde dismutase were decreased with the time. On the other hand, the activity and type conversion ratio (type D ${\to}$type O) of hepatic xanthine oxidase (XO) were gradually increased. From these changes of XO, the estimation of death time (mathematical equation) could be determined with the least square method. To clarify the cause of increasing XO activity, enzyme kinetics were examined. The Km values of XO were decreased with the time. In conclusion, the determination of liver XO activity might be used for the estimation of death time in the early postmortem period.
